Abstract
本发明公开了一种防治鱼类多子小瓜虫的中药组合物,包括以下原料:雷公藤1‑5份,苦楝子1‑5份,槟榔1‑5份,桉树叶3‑7份,樟树叶1‑5份,铁苋菜3‑7份,百部1‑5份,地锦草3‑7份。本发明还公开一种防治鱼类多子小瓜虫的中药制剂,其制备方法包括提取、合并提取液、静置、过滤、收集滤液、离心、浓缩、加入渗透剂等步骤,制得中药制剂。本发明能有效杀死小瓜虫成虫、包囊和幼虫,防治鱼类多子小瓜虫,使用方便,成本低,无毒副作用,无药物残留,绿色环保。

【技术领域】
本发明属于中药制备技术领域,具体涉及一种用于防治鱼类小瓜虫的中药组合物及中药制剂。
【背景技术】
中国是渔业大国,水产品总量持续领先,水产养殖量位居世界首位。鱼类是水产养殖中的大类,鱼类是水生变温动物,较之陆生恒温的家畜、家禽能量消耗较少,饲料转化效率高,产品中动物蛋白质含量也高,同时鱼肉中也含有丰富的维生素D、钙、磷等营养物质,对人身体健康有积极作用。寄生虫病是水产养殖中最常见的疾病,具有传播快、危害广、控制难度大的特点,其主要是通过破坏水产养殖动物的组织、器官、系统的途径危害水产养殖动物,造成水产养殖动物肉质下降甚至死亡,给水产养殖户带来经济损失。
水产中的寄生虫主要小瓜虫、车轮虫、单极虫、斜管虫、指环虫、三代虫、锚头蚤等,这些寄生虫每年都会给水产养殖造成很大的损失。其中小瓜虫主要寄生在鱼类的皮肤、鳍、鳃、头、口腔和眼睛等部位,形成胞囊呈白色小点状肉眼可见,严重时鱼身浑身都是小白点,所以也称为白点病。鱼类在感染了小瓜虫后还会引发一系列的细菌和病毒感染,造成大批量死亡,死亡率高达60-70%,严重时高达80-90%,对鱼类养殖带来严重的威胁,造成极大的经济损失。目前小瓜虫的治疗方法主要是将水温升高至30℃,利用小瓜虫不耐高温的特点将其杀死;或者将水位下降,并在太阳光或者紫外线灯光下照射将其杀死;但是这两种方法在大规模养殖时需要消耗巨大的人力物力,造成不必要的经济损失,并且效果不显著;投入化学药品也能将其杀死,例如亚甲蓝等,但是亚甲蓝具有毒性,会伤害到鱼类自身,虽能够深入渗透或系统作用于寄生在鱼类表皮下的小瓜虫,杀死自由生活阶段以及在体寄生阶段的小瓜虫,但亚甲蓝是一种苯胺染料,也不被批准用于食用鱼。
中草药组合物是目前在研究防治鱼类小瓜虫的优化方法,并且中草药药性较为温和,在杀死小瓜虫的同时不会伤害到鱼类。中国专利申请文献(专利申请号:CN201010251289.6)公开了一种杀灭多子小瓜虫的中草药复方组合物及制备方法和应用,其利用木瓜、槟榔、远志、肉桂、商陆、桉树叶制成中药复方组合物用于防治多子小瓜虫,能有效灭杀小瓜虫成虫、包囊、和幼虫,但是其疗效较慢,需要7-10天持续用药,没有能在较短的时间内杀死小瓜虫,导致在中度感染的金鱼和草鱼实验中存活率较低。
【发明内容】
本发明目的是为了提供一种防治鱼类多子小瓜虫的中药组合物及其制备方法,以解决目前防治小瓜虫使用物理方法消耗巨大,造成经济浪费,治愈率低,可能存在残留等问题:本发明中药组合物,使用方便,成本低,无毒副作用,无药物残留,绿色环保,并且能有效灭杀小瓜虫成虫、包囊和幼虫,不产生耐药性,能大规模使用,具有较高经济价值。
为解决上述技术问题,本发明采用以下技术方案:
一种防治鱼类多子小瓜虫的中药组合物,以重量份为单位包括以下原料:雷公藤1-5份,苦楝子1-5份,槟榔1-5份,桉树叶3-7份,樟树叶1-5份,铁苋菜3-7份,百部1-5份,地锦草3-7份。

进一步的,还包括天南星0.5-2份。
进一步的,还包括透骨草3-7份。
一种防治鱼类多子小瓜虫的中药制剂,应用了上述中药组合物,其制备方法包括以下步骤:
S1:将上述原料按比例称取,并磨碎成粉,装入容器中,然后按料液比为1:5-10加入水提取1-3次;
S2:合并提取液,然后静置10-14h,再使用7-10层纱布过滤,收集滤液,经离心后取上清液,按浓缩比为1:1-3进行浓缩,得中药组合物;
S3:向中药组合物中加入渗透剂,搅拌均匀,即得中药制剂。
进一步的,所述渗透剂包括冰片、水溶性氮酮、乙醇。
进一步的,所述的冰片的加入量为中药组合物质量的0.1-0.3%,所述的水溶性氮酮的加入量为中药组合物质量的1-3%,所述的乙醇的加入量为中药组合物质量的3-7%;所述的乙醇的体积分数为70-80%。
进一步的,所述渗透剂的制备方法为:将冰片加入到乙醇中溶解,再与水溶性氮酮混合均匀,即得。

苦楝子,别名:土楝子,金玲子,苦心子,楝枣子;为楝科植物楝Melia azedarachL.的果实;性味与归经:苦,寒,有小毒;归肝,胃经;功能主治:行气止痛,杀虫;主腕腹胁肋疼痛,疝痛,虫积腹痛,头藓,冻疮。
槟榔,别名:仁频、宾门、宾门药饯、白槟榔、橄榄子、洗瘴丹、大腹槟榔、槟榔子;为棕榈科常绿乔木植物棕榔Areca catechu L.的成熟种子;性味与归经:味苦,辛,性温,归胃,大肠经,功能主治:驱虫,消积,下气,行水,截疟;主虫积,食滞,脘腹胀痛,泻痢后重,脚气,水肿,疟疾。
桉树叶,别名:桉树叶、蓝桉叶、羊草果叶;为桃金娘科植物蓝桉的叶。全年可采,折取老叶,阴干或鲜用;性味与归经:味辛,苦,性寒;归肺,胃,脾,肝经;功能主治:疏风解表,清热解毒,化痰理气,杀虫止痒;主感冒,高热喘咳,百日咳,脘腹胀痛,腹泻,痢疾,钩,丝虫病,疟疾,风湿痛,湿疹,疥癣,烧烫伤,外伤出血。

铁苋菜,别名:人苋、血见愁、海蚌含珠、撮斗装珍珠、叶里含珠、野麻草;为大戟科铁苋菜属植物铁苋菜Acalypha australis L.,以全草入药,夏秋采集全草,去泥土,晒干;性味与归经:苦,涩,凉;归心,肺经;功能主治:清热解毒,利湿,收敛止血。用于肠炎,痢疾,吐血、衄血、便血、尿血,崩漏;外治痈疖疮疡,皮炎湿疹。

百部,别名:百条根,百奶,野天门冬,山百根;为百部科植物直立百部Stemonasessilifolia(Miq.)Miq.、蔓生百部Stemona japonica(Bl.)Miq.或对叶百部Stemonatuberosa Lour.的干燥块根;春、秋二季采挖,除去须根,洗净,置沸水中略烫或蒸至无白心,取出,晒干;性味与归经:甘,苦,微温;归肺经;功能主治:润肺下气止咳,杀虫;用于新久咳嗽,肺痨咳嗽,百日咳;外用于头虱,体虱,蛲虫病,阴痒。蜜百部润肺止咳;用于阴虚劳嗽。
本发明具有以下有益效果:
(1)本发明采用中药组合物,可直接使用在水塘中,能有效灭杀多子小瓜虫,常温下即可达到灭杀效果,减少人力物力消耗,减少经济损失;本发明使用的中药组合物不会产生毒素,没有任何毒素残留,不污染水资源,绿色环保,并且不会使小瓜虫产生耐药性,对小瓜虫幼虫、包囊都有积极的抑制和灭杀功效,可大规模使用,达到全面灭杀的效果;
(2)本发明中药组合物中,雷公藤、苦楝子、桉树叶、樟树叶、枫柳、百部具有杀虫功效,能对小瓜虫产生极高伤害,最终达到灭杀的效果;槟榔能有效驱虫,对常见的寄生虫也能起到一定的驱杀作用,并且在没有受到小瓜虫感染时也能进行使用,在感染没有形成规模时驱赶小瓜虫,达到预防的效果;雷公藤、樟树叶、铁苋菜、透骨草、地锦草、天南星有解毒功效,能够进一步巩固杀虫效果;对杀虫后的修复起到促进作用;雷公藤、苦楝子、桉树叶、樟树叶、铁苋菜、枫柳对皮肤的修复有促进效果,能够帮助鱼类尽快恢复健康;透骨草能舒筋活血,枫柳、天南星能行气祛风,多重功效之间相辅相成,对已经感染的鱼类进行治疗,有着较好的预防和治疗作用,增加存活率,提高养殖的存活率,多种药物配合使用能显著提高药物的利用率,提高药效;
(3)本发明中药制剂中添加了渗透剂,能加快中药有效成分渗透,更好的作用于寄生在鱼类表皮下面的小瓜虫;渗透剂中的冰片是一种小分子脂溶性萜类物质,具有“佐使”功能,可以引导其他药物进入机体,它能够改善细胞膜的通透性,加速药物的转运,促进药物快速到达作用部位;而且冰片还具有抑菌、抗炎作用,能够加快鱼类的修复过程;水溶性氮酮能改变脂质双分子层的致密性,增加脂质流动性,对药物活性成分有促进渗透作用,促进药物到达机体病变处,加强中药有限成分的应用效果,能有效降低药物用量,并且加快灭杀小瓜虫,达到快速治疗的效果。

实施例1
一种防治鱼类多子小瓜虫的中药组合物,以重量份为单位包括以下原料:雷公藤1份,苦楝子5份,槟榔5份,桉树叶3份,樟树叶1份,铁苋菜7份,百部1份,地锦草3份。
一种防治鱼类多子小瓜虫的中药制剂,应用了上述中药组合物,其制备方法包括以下步骤:
S1:将上述原料按比例称取并磨碎成粉,装入容器中,然后按料液比为1:5加入水提取1次,每次提取时间3h;
S2:合并提取液,然后静置14h,再使用7层纱布过滤,收集滤液,经转速为15000r/min离心10min后取上清液,按1:1比例浓缩,得中药组合物;
S3:向中药组合物中加入渗透剂,搅拌均匀,即得中药制剂。
所述渗透剂的制备方法为:将冰片加入到乙醇中溶解,再与水溶性氮酮混合均匀,即得;其中,冰片的加入量为中药组合物质量的0.1%,水溶性氮酮的加入量中药组合物质量的3%、70%体积分数的乙醇的加入量为中药组合物质量的7%。

一种防治鱼类多子小瓜虫的中药组合物,以重量份为单位包括以下原料:雷公藤3份,苦楝子2份,槟榔2份,桉树叶4份,樟树叶2份,铁苋菜5份,百部3份,地锦草5份,枫柳1-53份,天南星1份,透骨草4份。S1:将上述原料按比例称取并磨碎成粉,装入容器中,然后按料液比为1:8加入水提取2次,每次提取时间2h;
S2:合并提取液,然后静置12h,再使用8层纱布过滤,收集滤液,经转速为12000r/min离心11min后取上清液,按1:2比例浓缩,得中药组合物;
S3:向中药组合物中加入渗透剂,搅拌均匀,即得中药制剂。
所述渗透剂的制备方法为:将冰片加入到乙醇中溶解,再与水溶性氮酮混合均匀,即得;其中,冰片的加入量为中药组合物质量的0.1%,水溶性氮酮的加入量中药组合物质量的2.5%、75%体积分数的乙醇的加入量为中药组合物质量的5%。
实验例
(1)多子小瓜虫的活体传代
将感染有多子小瓜虫的金鱼与草鱼混养与50cm×50cm×50cm的水箱中,水温控制为(22±1)℃,增氧泵24h不间断供氧。每3日换水50%,感染约7d,鱼体表面出现密集的小白点即可,可用于多子小瓜虫的收集以及幼虫的培养。
(2)虫体的收集与培养
将严重感染多子小瓜虫的草鱼放置于玻璃缸中,并于玻璃缸底部放置10个培养皿,待成熟虫体黏附于培养皿底部后,收集虫体。一部分虫体用于多子小瓜虫幼虫的培养,一部分用于包囊的收集。
(3)体外杀幼虫试验
试验在96孔板上进行,于每个孔中加入1mL 400mg/L、200mg/L、100mg/L、50mg/L、25mg/L不同浓度的由实施例9制得的药液和100个多子小瓜虫幼虫,于用药后4h镜检统计各孔的多子小瓜虫死亡率。同时设置蒸馏水(0mg/L)为对照组,每组试验重复3次。
多子小瓜虫幼虫死亡判断标准:虫体纤毛不运动,胞质不流动,胞膜破裂,细胞核破碎。
用SPSS 22.0处理数据,以平均数±标准差表示,采用方差分析进行差异显著性析。

由表1可以看出,该中药组合物的药物浓度在200mg/L以上时对多子小瓜虫幼虫具有明显的灭杀效果,效果值达到100%,并且在浓度达100mg/L时,也能具有对小瓜虫的灭杀效果,证明了本发明对灭杀鱼类多子小瓜虫的有效性,并且,本发明的中药制剂在药物浓度低时也能产生效果,是优质灭杀小瓜虫的药剂。
(4)体外杀多子小瓜虫包囊的试验
试验在24孔板上进行,于每个孔中加入1mL 400mg/L、200mg/L、100mg/L、50mg/L、25mg/L不同浓度的由实施例9制得中药组合物药液和和30个多子小瓜虫包囊,于用药后6h,吸出药液,加入充分曝气的自然水。于22℃下培养24h后先镜检观察各孔的包囊存活及虫体孵化情况,之后将有幼虫孵化的细胞孔利用移液枪混匀,并吸取10μL液体用1%甲醛固定后计数,进而计算各孔的包囊孵化虫数。每孔取5次样品,取平均值,试验重复3次。
同时设置蒸馏水(0mg/L)为对照组。
平均每个包囊孵化虫数=每孔孵化虫体数/每孔包囊个数
多子小瓜虫包囊死亡判断标准:包囊破裂、未分裂或包囊未孵出幼虫。
用SPSS 22.0处理数据,以平均数±标准差表示,采用方差分析进行差异显著性析。

由表2可以看出,该中药组合物的药物浓度在200mg/L以上对多子小瓜虫包囊具有明显的灭杀效果,证明了本发明对灭杀多子小瓜虫包囊的有效性,并且,平均每个包囊孵化虫个数在浓度分别达到100mg/L和200mg/L时出现了断崖式的下降,达到非常好的效果;在药物浓度达400mg/L时,包囊的死亡率达到了100%。
(5)对比实验
将实施例1-9制成的中药组合物应用于活鱼治愈实验中。
将中度感染,鳃上有白点,鱼体表上有10-20个白点的金鱼和草鱼分为五组混养在水族箱中,每组金鱼20尾、草鱼10尾,水温控制在22±1℃,增氧泵24h不间断供氧,每日换水1/3,每次换水前在水里加入药剂,保证水中药剂保持在浓度为200mg/L,连续用药7天,在第1、2、3、5、7天记录小白点消失数据,第20天时统计存活率。
并设置蒸馏水(0mg/L)为对照组。
实验结果如下:

由表3中数据可以看出实施例1和2在第7天时治愈效果达到100%,但是在第5天时实施例2治愈效果只有89%;实施例3和4在原料中增加了枫柳,第1天的治愈效果就提高了至少3%,并且在第五天时治愈效果已经达到95%,并且存活率也有所上升,在80%左右;增加了枫柳和天南星的实施例5和实施例6则在第1天的治愈效果达到35%并且在第5天的治愈效果稳定在95%以上,20天之后存活率也能达到87%,明显的增强了中药组合物的药效;实施例7-9中,同时增加使用了枫柳、天南星和透骨草,可以看到在第5天时的治愈效果基本达到了痊愈,并且存活率更是保持在了90%以上,实施例9更是达到了97%的存活率。
在本发明中药组合物中,雷公藤、苦楝子都有着较强的杀虫能力,天南星、枫柳和透骨草配合使用,能增强舒筋活血、解毒作用,间接增强鱼免疫能力,从而提高感染了多子小瓜虫的金鱼和草鱼存活率,并且提取出来的提取液经过浓缩,使得药物有效成分的含量增加,在使用初期就能达到比较好的效果,在中度感染的鱼类还没有恶化为重度感染时就将病情遏制住,最终达到较高的存活率。

1.一种防治鱼类多子小瓜虫的中药组合物,其特征在于,以重量份为单位包括以下原料:雷公藤1-5份,苦楝子1-5份,槟榔1-5份,桉树叶3-7份,樟树叶1-5份,铁苋菜3-7份,百部1-5份,地锦草3-7份。
2.根据权利要求1所述的防治鱼类多子小瓜虫的中药组合物,其特征在于,还包括枫柳1-5份。
3.根据权利要求2所述的防治鱼类多子小瓜虫的中药组合物,其特征在于,还包括天南星0.5-2份。
4.根据权利要求3所述的防治鱼类多子小瓜虫的中药组合物,其特征在于,还包括透骨草3-7份。
5.一种防治鱼类多子小瓜虫的中药制剂,应用了如权利要求1-4任一项所述的中药组合物,其特征在于,制备方法包括以下步骤:
S1:将上述原料按比例称取,并磨碎成粉,装入容器中,然后按料液比为1:5-10加入水提取1-3次;
S2:合并提取液,然后静置10-14h,再使用7-10层纱布过滤,收集滤液,经离心后取上清液,按浓缩比为1:1-3进行浓缩,得中药组合物;
S3:向中药组合物中加入渗透剂,搅拌均匀,即得中药制剂。
6.根据权利要求5所述的防治鱼类多子小瓜虫的中药制剂,其特征在于,所述渗透剂包括冰片、水溶性氮酮、乙醇。
7.根据权利要求6所述的防治鱼类多子小瓜虫的中药制剂,其特征在于,所述的冰片的加入量为中药组合物质量的0.1-0.3%,所述的水溶性氮酮的加入量为中药组合物质量的1-3%,所述的乙醇的加入量为中药组合物质量的3-7%;所述的乙醇的体积分数为70-80%。
8.根据权利要求7所述的防止鱼类多子小瓜虫的中药制剂,其特征在于,所述渗透剂的制备方法为:将冰片加入到乙醇中溶解,再与水溶性氮酮混合均匀,即得。
9.根据权利要求5所述的防治鱼类多子小瓜虫的中药制剂,其特征在于,所述的步骤S1中,每次提取的时间为1-3h。
10.根据权利要求5所述的防治鱼类多子小瓜虫的中药组合物,其特征在于,所述的步骤S2中,离心转速为10000-15000r/min,离心时间为10-15min。
