Abstract
The invention discloses a quaternary ammonium salt washing-free disinfection gel with skin mildness, which comprises the following raw materials in parts by weight: 0.1-0.5 part of quaternary ammonium salt, 0.02-0.3 part of glycolipid surfactant, 0.5-1 part of gel matrix, 0.1-0.5 part of pH regulator and 97-99.3 parts of deionized water. The formula of the invention has the characteristics of quick drying, refreshing, no odor, no toxicity, no harm and good mildness.
Description
Technical Field
The invention relates to the technical field of daily-use chemical industry, in particular to a quaternary ammonium salt washing-free disinfection gel with skin mildness and a preparation method thereof.
Background
In hand disinfection products, the no-clean disinfection product has the advantages of use and carrying, and has wide consumption demands in various occasions, so that the no-clean disinfectant has wider consumption market compared with the traditional hand sanitizer and disinfectant.
The current disposable disinfection products are mainly divided into two types, namely ethanol and quaternary ammonium salt according to different effective components of the disinfection. The washing-free disinfectant taking ethanol as the main component has certain flammability risk, limits the use of the washing-free disinfectant on high-speed rails and airplanes, and has the advantages of low content of effective substances, good sterilization effect and safer and more reliable use compared with the disinfectant taking quaternary ammonium salt as the effective component. However, quaternary ammonium salts, as a cationic surfactant, neutralize the negative charges of the skin itself when they are in contact with the skin, thereby changing the charge properties of the skin surface and further affecting the barrier properties of the stratum corneum and skin permeability of the skin. The irritation of the quaternary ammonium salt component to skin is obviously greater than that of an anionic surfactant and a nonionic surfactant, and the quaternary ammonium salt component is mainly characterized by damaging the water retention effect of the horny layer of the skin and possibly causing the reduction of the water content of the skin, the increase of the water loss through skin, redness and swelling, pruritus and the like. Meanwhile, due to the characteristic that the no-clean disinfectant product is dried quickly after being used and does not need to be cleaned, the worry of consumers about skin irritation is more easily caused after the no-clean disinfectant product is used for a plurality of times.
The development history of the quaternary ammonium salt type no-clean disinfectant is short, and related researches on how to improve the mildness of the formula to reduce the irritation of the quaternary ammonium salt to the skin are lacked. In the current part of technologies, moisturizing and skin moistening components such as glycerin and propylene glycol are added in a formula so as to achieve a hand protection effect. However, related studies have found that humectants such as propylene glycol increase skin permeability and may further enhance skin irritation in the presence of quaternary ammonium salt components. In part of technologies, the skin mildness is improved by adding plant-derived extracts, but the mechanism is not clear, so that the development of quaternary ammonium salt type no-clean disinfectants with skin mildness has high technical requirements for further improving the mildness effect of disinfectants.
Disclosure of Invention
In view of the above problems in the prior art, the present applicant provides a quaternary ammonium salt no-clean disinfecting gel with skin mildness and a preparation method thereof. The formula of the invention has the characteristics of quick drying, refreshing, no odor, no toxicity, no harm and good mildness.

the quaternary ammonium salt is one or a mixture of a single-chain quaternary ammonium salt and a double-chain quaternary ammonium salt.
The single-chain quaternary ammonium salt is one or more of benzalkonium chloride, benzalkonium bromide, dodecyl trimethyl ammonium chloride, dodecyl trimethyl ammonium bromide, dodecyl-dimethyl-2-phenoxyethyl ammonium bromide, tetradecyl trimethyl ammonium chloride, tetradecyl trimethyl ammonium bromide, tetradecyl dimethyl benzyl ammonium chloride and tetradecyl dimethyl benzyl ammonium bromide.
The double-chain quaternary ammonium salt is one or more of bisoctyl dimethyl ammonium chloride, bisoctyl dimethyl ammonium bromide, bisdecyl dimethyl ammonium chloride, bisdecyl dimethyl ammonium bromide, bisdodecyl dimethyl ammonium bromide, bistetradecyl dimethyl ammonium chloride and bistetradecyl dimethyl ammonium bromide.
The glycolipid surfactant is one or more of rhamnolipid, sophorolipid, mannosylerythritol lipid, trehalose glycolipid, amber-trehalose glycolipid, and cellobiose.
The gel matrix is one or more of methylcellulose, hydroxymethyl cellulose, hydroxyethyl cellulose, hydroxypropyl methylcellulose and hydroxypropyl cellulose.
The pH regulator is organic alkali or basic amino acid. Preferably, the pH regulator is one or more of triethanolamine, monoethanolamine, ethylenediamine and arginine.
In a preferred scheme, the quaternary ammonium salt no-clean disinfection gel comprises the following raw materials in parts by weight:

(1) mixing 0.1-0.5 part of quaternary ammonium salt with 0.02-0.3 part of glycolipid surfactant, and stirring;
(2) adding 80 parts of deionized water into the product obtained in the step (1), stirring while adding, and fully dissolving all the components until the solution is uniform, clear and free of insoluble substances;
(3) heating the product obtained in the step (2) to a temperature higher than 50 ℃, keeping the temperature, adding 0.5-1 part of gel matrix, gradually cooling to room temperature while stirring, and keeping the stirring speed at 120rpm until gel is formed;
(4) and (4) adding 0.1-0.5 part of pH regulator into the product obtained in the step (3) to regulate the pH of the solution to 4-7, adding deionized water to make the mass part of the system reach 100 parts, and continuously stirring until the system is transparent and uniform, thus obtaining the quaternary ammonium salt washing-free disinfection gel.
The beneficial technical effects of the invention are as follows:
aiming at the characteristic that skin irritation is caused by electrostatic interaction between the quaternary ammonium salt cationic surfactant and the skin, the adsorption quantity of the quaternary ammonium salt on the skin is reduced by introducing the glycolipid surfactant and the quaternary ammonium salt for competitive adsorption, so that the skin mildness of the quaternary ammonium salt disinfection gel is endowed. The quaternary ammonium salt is shown in the effect of inhibiting the decrease of the quaternary ammonium salt to the water content of the skin, the effect of inhibiting the increase of the quaternary ammonium salt to the percutaneous water loss of the skin and the effect of reducing the increase of the quaternary ammonium salt to the friction force of the skin in the instrument detection, and the irritation to the skin of the rabbit is shown in the animal experiment.
The glycolipid surfactant is a nonionic surfactant derived from microbial fermentation, has polyhydroxy in the molecular structure, so that the binding force with oxygen-containing groups on the skin surface is stronger, and the glycolipid surfactant has stronger affinity and milder property for the skin compared with common nonionic surfactants. The glycolipid as a novel biosynthetic surfactant is applied to the aspects of medical treatment, medicaments and the like due to the unique biological property and degradability, and the invention finds that the irritation of the quaternary ammonium salt to the skin can be obviously weakened by introducing the glycolipid surfactant.
The formula of the quaternary ammonium salt type no-clean gel provided by the invention has high killing rate on common pathogenic bacteria such as escherichia coli, staphylococcus aureus, candida albicans and the like. The prepared gel type disinfectant is transparent and colorless, has no peculiar smell and has good stability.

Detailed Description
The present invention will be described in detail with reference to examples.
Examples 1 to 4
A preparation method of the quaternary ammonium salt no-clean disinfection gel with skin mildness comprises the following steps:
(1) 0.1-0.5 part of quaternary ammonium salt and 0.02-0.3 part of glycolipid surfactant are fully stirred until the mixture is uniformly mixed;
(2) adding 80 parts of deionized water into the product obtained in the step (1), stirring while adding, and fully dissolving all the components until the solution is uniform, clear and free of insoluble substances;
(3) heating the product obtained in the step (2) to a temperature higher than 50 ℃, keeping the temperature, adding 0.7 part of gel matrix, gradually cooling to room temperature while stirring, and keeping the stirring speed at 120 revolutions per minute until gel is formed; and (4) adding 0.1-0.5 part of pH regulator into the product obtained in the step (3) to adjust the pH of the solution to 4-7, adding deionized water to enable the mass part of the system to reach 100 parts, and continuously stirring until the system is transparent and uniform to obtain the disinfection gel.
The raw materials and the amounts of the raw materials used in examples 1 to 3 are shown in Table 1.

A method for preparing quaternary ammonium salt washing-free disinfection gel, which is as shown in the table 2, is as the following example 1-4, and the difference is that glycolipid surfactant is replaced.

(1) testing the disinfection effect;
the sterilization efficiency was tested by the standard suspension method of "disinfection and sanitation Specification (2002 edition)", 2.2.1.4, and the test results are shown in Table 3.

As can be seen from Table 3, in examples 1 to 4 of the present invention, the killing log of Escherichia coli and Staphylococcus aureus was 5 or more and the killing log of Candida albicans was 4 or more within 5 min. In the comparative example, the sterilization effect of the comparative example 1 with high quaternary ammonium salt content is equivalent to that of the example, and the sterilization log value of the comparative example 2 with 0.1% of quaternary ammonium salt content and the sterilization log value of the comparative example 3 are slightly lower than that of the example 2 with the same quaternary ammonium salt content of 0.1%, which shows that the introduction of the glycolipid has certain effect of promoting the sterilization and sterilization of the quaternary ammonium salt. The reason for this is that the glycolipid surfactant derived from microbial fermentation has a certain natural bacteriostatic function, thereby facilitating the microbial sterilization of the mixed bacteria in the fermentation process and becoming the dominant bacteria.

Skin moisture content was measured by a Corneometer brand multi-probe skin test system from CK, Germany (Courage + Khazaka electronic GmbH), skin moisture loss through skin was measured by a Tewameter brand skin moisture loss tester from CK, Germany, and friction was measured by a Fritionometer brand coefficient of friction tester from CK, Germany.
(2.2) test method
The human body experiment of 10 volunteers is adopted for measurement, four areas with the size of 1.5cm by 1.5cm are measured and divided in an arm, a cotton swab is used for dipping a disinfection gel sample to be measured and uniformly smearing the disinfection gel sample in 4 areas, the disinfection gel sample is waited for 1min, and smearing is carried out again after the gel is dried, and the total number of times is 10. Then, three indexes of skin water content, percutaneous water loss and friction are detected by using all probes. The test results are shown in table 4.

After the disinfectant gel is applied, the skin of a volunteer shows the trends of reduced skin water content, increased percutaneous water loss and increased skin friction, which represents that the disinfectant gel has irritation to the skin. The change degrees of the three indexes can reflect the skin irritation of the formula, and the smaller the change range of the three indexes, the higher the mildness. In order to more intuitively embody the change conditions of the three indexes, the skin water content reduction amplitude, the percutaneous water loss increase amplitude and the skin friction force increase amplitude of the embodiment and the comparative example are respectively drawn as bar graphs (1-3), and as can be seen from the lower graphs, the change degrees of the skin water content, the percutaneous water loss and the skin friction force of the comparative examples 1-3 are obviously greater than those of the embodiment, so that stronger skin irritation is embodied. It can be seen that the comparative example is less mild to the skin.
(3) Rabbit skin irritation test;
experimental methods A number of complete skin irritation experiments were performed as required in the 2.3.3 skin irritation experiment in Disinfection technical Specification (2002) of the Ministry of health. The final results are shown in table 5.

As can be seen from Table 5, examples 1-4 had good skin mildness, and showed no irritation to the rabbit skin after multiple applications over the observation period. In comparative examples 1-3, the glycolipid surfactant is replaced by the common polyhydroxy humectant, the conventional nonionic surfactant and other components, so that the neutralization effect on the irritation of the quaternary ammonium salt is not shown, and the prepared quaternary ammonium salt disinfection gel has obvious skin irritation. Compared with polyhydroxy humectant with similar chemical structure, conventional nonionic surfactant and the like, the glycolipid surfactant embodies the unique functions of improving the skin mildness of the quaternary ammonium salt disinfectant and relieving the skin irritation.

The quaternary ammonium salt no-clean disinfecting gel of claim 2, wherein the single-chain quaternary ammonium salt is one or more of benzalkonium chloride, benzalkonium bromide, dodecyltrimethylammonium chloride, dodecyltrimethylammonium bromide, dodecyl-dimethyl-2-phenoxyethylammonium bromide, tetradecyltrimethylammonium chloride, tetradecyltrimethylammonium bromide, tetradecyldimethylbenzylammonium chloride, and tetradecyldimethylbenzylammonium bromide.

The quaternary ammonium salt no-clean disinfecting gel of claim 2, wherein the double-chain quaternary ammonium salt is one or more of bisoctyl dimethyl ammonium chloride, bisoctyl dimethyl ammonium bromide, bisdecyl dimethyl ammonium chloride, bisdecyl dimethyl ammonium bromide, bisdodecyl dimethyl ammonium chloride, bistetradecyl dimethyl ammonium chloride, and bistetradecyl dimethyl ammonium bromide.
5. The quaternary ammonium salt no-clean disinfecting gel as claimed in claim 1, wherein the glycolipid surfactant is one or more of rhamnolipid, sophorolipid, mannosylerythritol lipid, trehalose glycolipid, amber-trehalose glycolipid, and cellobiose.
6. The quaternary ammonium salt no-clean disinfecting gel of claim 1, wherein the gel matrix is one or more of methyl cellulose, hydroxymethyl cellulose, hydroxyethyl cellulose, hydroxypropyl methyl cellulose and hydroxypropyl cellulose.
7. The quaternary ammonium salt no-clean disinfecting gel of claim 1, wherein the pH regulator is an organic base or a basic amino acid.
8. The quaternary ammonium salt no-clean disinfecting gel as claimed in claim 1, wherein the pH regulator is one or more of triethanolamine, monoethanolamine, ethylenediamine and arginine.
