技术领域
本发明涉及一种植物源精油,尤其涉及一种用于电子雾化棒的植物源抑菌 精油及其制备方法。
背景技术
近年来,随着国际控烟日益严峻和消费者健康意识普遍提升,世界各国相 继以立法形式限制传统卷烟销售,国际卷烟销量持续下滑,在此背景下,国内 外烟草公司不断推进新型烟草制品的发展,其中,电子烟作为新型烟草制品的 一种重要形式,取得了快速发展。电子烟是一种非燃烧的烟类替代型产品,有 着与香烟相似的外观,烟油在电子烟雾化器的作用下产生雾气类似于真烟烟气, 满足消费者的快感和使用习惯,但又与香烟有着本质上的不同,不燃烧、不含 焦油、一氧化碳等有害物质。根据实际消费者体验来看,电子烟虽然形状、手 感、烟雾等感觉像卷烟,但电子烟油产生的烟雾在香气、口味方面,与真正的卷烟感觉差距较大,这大大影响了电子烟的品质,同时也制约着电子烟的普及 及流行。为了解决这一技术问题,行业内大都采取向电子烟烟油内加入烟草提 取物,但是由传统方法制备出的提取物在电子烟烟油中所产生的感官质量并不 能很好满足消费者的要求,同时大部分消费者抽吸电子烟后会有口干舌燥等不 适现象,严重者甚至头晕、恶心,这一问题也亟待有所改善解决。
中国专利号201611216424.7公开了一种湿法粉碎制备中草药电子烟油的 方法,包括以下步骤:(1)准确称取干燥中草药100g,初粉碎后过5~50目筛 网,向初粉碎后的中草药中加溶剂,所述中草药与溶剂的质量比为1:5~10,混 合均匀后加入高速切割型湿法粉碎机中,于一定反应条件下粉碎,粉碎后出料, 所出料液即为中草药电子烟油基础油,其中所述一定反应条件为所述高速切割 型湿法粉碎机的转速为2500~8000r/min,粉碎温度为5~60℃,粉碎时间为 0.5~6h;(2)将质量百分比为70~100%的所述中草药电子烟油基础油与质量百 分比为0~30%香精香料搅拌混匀,即得中草药电子烟油;所述中草药电子烟油 基础油还包括具有生津止渴功效的药物提取物,所述药物提取物与中草药的质 量比为1~5:10;所述具有生津止渴功效的药物提取物由桑葚、石榴、仙草、乌 梅、橄榄、人面子、醋柳果、金棒锤、崖花子中的一种或两种混合提取制得, 利用该专利制备的中草药粉末电子烟油整体的中草药香气更加浓郁、甜腻感减 少,同时还兼具养生保健的功效,而且在烟油基础油中配以仙草、乌梅等具有 生津止渴功效的成分,可有效降低丙二醇、丙三醇等溶剂带来的甜腻感,缓解 消费者抽吸完电子烟后口干舌燥的症状,但是该种类型的电子烟油并没有抑菌 的效果,且中草药味重,不能满足消费者的需求。
发明内容
因此,针对上述的问题,本发明提供一种用于电子雾化棒的植物源抑菌精 华油及其制备方法,解决现有技术的电子烟油没有抑菌效果、无法缓解口腔异 味及发炎的问题。
为实现上述目的,本发明采用了以下技术方案:一种用于电子雾化棒的植 物源抑菌精油,包括以下重量份的各原料:丙二醇35-40份、丙三醇35-40份、 虎杖提取物3-4份、千里光提取物2-3份、万寿菊提取物2-3份、苦参提取物 3-4份、蛇床子提取物3-4份、薄荷提取物2-3份、黄芩提取物2-3份、金银花 提取物2-3份、黄柏提取物2-3份、食用香精1-2份、茶叶提取物2-3份。
进一步的改进是,所述虎杖提取物的制备方法为:将虎杖粉碎,过200目 筛,采用95%乙醇水浴回流提取三次,将三次得到的滤液合并,再对合并后的滤 液进行减压浓缩、过滤,滤液上聚酰胺柱,再经水洗、10%乙醇醇洗至无色,再 用30%乙醇冲洗,收集洗脱液,再次减压浓缩得到虎杖提取物。

进一步的改进是,所述黄柏提取物的制备方法为:取黄柏粉,加入10倍重 的去离子水,混合均匀后加入纤维素酶,所述纤维素酶的用量为:每1g黄柏粉 加入50万活力单位纤维素酶,加热至40-50℃,恒温30-45min后再升温至 90-95℃,并恒温1-1.5h,过滤,取滤液,再减压浓缩至滤液体积的1/3-1/2, 再采用无水乙醇沉淀除杂,最后再将无水乙醇去除,即得到黄柏提取物。

千里光:具有清热解毒、杀虫、明目、凉血、生肌及驱风除湿等功效,千 里光具有光谱的抗菌作用,特别对金黄色葡萄球菌、肺炎双球菌、大肠杆菌、 沙眼病毒等具有较强的抑制作用。
黄芩:具有清热燥湿、泄火清心、凉血等功效,黄芩的提取物,例如黄酮 类化合物,具有抗菌、抗病毒、抗氧化等作用。
黄柏:具有清热解毒、泄火燥湿、解毒疗疮等功能,用于治疗湿热泻痢、 黄疸、湿疹瘙痒等症。黄柏的主要要用成分为小檗碱,而小檗碱为抗菌药物,是 《国家基本药物目录》选列的品种,它对溶血性链球菌、金黄色葡萄球菌、淋 球菌等具有抗菌作用,并具有增强白血球吞噬作用,对结核杆菌、鼠疫杆菌也 有不同程度的抑制作用;广泛用于治疗胃肠炎、细菌性痢疾等。
苦参:苦参属豆科槐属植物,是我国历史悠久的传统中药之一,苦参根具 有清热燥湿、杀虫、利尿等功效;临床研究证实,苦参含有多种化学成分并分 别具有不同的化学活性,苦参传统药理以抑菌、抗心律失常为主;现代药理研 究有抗菌、抗病毒、抗肝纤维化等作用。
表没食子儿茶素没食子酸酯(EGCG)也称为表没食子儿茶素-3-没食子酸酯, 是表没食子儿茶素和没食子酸的酯,并且是通常存在于茶树(Camellia Sinensis)等中的一种儿茶素。表没食子儿茶素没食子酸酯(EGCG)在化学上表示 为(2R,3R)-2-(3,4,5-三羟基苯基)-3,4-二氢-1(2H)-苯并吡喃-3,5,7- 三醇-3-[3,4,5-三羟基苯甲酸酯)。化学式为C22H18O11,分子量为 458.37g/mol,结构式为:
通过采用前述技术方案,本发明的有益效果为:
1、本发明的用于电子雾化棒的植物源抑菌精油,以植物为主要原料,成分 中不含尼古丁,组分对人体安全无害。原料中植物提取物含有抗菌成分,例如, 虎杖提取液中含有大黄素、黄素葡萄糖甙、白藜芦醇甙,对金黄色葡萄球菌及 肺炎双球菌有抑制作用,白藜芦醇对深红色发癣菌、趾间发癣菌具有抗菌效果, 虎杖提取液中的黄酮类、芪三酚苷及葡萄欧鼠李苷也均均有抗菌的作用;茶叶 提取物中含有EGCG,能够缓解使用者口腔异味及发炎的问题。
2、本发明抑菌机理为:细胞壁最为微生物细胞最外的一层外壁,具有阻拦 大分子有害物质进入细胞、抵抗渗透压的功能,而细胞膜是维持细胞完整性及 保持正常物质能量代谢的基础,本发明的原料中的植物提取物能够作用于细胞 壁和细胞膜,导致膜结构完整性被破坏,进而影响细胞代谢并改变细胞通透性, 再者,本发明的植物提取物中的活性成分对微生物的遗传物质可能还具有干扰 作用,对细菌内基因转录过程进行抑制,使得细菌细胞壁无法正常合成。
3、本发明的千里光提取物的制备,采用乙醇回流提取结合微波萃取,萃取 率过程较快,效率较高。
4、黄芩提取物的制备采用植物水解酶,较为环保,植物水解酶能够将黄芩 植物细胞壁分解破坏进而使有效物质溶出,滤液采用氯化钙去除絮凝物,能够 除去胶体、蛋白质等大分子杂质,得到的黄芩提取物中杂质少。
5、黄柏提取物的制备过程中添加纤维素酶,在40-50℃的温度下反应 30-45min后再升温至90-95℃,并恒温1-1.5h,能够将黄柏中的小檗碱溶出, 再采用乙醇除杂,得到的提取物中含有叫纯的小檗碱。
6、本发明的茶叶提取物的制备方法为:将茶叶放入沸水中浸泡2-4h,再放 入茶叶重的0.2-0.5%的纤维素酶和0.3-0.5%的果胶酶,反应4-5h,过滤,将滤 液和滤渣分离,将滤液经H-600树脂吸附洗脱,得到含EGCG的茶叶提取物,以 纤维素酶和果胶酶为反应原料,原料环保无毒且能够将茶叶中的EGCG有效的溶 出,该成分能够缓解口腔异味,其原理是:口腔中产生异味的主要化合物是挥 发性含硫化合物、含氮化合物,其中硫化氢、甲硫醇等硫化物气味最重,EGCG 能够与这些化合物发生反应,进而去除异味。

现结合具体实施方式对本发明进一步说明。
实施例一
一种用于电子雾化棒的植物源抑菌精油,包括以下重量份的各原料:丙二 醇35份、丙三醇35份、虎杖提取物3份、千里光提取物2份、万寿菊提取物2 份、苦参提取物3份、蛇床子提取物3份、薄荷提取物2份、黄芩提取物2份、 金银花提取物2份、黄柏提取物2份、食用香精1份、绿茶提取物2份。
所述虎杖提取物的制备方法为:将虎杖粉碎,过200目筛,采用95%乙醇水 浴回流提取三次,将三次得到的滤液合并,再对合并后的滤液进行减压浓缩、 过滤,滤液上聚酰胺柱,再经水洗、10%乙醇醇洗至无色,再用30%乙醇冲洗, 收集洗脱液,再次减压浓缩得到虎杖提取物。所述千里光提取物的制备方法为: 将千里光粉碎成粉末状,过筛,加入加入体积浓度为90%的乙醇回流提取,过滤, 减压干燥,再经微波萃取,微波萃取时加入体积浓度为30%的乙醇,微波功率 800W,温度80℃,时间15min。所述黄芩提取物的制备方法为:将黄芩粉碎成 粉末状,过20目筛后加入水、植物水解酶,所述水的用量为黄芩重的10倍, 所述植物水解酶的用量为黄芩重的0.1%,搅拌混合均匀,在40℃反应30min, 过去取滤液,在滤液中加入氯化钙,所述氯化钙的用量为滤液重的3wt%,去除 絮凝物,取滤液,再滤液中加入体积浓度为60%的乙醇醇洗30min,得到黄芩提 取物。所述黄柏提取物的制备方法为:取黄柏粉,加入10倍重的去离子水,混 合均匀后加入纤维素酶,所述纤维素酶的用量为:每1g黄柏粉加入50万活力 单位纤维素酶,加热至40℃,恒温30min后再升温至90℃,并恒温1h,过滤, 取滤液,再减压浓缩至滤液体积的1/3,再采用无水乙醇沉淀除杂,最后再将无 水乙醇去除,即得到黄柏提取物。所述绿茶提取物的制备方法为:将茶叶放入 沸水中浸泡2h,再放入茶叶重的0.2%的纤维素酶和0.3%的果胶酶,反应4h, 过滤,将滤液和滤渣分离,将滤液经H-600树脂吸附洗脱,得到含EGCG的绿茶 提取物。
上述万寿菊提取物、苦参提取物、蛇床子提取物、薄荷提取物、金银花提 取物的制备方法均为,将植物原料,即万寿菊、苦参、蛇床子、薄荷、金银花 在常温下用水浸泡48h,加热至100℃,过滤,降至常温,取滤液,进行浓缩处 理,即得到提取物。
一种用于电子雾化棒的植物源抑菌精油的制备方法,包括以下制备步骤: 将虎杖提取物、千里光提取物、万寿菊提取物、苦参提取物、蛇床子提取物、 薄荷提取物、黄芩提取物、金银花提取物、黄柏提取物、食用香精、绿茶提取 物加热至45℃,加入丙二醇、丙三醇,送入搅拌罐中搅拌溶解均匀后进行罐装。
实施例二
一种用于电子雾化棒的植物源抑菌精油,包括以下重量份的各原料:丙二 醇38份、丙三醇38份、虎杖提取物3.5份、千里光提取物2.5份、万寿菊提 取物2.5份、苦参提取物3.5份、蛇床子提取物3.5份、薄荷提取物2.5份、 黄芩提取物2.5份、金银花提取物2.5份、黄柏提取物2.6份、食用香精1.5 份、绿茶提取物2.4份。
所述虎杖提取物、黄芩提取物、黄柏提取物、绿茶提取物、万寿菊提取物、 苦参提取物、蛇床子提取物、薄荷提取物、金银花提取物的制备方法均与实施 例一相同。
实施例三
一种用于电子雾化棒的植物源抑菌精油,包括以下重量份的各原料:丙二 醇40份、丙三醇40份、虎杖提取物4份、千里光提取物3份、万寿菊提取物3 份、苦参提取物4份、蛇床子提取物4份、薄荷提取物3份、黄芩提取物3份、 金银花提取物3份、黄柏提取物3份、食用香精2份、绿茶提取物3份。
所述虎杖提取物、黄芩提取物、黄柏提取物、绿茶提取物、万寿菊提取物、 苦参提取物、蛇床子提取物、薄荷提取物、金银花提取物的制备方法均与实施 例一相同。
一种用于电子雾化棒的植物源抑菌精油,其制备方法与实施例一相比,在 灌装前进行了紫外灯照射杀菌处理,采用紫外灯照射处理能够避免将植物提取 物中的活性物质成分破坏且能够将液体中存在的细菌杀除。
紫外灯照射处理的具体过程为:精油送入带紫外灯的存储装置,紫外灯设 置在存储装置的上方及存储装置的周测,使精油充分的被紫外灯照射,每个存 储装置存储100L的精油,每个存储装置的上方设置10个紫外灯,周测设置20 个紫外灯,每个存储装置100L的精油紫外灯照射1-2h即完成杀菌消毒处理。 存储装置具有内壁和外壁,紫外灯设置在内壁和外壁之间,且内壁为透明材质。
委托广州质量监督检测研究院对本发明的植物源抑菌精油进行检测,检测 过程及结果如下:
一、检验依据:《消毒技术规范》(2002年版)2.3.1。
二、受试样品:本发明的实施例一的精油。
三、实验动物和饲养环境:SPF级KM小鼠20只,雌雄各半,体重18-22g, 由广东省医学实验动物中心提供,实验动物生产许可证号:SCXK(粤)2018-0002, 质量合格证号:No.44007200076924,饲料由广东省医学实验动物中心提供,动 物购进后在广州质量监督检测研究院屏障环境动物房饲养,检疫合格后使用, 实验动物使用许可证号:SYXK(粤)2018-0137,动物房温度:20-26℃,相对 湿度:40-70%。
四、检测方法
1.剂量设计及试验分组:采用一次最大限度试验,设5000mg/kg体重一个 剂量组,实验时称取样品5.0g,加纯水至10.0ml混匀后供试;
2、实验动物隔夜禁食不禁水后,按10.0ml/kg体重灌胃,一次给样,样品 剂量为5000mg/kg体重,给样后立即观察并记录实验动物的中毒表现、死亡数 和死亡时间,观察期14d;
3、结果评价:在14天内无实验动物死亡,可判定LD50>5000mg/kg体重。
五、试验结果
染毒后,在观察期内未发现小鼠有中毒表现及死亡,观察期结束后对存货 动物解剖,肉眼未见异常,试验结果见下表:

六、结论
样品对雌性KM小鼠急性经口毒性LD50>5000mg/kg体重,属实际无毒级, 符合《消毒技术规范》(2002年版)2.3.13.1(1)要求。
对小鼠骨髓嗜多染红细胞微核实验,实验过程、方法及结果如下:
一、材料和动物
1、受试样品:本发明的实施例一的精油。根据该试样对雌性性KM小鼠的急 性经口毒性LD50>5000mg/kg体重,设5000mg/kg体重、2000mg/kg体重、500mg/kg 体重三个样品剂量组,实验时称取样品5.0g、2.0g、0.5g,分别加纯净水至10.0ml 混匀后供试。
2、阳性对照物:以环磷酰胺最为试验的阳性对照物,购自上海源叶生物科 技有限公司,批号:SJ0121RA14。
3、实验动物和饲养环境:SPF级KM小鼠50只,雌雄各半,体重25-30g, 由广东省医学实验动物中心提供,实验动物生产许可证号:SCXK(粤)2018-0002, 质量合格证号:No.44007200078540,饲料由广东省医学实验动物中心提供,动 物购进后在广州质量监督检测研究院屏障环境动物房饲养,检疫合格后使用, 实验动物使用许可证号:SYXK(粤)2018-0137,动物房温度:20-26℃,相对 湿度:40-70%。
二、检验方法:
1、检验依据:《消毒技术规范》(2002年版)2.3.8.4.
2、检验方法:a、分组:取经检疫合格雌雄小鼠各25只,随机分为5组, 每组5只;b、染毒:用经口灌胃方式给予各组动物相应的受试物,共染毒2次, 间隔24h,每次灌胃量为10ml/kg体重,于末次染毒6h后采集标本;c、对照组 的处理:阳性对照组选用剂量为40mg/kg体重的环磷酰胺,临用前用纯水配制 成浓度为4mg/kg;阴性对照组选用水,操作程序通试验组;d、取材:颈椎脱臼 处死动物,取出胸骨,去除肌肉等组织,横向剪开胸骨,暴露骨髓腔,然后用 止血钳挤出骨髓液,将骨髓液滴在载玻片一端的小妞血清液滴里,仔细混匀,自然晾干,放入甲醇液中固定5min,Giemsa染色15min,立即用1/15mol/L磷 酸盐缓冲液冲洗,晾干;e、镜检:每只动物观察1000个嗜多染红细胞(PCE), 记录出现微核的PCE数,以千分率表示,此外,数200个PCE,同时记数所见的 NCE,计算PCE/NCE比例;f、统计:用SPSS软件建立数据库,用u检验按雌雄 分别比较受试物各剂量组与阴性(水)对照组的微核细胞率。
三、试验结果
阳性对照组微核细胞率与阴性对照组比较,微核率提高,差异有统计学意 义(P<0.01),试验系统成立。样品各剂量组微核细胞率与阴性对照组比较,无 显著性差异(P>0.05),试验结果为阴性。
PCE/NCE>0.1,提示该受试物对骨髓无明显抑制作用,该试样对小鼠骨髓嗜 多染红细胞无致微核作用,试验结果见下表:

注:**表示与阴性对照组比较P<0.01;微核细胞率‰和PCE/NCE均以小鼠为单位 统计,表示为均值±标准差。
四、结论
样品在剂量500mg/kg体重、200mg/kg体重、5000mg/kg体重时,雌、雄 性KM小鼠结果均为阴性,无致微核作用,符合《消毒技术规范》(2002年版) 2.3.13.2(2)要求。
本发明的茶叶提取物的制备方法为:将茶叶放入沸水中浸泡2-4h,再放入 茶叶重的0.2-0.5%的纤维素酶和0.3-0.5%的果胶酶,反应4-5h,过滤,将滤液 和滤渣分离,将滤液经H-600树脂吸附洗脱,得到含EGCG的茶叶提取物。采用 该方法可以有效的提取茶多酚,茶多酚中含有咖啡因、EGCG、EGC、EC等物质, EGCG的含量最高,约占80%左右,传统的茶叶提取物的制备方法是:将茶叶用 沸水煮沸或者用乙醇-水作为溶剂,进行浸提处理,采用这种方式不仅杂质含量 高,茶多酚的有效成分低,一般含有植物多糖、色素、咖啡碱、树脂和氨基酸 等多种物质,茶多酚的含量仅有30%左右,为了提高茶多酚的含量,现有技术有 的采用氯仿作为溶剂,而这种物质有毒,最终的产品使用不安全。而本发明是 用于电子雾化棒的精油,是供吸食的产品,采用纤维酶和果胶酶为反应原料, 较为安全,且茶多酚的提取率高达85%,茶多酚提取率的计算公式为:
茶多酚提取率(%)=茶多酚溶出量(g)/茶叶质量(g)*100%。
另外,采以3.9%D/E中和肉汤为中和剂,样品原液(实施例一)作用2min, 对金黄色葡萄球菌的平均杀菌率>99.99%,对大肠杆菌的平均杀菌率均>99.99%, 对铜绿假单胞菌的平均杀菌率>99.99%,均具有杀菌作用,符合GB15979-202《一 次性使用卫生用品卫生标准》的规定。以3.9%D/E中和肉汤为中和剂,样品原 液作用2min,对白色念珠菌的平均杀菌率>99.99%,具有杀菌作用,符合 GB15979-202《一次性使用卫生用品卫生标准》的规定。
尽管结合优选实施方案具体展示和介绍了本发明,但所属领域的技术人员 应该明白,在不脱离所附权利要求书所限定的本发明的精神和范围内,在形式 上和细节上可以对本发明做出各种变化,均为本发明的保护范围。

1.一种用于电子雾化棒的植物源抑菌精油,其特征在于,包括以下重量份的各原料:丙二醇35-40份、丙三醇35-40份、虎杖提取物3-4份、千里光提取物2-3份、万寿菊提取物2-3份、苦参提取物3-4份、蛇床子提取物3-4份、薄荷提取物2-3份、黄芩提取物2-3份、金银花提取物2-3份、黄柏提取物2-3份、食用香精1-2份、茶叶提取物2-3份。
2.根据权利要求1所述的一种用于电子雾化棒的植物源抑菌精油,其特征在于,所述虎杖提取物的制备方法为:将虎杖粉碎,过200目筛,采用95%乙醇水浴回流提取三次,将三次得到的滤液合并,再对合并后的滤液进行减压浓缩、过滤,滤液上聚酰胺柱,再经水洗、10%乙醇醇洗至无色,再用30%乙醇冲洗,收集洗脱液,再次减压浓缩得到虎杖提取物。
