技术领域
本发明涉及通过低分子化合物由源自内胚层组织或器官的细胞制备干细胞/祖细胞的制备方法,特别是,通过低分子化合物由成熟胰腺外分泌细胞制备胰腺干细胞/祖细胞的制备方法,以及涉及包含该低分子化合物的将成熟胰腺外分泌细胞诱导为胰腺干细胞/祖细胞的诱导剂等。
背景技术
干细胞生物学的进步使其在再生医疗中的应用受到了重大的瞩目,然而至今还尚未实现。作为最被期待的细胞源之一的人工多能性干细胞(iPS细胞)依然存在形成肿瘤的风险性,没有应用于实际临床实践的前景(Cell.2014Oct 9;159(2):428-39)(CellMetab.2016Apr 12;23(4):622-634.)。另一方面,通过最近的研究表明,虽然将不同谱系的细胞直接重编程(Direct Reprogramming)为祖细胞样细胞是可能的,但是由于直接重编程与iPS细胞的情况一样伴随有基因修饰,因此依然存在无法预期的风险,不能应用于再生医疗。(Nat Commun.2016Jan 6;7:10080.)(Cell Stem Cell.2016Mar 3;18(3):410-21.)(Nat Biotechnol.2014Dec;32(12):1223-30.)
最近,在胰脏受到伤害的情况下,能够由成熟胰腺外分泌细胞中分离出具有增殖性和两能性的胰腺干细胞/祖细胞这一惊人的发现被报道了(EMBO J.2013Oct 16;32(20):2708-21.)。这些革新的发现,不仅为胰腺干细胞理论,而且为胰腺再生研究也提供了深刻见解。也就是说,如果能在体外再现这样的重编程(Reprogramming),则如此得到的干细胞/祖细胞就可能成为再生医疗中的新的细胞源。
然而,在不伴随基因修饰的条件下,将成熟细胞重编程为干细胞/祖细胞方法尚未被发现。
本发明者和其他的团体以前曾报道某种低分子抑制剂的组合有助于干细胞的多能性的诱导和维持方面(Proc Natl Acad Sci U S A.2010Aug 10;107(32):14223-8.)(Cell Stem Cell.2017Jan 5;20(1):41-55)(Cell Stem Cell.2016 Oct 6;19(4):449-461)。
另外本发明者,成功通过低分子化合物由成熟肝细胞制备肝干细胞/祖细胞(WO2017/119512)。
然而,并没有关于这些低分子抑制剂在将肝细胞以外的成熟细胞重编程为干细胞/祖细胞时有效的报道。

发明内容
本发明所要解决的技术问题
本发明的目的是提供一种在不伴随基因修饰的条件下,将成熟细胞高效地重编程为干细胞/祖细胞的方法。
解决技术问题的技术手段
本发明者们为达成所述的目的锐意研究的结果,发现了如果在TGFβ受体抑制剂、糖原合成酶激酶3(GSK3)抑制剂以及Rho激酶(ROCK)抑制剂的存在下培养哺乳动物的成熟细胞(例如成熟胰腺外分泌细胞),则能将胰腺外分泌细胞重编程为增殖性且能分化为胰腺内分泌细胞的细胞。显示如果将如此得到的源自成熟胰腺外分泌细胞的胰腺干细胞/祖细胞移植到糖尿病模型小鼠中,则能够作为成熟胰腺内分泌细胞正常存活。
本发明者们,基于这些发现进一步研究的结果,完成了本发明。
也就是说,本发明如下。
(1)一种由源自哺乳动物的内胚层组织或除肝脏之外的器官的细胞制作该细胞的干细胞/祖细胞的方法,其包含:
在体外使所述源自内胚层组织或器官的细胞与TGFβ受体抑制剂接触的步骤。
(2)根据项(1)所述的方法,其进一步包含:
在体外使所述源自内胚层组织或器官的细胞与GSK3抑制剂和/或ROCK抑制剂接触的步骤。

(ii)测定所述细胞中受试化合物的代谢的步骤。
(22)一种细胞分泌的酶的分泌诱导物质的筛选方法,其包含:
(i)使通过项(1)~(9)中任一项所述的方法所得到的干细胞/祖细胞、项(10)所述的胰腺干细胞/祖细胞或通过项(20)所述的方法诱导得到的细胞与受试化合物接触的步骤;和
(ii)测定所述细胞中分泌得到的物质的步骤。
(23)一种评价受试化合物对哺乳动物的内胚层组织或器官毒性的方法,其包含:

发明的效果
根据本发明,能在不伴随基因修饰的条件下,安全且迅速地将成熟细胞(例如胰腺外分泌细胞)诱导为具有自我复制能力和分化为胰腺内分泌细胞的分化能力(两能性)的胰腺干细胞/祖细胞。

本发明的具体实施方式
1.概要
本发明提供由源自哺乳动物的内胚层组织或除肝脏之外的器官的细胞制作该细胞的干细胞/祖细胞的方法,该方法包含在体外使所述源自内胚层组织或器官的细胞与TGFβ受体抑制剂接触的步骤。
如上所述,本发明者成功实现了使用低分子化合物,由成熟肝细胞制备肝干细胞/祖细胞的方法(WO2017/119512)。然而,对于肝细胞以外的内胚层器官或组织,能否由成熟细胞重编程得到干细胞或祖细胞(以下称为“干细胞/祖细胞”)尚不明确。
本发明者,使用了胰脏作为肝细胞以外的细胞,通过低分子化合物成功重编程得到了胰腺干细胞或胰腺祖细胞(以下称为“胰腺干细胞/祖细胞”)。
本说明书中,“干细胞”是指具有自我复制能力和分化为各种各样的细胞的多向分化能力的细胞,“祖细胞”是指,由干细胞产生,处于向构成身体的特定的最终分化细胞分化的中间阶段的细胞。在本说明书中,干细胞或祖细胞总称为“干细胞/祖细胞”,重编程的对象细胞是胰脏细胞的话则以“胰腺干细胞/祖细胞”表示。
本发明中,“内胚层”是后生动物的发育过程中产生的三胚层之一,发育学上而言,原肠形成的时期(囊胚期)构成原肠壁的全部或一部分。内胚层是消化管的主要部分和其附属腺(肝脏·胰脏),在甲状腺、肺等的呼吸器官发达。
本发明中,作为“内胚层组织或器官”,例如消化管(食道、胃、小肠、大肠)、肺、胰脏、甲状腺、甲状旁腺、喉头、气管、支气管、膀胱、尿道、前列腺等(但是,肝脏除外)。
但是,肝脏中存在2000种类以上的酶,不间断地进行着使各种各样的物质发生化学变化的代谢反应。这是肝脏被称为“生物体的化学工场”的原因,与例如只有制造胰岛素这种单一激素物质的功能的胰脏β细胞或属于内胚层的一系列消化器官类细胞等肝脏以外的内胚层组织具有差异巨大。肝脏是体内再生能力最高的细胞、脏器,肝脏的重编程难度和其他内胚层组织相比,肝脏以外的内胚层组织的重编程难度远高于肝脏的重编程难度。也就是说,完成了向成熟细胞的分化的源自内胚层的细胞的增殖能力、代谢能力、液性因子的分泌能力等根据各个脏器的不同具有固有的特征,在成熟肝细胞中发现的现象是成熟肝细胞特异性现象还是源自内胚层的成熟细胞中普遍的现象仍然是未知的。
本发明发现了对于肝脏以外的内胚层组织或器官,也可以由成熟细胞重编程得到干细胞/祖细胞。
本发明虽然以使源自哺乳动物的内胚层组织或除肝脏之外的器官的细胞与TGFβ受体抑制剂在体外接触为特征,但也包含使之与糖原合成酶激酶3(GSK3)抑制剂和Rho激酶(ROCK)抑制剂的任一种或两种接触的同时与TGFβ受体抑制剂接触。如此一来,就能够由源自内胚层组织或器官的成熟细胞制备该内胚层组织或器官的干细胞/祖细胞。本发明的方法称为“本发明的重编程法”。
2.由成熟细胞向干细胞/祖细胞的诱导
本发明提供由源自哺乳动物的内胚层组织或除肝脏之外的器官的细胞,制作该细胞的干细胞/祖细胞的方法,该方法包含在体外使所述源自内胚层组织或器官的细胞与TGFβ受体抑制剂接触的步骤。
本发明中,由成熟胰腺外分泌细胞制备或诱导胰腺干细胞/祖细胞包含使哺乳动物的胰腺外分泌细胞与包含TGFβ受体抑制剂、糖原合成酶激酶3(GSK3)抑制剂以及Rho激酶(ROCK)抑制剂的低分子信号传递通路抑制剂在体外接触的步骤。
作为用作本发明的重编程法的起始材料的源自内胚层组织或除肝脏之外的器官的细胞(称为“成熟细胞”),举例而言,有消化管上皮细胞、肺胞上皮细胞、胰腺实质细胞、甲状腺滤胞上皮细胞、尿路上皮细胞、前列腺上皮细胞。作为起始材料使用胰腺实质细胞时,可以使用胰腺外分泌细胞、胰腺内分泌细胞等。“胰腺外分泌细胞”被分类为腺泡细胞和胰腺管细胞,被认为其中任一种或两种可以作为胰腺干细胞/祖细胞的来源。另外,“胰腺内分泌细胞”被分类为α细胞、β细胞、δ细胞、ε细胞、PP细胞,被认为其中任一种或全部可以作为胰腺干细胞/祖细胞的来源。
本发明的重编程法使用的细胞可以为任意源所提供,举例可举出哺乳动物(例如,人、大鼠、小鼠、豚鼠、兔子、羊、马、猪、牛、猴子等,优选人、大鼠、小鼠)的胚性干细胞(ES细胞)或者iPS细胞等的多能性干细胞。
然而,考虑到本发明的主要课题:在不伴随基因修饰的情况下,安全且迅速地提供干细胞/祖细胞,则例如使用胰腺外分泌细胞时,使用从哺乳动物摘出的胰脏中分离纯化的胰腺外分泌细胞较为理想。
例如大鼠的情况下,虽然优选使用从10-20周龄的成体大鼠摘出的胰脏,也可以使用来自2个月龄以下的幼年大鼠的胰脏。人的情况下,可以通过活检或外科手术得到。例如,胰腺实质、消化管上皮、肺胞上皮、前列腺上皮等,任一种都可以通过活检采集。外科手术时,可以使用切除的成人的胰脏组织片,也可以使用从死亡胎儿切除的胰脏。或者,也可以使用将从这些摘出的胰脏中分离纯化的胰腺外分泌细胞冷冻而获得的细胞(冷冻胰腺外分泌细胞)。
为了纯化源自哺乳动物的内胚层组织或器官的细胞,分离组织后,进行酶处理之后,通过过滤、远心分离等进行纯化。
作为从哺乳动物的胰脏或其组织片纯化胰腺外分泌细胞的方法,分离胰腺组织后,用胶原酶消化胰脏,通过过滤,远心等除去朗格汉斯岛、非实质细胞、细胞片以纯化胰腺外分泌细胞。
使如上所述调制的成熟细胞(例如胰腺外分泌细胞),与包含TGFβ受体抑制剂的1种以上的低分子信号传递通路抑制剂在体外接触。
作为本发明使用的TGFβ受体抑制剂,只需具有抑制转化生长因子(TGF)β受体的机能的作用,没有特别的限制,举例而言,有2-(5-苯并[1,3]二氧杂-4-基-2-叔丁基-1H-咪唑-4-基)-6-甲基吡啶、3-(6-甲基吡啶-2-基)-4-(4-喹啉基)-1-苯基硫代氨基甲酰基-1H-吡唑(A-83-01)、2-(5-氯-2-氟苯基)蝶啶-4-基)吡啶-4-基胺(SD)-208)、3-(吡啶-2-基)-4-(4-醌基)]-1H-吡唑、2-(3-(6-甲基吡啶-2-基)-1H-吡唑-4-基))-1,5-萘啶(以上,Merck公司)、SB431542(Sigma Aldrich公司)等。优选举出A-83-01。TGFβ受体抑制剂也包含TGFβ受体拮抗剂。
这些TGFβ受体抑制剂可以只使用1种化合物,也可以将2种以上的化合物组合使用。
作为TGFβ受体抑制剂以外的低分子信号传递通路抑制剂,优选举出GSK3抑制剂和ROCK抑制剂。
作为本发明使用的GSK3抑制剂,只需具有抑制糖原合成酶激酶(GSK)3的机能,没有特别限制,举例而言,可举出SB216763(Selleck公司)、CHIR98014、CHIR99021(以上,Axonmedchem公司)、SB415286(Tocris Bioscience公司)、Kenpaullone(Cosmo·Bio公司)等。优选举出CHIR99021。
这些GSK3抑制剂可以只使用1种化合物,也可以将2种以上的化合物组合使用。
作为本发明使用的ROCK抑制剂,只需具有抑制Rho结合激酶的机能的作用,没有特别限制。作为ROCK抑制剂,举例而言,有GSK269962A(Axon medchem公司)、Fasudilhydrochloride(Tocris Bioscience公司)、Y-27632、H-1152(以上、和光纯药工业公司)等。优选举出Y-27632。
这些ROCK抑制剂可以只使用1种化合物,也可以将2种以上的化合物组合使用。
GSK3抑制剂和ROCK抑制剂,分别单独或两种组合和TGFβ受体抑制剂一起与成熟细胞(例如胰腺外分泌细胞)接触时,与只和TGFβ受体抑制剂接触时相比,干细胞/祖细胞的诱导效率(也称为“重编程效率”)显著上升。因此,在本发明的重编程法中,优选除TGFβ受体抑制剂外,进一步使GSK3抑制剂和/或ROCK抑制剂与成熟细胞(例如胰腺外分泌细胞)接触。
本发明中,TGFβ受体抑制剂和GSK3抑制剂和/或ROCK抑制剂的组合如下所示。
(a)作为TGFβ受体抑制剂的A-83-01(A)和作为GSK3抑制剂的CHIR99021(C)的组合(AC)。
(b)作为TGFβ受体抑制剂的A-83-01(A)和作为ROCK抑制剂的Y-27632(Y)的组合(YA)。
(c)作为TGFβ受体抑制剂的A-83-01(A)、作为GSK3抑制剂的CHIR99021(C)和作为ROCK抑制剂的Y-27632(Y)的组合(YAC)。
TGFβ受体抑制剂与GSK3抑制剂和ROCK抑制剂组合使用时,与TGFβ受体抑制剂和GSK3抑制剂组合使用时相比,未观察到重编程效果有显著的差异,前者与后者相比,得到的干细胞/祖细胞的增殖能力更优异。因此,在本发明的实施方式中,更优选使TGFβ受体抑制剂、GSK3抑制剂和ROCK抑制剂的组合与成熟细胞(例如胰腺外分泌细胞)接触。
在本发明的重编程法中,可以将GSK3抑制剂、ROCK抑制剂以外的低分子信号传递通路抑制剂与TGFβ受体抑制剂组合。作为这样的抑制剂,例如有MEK抑制剂等,没有限制。作为MEK抑制剂,只需具有抑制MEK(MAP激酶-ERK激酶)的机能的作用,没有特别限制,举例而言,有AZD6244、CI-1040(PD184352)、PD0325901、RDEA119(BAY86-9766)、SL327、U0126-EtOH(以上、Selleck公司)、PD98059、U0124、U0125(以上,Cosmo·Bio公司)等。
在本发明的重编程法中,成熟细胞(例如胰腺外分泌细胞)和包含TGFβ受体抑制剂的低分子信号传递通路抑制剂的接触可以通过在这些抑制剂的存在下培养成熟细胞进行。具体而言,向培养基中添加有效浓度的这些抑制剂进行培养。作为这里的培养基,可以将在动物细胞的培养中广泛使用的培养基作为基础培养基使用。也可以使用市售的基础培养基,作为这些培养基,举例而言,有最少必需培养基(MEM)、dulbecco改良最少必需培养基(DMEM)、RPMI1640培养基、199培养基、Ham’s F12培养基、William's E培养基等,这些培养基可以单独或2种以上组合使用,没有特别限制。
作为加入培养基的添加剂,举例而言,有各种氨基酸(例如,L-谷氨酰胺、L-脯氨酸等)、各种无机盐(亚硒酸盐、NaHCO3等)、各种维生素(烟酰胺、抗坏血酸衍生物等)、各种抗生物质(例如,青霉素、链霉素等)、抗真菌剂(例如两性霉素等)、缓冲剂(HEPES等)等。
另外,培养基中可以添加5-20%的血清(FBS等),也可以是无血清培养基。无血清培养基的情况下,可以添加血清代替物(BSA、HAS、KSR等)。进一步,通常添加生长因子、细胞因子、激素等因子。作为这些因子,例如有上皮生长因子(EGF)、胰岛素、转铁蛋白、氢化可的松21-半琥珀酸或其盐、地塞米松(Dex)等,但不限于此。
TGFβ受体抑制剂的向培养基中的添加浓度,举例而言,为0.01-10μM,优选从0.1-1μM的范围中适宜选择。
GSK3抑制剂的向培养基中的添加浓度,举例而言,为0.01-100μM,优选从1-10μM的范围中适宜选择。
ROCK抑制剂的向培养基中的添加浓度,举例而言,为0.0001-500μM,优选从1-50μM的范围适宜选择。
这些抑制剂为水不溶性或水难溶性的化合物时,可以溶解在少量的低毒性的有机溶媒(例如,DMSO等)中后,添加到培养基中达到所述的最终浓度。
培养中使用的培养容器,只需适用于贴壁培养,没有特别限制,举例而言,有皿、培养皿、组织培养用培养皿、多孔培养皿、微孔板、微孔培养板、多孔板、多孔培养板、腔室载玻片、细胞用浅底盘、离心管、托盘、培养包等。培养容器可以使用为了进行细胞的悬浮培养而表面加工成细胞无法贴壁的培养容器。或者,在贴壁培养时,可以使用为提高内表面和细胞的贴壁性而用细胞支持用基质包被的容器。作为这样的细胞支持用基质,举例而言,有胶原、明胶、基质胶、聚-L-赖氨酸、层粘连蛋白、纤连蛋白等。优选胶原或基质胶。
成熟细胞可以以102~106细胞/cm2,优选以103~105细胞/cm2的细胞密度接种到培养容器上。
胰腺外分泌细胞的情况下,可以以102~106细胞/cm2,优选以103~105细胞/cm2的细胞密度接种到培养容器上。培养可以在CO2培养箱中,在1-10%,优选2-5%,更优选约5%的CO2浓度的氛围中,在30-40℃,优选35-37.5℃,更优选约37℃的温度下进行。作为培养时间,举例而言为1-4周,优选1-3周。每1-3天更换新鲜培养基。
如上所述,通过使成熟细胞(胰腺外分泌细胞)与TGFβ受体抑制剂,以及任意地与GSK3抑制剂和/或ROCK抑制剂接触,可以重编程为成熟细胞干细胞/祖细胞。例如,使用作为TGFβ受体抑制剂的A-83-01(A),与作为GSK3抑制剂的CHIR99021(C)和作为ROCK抑制剂的Y-27632(Y)组合(YAC),以培养小鼠初代成熟胰腺外分泌细胞时,通过30的培养约增殖3000倍,与在不存在YAC条件下培养时相比显著增加。
本说明书中“胰腺干细胞/祖细胞”(也称为“PSC”)是指,具有(a)自我再生能力,且具有(b)能分化为胰腺内分泌细胞(例如构成朗格汉斯岛的细胞)或胰腺外分泌细胞的性质的细胞,是指干细胞或祖细胞。
胎儿胰脏的胰腺胚细胞也包含在胰腺干细胞/祖细胞(PSC)中。
在优选的一实施方式中,通过本发明的重编程方法得到的PSC,在所述(a)和(b)性质之外,还具有(c)表达作为关键因子的Pdx1和Nkx6.1之外,还表达Gata4、Hes1、Sox9、Foxa2、CK19、CD133等。但是,通过本发明的重编程方法得到的PSC,不表达其他已知的PSC表达的Lgr5。因此,本发明的PSC是新的PSC。
本发明的PSC,进一步具有以下1个以上的特征。
(d)至少10传代,优选20传代以上,表观上的增殖速度不降低。
(e)至少10传代,优选20传代以上,保持向胰腺内分泌细胞分化的分化能力。
(f)与胰腺外分泌细胞相比核/细胞质(N/C)比较高。
(g)选自Pdx1、Nkx6.1中的1种以上的PSC标记物基因的表达和胰腺外分泌细胞相比上升。
(h)选自Pdx1、Nkx6.1中的一种以上的蛋白质的表达与胰腺外分泌细胞相比上升。
优选实施方式中,本发明的PSC具有所述(d)~(h)所有的特征。
如上所述,通过使成熟细胞(例如胰腺外分泌细胞)与TGFβ受体抑制剂、GSK3抑制剂和/或ROCK抑制剂接触,能够向干细胞/祖细胞(例如由胰腺外分泌细胞向PSC诱导)诱导。
因此,本发明提供由源自哺乳动物的内胚层组织或除肝脏之外的器官的细胞,诱导该细胞的干细胞/祖细胞的方法,该方法包含在体外使所述源自内胚层组织或器官的细胞和TGFβ受体抑制剂、或TGFβ受体抑制剂和GSK3抑制剂和/或ROCK抑制剂的组合接触的步骤。
特别是提供由胰腺外分泌细胞诱导PSC时,通过使胰腺外分泌细胞与TGFβ受体抑制剂、GSK3抑制剂和/或ROCK抑制剂接触,而由胰腺外分泌细胞诱导PSC的方法。本发明还提供包含TGFβ受体抑制剂的、由胰腺外分泌细胞诱导的PSC的诱导剂。本发明的PSC诱导剂优选包含TGFβ受体抑制剂和GSK3抑制剂和/或ROCK抑制剂的组合,更优选包含TGFβ受体抑制剂和GSK3抑制剂和ROCK抑制剂的组合。
TGFβ受体抑制剂、GSK3抑制剂和ROCK抑制剂可以直接作为PSC诱导剂使用,也可以溶解于适当的溶媒中作为液剂使用。或者,这些抑制剂也可以和所述的用于由胰腺外分泌细胞诱导PSC的培养基组合形成试剂盒。
3.干细胞/祖细胞维持·扩增
如所述方法得到的本发明的干细胞/祖细胞,通过在用胶原或基质胶进行了包被的培养容器上,分别在TGFβ受体抑制剂、GSK3抑制剂和ROCK抑制剂的存在下传代培养,可以高效地维持·扩增。
作为培养容器,可以使用和用于由成熟细胞诱导干细胞/祖细胞的培养容器同样的培养容器。
使用了胰腺外分泌细胞时,通过本发明的方法所得到的PSC,通过在用胶原或基质胶进行了包被的培养容器上,分别在TGFβ受体抑制剂、GSK3抑制剂和ROCK抑制剂的存在下传代培养,可以高效地维持·扩增。
作为培养容器,可以使用和用于由胰腺外分泌细胞诱导PSC的培养容器同样的培养容器。
将如所述的方法得到的初代PSC,在达到70-100%汇合的阶段,以103~105细胞/cm2的密度接种到所述胶原或基质胶进行了包被的培养容器上。作为培养基,可以同样地使用PSC的诱导培养中所述的培养基。TGFβ受体抑制剂、GSK3抑制剂、ROCK抑制剂的添加浓度,也可以从PSC的诱导培养中所述的浓度范围适宜选择。培养温度、CO2浓度也参照PSC诱导培养的条件。在达到70-100%汇合阶段,将细胞用胰蛋白酶处理并解离,进行传代。能够在约5-8传代安定地获得PSC。10传代以上传代后,可以通过通常方法进行克隆化。
如上所述,TGFβ受体抑制剂、GSK3抑制剂、ROCK抑制剂,不仅在干细胞/祖细胞(例如PSC)的诱导培养中,而且在维持·扩增培养中也添加到培养基中。因此,本发明还提供含有TGFβ受体抑制剂、GSK3抑制剂、ROCK抑制剂的干细胞/祖细胞(例如PSC)的维持·扩增剂。
4.由干细胞/祖细胞向成熟细胞的分化
由干细胞/祖细胞向成熟细胞的分化诱导,例如可以通过使用YAC添加SHM(Smallhepatocyte medium)培养基在超低吸附培养皿上悬浮培养7天来进行。或者,也可以使用用添加了佛波醇12,13-二丁酸酯、LDN193189、角质细胞生长因子、SANT1、维A酸、XXI、β细胞素等的培养液进行培养的方法(Cell.2014Oct 9;159(2):428-39.)等。
对于由PSC向胰腺内分泌细胞的分化诱导,也可以适用和所述同样的方法。或者,也可以使用用添加了佛波醇12,13-二丁酸酯、LDN193189、角质细胞生长因子、SANT1、维A酸、XXI、β细胞素等的培养液进行培养的方法(Cell.2014Oct 9;159(2):428-39.)等。
将本发明的PSC分化诱导从而得到的胰腺外分泌细胞,具有成熟胰腺内分泌细胞典型的胰岛素产生功能,也检测出反映胰岛素分泌的C肽分泌。另外,作为胰腺内分泌细胞的关键转录因子的Ngn3、葡萄糖转运蛋白(GLUT2)的mRNA上升也被观察到了。也就是说,本发明的PSC能够分化为机能性的胰腺内分泌细胞。
5.本发明的干细胞/祖细胞的用途
如上述4.所述的,由本发明的干细胞/祖细胞再分化的成熟细胞,举例而言,可以用于受试化合物的代谢、细胞或组织毒性的评价等。
受试化合物的代谢、毒性的评价,一直以来使用动物模型等,能够一次评价的受试化合物的数量存在制限,另外存在动物模型等得到的评价无法直接适用于人这样的问题。因此,使用人癌细胞株、初代正常人培养细胞的评价方法被采用。然而,就人癌细胞株而言,用人癌细胞株得到的评价有无法适用于人正常细胞的可能性。另外,初代正常人培养细胞则在稳定供给、成本的方面存在问题。进一步而言,将初代正常人培养细胞永生化的细胞株,与未永生化时相比,其活性被观察到降低。通过利用以本发明的方法制造的细胞,可以解决这些问题。
因此,本发明提供评价受试化合物在哺乳动物体内的代谢的方法,该方法包含使通过本发明的方法所得到的干细胞/祖细胞、胰腺干细胞/祖细胞或通过本发明的诱导方法所得到的细胞与受试化合物接触的步骤;和测定所述细胞中受试化合物的代谢的步骤。
因此,在本发明的方法中,使用胰腺外分泌细胞时,使通过本发明的方法制造的胰腺外分泌细胞与受试化合物接触。接下来,测定与胰腺外分泌细胞接触的受试化合物的代谢。
作为本发明中使用的受试化合物,没有特别的限制。举例而言,可举出:生物体异物、天然化合物、有机化合物、无机化合物、蛋白质、肽等的单一化合物,以及化合物库、基因库的表达产物、细胞提取物、细胞培养上清液、发酵微生物产物、海洋生物提取物、植物提取物等,且不限于此。
作为生物体异物,例如有药剂或食品的候补化合物,现有的药剂或食品,但不限于此,只要对生物体而言是异物,就包含在本发明的生物体异物中。更具体而言,可以举出利福平(Rifampin)、地塞米松(Dexamethasone)、苯巴比妥(Phenobarbital)、环格列酮(Ciglirazone)、苯妥英(Phenytoin)、依法韦仑(Efavirenz)、斯伐他汀(Simvastatin)、β-萘黄酮(β-Naphthoflavone)、奥美拉唑(Omeprazole)、克霉唑(Clotrimazole)、3-甲基胆蒽(3-Methylcholanthrene)等。
成熟细胞(例如胰腺外分泌细胞)和受试化合物的接触,通常,通过将受试化合物添加到培养基或培养液中进行,但不限定于此方法。受试化合物为蛋白质等时,可以通过将表达该蛋白质的DNA载体导入该细胞来进行接触。
受试化合物的代谢可以用本领域技术人员周知的方法进行测定。例如检测出受试化合物的代谢产物时,判定受试化合物被代谢。
例如,通过受试化合物的接触,胰岛素等的酶基因的表达被诱导时或这些酶的活性上升时,判定受试化合物被代谢。
对于胰脏以外的组织或器官,也可以以存在于该组织或器官的基因的表达作为指标,判定受试化合物被代谢。
另外本发明提供细胞分泌的酶的分泌诱导物质的筛选方法,该方法包含使通过本发明的方法所得到的干细胞/祖细胞、本发明的胰腺干细胞/祖细胞或通过本发明的诱导方法诱导得到的细胞与受试化合物接触的步骤;和测定所述细胞中分泌得到的物质的步骤。
例如,可以以使被检物质与胰腺干细胞/祖细胞或诱导得到的胰腺细胞接触后,细胞分泌的各种酶是否收到分泌诱导作为指标,筛选分泌调整物质。
另外本发明提供受试化合物对哺乳动物的内胚层组织或器官毒性的评价方法。在此方法中,包含使通过本发明的方法所得到的干细胞/祖细胞、胰腺干细胞/祖细胞或通过本发明的诱导方法诱导得到的细胞与受试化合物接触的步骤;和测定在与受试化合物接触的细胞中是否产生损伤和其程度的步骤。
本发明中,评价受试化合物的胰腺毒性时,使通过本发明的方法制造的胰腺外分泌细胞与受试化合物接触。接下来,测定与受试化合物接触了的胰腺外分泌细胞的损伤的程度。损伤的程度,可以以例如胰腺外分泌细胞的生存率、胰腺损伤标记物为指标测定。
例如,通过向胰腺外分泌细胞的培养液中添加受试化合物,胰腺外分泌细胞的生存率降低时,判定该受试化合物具有胰腺毒性,生存率没有显著变化时,则判定该受试化合物没有胰腺毒性。
需要说明的是,通过使用已判明有无胰腺毒性的化合物作为对照,能够更正确地评价受试化合物是否具有胰腺毒性。
进一步而言,本发明提供包含通过本发明的方法所得到的干细胞/祖细胞、胰腺干细胞/祖细胞或通过本发明的诱导方法诱导得到的细胞的内胚层组织或器官损伤改善剂。进一步而言,本发明提供包含以通过本发明的方法所得到的干细胞/祖细胞、胰腺干细胞/祖细胞或通过本发明的诱导方法诱导得到的细胞的有效量,向具有内胚层组织或器官损伤的哺乳动物施用的步骤的方法。
例如改善胰腺损伤,通过将本发明的PSC移植到具有慢性胰腺损伤的免疫缺陷小鼠,能够发挥与移植初代成熟胰腺外分泌细胞时同等的胰腺再生能力。因此,本发明还提供含有本发明的PSC的胰腺损伤改善剂。
本发明的PSC可以应需要使用表面抗原标记物的抗体,通过流式细胞仪纯化后使用。PSC可以在适当的等渗缓冲液(例如,PBS)中悬浮以制剂化。应需要,可以进一步含有医药上容许的添加物。PSC悬浮液根据胰腺疾病的种类、胰腺损伤的严重程度等而不同,例如成人的情况下,可以移植108-1011细胞。
接下来将以以下的实施例对本发明进行更加具体的说明,但本发明不限于这些实施例。

胰腺祖细胞的构建
小鼠或大鼠的胰腺外分泌细胞按照既知的手法进行了胶原酶处理以及分离(Reichert et al.Cold Spring Harb Protoc.2015Jun 1;2015(6):558-61)。通过双硫腙(DTZ)染色确认了分离的细胞中不含有胰岛素分泌细胞。将分离而得的胰腺外分泌细胞,用添加了3种低分子化合物(Y-27632[10μM],A-83-01[0.5μM],CHIR99021[3μM];以下YAC)的小型肝细胞培养培养基(Small hep atocyte medium,SHM),在I型胶原包被培养皿上平板培养。
SHM培养基:向DMEM/F12 472.7mL中加入了以下的添加物的培养基(Katsuda etal.Bio Protoc.2018Jan 20;8(2))
2M HEPES,1.25mL
30g/L L-脯氨酸,500μL
100x antibiotic/antimycotic,5ml
5N NaOH,250μL(为将调整为pH7.5)
5%BSA,5mL
10μg/mL EGF,500μL
100x ITS-X,5mL
10-4M地塞米松,500μL
1M烟酰胺,5mL
100mM Asc2P,5mL
PSC的传代培养
在初代培养的第14天,将在YAC存在下培养的细胞用胰蛋白酶处理并回收,以9×103细胞/cm2接种到了添加了YAC的SHM中。小鼠的情况下,在I型胶原包被培养皿上,大鼠的情况下,在基质胶进行了包被的培养皿上培养细胞。使用CELLBANKER(登录商标)1(宝酒造,大津)调制冷冻原种(stock)。至少10传代后,使用BD FACSAria II细胞分选仪实施了PSC的克隆。
在低细胞密度下的低速度拍摄
将初代胰腺外分泌细胞向胶原包被的35mm板(IWAKI)上,在YAC的存在下或不存在的条件下以1×102细胞/cm2接种。在第一天更换培养基。第2次的培养基更换后,使用BZ9000All-in-One荧光显微镜(Keyence,大阪)进行了低速度拍摄。相位差图像是从第2天到第6天以30分钟的间隔拍摄300次,对于各个解析范围分别制作动画。接下来,在拍摄期间对各个细胞进行追踪,计量了源自目标细胞的最终的细胞数。另外,对源自各个细胞的凋亡细胞的总数也进行了计数,凋亡频率以总凋亡细胞/最初的总细胞数(低速度拍摄开始时的计数)进行了定量。
定量RT-PCR
从胰腺外分泌细胞以及PSC细胞中,使用miRNeasy Mini Kit(QIAGEN)分离了总RNA。逆转录反应使用High-Capacity cDNA Reverse Transcription Kit(Lifetechnologies),依照制造者的说明书实施。以得到的cDNA作为模板,使用PlatinumSYBR Green qPCR SuperMix UDG(Invitrogen)进行了PCR。靶基因的表达水平用作为内在性对照的β-肌动蛋白进行了标准化。
免疫细胞化学、免疫组织化学
细胞用4%多聚甲醛固定15分钟。用封闭液(Blocking One)(nacalai tesque,京都),在4℃孵育30分钟后,使细胞和一次抗体在室温下孵育1小时或在4℃下孵育过夜。接下来,使用Alexa Fluor 488或Alexa Fluor594-结合二次抗体(Lifetechnologies),检测了一次抗体。核用Hoechst 33342(Dojindo)进行了共染色。
组织样本用福尔马林固定,进行了石蜡包埋。脱蜡和再水合之后,通过将标本在1/200稀释的ImmunoSaver(日新EM,东京)中,98℃下煮沸45分钟,来进行了热诱导性表位恢复。接下来,将标本用0.1%Triton-X 100处理并进行了膜通透化。用封闭试剂(nacalaitesque)在4℃下,处理30分钟后,使标本在室温下和一次抗体孵育1小时。对这些切片,通过按照制造者的指示使用ImmPRESS IgG-peroxidase kit(Vector Labs)和metal-enhancedDAB substrate kit(Life Technologies)进行了染色。用苏木精进行对比染色后,对标本进行了脱水封入。
PSC的胰腺内分泌细胞诱导
将细胞悬浮在添加了5%FBS的SHM+YAC中(细胞浓度:1×104~1×106细胞/cm2),在超低吸附培养皿上培养7天。培养基更换在第4天进行。
2.结果
1)胰腺祖细胞的构建
通过向含有胰腺外分泌细胞的培养基中添加YAC,如图1所示,构建了具有高增殖能力的小型上皮细胞。该细胞可以传代20次以上,也可以单细胞培养。另外从可以向具有胰岛素合成能力的内分泌类细胞分化来看,可以认为具有作为胰腺祖细胞的特征。
2)诱导的胰腺祖细胞的特性
构建的细胞,如图2、3所示,表达包含作为胰腺祖细胞中的关键转录因子的Pdx1和Nkx6.1的干细胞标记物。
3)由胰腺祖细胞向胰岛素产生细胞的分化诱导
构建的胰腺祖细胞,通过用YAC添加SHM培养基在超低吸附培养皿上悬浮培养7天,如图4所示细胞一边积聚成簇状一边增殖,该簇为DTZ染色阳性,另外使用免疫染色检测出了胰岛素的染色性。从中可以看出,胰腺祖细胞向胰岛素产生细胞进行了分化。实际上,也观察到了除了胰岛素之外,作为胰腺内分泌细胞的关键转录因子的Ngn3、葡萄糖转运蛋白(GLUT2)的mRNA的上升(图5)。另外反映胰岛素分泌的C肽分泌也被检出(图5)。
另外,通过向YAC中加入ALK5抑制剂II(Enzo,ALX-270-445-M001)悬浮培养7天,能够进一步提高胰岛素、GLUT2的mRNA表达(图6)。
此外,向胰腺内分泌细胞的分化诱导后,在培养基的葡萄糖浓度的低浓度(3mM)和高浓度(20mM)的条件下进行比较,发现高浓度葡萄糖下的胰岛素mRNA、培养基中C肽的上升,显示了其获得了葡萄糖浓度应答性(图7)。
4)用动物的实施例
使用链脲佐菌素给药糖尿病模型小鼠,将进行了向胰岛素产生细胞的分化诱导处理的胰腺祖细胞封入Matrigel并移植到了胰腺被膜下。如图8所示,移植3天后血糖值得到了改善。此外,在移植部位中移植细胞簇的大部分分化为腺管状的细胞,而一部分的细胞为胰岛素、Pdx1、Nkx6.1阳性,观察到了向β细胞分化的样态(图9)。
工业实用性
根据本发明,能够在不伴随基因修饰的情况下,安全且迅速地由胰腺外分泌细胞诱导为具有自我再生能力和向胰腺外分泌细胞的分化能力(两能性)的胰腺干细胞/祖细胞,因此从可应用于药物评价系统、胰腺再生医疗的这一点而言,本发明的方法极具有用性。
Claims (26)
1.一种由源自哺乳动物的内胚层组织或除肝脏之外的器官的细胞制作该细胞的干细胞/祖细胞的方法,其包含:
在体外使所述源自内胚层组织或器官的细胞与TGFβ受体抑制剂接触的步骤。
2.权利要求1所述的方法,其进一步包含:
在体外使所述源自内胚层组织或器官的细胞与GSK3抑制剂和/或ROC K抑制剂接触的步骤。
3.权利要求1所述的方法,其进一步包含:
在体外使所述源自内胚层组织或器官的细胞与GSK3抑制剂和ROCK抑制剂接触的步骤。
4.权利要求1~3中任一项所述的方法,其中,
内胚层组织或器官为消化道、肺、甲状腺、胰脏、分泌腺、腹膜、胸膜、喉头、咽鼓管、气管、支气管、膀胱、尿道或输尿道。
5.权利要求1~4中任一项所述的方法,其中,
内胚层组织或器官为胰脏。
6.权利要求5所述的方法,其中,
源自胰脏的细胞为胰腺外分泌细胞。
7.权利要求6所述的方法,其中,
所述胰腺外分泌细胞和所述TGFβ受体抑制剂的接触是通过在该抑制剂的存在下培养该胰腺外分泌细胞而实施的。
8.权利要求6或7所述的方法,其中,
所述胰腺外分泌细胞和所述GSK3抑制剂和/或所述ROCK抑制剂的接触是通过在该抑制剂的存在下培养该胰腺外分泌细胞而实施的。
9.权利要求1~8中任一项所述的方法,其中,
哺乳动物为人、大鼠或小鼠。
10.一种源自哺乳动物的胰腺外分泌细胞的胰腺干细胞/祖细胞,其具有以下特征:
(a)具有自我再生能力
(b)能分化为胰腺内分泌细胞
(c)表达Pdx1和Nkx6.1,但不表达胰岛素。
11.一种将源自哺乳动物的内胚层组织或除肝脏之外的器官的细胞向该细胞的干细胞/祖细胞进行诱导的诱导剂,其包含:
TGFβ受体抑制剂。
12.权利要求11所述的诱导剂,其进一步包含:
GSK3抑制剂和/或ROCK抑制剂的组合。
13.权利要求11所述的诱导剂,其进一步包含:
GSK3抑制剂和ROCK抑制剂的组合。
14.权利要求11~13中任一项所述的诱导剂,其中,
内胚层组织或器官为消化道、肺、甲状腺、胰脏、分泌腺、腹膜、胸膜、喉头、咽鼓管、气管、支气管、膀胱、尿道或输尿道。
15.权利要求11~13中任一项所述的诱导剂,其中,
内胚层组织或器官为胰脏。
16.权利要求15所述的诱导剂,其中,
源自胰脏的细胞为胰腺外分泌细胞。
17.权利要求11~16中任一项所述的诱导剂,其中,
哺乳动物为人、大鼠或小鼠。
18.权利要求11~17中任一项所述的诱导剂,其作为通过权利要求1~9中任一项所述的方法所得到的干细胞/祖细胞或权利要求10所述的胰腺干细胞/祖细胞的维持剂或扩增剂而使用。
19.一种维持或扩增胰腺干细胞/祖细胞的方法,其是维持或扩增通过权利要求1~9中任一项所述的方法所得到的干细胞/祖细胞或权利要求10所述的胰腺干细胞/祖细胞的方法,其中,
在用胶原或基质胶进行了包被的培养容器上,在TGFβ受体抑制剂、GSK3抑制剂和ROCK抑制剂的存在下,传代培养该干细胞/祖细胞或胰腺干细胞/祖细胞。
20.一种方法,其是将通过权利要求1~9中任一项所述的方法所得到的干细胞/祖细胞或权利要求10所述的胰腺干细胞/祖细胞向源自哺乳动物的内胚层组织或器官的细胞或胰腺外分泌细胞进行诱导的方法,其中,所述方法包含:
在TGFβ受体抑制剂、GSK3抑制剂和ROCK抑制剂的存在下培养所述干细胞/祖细胞或胰腺干细胞/祖细胞的步骤。
21.一种评价受试化合物在哺乳动物体内的代谢的方法,其包含:
(i)使通过权利要求1~9中任一项所述的方法所得到的干细胞/祖细胞、权利要求10所述的胰腺干细胞/祖细胞或通过权利要求20所述的方法所得到的细胞与受试化合物接触的步骤;和
(ii)测定所述细胞中受试化合物的代谢的步骤。
22.一种细胞分泌的酶的分泌诱导物质的筛选方法,其包含:
(i)使通过权利要求1~9中任一项所述的方法所得到的干细胞/祖细胞、权利要求10所述的胰腺干细胞/祖细胞或通过权利要求20所述的方法诱导得到的细胞与受试化合物接触的步骤;和
(ii)测定所述细胞中分泌得到的物质的步骤。
23.一种评价受试化合物对于哺乳动物的内胚层组织或器官毒性的方法,其包含:
(i)使通过权利要求1~9中任一项所述的方法所得到的干细胞/祖细胞、权利要求10所述的胰腺干细胞/祖细胞或通过权利要求20所述的方法所得到的细胞与受试化合物接触的步骤;和
(ii)测定在与受试化合物接触的细胞中是否产生损伤和其程度的步骤。
24.一种内胚层组织或器官损伤改善剂,其包含:
通过权利要求1~9中任一项所述的方法所得到的干细胞/祖细胞、权利要求10所述的胰腺干细胞/祖细胞或通过权利要求20所述的方法所得到的细胞。
25.一种哺乳动物的内胚层组织或器官损伤的改善方法,其包含:
对于具有该组织或器官损伤的哺乳动物,以有效量施用通过权利要求1~9中任一项所述的方法所得到的干细胞/祖细胞、权利要求10所述的胰腺干细胞/祖细胞或通过权利要求20所述的方法诱导得到的细胞。
26.一种细胞,其是通过权利要求1~9中任一项所述的方法所得到的干细胞/祖细胞、权利要求10所述的胰腺干细胞/祖细胞或通过权利要求20所述的方法所得到的细胞,
所述细胞为用作内胚层组织或器官损伤改善剂的细胞。
