Description
Technical Field
The invention relates to the technical field of magnolia variegata seed treatment, in particular to a method for promoting the germination of magnolia variegata seeds.
Background
Indigofera tinctoria (Indigofera amblyyantha) is perennial shrub of the genus Indigofera of the family Leguminosae (Leguminosae sp.). The root system of the flowery knotweed can effectively intercept water drops and prevent soil from scouring, and the flowery knotweed is a water and soil conservation plant with strong resistance. The root nodule of the magnolia liliiflora can fix nitrogen, so that the nitrogen content of the soil is increased to improve the soil, and the magnolia liliiflora can grow in the soil nutrient-deficient area. Therefore, the flowery knotweed herb is usually used as a pioneer recovery species and a water and soil conservation plant for ecological restoration, has the functions of water and soil conservation, greening, soil quality improvement and the like, and has low germination rate. The reason for the low germination rate of the magnolia liliiflora is related to the seed hard-seed rate and the seed dormancy, the seed dormancy needs to be broken if the germination rate is improved and the seedling emergence is ensured to be uniform, the hard shell of the seed is broken by traditional common sulfuric acid and other medicaments, so that the seed coat absorbs water and breathes air, the seed germination is promoted, and the growth of the germinated radicle and the embryo has certain influence.

In order to achieve the purpose, the invention adopts the following technical scheme:
according to the first aspect of the invention, the method for promoting the germination of the magnolia variegata seeds is to soak the magnolia variegata seeds in the green ratio multi-solution with the concentration of 6-9ml/L for 8-12 hours at the temperature of 60-65 ℃.
Preferably, the method is to soak the seed of the magnolia variegata by using a green ratio multiple solution with the concentration of 6ml/L and the temperature of 65 ℃ for 12 hours.
Preferably, the LVBIODU solution is purchased from Zhan China investment Co.
In a second aspect of the present invention, there is provided a magnolia biondii seed prepared by the above method.
In a third aspect of the invention, the use of the above method for promoting germination of magnolia variegata seeds is provided.
Preferably, germination of the magnolia biondii seeds is promoted by at least one of the following pathways (1) to (3):
(1) the germination rate of the magnolia variegata seeds is improved;
(2) the germination vigor of the magnolia variegata seeds is improved;
(3) improving the germination index of the magnolia variegata seeds;
(4) improving the vitality index of the magnolia variegata seeds.
The invention has the beneficial effects that:
the method has the advantages that the magnolia variegata seeds are soaked in the green ratio multi-solution with specific concentration at specific temperature and time, so that the germination rate, the germination vigor, the germination index and the vitality index of the magnolia variegata seeds can be obviously improved, and the germination performance of the magnolia variegata seeds is obviously improved.
Detailed Description
It should be noted that the following detailed description is exemplary and is intended to provide further explanation of the disclosure. Unless defined otherwise, all technical and scientific terms used herein have the same meaning as commonly understood by one of ordinary skill in the art to which this application belongs.
As described in the background section, the flowery knotweed is generally used as a pioneer recovery species and a water and soil conservation plant for ecological restoration, and has the functions of water and soil conservation, greening, soil quality improvement and the like, but the flowery knotweed has low germination rate. The reason for the low germination rate of the magnolia variegata is related to the seed hard-seed rate and the seed dormancy, and the dormancy of the seeds must be broken to improve the germination rate and ensure the uniformity of the seedling emergence, so that the hard shells of the seeds are broken, the seed coats absorb water and ventilate, and the seed germination is promoted.
According to the invention, through research, the method can obviously improve the germination rate, the germination vigor, the germination index and the vigor index of the magnolia biondii seeds and obviously improve the germination performance of the magnolia biondii seeds by soaking the magnolia biondii seeds in the green ratio multi-solution with the concentration of 6ml/L and the temperature of 65 ℃ for 12 hours.
In order to make the technical solutions of the present application more clearly understood by those skilled in the art, the technical solutions of the present application will be described in detail below with reference to specific embodiments.
The test materials used in the examples of the present invention are all conventional in the art and commercially available.
Example 1: a method for promoting germination of the seed of the magnolia variegata is to soak the seed of the magnolia variegata for 12 hours at a temperature of 65 ℃ by using a green ratio multi-solution with a concentration of 6 ml/L. The multiple green ratio solution is a Xiongdaoda multiple green ratio solution, and the original country: italy, the linkage in china: just reaching the investment company of China.
1. Germination test in culture dish under phytotron conditions
(1) Seed disinfection: sterilizing the seeds in 5% sodium hypochlorite solution for 5-8min, pouring out sodium hypochlorite, and washing with sterilized water for 3 times;
(2) seed soaking: averagely dividing the same batch of the kalanchoe odorata seeds into 16 groups, respectively soaking the seeds by green multi-ratio solutions of 0ml/L, 3ml/L, 6ml/L and 9ml/L, and respectively dividing the green multi-ratio solutions at each concentration into 4 groups according to seed soaking time, wherein the seed soaking time is 4 hours, 8 hours, 12 hours and 24 hours;
(3) and (3) germination: the seeds after seed soaking were placed in a petri dish for germination in a 25 ℃ culture room, the number of germinated seeds was recorded every day, and the germination vigor was measured at 3 d.
(4) Determining the germination rate, the germination vigor, the germination index and the vitality index of the seed germination;
the germination rate is (number of germinated seeds at the final germination stage/total number of tested seeds) × 100%;
germination potential (number of seeds germinated on 3 d/total number of seeds tested) × 100%;
germination index ═ Σ (Gt/Dt);
the vitality index is multiplied by the germination index multiplied by G;
in the above formula: gt is the number of sprouts in t days; dt is the corresponding number of germination days; g is the weight of the seedling at the end of germination. The results are shown in tables 1 to 4.
