Abstract
本发明涉及一种饲料用鸭油脂肪酸甘油酯的制备方法,包括以下步骤:甘油与鸭油于反应器中混合,加入碱性催化剂和相转移剂,在1~2kPa真空下放入油浴,以5~10℃/min的速度升温至80~85℃,搅拌下反应2~4min,然后以10~15℃/min的速度升温至180~220℃,搅拌下反应2~4h后,立即用磷酸中和至pH 6~7,并以25~30℃/min的速度降温至100~110℃,固化得到蜡状产品。本发明方法针对油脂与甘油混溶性差的特点,采用相转移剂增加油脂与甘油的混溶性,促进反应进行,合理设置脂肪酸甘油酯合成工艺中反应温度、反应时间、反应物用量比,选择合适的催化剂、相转移剂及用量关系,提高了鸭油甘油解的效率,最终所得单甘酯和甘二酯的质量分数可达80%以上。
Description
技术领域
本发明涉及一种饲料用鸭油脂肪酸甘油酯的制备方法,属于脂肪酸甘油酯制备领域。
背景技术
油脂是饲料中能量和必需脂肪酸的重要来源,不仅能提供动物自身体内不能合成或合成很少的必需脂肪酸,还能改善饲料的适口性和外观特性,并促进脂溶性维生素的有效利用,使畜禽的生产性能得到提高,因此油脂在畜禽生产中具有重要作用。然而普通油脂主要为甘三酯,作为饲料使用存在消化率低、油脂品质差、油脂氧化酸败严重、乳化效果不好等问题,致使饲料营养价值低、口感差、颗粒与油脂不均匀,从而影响动物食用及健康生长。因此,在饲料油脂使用过程中把控油脂品质、提高油脂消化利用率及饲料的乳化性尤为重要,而脂肪酸甘油酯则因消化率高、乳化效果好,在饲料中广泛应用。
鸭油作为一种重要的动物油脂,其胆固醇含量相对其他动物油含量比较低,是动物油中比较利于健康的品种。此外,鸭油的饱和脂肪酸、单不饱和脂肪酸(主要为油酸)、多不饱和脂肪酸的比例较好(27:56:17),与橄榄油接近,营养价值较高,目前主要用作饲用油脂,然而鸭油与其他饲用油脂类似,存在消化率和乳化性不高的问题,如将鸭油进行甘油解,不但能保留鸭油原有的脂肪酸,还能提高鸭油的乳化性和消化率,更好的用于饲料中。目前,油脂的甘油解主要有化学法和酶法,其中,酶催化合成法虽然具有选择性高,反应条件温和,对设备要求不高等优点,但是其产率较低,且脂肪酶价格较高,工业化生产受到一定限制。而化学法由于甘油与油脂的混溶性较差,因此在工业上也受到一定程度限制。
发明内容
针对现有技术的不足,本发明的目的是提供一种饲料用鸭油脂肪酸甘油酯的制备方法。
为了实现上述目的,本发明所采用的技术方案是:
一种饲料用鸭油脂肪酸甘油酯的制备方法,包括以下步骤:
将甘油与鸭油于反应器中混合,加入碱性催化剂和相转移剂,在1~2kPa真空下放入油浴,以5~10℃/min的速度升温至80~85℃,搅拌下反应2~4min,然后继续以10~15℃/min的速度升温至180~220℃,搅拌下反应2~4h后,立即用磷酸中和至pH 6~7,并以25~30℃/min的速度降温至100~110℃,固化得到蜡状产品;所述甘油与鸭油的质量比为1:2.5~4.5。
所述碱性催化剂为乙酸钾、K3PO4或NaOH。
所述碱性催化剂的质量为鸭油质量的0.2~0.6%。
所述相转移剂的用量为鸭油质量的0~0.4%。
所述相转移剂为单辛酸甘油酯。
本发明有益效果:
本发明方法针对油脂与甘油混溶性差的特点,采用相转移剂增加油脂与甘油的混溶性,促进反应进行,合理设置脂肪酸甘油酯合成工艺中反应温度、反应时间、反应物用量比,选择合适的催化剂、相转移剂及用量关系,实现了对化学法合成鸭油脂肪酸甘油酯工艺的优化,提高了鸭油甘油解的效率,最终所得单甘酯和甘二酯的质量分数可达80%以上。
本发明基于油脂甘油解分步反应的原理,先生成1,3-甘油二酸酯和1,2-甘油二酸酯,再通过第二步酯交换反应生成α-或β-单甘脂,随着反应的进行,体系中形成了单甘酯、甘二酯、甘三酯和甘油的多相混合物,过量的甘油有利于单甘脂和甘二酯的生成。
本发明食品级催化剂乙酸钾(KOAc)、K3PO4或NaOH的加入,不但可以促进鸭油酯键的断裂,加速脂肪酸酯与甘油的酯交换反应速度,而且食品级催化剂的使用使饲料制品更加安全;同时,KOAc还可以有效保护产品的颜色,从而得到颜色更浅的产品。
然而,由于甘油与鸭油溶解性的差异使得该反应为界面反应,速率较低。本发明相转移剂单辛酸甘油酯结构中既有长碳链的脂肪酸部分,又有亲水的甘油部分,加入至反应体系中作为相转移剂,不但可以促进不同相间的互相转移,有效加速甘油与鸭油的融合,还不影响最终产品的结构,从而达到缩短反应时间、降低反应温度,得到更高质量的鸭油脂肪酸甘油酯产品的目的。
具体实施方式
以下结合实施例对本发明的具体实施方式作进一步详细说明。
实施例
一种饲料用鸭油脂肪酸甘油酯的制备方法,包括以下步骤:
将甘油与鸭油于反应器中混合,加入碱性催化剂KOAc(乙酸钾)、K3PO4或NaOH(条件①)、相转移剂单辛酸甘油酯,其中甘油与鸭油的质量比为1:2.5~4.5(条件②),碱性催化剂的质量为鸭油质量的0.2~0.6%(条件③),单辛酸甘油酯的质量为鸭油质量的0~0.4%(条件④);于1~2KPa真空下放入油浴中,以5~10℃/min的速度升温至80~85℃,搅拌反应2~4min,然后继续以10~15℃/min的速度升温至180~220℃(条件⑤),搅拌下反应2~4h(条件⑥)后,立即用磷酸中和至pH6~7,并以25~30℃/min的速度降温至100~110℃,固化得到蜡状产品。
具体实施例如下表实施例1~10所示,采用薄层色谱法对所得蜡状产品的成分及含量进行分析,结果如下。
由上表可知,脂肪酸甘油酯合成工艺中反应温度、反应时间、反应物用量比、催化剂、相转移剂等条件均影响最终产品中单甘酯、甘二酯及甘三酯的含量占比。因此,通过合理调整各参数的数值范围,可提高鸭油的甘油解效率,最终所得单甘脂和甘二酯的质量分数可达80%以上。

1.一种饲料用鸭油脂肪酸甘油酯的制备方法,其特征在于,包括以下步骤:
将甘油与鸭油于反应器中混合,加入碱性催化剂和相转移剂,在1~2kPa真空下放入油浴,以5~10℃/min的速度升温至80~85℃,搅拌下反应2~4min,然后继续以10~15℃/min的速度升温至180~220℃,搅拌下反应2~4h后,立即用磷酸中和至pH 6~7,并以25~30℃/min的速度降温至100~110℃,固化得到蜡状产品;所述甘油与鸭油的质量比为1:2.5~4.5。
2.如权利要求1所述的制备方法,其特征在于,所述碱性催化剂为乙酸钾、K3PO4或NaOH。
3.如权利要求2所述的制备方法,其特征在于,所述碱性催化剂的质量为鸭油质量的0.2~0.6%。
4.如权利要求1所述的制备方法,其特征在于,所述相转移剂的用量为鸭油质量的0~0.4%。
