At present, microneedles are mainly used, such as skin care products applied after a minimally invasive channel is formed on the skin through metal microneedles, soluble microneedles are directly used, and the like. But the operation of the metal micro-needle requires professional personnel, and the home use of the consumer is inconvenient; the soluble micro-needle patch has relatively single efficacy, cannot be used in a large area, and the planar array layout of the soluble micro-needle patch is difficult to fit with the 3D configuration of a human face and some parts are difficult to fit. CN109381422A discloses the use of spicules and nanoparticles to form a composition for percutaneous absorption. However, the spicule is adopted, the spongiform spicule is a siliceous or calcareous material as the main component of the spicule, the siliceous or calcareous material is brittle and is not a biocompatible material, if the spicule is broken in the skin, the symptoms such as allergy and the like are easy to cause, and meanwhile, most of the spicule materials are also in a single-axis form and are difficult to effectively penetrate into the skin to establish a micro-channel.
In view of the above, the inventor of the present invention creatively designs a mask containing sea urchin-type microneedle particles with a hydro-acupuncture effect, aiming at the problems associated with pain during the injection of hydro-acupuncture, skin trauma after injection, microneedle structure and bone needle material
Disclosure of Invention
The invention mainly aims to provide a mask containing sea urchin type microneedle particles, which comprises a bonding layer, an essence layer containing the sea urchin type microneedle particles and a moisturizing and water replenishing layer, wherein the essence layer containing the sea urchin type microneedle particles is stacked on the bonding layer; a plurality of sea urchin type microneedle particles are dispersed in the essence layer; and a moisturizing layer laminated on the essence layer.
In one embodiment, the invention also provides a mask combination set containing the echinoid type microneedle particles, which comprises a mask and an essence bottle, wherein the mask comprises a fitting layer, an essence layer containing the echinoid type microneedle particles and a moisturizing and hydrating layer, wherein the essence layer containing the echinoid type microneedle particles is laminated on the fitting layer; a plurality of sea urchin type microneedle particles are dispersed in the essence layer; and the moisturizing and water replenishing layer is laminated on the essence layer, essence is filled in the essence bottle, and the essence contains nano-liposome wrapped with functional components.
In one embodiment, the sea urchin type microneedle particle is composed of a central structure and microneedles carried on the central structure.
In one embodiment, the central structure of the sea urchin type microneedle particle is one or more of a spheroid, a polyhedron, a cylinder, a tetrahedron, a hexahedron, and an octahedron.
In one embodiment, the sea urchin type microneedle particle has a central structure particle size of 80-2000 um.
In one embodiment, the number of microneedles carried on the central structure of the sea urchin type microneedle particle is 4 to 100, preferably 4 to 50, and more preferably 4 to 20.
In one embodiment, the shape of the microneedle is selected from one or more of a conical shape, a triangular pyramid shape, and a quadrangular pyramid shape.
In one embodiment, the length of the microneedle is 100-.
In one embodiment, the adhesive layer, the essence layer and the moisture-keeping and water-supplementing layer are selected from any one of non-woven fabrics, cotton fabrics, silk fabrics, down feathers, biological fiber fabrics, reticular fiber fabrics, tencel fabrics and wood pulp fabrics.
In one embodiment, the essence contains nano liposome vesicles or nano particles coated with functional ingredients, wherein the functional ingredients are selected from one or more of anti-aging, anti-wrinkle, whitening, freckle removing, acne removing, oil controlling, moisturizing and moisture keeping active ingredients.
The invention also provides sea urchin type microneedle particles, which consist of a central structure and microneedles loaded on the central structure; the central structure of the sea urchin type microneedle particles is one or more of spheroids, polyhedrons, cylinders, tetrahedrons, hexahedrons and octahedrons; the central structure particle size of the sea urchin type microneedle particles is 80-2000 um; the number of the micro-needles carried on the central structure of the sea urchin type micro-needle particle is 4-100, preferably 4-50, and more preferably 4-20; the shape of the microneedle is selected from one or more of a cone, a triangular pyramid and a rectangular pyramid; the length of the microneedle is 100-500um, preferably 200-400 um.
The invention also provides application of the sea urchin type microneedle particles in preparation of cosmetics, and the microneedle particles can be used for opening and providing skin surface channels in the cosmetics and improving the speed and the quantity of functional substances in the cosmetics entering the skin.
In one embodiment, the sea urchin type microneedle particles of the present invention can also be used in combination with nanoliposomes coated with an effective ingredient.
In one embodiment, the sea urchin type microneedle particles of the present invention are used for preparing a mask.
In one embodiment, the sea urchin type microneedle particles are used in combination with nanoliposomes wrapped with functional ingredients for preparing a facial mask.
In one embodiment, the sea urchin type microneedle particles of the present invention are prepared by hot stamping, and the material of the sea urchin type microneedle particles is selected from one or more of biocompatible materials, preferably soluble materials, more preferably polymethylmethacrylate, polylactic acid, poly (lactic-co-glycolic acid), polyglycolic acid, gelatin, and soluble starch.
The mask containing the sea urchin type microneedle particles does not contain liquid essence before use, when the mask is used, the liquid essence in an essence bottle is poured into the mask containing the sea urchin type microneedle particles, the mask is directly pasted on the face after being soaked, then the face of the mask is lightly pressed by hands, a skin conveying channel is opened by the sea urchin type microneedle particles through physical pressure, meanwhile, the liquid essence enters deep skin through the conveying channel opened by the sea urchin type microneedle particles, and the powerful anti-aging and wrinkle-removing effects are rapidly exerted.
Advantageous effects
The sea urchin type microneedle particle can play a role in multiple faces, the problem that part of face and nose skins cannot have the effect of inserting microneedles due to the 3D three-dimensional structure of the face is avoided, meanwhile, the nanoscale microneedle has small stimulation and basically no pain, and the defects that the water laser needle needs anesthesia, is strong in pain, is easy to infect and the like are overcome. The essence is added with nano liposome vesicles and nano particles which are wrapped with functional components, so that the rapid epidermal permeability of functional substances and the controlled and sustained release in deep skin can be further enhanced, and the effect of exerting the effects for a long time is achieved. The sea urchin type microneedle particles and the nanoliposome vesicles are adopted for synergistic interaction, the operation is simple and convenient, the cost is low, and the sea urchin type microneedle particles and the nanoliposome vesicles have great commercial value and market potential.

Detailed Description
The present invention will be further described with reference to specific embodiments, but the present invention is not limited to the examples in any way. The starting reagents employed in the examples of the present invention are, unless otherwise specified, those that are conventionally purchased.
Example 1
Preparing polymethyl methacrylate sea urchin microneedle particles by adopting a hot stamping method: drying the polymethyl methacrylate material at 70-75 ℃ to constant weight, and preparing the dried polymethyl methacrylate material into a sheet by an injection molding machine, wherein the sheet is used as a raw material for preparing sea urchin microneedle particles by a hot stamping process. The mould used in the hot stamping process corresponds to an intaglio stainless steel mould with a three-dimensional shape of sea urchin microneedles. The hot stamping temperature is 116 ℃, the hot stamping pressure is 12.00MPa, the temperature is kept for 1min after stamping loading, the temperature is reduced to 80 ℃, the unloading is carried out, and finally the sea urchin microneedle is obtained after demoulding. In this embodiment, the central structure of the sea urchin type microneedle is a spheroid, the central structure particle size is 1300um, the number of microneedles on the central structure particle is 8, the microneedles are conical, and the length of the microneedles is 250 um.
As shown in fig. 1 and 2, a mask 1 containing sea urchin-type microneedle particles according to the present invention includes: laminating layer 11, the quintessence layer 12 that contains sea urchin type micropin granule, a plurality of sea urchin type micropin granule 121, moisturizing layer 13, wherein, the quintessence layer 12 that contains sea urchin type micropin granule stack on laminating layer 11, a plurality of sea urchin type micropin granule 121 disperse in quintessence layer 12, moisturizing layer 13 stack on quintessence layer 12. Wherein the material of the attaching layer is selected from silk cloth, the material of the essence layer is selected from non-woven fabric, and the material of the moisturizing layer is selected from biological fiber cloth. The preparation method of the facial mask can be prepared according to the conventional processing and manufacturing method of the facial mask in the field, and comprises the steps of cutting, laminating, pasting and packaging of facial mask materials.
The sea urchin type microneedle mask 1 does not contain liquid essence before use, when the sea urchin type microneedle mask is used, the liquid essence (formula shown in table 1) in an essence bottle (shown in figure 4) is poured into the mask 1 containing sea urchin type microneedle particles, the mask is directly pasted on the face after being soaked, then the face of the mask is lightly pressed by hands, a skin conveying channel is opened by the sea urchin type microneedle through physical pressure, and meanwhile, the liquid essence enters the skin through the conveying channel opened by the sea urchin type microneedle, so that the anti-aging and wrinkle-removing effects with strong effects are rapidly exerted.

Wherein the functional substance is a nano liposome vesicle composition wrapped with carnosine active peptide molecules. The preparation method of the composition comprises the following steps: weighing the formula amount (in percentage by weight), dissolving 10% of phospholipid and 4% of cholesterol in a proper amount of ethanol in a constant-temperature water bath at 65 ℃, cooling to 45 ℃, magnetically stirring for 15min, sequentially adding 3% of propylene glycol, 10% of glycerol, 4% of carnosine and water at a constant speed, stirring while adding, and quickly and uniformly mixing to obtain the colostrum. Shearing the colostrum at 8000rpm for 10min to obtain micron-sized dispersoid, and performing 3 times of circular homogenization treatment at 120MPa in a high-pressure homogenizer to obtain carnosine nano liposome vesicle composition.
Example 2
Preparing polylactic acid sea urchin microneedle particles by adopting a hot stamping method: drying the lactic acid material at 70-75 ℃ to constant weight, preparing the lactic acid material into a sheet by an injection molding machine, and using the sheet as a raw material for preparing sea urchin microneedle particles by a hot stamping process. The mould used in the hot stamping process corresponds to an intaglio stainless steel mould with a three-dimensional shape of sea urchin microneedles. The hot stamping temperature is 135 ℃, the hot stamping pressure is 18.00MPa, the temperature is kept for 1min after stamping loading, the temperature is reduced to 80 ℃, the unloading is carried out, and finally the sea urchin microneedle is obtained after demoulding. In this example, the central structure of the sea urchin type microneedle is a hexagonal body, the central structure particle size is 1500um, the number of microneedles on the central structure particle is 6, the microneedles are conical, and the length of the microneedles is 300 um.
The preparation of the mask containing sea urchin type microneedle particles and the formula of the essence were the same as in example 1.
Example 3
The preparation method of the echinoid type microneedle and the preparation method of the mask containing the echinoid type microneedle particles are the same as those in example 1, except that the functional substance carnosine active peptide molecules in the formula of the essence are directly added into the formula and are not added in the form of a nano liposome vesicle composition.
Example 4
The preparation method of the sea urchin type microneedle particles and the preparation method of the mask containing the sea urchin type microneedle particles are the same as those in example 2, except that the functional substance carnosine active peptide molecules in the formula of the essence are directly added into the formula and are not added in the form of a nano liposome vesicle composition.
Comparative example 1
Reference example 1 a three-layer composite mask and essence were prepared, except that the essence layer of the mask did not contain echinus type microneedle particles.
Comparative example 2
Reference example 3 a three-layer composite mask and essence were prepared, except that the essence layer of the mask did not contain echinus type microneedle particles.
The invention utilizes an in vitro transdermal test to verify the transdermal effect of carnosine in the essence of different technical schemes (see table 2), and specifically comprises the following steps: a vertical Franz diffusion cell method is adopted to carry out a transdermal experiment of in-vitro rat skin, the intact rat abdominal skin is fixed between a receiving cell and a supply cell, the mask samples of the practical examples 1-4 and the comparative examples 1-2 are taken, cut into circular patches with the diameter of 1cm, and the circular patches are attached to a supply chamber, and are pressed to enable sea urchin microneedles in the patches to penetrate into the rat abdominal skin, so that essence liquid corresponding to the samples is respectively absorbed and dripped on the patches. Normal saline containing 20% propylene glycol by mass is used as receiving liquid, and the mixture is stirred at 37 ℃ for 300 r/min. Sucking 0.5mL of receiving solution in 1,2,4,6,8,10 and 12 hours, supplementing 0.5mL of isothermal blank receiving solution, detecting by HPLC, and calculating the cumulative permeation amount of the medicament at each time, wherein the calculation formula is as follows:
where Qn is the cumulative drug permeation, Cn is the drug concentration measured at the nth point, Ci is the drug concentration measured at the ith point, V0 is the volume of the diffusion cell, i.e., the amount of release medium added, and Vi is the amount of each sample. The cumulative permeation Q per unit area is Qn/S, S is the area of the diffusion cell, and 3.14cm 2. And after the permeation experiment is finished, taking down the skin, sucking the residual liquid of the sample by ultrapure water, shearing, transferring the sample into a tissue homogenizer, fully grinding the sample into homogenate, adding a proper amount of receiving solution, transferring the homogenate into a centrifuge tube, centrifuging at 7000r/min for 20min, filtering the supernatant by a 0.45-micrometer filter membrane, analyzing by HPLC, and calculating the carnosine retention in the skin. HPLC analysis method of carnosine, which selects a Sepax HP-Amino chromatographic column, uses acetonitrile and 30mmol/L dipotassium hydrogen phosphate aqueous solution (phosphoric acid is used for adjusting pH to 6.8, the volume ratio is 65: 35) as a mobile phase for analysis, carnosine is detected at 214nm, the column temperature: 30 ℃, flow rate: 1mL/min, sample size: 10 μ L.

As can be seen from the data of table 2, the cumulative permeation amount of carnosine after use of the mask to which the echinoid type microneedle particles were added (examples 1 to 4) was significantly higher than that of the general mask; the skin retention of carnosine after encapsulation using nanoliposome vesicles (examples 1-2, comparative example 1) was significantly higher than that of the example in which carnosine was directly added (examples 3-4, comparative example 2) in terms of the skin retention of carnosine. The effect of the matching use of the sea urchin type microneedle particles and the wrapped carnosine nano liposome vesicles (example 1-2) is obvious (P is less than 0.05), and is higher than that of the single use of the sea urchin type microneedle particles (example 3-4) or the carnosine nano liposome (comparative example 1), so that the synergistic effect of the two modes is shown, and the cumulative permeation amount and retention amount of the effective substances can be obviously improved by matching the sea urchin type microneedle particles and the wrapped carnosine nano liposome, so that the bioavailability of the effective substances is improved.
To further verify the efficacy of the microneedle mask of the present invention, skin irritation test and wrinkle removal efficacy test were also performed in the present invention.
1) And (3) irritation test:
human body patch test: 30 volunteers were randomly recruited and randomly divided into 6 groups, and 6 mask samples were used as subjects in examples 1 to 4 and comparative examples 1 to 2, respectively. Cutting a test object into a circular patch with the diameter of 1cm, fully soaking the essence, then pasting the patch on the curved side of the forearm of a subject, pressing 3 times with the palm to ensure that the microneedle of the sandwiched sea urchin of the patch pierces the skin, continuously pasting for 24h, observing skin reactions 30min, 24h and 48h after the patch of the test object is removed, and recording the result according to the skin reaction grading standard in technical Specification for safety of cosmetics (2015 edition) (see Table 3). The result of the human body patch test shows that: at 30min, 24h and 48h after patch removal, all subjects showed no skin local irritation reactions such as erythema, infiltration, edema, pimple, etc.

2) Evaluation of wrinkle-removing efficacy:
wrinkle removal test: 60 volunteers were randomly recruited as subjects, and divided into 6 groups, each corresponding to one of the subjects. The test points for the elasticity data were the skin under the eyes and the left and right canthus of the subject. After cleaning, use the skin elasticity testerThe MPA 580 tests the elasticity index of the skin test area and records the experimental data, then applies the mask to the face, and again tests the elasticity index of the subject's face test area and records the experimental data at the corresponding time.
The skin elasticity before, 10 minutes, half an hour, and seven days after use was compared and analyzed using the R2 value data (R2 ═ elastic-plastic weight of the springback portion/total amount of elasticity of the stretched portion. the closer the value was to 1 (100%), the better the skin elasticity was indicated. The results are shown in Table 4.

As can be seen from the data of table 4, the elasticity of the mask to which the sea urchin type microneedle particles were added (examples 1 to 4) after use was significantly higher than that of the general mask; although the elasticity index was decreased at day 7, the decrease was lower than that of the general mask, which was associated with higher skin penetration and retention of the effective ingredient after the skin channel was physically opened using the sea urchin-type microneedle particles. And after the nano liposome vesicle is used for wrapping carnosine, the skin permeability and the retention capacity are further improved, so that the skin still keeps a higher elasticity index 7 days after the use. For the common facial mask without microneedle particles or the examples or comparative examples without using nanoliposomes to wrap the functional components (see examples 3-4 and comparative examples 1-2), the functional components are not established to enter the skin channel, and the skin permeability is not high; without the use of nanoliposomes to encapsulate the active ingredients, skin permeability and long-term retention of the active ingredients into the skin are affected, the skin elasticity index is increased only in a limited way, and the effect cannot be maintained for a long time. The synergistic effect of the echinus type microneedle particles in cooperation with the carnosine nano liposome is further proved from the effects, the echinus type microneedle particles in cooperation with the carnosine nano liposome can remarkably improve the accumulated permeability and retention of the functional substances, improve the bioavailability of the functional substances, enable the skin to keep elasticity for a long time and achieve the effects of resisting aging and removing wrinkles.
It should be understood that the above-described embodiments of the present invention are merely examples for clearly illustrating the present invention, and are not intended to limit the embodiments of the present invention. Other variations and modifications will be apparent to persons skilled in the art in light of the above description. And are neither required nor exhaustive of all embodiments. Any modification, equivalent replacement, and improvement made within the spirit and principle of the present invention should be included in the protection scope of the claims of the present invention.
Claims (10)
1. The sea urchin type microneedle particle is characterized by consisting of a central structure and microneedles loaded on the central structure.
2. The sea urchin-type microneedle particle of claim 1, wherein the sea urchin-type microneedle particle has a central structure of a spheroid, polyhedron or cylinder; preferably the polyhedron is tetrahedral, hexahedral or octahedral.
3. The sea urchin-type microneedle particle of claim 1, wherein the sea urchin-type microneedle particle has a central structure particle size of 80 to 2000 um.
4. The sea urchin-type microneedle particle of claim 1, wherein the number of microneedles carried on a central structure of the sea urchin-type microneedle particle is 4 to 100; the sea urchin type microneedle particles are made of a biocompatible material, preferably a soluble material, more preferably one or more of polymethyl methacrylate, polylactic acid, poly (lactic-co-glycolic acid), polyglycolic acid, gelatin and soluble starch.
5. The sea urchin-type microneedle particle of any one of claims 1 to 4, wherein the microneedle has a length of 100 and 500 um.
6. Use of sea urchin-type microneedle particles having the structure as set forth in any one of claims 1 to 5 in the preparation of cosmetics.
7. The use of claim 6, wherein the sea urchin type microneedle particles are used in combination with nanoliposomes coated with an effective component.
8. The use according to claim 6, the sea urchin type microneedle particles are used for preparing a facial mask, and preferably the sea urchin type microneedle particles are used in combination with nanoliposomes wrapped with functional ingredients for preparing the facial mask.
9. A facial mask containing sea urchin type microneedle particles is characterized in that: the mask pack (1) comprises: a bonding layer (11); an essence layer (12) containing sea urchin-type microneedle particles, which is laminated on the adhesive layer (11); a plurality of sea urchin type microneedle particles (121) are dispersed in the essence layer (12); a moisture-retaining and water-replenishing layer (13) laminated on the essence layer (12) containing sea urchin-type microneedle particles, the structure of which is as set forth in any one of claims 1 to 5.
10. A facial mask combination suit containing sea urchin type microneedle particles is characterized in that: the facial mask combination set comprises a facial mask (1) and an essence bottle, wherein the facial mask (1) comprises: a bonding layer (11); an essence layer (12) containing sea urchin-type microneedle particles, which is laminated on the adhesive layer (11); a plurality of sea urchin type microneedle particles (121) are dispersed in the essence layer (12); a moisture-retaining and water-replenishing layer (13) laminated on the essence layer (12) containing sea urchin-type microneedle particles, the structure of which is as set forth in any one of claims 1 to 5; the essence bottle is filled with essence which contains nano-liposome wrapped with functional components.
