Second, Chinese medicinal composition antioxidation experiment
2.1FRAP method: the total antioxidant capacity detection kit (FRAP method) can detect the total antioxidant capacity of various body fluids such as plasma, serum, saliva, urine and the like, cell or tissue lysate, plant or Chinese herbal medicine extract or various antioxidant solutions.
The principle is as follows: under acidic condition, antioxidant can reduce Ferric-tripyridyltriazine (Fe)3+TPTZ) produces Fe in blue2+TPTZ, followed by determination of blue Fe at 593nm2+TPTZ gives the total antioxidant capacity in the sample. The detection of the total antioxidant capacity of the plant or Chinese herbal medicine extract or various antioxidant solutions can be used for detecting the strength of the antioxidant capacity of various solutions and screening drugs with strong antioxidant capacity. This study was performed with Trolox as a control. Trolox is an analogue of vitamin E, has good water solubility and strong oxidation resistance, and is similar to vitamin E.
The test results of example 1 and comparative examples 1 to 4 are shown in the following table 3:
TABLE 3
Examples | Example 1 | Comparative example 1 | Comparative example 2 | Comparative example 3 | Comparative example 4 |
Antioxidant capacity | 70% | 12% | 16% | 6% | 7% |
The results show that example 1 has an antioxidant capacity of 70% of that of the positive control (1.5mM Trolox), and all the comparative examples have a lower antioxidant capacity.
2.2ABTS method: the Total Antioxidant Capacity Assay Kit with ABTS method adopts 2,2' -azino-bis (3-ethylben-thiazoline-6-sulfonic Acid (ABTS) as a color developing agent, and can be used for detecting the Total Antioxidant Capacity of various body fluids such as plasma, serum, saliva and urine, lysates such as cells and tissues, extracts of plants or Chinese herbal medicines or various Antioxidant (Antioxidant) solutions+ABTS in the Presence of antioxidants+Is inhibited, and ABTS is measured+The absorbance can be measured and the total antioxidant capacity of the sample can be calculated. Trolox is an analogue of vitamin E, has the antioxidant capacity similar to that of vitamin E, and is used as a reference for the total antioxidant capacity of other antioxidants. For example, Trolox has a total antioxidant capacity of 1, and the antioxidant capacity of other substances at the same concentration is expressed as a multiple of the antioxidant capacity of Trolox.
