Abstract
本发明提供一种基于pH响应的DNA纳米机器建立可编程细胞间通讯的方法,使用三链DNA结构使细胞表面功能化,赋予细胞响应pH变化的功能而进行细胞组装,固定在细胞表面的三链DNA纳米结构可以响应生理条件下的pH变化,实现三链结构与双链结构的相互转换,从而连接上另一组通过互补ssDNA修饰的细胞。根据本发明提供的一种基于pH响应的DNA纳米机器建立可编程细胞间通讯的方法,利用三链DNA结构赋予细胞可编程pH响应型通讯的能力,具有用量少、操作简便快速和灵敏度高等优点。
Description
技术领域
本发明涉及生物检测技术领域,更具体地涉及一种基于pH响应的DNA纳米机器建立可编程细胞间通讯的方法。
背景技术
细胞间的通讯可以通过细胞识别来调控生理和病理过程,目前已经能够通过多种纳米材料,如仿生聚合物,ssDNA和框架核酸来实现人为编程和调控细胞通讯。但是,目前仍未实现响应外界刺激而导致的信息分子的传递。因此,利用pH值变化等胞外微环境触发的人工仿生系统,可以促进细胞增殖和迁移,促进药物高效传递。DNA在细胞膜工程化中显示出巨大的优势,通过DNA杂交反应,可以方便地构建同质或者是异质细胞群。三链DNA由于其对pH精确的响应性,已被广泛的应用于pH纳米传感器和药物释放纳米机器中。
发明内容
本发明的目的是提供一种基于pH响应的DNA纳米机器建立可编程细胞间通讯的方法,从而解决现有技术仍未实现响应外界刺激而导致的信息分子的传递的问题。
为了解决上述技术问题,本发明采用以下技术方案:
提供一种基于pH响应的DNA纳米机器建立可编程细胞间通讯的方法,包括以下步骤:S1:提供一种含有60%TAT的三链DNA结构,所述三链DNA结构的5’端修饰由数个碱基形成的ssDNA-1,茎环结构中插有淬灭基团BHQ1;S2:提供一种细胞,并在细胞膜上插入胆固醇修饰的与所述三链DNA结构5’端的ssDNA-1互补配对的ssDNA-2,将该细胞与来自步骤S1的三链DNA结构混合孵育后,通过测量细胞的荧光强度,分析细胞在不同pH环境下的荧光强度变化,确定所述三链DNA结构成功锚定在细胞膜上,并且具有可开关的特性;S3:将锚定了所述三链DNA结构的细胞与另一组修饰了ssDNA-2的细胞进行共孵育,通过流式细胞荧光分选技术确定不同pH环境下细胞的组装效率;S4:将所述三链DNA结构拆分为一个茎环结构和一条ssDNA-1,分别利用胆固醇修饰固定在细胞膜上,并且在不同pH环境下将两组细胞进行共孵育,通过流式细胞荧光分选技术确定细胞组装效率;S5:对步骤S4的其中一组细胞使用CellTracke DeepRed进行胞质染色,经三链DNA结构介导的细胞组装后,通过共聚焦荧光显微镜分析邻近细胞的胞内荧光强度即可确定细胞间的分子传递。
步骤S2包括:通过检测AL488的荧光强度在pH6.5-7.5之间的变化来测试所述三链DNA结构在细胞膜上开关的可逆性。
步骤S3和步骤S4均包括:使用流式细胞仪对细胞进行分选,统计488纳米和633纳米通道双阳性的细胞群体数量并成像。
步骤S5包括:荧光分析时所采用的激发光波长为633纳米,记录650-690纳米处的荧光发射强度。
根据本发明,首先要提供一种含有60%TAT三链结构的DNA纳米开关,可通过荧光共振能量转移来表征DNA纳米开关的pH响应性。应当知晓的是,60%TAT结构开关示意图参考如图4所示(J Am Chem Soc,2019,141,18910-18915),即开关部分含T-A·T互补配对的结构占比为60%,该占比直接决定三链DNA的pH响应范围,该三链DNA结构的设计属于本领域公开的技术,具体设计方法可参考文献J Am Chem Soc,2014,136,5836-5839.。pH环境可以为4.5,5.1,5.5,6.1,6.5,6.8,7.1,7.5,8.0,8.5和9.4。
应当理解的是,细胞膜修饰DNA的方法为本领域的公知技术,具体方法可参考文献(J Am Chem Soc,2020,142,8800-8808.)。如图1中的a和b所示,两个深色小球分别代表两组进行不同DNA修饰的细胞,虚线框内放大了细胞膜上DNA在不同pH环境下(a:pH7.5,b:pH6.5)的结构状态。
根据本发明的一个优选方案,步骤S1中所述的三链DNA结构的序列自5’-3’为:
AAGGAAGAAGTTT(BHQ-1)ACTTCTTCCTTCTTTGTTCCTTCTTC-Alexa Flour488,其中,3’端修饰了pH不敏感的基团Alexa Flour488,茎环结构中插入了淬灭基团BHQ1。
步骤S1中提供的所述三链DNA结构的5’端碱基数量优选为20~30个。
根据本发明的一个优选方案,所述三链DNA结构5’端延长的ssDNA-1的碱基序列为ACCACCACCACCACCACCAA。当pH小于6.5时,荧光基团和淬灭基团靠近,细胞膜上荧光强度低,当pH大于7.5时,荧光基团远离淬灭基团,荧光强度增高。
正如本发明的背景技术部分所述,现有技术忽略了细胞的环境响应型通讯,而本发明的创造性即主要体现在步骤S3,S4以及S5,通过这些步骤赋予了细胞响应环境pH变化进行自组装的功能,并在自组装后产生通讯的能力。
根据本发明提供的方法,使用三链DNA结构使细胞表面功能化,赋予细胞响应pH变化的功能而进行细胞组装,固定在细胞表面的三链DNA纳米结构可以响应生理条件下的pH变化,实现三链结构与双链结构的相互转换,从而连接上另一组通过互补ssDNA修饰的细胞。
综上所述,根据本发明提供的一种基于pH响应的DNA纳米机器建立可编程细胞间通讯的方法,利用三链DNA赋予细胞可编程pH响应型通讯的能力,具有用量少、操作简便快速和灵敏度高等优点。

具体实施方式
以下结合具体实施例,对本发明做进一步说明。应理解,以下实施例仅用于说明本发明而非用于限制本发明的范围。
实施例1Ramos细胞膜上修饰pH响应型三链DNA:
修饰方法如下:
1)将下表中两种序列的DNA粉末经4000rpm转速下离心1分钟,分别使用适当的超纯水溶解,最终定量为100μM。所用DNA序列如下:
2)25万Ramos细胞在1毫升的1640培养基中混匀,放入24孔板中过夜培养12小时,消化后在1200rpm转速下使用1毫升PBS离心3分钟,清洗3次。
3)使用100μL PBS重悬细胞,加入终浓度为500nM的GGT-chol,室温孵育30分钟。
4)在1200rpm转速下使用1毫升PBS离心3分钟,清洗3次。每组细胞分别使用100μLpH6.5,pH 7.5的PBS重悬细胞,加入终浓度为1μM的ACC-DTN-BHQ-AL488,室温孵育30分钟。
5)在1200rpm转速下使用1毫升DPBS离心3分钟,清洗3次。使用100μL DPBS重悬细胞,用流式细胞仪488纳米通道检测细胞荧光。
6)将来自于步骤4)的处于pH 7.5环境下的细胞在1200rpm转速下使用1毫升pH6.5的DPBS离心3分钟,清洗3次。使用100μL pH 6.5的DPBS重悬细胞,用流式细胞仪488纳米通道检测细胞荧光。再将处于pH 6.5环境下的细胞在1200rpm转速下使用1毫升pH 7.5的DPBS离心3分钟,清洗3次。使用100μL pH 7.5的DPBS重悬细胞,用流式细胞仪488纳米通道检测细胞荧光。以上操作重复3次。
本实施例步骤中的示意图如图1中的a,c所示。
本实施例步骤5)中所得到的细胞荧光强度分析如图1中的b所示。图1中的b为细胞膜上三链DNA平均荧光统计图,从图中可知荧光标记的三链DNA已经成功组装在细胞膜上并且保留了其pH依赖性。
本实施例步骤6)中所得到的细胞荧光强度分析如图1中的d所示。图1中的d为三链DNA纳米开关在细胞膜上的可逆构象变化分析图,通过检测AL488的荧光强度在pH6.5-7.5之间的变化来测试三链开关在细胞膜上的可逆性。
实施例2
三链DNA介导的pH=7.5时的细胞组装。
1)将下表中三种序列的DNA粉末经4000rpm转速下离心1分钟,使用适当的超纯水溶解,最终定量为100μM。所用DNA序列如下:
2)25万Ramos细胞在1毫升的1640培养基中混匀,放入24孔板中过夜培养12小时,消化后在1200rpm转速下使用1毫升PBS离心3分钟,清洗3次。
3)使用100μL PBS重悬细胞,用Celltrake Green对其进行胞质染色。而后加入终浓度为500nM的GGT-chol,室温孵育30分钟。
4)在1200rpm转速下使用1毫升PBS离心3分钟,清洗3次。使用100μL的PBS重悬细胞,加入终浓度为1μM的ACC-DTN,室温孵育30分钟。
5)另一组空白细胞用Celltrake Deep Red对其进行胞质染色。而后加入终浓度为500nM的chol-GAA,室温孵育30分钟。
6)两组细胞分别在1200rpm转速下使用1毫升DPBS离心3分钟,清洗3次。使用100μL的DPBS重悬细胞后,混匀,室温孵育30分钟。
7)使用流式细胞仪对细胞进行分选,统计488纳米和633纳米通道双阳性的细胞群体数量并成像。
本实施例步骤中的示意图如图2中的a所示。
在本实施例中,细胞的组装效率如图2中的b所示,在pH6.5时仅观察到少量细胞组装,而当溶液pH值为7.5时,细胞组装的效率更高且提升了7倍。多光谱流式细胞成像如图2中的c所示,交错的细胞链也证明了细胞团是由三链DNA所介导形成的。
实施例3
三链DNA介导的pH=6.5时的细胞组装。
1)将下表中两种序列的DNA粉末经4000rpm转速下离心1分钟,使用适当的超纯水溶解,最终定量为100μM。所用DNA序列如下:

5)两组细胞分别在1200rpm转速下使用1毫升DPBS离心3分钟,清洗3次。使用100μL的DPBS重悬细胞后,混匀,室温孵育30分钟。
6)使用流式细胞仪对细胞进行分选,统计488纳米和633纳米通道双阳性的细胞群体数量并成像。
本实施例步骤中的示意图如图2中的d所示。
在本实施例中,细胞的组装效率如图2中的e所示,在pH7.5时仅观察到少量细胞组装,而当溶液pH值为6.5时,细胞组装的效率更高且提升了6倍。多光谱流式细胞成像如图2中的f所示,交错的细胞链也证明了细胞团是由三链DNA所介导形成的。
实施例4

2)25万Ramos细胞在1毫升的1640培养基中混匀,放入24孔板中过夜培养12小时,消化后在1200rpm转速下使用1毫升PBS离心3分钟,清洗3次。
3)使用100μL PBS重悬细胞,加入终浓度为500nM的chol-tri488,室温孵育30分钟。
4)另一组空白细胞用Celltrake Deep Red对其进行胞质染色。而后加入终浓度为500nM的AAG-chol,室温孵育30分钟。
5)两组细胞分别在1200rpm转速下使用1毫升DPBS离心3分钟,清洗3次。使用100μL的DPBS重悬细胞后,混匀,室温孵育30分钟。
6)使用共聚焦荧光显微镜对细胞进行荧光成像,所采用的激发光波长为488/633纳米,记录500-600/650-690纳米处的荧光发射强度。
本实施例步骤中的示意图如图3中的a所示。
本实施例中,将一组细胞作为供体,在胞浆上用红色荧光素染色;另一组细胞作为受体,在细胞膜上用绿色荧光素染色。通过三链在酸性环境pH6.5的条件下将细胞连接到一起,临近的细胞之间小分子能够从供体细胞的胞质到达另一个细胞。共聚焦荧光成像图如图3中的b所示,受体细胞(细胞2)和供体细胞(细胞1)组装后呈红色荧光信号,而未组装的受体细胞(细胞3)则完全没有红色荧光信号。荧光定量分析图如图3中的b,c所示,细胞1到细胞2的红色荧光逐渐减弱,而细胞3的红色荧光几乎为零。同时,由于细胞2和细胞3的细胞膜被染色,两个细胞均能观察到绿色荧光信号,而细胞1中没有绿色荧光信号。这表明了通过三链DNA构建了响应型细胞组装后,建立了细胞间的分子转移。

Claims (5)
1.一种基于pH响应的DNA纳米机器建立可编程细胞间通讯的方法,其特征在于,包括以下步骤:
S1:提供一种含有60%TAT的三链DNA结构,所述三链DNA结构的序列为:(5’-3’)AAGGAAGAAGTTT(BHQ-1)ACTTCTTCCTTCTTTGTTCCTTCTTC-Alexa Flour488,所述三链DNA结构的5’端修饰由数个碱基形成的ssDNA-1,3’端修饰了pH不敏感的基团Alexa Flour488,茎环结构中插有淬灭基团BHQ1,所述ssDNA-1的碱基数量为20~30个;
S2:提供一种细胞,并在细胞膜上插入胆固醇修饰的与所述ssDNA-1互补配对的ssDNA-2,将该细胞与来自步骤S1的三链DNA结构混合孵育后,通过测量细胞的荧光强度,分析细胞在pH6.5-7.5之间的不同pH环境下的荧光强度变化,确定所述三链DNA结构成功锚定在细胞膜上,并且具有可开关的特性;
S3:将锚定了所述三链DNA结构的细胞与另一组修饰了ssDNA-2的细胞进行共孵育,通过流式细胞荧光分选技术确定在pH6.5-7.5之间的不同pH环境下细胞的组装效率;
S4:将所述三链DNA结构拆分为一个茎环结构和一条ssDNA-1,分别利用胆固醇修饰固定在两组细胞的细胞膜上,并且在pH6.5-7.5之间的不同pH环境下将两组细胞进行共孵育,通过流式细胞荧光分选技术确定细胞组装效率;
S5:对步骤S4的其中一组细胞使用CellTracke DeepRed进行胞质染色,经三链DNA结构介导的细胞组装后,通过共聚焦荧光显微镜分析邻近细胞的胞内荧光强度即可确定细胞间的分子传递。
2.根据权利要求1所述的基于pH响应的DNA纳米机器建立可编程细胞间通讯的方法,其特征在于,步骤S2包括:通过检测AL488的荧光强度在pH6.5-7.5之间的变化来测试所述三链DNA结构在细胞膜上开关的可逆性。
3.根据权利要求1所述的基于pH响应的DNA纳米机器建立可编程细胞间通讯的方法,其特征在于,步骤S3和步骤S4均包括:使用流式细胞仪对细胞进行分选,统计488纳米和633纳米通道双阳性的细胞群体数量并成像。
4.根据权利要求1所述的基于pH响应的DNA纳米机器建立可编程细胞间通讯的方法,其特征在于,步骤S5包括:荧光分析时所采用的激发光波长为633纳米,记录650-690纳米处的荧光发射强度。
5.根据权利要求1所述的基于pH响应的DNA纳米机器建立可编程细胞间通讯的方法,其特征在于,所述三链DNA结构5’端延长的ssDNA-1的碱基序列为ACCACCACCACCACCACCAA。
