Abstract
The invention discloses a method for prolonging the flowering phase of hemerocallis fulva flowers, which comprises the following steps: (1) preparing an in-vitro culture medium of the hemerocallis fulva flowers for later use, wherein 6-BA is added into the in-vitro culture medium, and the concentration of the BA is 10-20 mg/L; (2) picking the blossom of hemerocallis fulva from the living body of the hemerocallis fulva, removing the part below the receptacle, and keeping the receptacle; (3) the method is simple to operate and low in cost, can remarkably delay the withering rate of the hemerocallis flowers, prolong the opening time of the hemerocallis flowers, and increase the ornamental time of the hemerocallis flowers, and has obvious social and economic benefits and wide application prospect, thereby being beneficial to popularization and application.
Description
Technical Field
The invention relates to the technical field of plant cultivation, in particular to a method for prolonging the flowering phase of hemerocallis fulva flowers.
Background
Hemerocallis fulva (Hemerocallis fulva) is perennial root flower of Hemerocallis of Liliaceae (Liliaceae), has about 14 natural species, various varieties and rich flower colors and flower types, and is a good greening and courtyard ornamental plant. The natural seeds of hemerocallis fulva in China are 11, the resources of the seeds are rich, the functions are wide, and the seeds can be seen as flowers, eaten as vegetables and used as medicines. The variety of hemerocallis occupies an important position in the gardens and greening in the world. At present, the market demand for the hemerocallis is continuously increased, and the research and the application of the hemerocallis are more and more paid attention by researchers. Although the flowers of the hemerocallis fulva are beautiful, the flowering period of the hemerocallis fulva is short, the time from the day of opening to withering of most varieties is less than 24 hours, the withering rate of the hemerocallis fulva flowers is fast, and the ornamental time is short. Therefore, there is an urgent need to develop a method for prolonging the flowering period of hemerocallis fulva flowers to solve the above technical problems.
In view of the above, the present invention is particularly proposed.
Disclosure of Invention
The invention aims to provide a method for prolonging the flowering phase of hemerocallis fulva flowers, which is simple to operate, low in cost, capable of remarkably delaying the withering rate of the hemerocallis fulva flowers, prolonging the opening time of the hemerocallis fulva flowers, increasing the available viewing time of the hemerocallis fulva flowers, obvious in social and economic benefits, wide in application prospect and beneficial to popularization and application.
In order to achieve the above object, the present invention provides a method for prolonging the flowering phase of hemerocallis fulva flowers, comprising the following steps:
(1) preparing an in-vitro culture medium of the hemerocallis fulva flowers for later use, wherein 6-BA is added into the in-vitro culture medium, and the concentration of the BA is 10-20 mg/L;
(2) picking the blossom of hemerocallis fulva from the living body of the hemerocallis fulva, removing the part below the receptacle, and keeping the receptacle;
(3) immediately soaking the receptacle of the daylily flower in the in-vitro culture medium for culture, wherein the soaking depth is 1-2 cm.
Preferably, the essential components of the in vitro culture medium include sucrose, sodium benzoate and MES.
Preferably, the concentration of sucrose, the concentration of sodium benzoate and the concentration of MES in the in vitro culture medium are respectively 15-25g/L, 0.5-1.5g/L and 20-40mmol/L respectively.
Preferably, the concentration of sucrose, the concentration of sodium benzoate and the concentration of MES in the in vitro culture medium are respectively 20g/L, 1.0g/L and 30mmol/L respectively.
Preferably, the pH of the in vitro culture medium is 5.6-6.0.
The method for prolonging the flowering phase of the hemerocallis fulva flowers has the following beneficial effects.
1. According to the method, the in-vitro culture of the hemerocallis fulva flowers can obviously delay the withering rate of the hemerocallis fulva flowers, prolong the service life of the hemerocallis fulva flowers and prolong the ornamental time of the hemerocallis fulva flowers.
2. The variety of the hemerocallis fulva is various in China, the flowering period of hemerocallis fulva flowers is prolonged, more social and economic benefits can be brought, and future application and popularization of the hemerocallis fulva are facilitated.

Detailed Description
The present invention will be further described with reference to the following specific embodiments and accompanying drawings to assist in understanding the contents of the invention.
Example 1
A method for prolonging the flowering phase of hemerocallis fulva flowers comprises the following steps:
(1) preparing an in-vitro culture medium for hemerocallis fulva flowers for later use, wherein the components of the in-vitro culture medium comprise sucrose, sodium benzoate, MES and 6-BA, the concentration of the sucrose is 25g/L, the concentration of the sodium benzoate is 0.5g/L, the concentration of the MES is 40mmol/L, the concentration of the 6-BA is 10mg/L, and the pH value of the in-vitro culture medium is 6.0;
(2) picking full-bloom hemerocallis middendorfii flowers in the field at about 9:30 in the morning, picking the full-bloom hemerocallis middendorfii flowers from the living bodies of the hemerocallis middendorfii plants, removing the parts below the receptacle, and keeping the receptacle;
(3) immediately soaking the receptacle of the hemerocallis fulva flowers in the in-vitro culture medium for culture, wherein the soaking depth is 1cm, 3 repetitions are set for each treatment of the hemerocallis fulva flowers, and the hemerocallis fulva flowers with good vigor are selected for each repetition.
(4) The survival time of day lily flowers was observed and recorded. The growth condition of the hemerocallis fulva flowers is as follows: at about 20:00 night, the petals begin to curl, about 20:30, the flowers are basically withered, about 22:00, and the flowers are completely withered. The flowers were treated from 9:30 in the morning until the flowers withered and the survival time of the flowers was 12.5 h. The growth condition of the day lily flower of the embodiment 1 of the invention in one day is shown in fig. 1.
Example 2
A method for prolonging the flowering phase of hemerocallis fulva flowers comprises the following steps:
(1) preparing an in-vitro culture medium for hemerocallis fulva flowers for later use, wherein the components of the in-vitro culture medium comprise sucrose, sodium benzoate, MES and 6-BA, the concentration of the sucrose is 15g/L, the concentration of the sodium benzoate is 1.5g/L, the concentration of the MES is 20mmol/L, the concentration of the 6-BA is 20mg/L, and the pH value of the in-vitro culture medium is 5.6;
(2) picking full-bloom hemerocallis middendorfii flowers in the field at about 9:30 in the morning, picking the full-bloom hemerocallis middendorfii flowers from the living bodies of the hemerocallis middendorfii plants, removing the parts below the receptacle, and keeping the receptacle;
(3) immediately soaking the receptacle of the hemerocallis fulva flowers in the in-vitro culture medium for culture, wherein the soaking depth is 2cm, 3 repetitions are set for each treatment of the hemerocallis fulva flowers, and the hemerocallis fulva flowers with good vigor are selected for each repetition.
(4) The survival time of day lily flowers was observed and recorded. The growth condition of the hemerocallis fulva flowers is as follows: at night, the flowers begin to wither at about 20:30, at about 21:00, the flowers basically wither, at about 22:00, and the flowers completely wither. The flowers were treated from 9:30 in the morning until the flowers withered and the survival time of the flowers was 12.5 h. The growth condition of the day lily flower of the embodiment 2 of the invention in one day is shown in fig. 1.
Example 3
A method for prolonging the flowering phase of hemerocallis fulva flowers comprises the following steps:
(1) preparing an in-vitro culture medium for hemerocallis fulva flowers for later use, wherein the components of the in-vitro culture medium comprise sucrose, sodium benzoate, MES and 6-BA, the concentration of the sucrose is 20g/L, the concentration of the sodium benzoate is 1.0g/L, the concentration of the MES is 30mmol/L, the concentration of the 6-BA is 15mg/L, and the pH value of the in-vitro culture medium is 5.8;
(2) picking full-bloom hemerocallis middendorfii flowers in the field at about 9:30 in the morning, picking the full-bloom hemerocallis middendorfii flowers from the living bodies of the hemerocallis middendorfii plants, removing the parts below the receptacle, and keeping the receptacle;
(3) immediately soaking the receptacle of the hemerocallis fulva flowers in the in-vitro culture medium for culture, wherein the soaking depth is 1.5cm, 3 repetitions are set for each treatment of the hemerocallis fulva flowers, and the hemerocallis fulva flowers with good vigor are selected for each repetition.
(4) The survival time of day lily flowers was observed and recorded. The growth condition of the hemerocallis fulva flowers is as follows: at night, about 19:00, the petals are obviously curled, at 20:30, the flowers are obviously withered, about 21:00, the withering degree is about the same as 20:30, about 22:00, the flowers are basically withered, about 22:30, and the flowers are completely withered. The flowers were treated from 9:30 in the morning until the flowers withered and the survival time of the flowers was 13 h. The growth condition of the day lily flower of the embodiment 3 of the invention in one day is shown in fig. 3.
Comparative example
A method for prolonging the flowering phase of hemerocallis fulva flowers comprises the following steps:
(1) preparing an in-vitro culture medium for hemerocallis fulva flowers for later use, wherein the components of the in-vitro culture medium comprise sucrose, sodium benzoate and MES, the concentration of the sucrose is 20g/L, the concentration of the sodium benzoate is 1.0g/L, the concentration of the MES is 30mmol/L, and the pH value of the in-vitro culture medium is 5.8;
(2) picking full-bloom hemerocallis middendorfii flowers in the field at about 9:30 in the morning, picking the full-bloom hemerocallis middendorfii flowers from the living bodies of the hemerocallis middendorfii plants, removing the parts below the receptacle, and keeping the receptacle;
(3) immediately soaking the receptacle of the hemerocallis fulva flowers in the in-vitro culture medium for culture, wherein the soaking depth is 1.5cm, 3 repetitions are set for each treatment of the hemerocallis fulva flowers, and the hemerocallis fulva flowers with good vigor are selected for each repetition.
(4) The survival time of day lily flowers was observed and recorded. The growth condition of the hemerocallis fulva flowers is as follows: at about 20:00 night, the flowers start to wither, and at about 20:30, the flowers completely wither. From 9:30 in the morning until the flowers wither, the survival time of the flowers is 11 h. The growth state of the day lily flower of the control example in one day is shown in fig. 4.
By comparison, the method for prolonging the flowering phase of the hemerocallis fulva flowers can be used for treating the hemerocallis fulva flowers, so that the survival time of the hemerocallis fulva flowers is remarkably longer than that of a control group, the withering rate of the hemerocallis fulva flowers can be remarkably reduced, and the survival time of the hemerocallis fulva flowers can be prolonged.
According to the method, the in-vitro culture of the hemerocallis fulva flowers can obviously delay the withering rate of the hemerocallis fulva flowers, prolong the service life of the hemerocallis fulva flowers and prolong the ornamental time of the hemerocallis fulva flowers. The variety of the hemerocallis fulva is various in China, the flowering period of hemerocallis fulva flowers is prolonged, more social and economic benefits can be brought, and future application and popularization of the hemerocallis fulva are facilitated.

(1) preparing an in-vitro culture medium of the hemerocallis fulva flowers for later use, wherein 6-BA is added into the in-vitro culture medium, and the concentration of the BA is 10-20 mg/L;
(2) picking the blossom of hemerocallis fulva from the living body of the hemerocallis fulva, removing the part below the receptacle, and keeping the receptacle;
(3) immediately soaking the receptacle of the daylily flower in the in-vitro culture medium for culture, wherein the soaking depth is 1-2 cm.
2. The method of claim 1, wherein the essential components of said in vitro culture medium comprise sucrose, sodium benzoate, and MES.
3. The method of claim 2, wherein the concentration of sucrose, sodium benzoate and MES in said in vitro culture medium is 15-25g/L, 0.5-1.5g/L, respectively.
4. The method of claim 3, wherein the concentration of sucrose, sodium benzoate and MES in said in vitro culture medium are respectively 20g/L, 1.0g/L and 1.0g/L, respectively.
5. The method of claim 1, wherein the pH of said in vitro culture medium is between 5.6 and 6.0.
