Example 8
The experimental results were verified for the compositions obtained in any of examples 1 to 7:
and (3) hair growth effect verification:
experimental animals: c57BL/6 mice, half male and female, clean grade, 7-8 weeks old, body weight 20-22g, total 40, provided by the animal center of Xinjiang medical university; feeding the mice in a laboratory for 7 days in a conventional way, and starting an experiment after the mice adapt to the environment;
the experimental method comprises the following steps: grouping animals, modeling and administering;
grouping: 40C 57BL/6 mice were randomly divided into five groups, namely a model group, a positive control group, a traditional Chinese medicine compound group and a blank group. The first three components are correspondingly coated with distilled water, and the baofantang six-ingredient alopecia preventing and hair growing tincture is prepared;
molding: the mice were anesthetized with ether, a paraffin/rosin mixture (1:1) was melted, smeared on the back of the mice, and after solidification and hardening, the mixture was removed. And (3) the successful standard of molding: the mouse has no abnormal expression, the surface of the back skin is smooth, no root hair and wound are preferred, the depilation area is about 2cm multiplied by 2cm, and the hair of the mouse is induced to enter the growth period from the resting period;
administration: coating corresponding medicines on the depilatory area twice a day from the next day after depilation;
detecting the apoptosis number of hair follicle cells:
at 15d after depilation, all mice were sacrificed by cervical dislocation. Fixing skin at the same part of a depilation region with 10% formaldehyde, dehydrating, embedding in paraffin, slicing, spreading on a polylysine glass slide, and performing in-situ hybridization by using an in-situ hybridization technique as an end marking method (detecting the growth/apoptosis condition of cells in hair follicles);
the statistical method comprises the following steps:
the experimental data were analyzed using statistical software SPSS 17.0, and the measured data were expressed as mean ± standard deviation and tested using the t-test method. The counting data are expressed by a rate, and the counting data are considered to have statistical significance by adopting a Chi 2 test and P is less than 0.05;
as a result:
the detection result of the apoptosis of the hair follicle cells in the hair removal area on the back of each group of mice shows that the numbers of the apoptosis of the hair follicle cells in the blank group and the model control group are respectively (41.56 +/-0.32) and (42.77 +/-0.68) per visual field at 15d after hair removal; the numbers of apoptosis of hair follicle cells of mice in the traditional Chinese medicine compound group and the positive control group are respectively (16.54 +/-0.33) and (17.32 +/-0.37) per visual field. The apoptosis number of the traditional Chinese medicine compound group and the positive control group is obviously less than that of the blank group and the model group, and P is less than 0.05, so that the statistical significance is achieved. Meanwhile, the traditional Chinese medicine compound group and the positive control group as well as the blank group and the model group have no statistical significance when P is more than 0.005. The conclusion is drawn that the traditional Chinese medicine compound has obvious inhibition effect on the apoptosis of hair follicle cells, has similar effect to the Baofitang six-ingredient alopecia preventing and hair growing tincture on the hair growing, and has no obvious correlation between the inhibition effect and the solvent. The traditional Chinese medicine composition is proved to have the hair growth effect.
Example 9
And (3) hair blackening effect verification:
experimental animals: c57BL/6 mice, half male and female, clean grade, 7-8 weeks old, body weight 20-22g, total 40, provided by the animal center of Xinjiang medical university; feeding the mice in a laboratory for 7 days in a conventional way, and starting an experiment after the mice adapt to the environment;
the experimental method comprises the following steps: animal grouping, modeling, administration:
grouping: 40C 57BL/6 mice were randomly divided into four groups, namely a model group, a positive control group, a Chinese medicinal compound group and a blank group. The first three components are coated with distilled water and polygonum multiflorum extracting solution correspondingly, and the blank group is not coated with any medicine;
molding: anesthetizing the mouse with ether, heating and melting paraffin/rosin mixture (1:1), smearing on the back of the mouse, solidifying and hardening, and removing the mixture; 12h after unhairing, selecting a mouse without a wound on the back, smearing a hydrogen peroxide solution with the concentration of 6% on the unhairing position, 2 times a day for 7d, and making a model successfully according to the standard: the mouse has no abnormal expression, white villi grow on the surface of back skin, no black miscellaneous hair, no black skin and wound are preferably on the back, the depilation area is about 2cm multiplied by 2cm, melanocytes in hair follicles are damaged, and light-colored hair is induced to grow;
administration: respectively coating corresponding medicines on the hair growing areas with light color twice a day after the modeling is successful;
and (3) recovery detection of hair follicle melanocytes:
15d after the modeling is successful, all mice are killed by a cervical dislocation method, the skin at the same position of a depilation region is taken, 10% formaldehyde is used for fixing, dehydration and paraffin embedding are carried out, two longitudinal sections and two transverse sections of the skin are taken for each section, HE dyeing is respectively carried out, and the distribution condition of melanin of each group of the longitudinal sections is observed under a microscope;
the statistical method comprises the following steps:

the results of the back longitudinal section of each group of mice show that the number of melanin hair follicles in the blank group is (110.43 +/-16.22) per visual field, the number of melanin hair follicles in the traditional Chinese medicine compound group is (133.05 +/-11.52) per visual field, the number of melanin hair follicles in the positive control group is (134.77 +/-12.09) per visual field, and the number of melanin hair follicles in the model group is (45.44 +/-9.02) per visual field; compared with the traditional Chinese medicine compound group, the blank group has statistical significance when P is less than 0.05; comparing the blank group with the positive control group, wherein P is less than 0.01, and the statistical significance is achieved; compared with the model group, the blank group has no statistical significance when P is more than 0.05; compared with the positive control group, the traditional Chinese medicine compound group has statistical significance when P is less than 0.01. The conclusion is that the traditional Chinese medicine compound has obvious promotion effect on the growth of the melanocyte of the hair follicle, is similar to the effect of the polygonum multiflorum extracting solution on blacking the hair, and has no obvious relationship between the promotion effect and the solvent. The traditional Chinese medicine composition is proved to have the hair blacking effect.
Typical cases (the following experiments were performed with the patient's own consent):
case 1:
mr. Wang, Man, age 64, Xinjiang Shihe university, diagnosed hair loss, thinning, white hair. Symptoms and signs (clinical manifestations) are that the hair is shed and sparse ten years ago, and the hair is whitened to more than 70%; many treatments have not been effective and have become more serious over the years. After the traditional Chinese medicine composition prepared in the example 2 is used for 4 months in 2020, the hair is normally cleaned, and then the hair is smeared on the root of the hair, and is properly massaged to promote absorption, 1 time in two days with the dosage of about 4-5ml each time, and the hair does not need to be washed after use, and is naturally dried in the air, so that the hair is kept on the scalp and the nutrition of the hair root is kept. Case 2:
zhao teacher, man, 56 years old, Shihe university, pharmacist, diagnosed baldness and canities in 9 months of 2020. the hair is white, the study pressure is huge, and the white hair accounts for about one third of the whole hair after being usually left up all night; many medicines have no effect when being taken orally or externally used as hair blackening liquid; after the traditional Chinese medicine composition provided by the embodiment 2 is used, the hair is normally cleaned, the hair is smeared on the root of the hair, and is properly massaged to promote absorption, the dosage is about 4-5ml every time after 1 time every two days, the hair does not need to be washed after use, the hair is naturally dried in the air, the hair is kept on the scalp and the nutrition of the hair root is kept, the white hair disappears after 2 months, and the hair does not relapse at present after follow-up.
By observing 100 patients, the traditional Chinese medicine composition provided by the invention is used, the cure rate is 64%, the effective rate is 95%, and the ineffective rate is 5%.

The traditional Chinese medicine composition with the effects of growing and blackening hair is characterized by being prepared from 25-45 parts by weight of radix bupleuri, 10-23 parts by weight of semen pruni armeniacae, 20-35 parts by weight of nigella sativa seeds, 12-20 parts by weight of pomegranate seeds, 10-25 parts by weight of black sesame seeds, 3-8 parts by weight of beet and 2-4 parts by weight of medically acceptable auxiliary materials, namely menthol, 2-3 parts by weight of tetrahydropiperine, 2-4 parts by weight of azone and 5-10 parts by weight of simethicone.

The traditional Chinese medicine composition with the effects of growing and blacking hair according to claim 1, which is prepared from 25-30 parts by weight of radix bupleuri, 10-18 parts by weight of semen pruni armeniacae, 20-30 parts by weight of nigella sativa seeds, 12-15 parts by weight of pomegranate seeds, 10-20 parts by weight of black sesame seeds, 3-5 parts by weight of beet and 2-4 parts by weight of pharmaceutically acceptable auxiliary materials such as menthol, 2-3 parts by weight of tetrahydropiperine, 2-4 parts by weight of azone and 5-10 parts by weight of simethicone.

The traditional Chinese medicine composition with the effects of growing and blacking hair according to claim 1, which is prepared from 30 parts by weight of radix bupleuri, 15 parts by weight of semen pruni armeniacae, 25 parts by weight of nigella sativa seeds, 12 parts by weight of pomegranate seeds, 15 parts by weight of black sesame seeds, 4 parts by weight of beet and 4 parts by weight of medically acceptable auxiliary materials such as menthol, 3 parts by weight of tetrahydropiperine, 4 parts by weight of azone and 7 parts by weight of simethicone.
4. The preparation method of the traditional Chinese medicine composition with the effects of growing and blacking hair according to claim 1 is characterized by comprising the following steps:
a. weighing the raw materials according to the formula ratio, drying 25-45 parts of radix bupleuri, 10-23 parts of semen pruni armeniacae, 20-35 parts of nigella sativa seeds, 12-20 parts of pomegranate seeds, 10-25 parts of black sesame seeds and 3-8 parts of beet at the temperature of 80-90 ℃, and crushing by using a crusher; sieving with 200-300 mesh sieve to remove insoluble solid to obtain mixture;
b. using CO for the mixture obtained in the step a2Extracting with a supercritical extractor under 40-70MPa at 40-60 deg.C for 180 min to obtain liposoluble fraction and residue;
c. b, adding 20-40 times of edible alcohol solution with the mass fraction of 50-95% into the medicine residues obtained in the step b, boiling for 30-60 minutes under reduced pressure, extracting for 3 times, extracting for 2-4 hours each time, and combining the extracting solutions;
d. concentrating the extracting solution obtained in the step c to the density of 1.00-1.80g/ml, and then uniformly mixing the fat-soluble part obtained in the step b with the concentrated solution to obtain a stock solution A;
e. adding 2-4 parts of menthol, 2-3 parts of tetrahydropiperine, 2-4 parts of azone and 5-10 parts of simethicone into 5-8 parts of alkyl glycoside with average polymerization degree of 1.4-1.6, solid content of more than or equal to 50% and density of 1.05-1.15, and uniformly mixing by using a stirrer to obtain stock solution B;
f. and d, uniformly mixing the stock solution A obtained in the step d with the stock solution B obtained in the step e to obtain the traditional Chinese medicine composition with the functions of growing and blacking hair.
