本发明公开了一种冷冻制片技术检测橡胶草植物根组织乳管的方法,包括选取待测橡胶草完整的植物组织放入固定液中;对所述固定液进行真空抽气,抽气时间控制1‑3min;在4℃放置6‑8h;取出所述固定液中的所述植物组织浸洗三次,每次30min;所述植物组织通过蔗糖溶液逐级脱水;将脱水后的所述植物组织吸附多余水分后用Biosharp包埋剂包埋,在‑20℃放置30‑45min,将所述根组织采用冷冻切片机切片后加10%甘油封片,10min甘油凝固封片得到玻片标本后,得到橡胶草根组织中乳管分布情况。本发明能快速检测橡胶草根内乳管分布和数量情况,此方法大大缩减了检测周期,能直观反应橡胶草乳管生长状况。

Description

一种冷冻制片技术检测橡胶草植物根组织乳管的方法
技术领域
本发明属于植物组织鉴定技术领域,尤其涉及一种冷冻制片技术检测橡胶草植物根组织乳管的方法。
背景技术
橡胶草其中甲羟戊酸(MVA)途径中用于调节天然橡胶生物合成,橡胶草是二倍体(2n=16),具有相对简单的基因组,长度为1.29Gb,其中包含46,731个预测的蛋白质编码基因,橡胶草根含有高分子量橡胶分子,其特性与巴西橡胶树的橡胶胶乳相当,可以作为一种替代橡胶作物,橡胶草根的橡胶含量高,占其总干重的3%至28%,它们也是菊粉的新来源(线性β-(2-1)-连锁果聚糖),是食品工业和生物乙醇生产的重要材料。因此可以用作发明小麦基因功能的理想模型植物。橡胶草根组织中含有大量乳管细胞能够合成天然橡胶胶乳,而橡胶草含胶量与乳管数量成正比,因此,橡胶草根组织乳管细胞检测可以初步判断橡胶草的乳管发育、含胶量和最佳收获期等重要指标。但是,现有技术中缺少对检测橡胶草植物根组织乳管的制片方法,因此需要一种冷冻制片技术检测橡胶草植物根组织乳管的方法。
发明内容
本发明提供一种排除蒲公英与橡胶草杂种干扰,冷冻制片技术检测橡胶草植物根组织乳管的方法。
本发明包括以下步骤:
A选取待测橡胶草完整的植物组织放入固定液中;
B对所述固定液进行真空抽气,抽气时间控制1-3min;在4℃放置6-8h;
C取出所述固定液中的所述植物组织浸洗三次,每次30min;
D所述植物组织通过蔗糖溶液逐级脱水;
E将脱水后的所述植物组织吸附多余水分后用Biosharp包埋剂包埋,在-20℃放置30-45min,
F将所述根组织采用冷冻切片机切片后加10%甘油封片,10min甘油凝固封片得到玻片标本后,得到橡胶草根组织中乳管分布情况。
进一步地,所述固定液包括4%多聚甲醛+0.5%戊二醛
+0.1molPBS((100mL)1mol磷酸二氢钠(31.6mL)。
进一步地,所述逐级脱水的方法包括0.1molPBS,pH=7.2)2%1h→6%1h→8%1h→10%1h→12%1h→15%1h,
进一步地,所述浸洗液为0.1molPBS
进一步地,在步骤E中可去除完全脱水的根组织。
本发明的有益效果为:
本发明能快速检测橡胶草根内乳管分布和数量情况,此方法大大缩减了检测周期,能直观反应橡胶草乳管生长状况。

具体实施方式
以下对本发明的原理和特征进行描述,所举实例只用于解释本发明,并非用于限定本发明的范围。
如图1所示,在本实施例子中包括以下步骤:
本发明包括以下步骤:
A选取待测橡胶草完整的植物组织放入固定液中;
B对所述固定液进行真空抽气,抽气时间控制1-3min;在4℃放置6-8h;
C取出所述固定液中的所述植物组织浸洗三次,每次30min;
D所述植物组织通过蔗糖溶液逐级脱水;
E将脱水后的所述植物组织吸附多余水分后用Biosharp包埋剂包埋,在-20℃放置30-45min,
F将所述根组织采用冷冻切片机切片后加10%甘油封片,10min甘油凝固封片得到玻片标本后,得到橡胶草根组织中乳管分布情况。配置固定液:4%多聚甲醛+0.5%戊二醛+0.1molPBS((100mL)1mol磷酸二氢钠(31.6mL);(2)1mol磷酸氢二钠(68.4mL));
橡胶草根组织材料收取,放入固定液中;真空抽气,根据根组织的大小,以及软硬程度,抽气时间控制1-3min;随后放置4℃固定过夜(6-8h);固定好后,用0.1molPBS浸洗三次,每次30min;通过蔗糖溶液(0.1molPBS,pH=7.2)2%1h→6%1h→8%1h→10%1h→12%1h→15%1h,逐级脱水;
去除完全脱水的根组织,滤纸吸干后,用Biosharp包埋剂包埋,在-20℃放置30-45min,随后在冷冻切片机舱内操作;
冷冻切片机设置切片厚度,切好后,放置在载玻片上,再用盖玻片盖好,周边气泡,滴加10%甘油封片,10min甘油凝固封片,标记好样品信息,放置在制片盒内暂存;电子显微下观察橡胶草根组织中乳管分布及数量。
如图2所示,冷冻制片结果显示,乳管细胞在不同品种的橡胶草此冷冻制片中的分布清晰,能正确定位乳管在根中的具体位置,与传统的石蜡切片检测相比,此方法步骤简单直观,检测用时更短,只需24h即可观察到较明显的结果。
橡胶草冷冻制片技术可以较为迅速的检测跟组织内乳管细胞,并且此方法能应用于商业化种植时橡胶含量测定前提指标,为橡胶草最适收获期提供一定的理论参考价值。本发明可以通过植物冷冻切片,能够快速检测橡胶草根内的乳管分布情况,既适于从不同品种和不同发育时期的橡胶草植物材料的乳管检测,还可用于野外采集野生橡胶草植物获得较高产量品种筛选。

A选取待测橡胶草完整的植物组织放入固定液中;
B对所述固定液进行真空抽气,抽气时间控制1-3min;在4℃放置6-8h;
C取出所述固定液中的所述植物组织浸洗三次,每次30min;
D所述植物组织通过蔗糖溶液逐级脱水;
E将脱水后的所述植物组织吸附多余水分后用Biosharp包埋剂包埋,在-20℃放置30-45min;
F将所述根组织采用冷冻切片机切片后加10%甘油封片,10min甘油凝固封片得到玻片标本后,得到橡胶草根组织中乳管分布情况。
2.根据权利要求1所述一种冷冻制片技术检测橡胶草植物根组织乳管的方法,其特征在于,所述固定液包括4%多聚甲醛+0.5%戊二醛+0.1molPBS((100mL)1mol磷酸二氢钠(31.6mL)。
3.根据权利要求1所述一种冷冻制片技术检测橡胶草植物根组织乳管的方法,其特征在于,所述逐级脱水的方法包括在pH=7.2条件下采用0.1molPBS 2%1h→6%1h→8%1h→10%1h→12%1h→15%1h。
4.根据权利要求1所述一种冷冻制片技术检测橡胶草植物根组织乳管的方法,其特征在于,所述浸洗液为0.1molPBS。
5.根据权利要求1所述一种冷冻制片技术检测橡胶草植物根组织乳管的方法,在步骤E中可去除完全脱水的根组织。
