Abstract
Description
しかしながら、遺伝子改変を伴わずに、成熟細胞を幹/前駆細胞にリプログラムする方法は全く知られていない。
また本発明者は、低分子化合物による成熟肝細胞から肝幹/前駆細胞を作製することに成功した(WO2017/119512)
しかしながら、これらの低分子阻害薬の、肝細胞以外についての成熟細胞から幹/前駆細胞へのリプログラミングに対する寄与に関する報告は皆無である。
本発明者らは、これらの知見に基づいてさらに研究を重ねた結果、本発明を完成させるに至った。
(1)哺乳動物の内胚葉組織又は器官(肝臓を除く)由来の細胞から、当該細胞の幹/前駆細胞を作製する方法であって、インビトロで前記内胚葉組織又は器官由来の細胞にTGFβ受容体阻害薬を接触させることを含む、前記方法。
(2)さらに、GSK3阻害薬及び/又はROCK阻害薬を、インビトロで前記内胚葉組織又は器官由来の細胞に接触させることを含む、(1)に記載の方法。
(3)さらに、GSK3阻害薬及びROCK阻害薬を、インビトロで前記内胚葉組織又は器官由来の細胞に接触させることを含む、(1)に記載の方法。
(4)内胚葉組織又は器官が、消化管、肺、甲状腺、膵臓、分泌腺、腹膜、胸膜、喉頭、耳管、気管、気管支、膀胱、尿道又は尿管である、(1)〜(3)のいずれか1項に記載の方法。
(5)内胚葉組織又は器官が膵臓である(1)〜(4)のいずれか1項に記載の方法。
(6)膵臓由来の細胞が膵外分泌細胞である(5)に記載の方法。

(22)細胞が分泌する酵素の分泌誘導物質のスクリーニング方法であって、
(i)(1)〜(9)のいずれか1項に記載の方法により得られた幹/前駆細胞、(10)に記載の膵幹/前駆細胞、又は(20)に記載の方法により誘導された細胞に被検化合物を接触させる工程、及び
(ii)前記細胞において分泌された物質を測定する工程、
を含む、前記方法。
(23)哺乳動物に対する被検化合物の内胚葉組織又は器官毒性を評価する方法であって、
(i)(1)〜(9)のいずれか1項に記載の方法により得られた幹/前駆細胞、(10)に記載の膵幹/前駆細胞、又は(20)に記載の方法により得られた細胞に被検化合物を接触させる工程、及び
(ii)被検化合物と接触した細胞における障害の有無又はその程度を測定する工程
を含む、前記方法。
(24)(1)〜(9)のいずれか1項に記載の方法により得られた幹/前駆細胞、(10)に記載の膵幹/前駆細胞、又は(20)に記載の方法により得られた細胞を含む、内胚葉組織又は器官障害改善剤。

本発明は、哺乳動物の内胚葉組織又は器官(肝臓を除く)由来の細胞から、当該細胞の幹/前駆細胞を作製する方法であって、インビトロで前記内胚葉組織又は器官由来の細胞にTGFβ受容体阻害薬を接触させることを含む、前記方法を提供する。
前記の通り、低分子化合物を用いて、成熟肝細胞から肝幹/前駆細胞を作製する方法は、本発明者により成功している(WO2017/119512)。しかしながら、肝細胞以外の内胚葉器官又は組織については、成熟細胞から幹細胞又は前駆細胞(以下「幹/前駆細胞」という)にリプログラミングできるか否かは不明である。
本明細書において、「幹細胞」とは、自己複製能と、様々な細胞に分化する多分化能とを持つ細胞を意味し、「前駆細胞」とは、幹細胞から発生し、体を構成する特定の最終分化細胞へ分化する中間段階にある細胞を意味する。本明細書では、幹細胞又は前駆細胞を総称して、「幹/前駆細胞」と呼び、リプログラミングの対象となった細胞が膵臓細胞であれば「膵幹/前駆細胞」と表示する。
本発明において、「内胚葉組織又は器官」としては、例えば消化管(食道、胃、小腸、大腸)、肺、膵臓、甲状腺、副甲状腺、喉頭、気管、気管支、膀胱、尿道、前立腺などである(但し、肝臓は除く)。
本発明は、哺乳動物の内胚葉組織又は器官(肝臓を除く)由来の細胞に、TGFβ受容体阻害薬を、インビトロで接触させることを特徴とするが、グリコーゲン合成酵素キナーゼ3(GSK3)阻害薬及びRhoキナーゼ(ROCK)阻害薬のいずれか一方又は両者を、TGFβ受容体阻害薬とともに接触させることも含む。これにより、内胚葉組織又は器官由来の成熟細胞から、当該内胚葉組織又は器官の幹/前駆細胞を作製することができる。本発明の方法を、「本発明のリプログラミング法」ともいう。
本発明は、哺乳動物の内胚葉組織又は器官(肝臓を除く)由来の細胞から、当該細胞の幹/前駆細胞を作製する方法であって、インビトロで前記内胚葉組織又は器官由来の細胞にTGFβ受容体阻害薬を接触させることを含む、前記方法を提供する。
本発明において、成熟膵外分泌細胞からの膵幹/前駆細胞を作製又は誘導するには、哺乳動物の膵外分泌細胞に、TGFβ受容体阻害薬、グリコーゲン合成酵素キナーゼ3(GSK3)阻害薬、およびRhoキナーゼ(ROCK)阻害薬を含む低分子シグナル伝達経路阻害薬を、インビトロで接触させることを含む。
例えばラットの場合、10−20週齢の成体ラットから摘出した膵臓を用いることが好ましいが、2ヶ月齢以下の幼若ラット由来の膵臓を用いてもよい。ヒトの場合、生検又は外科手術により得ることができる。例えば、膵実質、消化管上皮、肺胞上皮、前立腺上皮などいずれも生検により採取することができる。外科手術の場合は、切除した成人の膵臓組織片を用いることも、死亡胎児から切除した膵臓を用いることもできる。あるいは、これら摘出した膵臓から単離精製された膵外分泌細胞を凍結した細胞(凍結膵外分泌細胞)を用いることもできる。
哺乳動物の膵臓又はその組織片から膵外分泌細胞を精製する方法としては、膵組織を単離した後、コラゲナーゼで膵臓を消化し、濾過、遠心等によってランゲルハンス島や非実質細胞、細胞片を除去して膵外分泌細胞を精製する。
本発明に用いられるTGFβ受容体阻害薬としては、トランスフォーミング増殖因子(TGF)β受容体の機能を阻害する作用を有するものであれば特に限定されることはなく、例えば、2−(5−ベンゾ[1,3]ジオキソール−4−イル−2−tert−ブチル−1H−イミダゾール−4−イル)−6−メチルピリジン、3−(6−メチルピリジン−2−イル)−4−(4−キノリル)−1−フェニルチオカルバモイル−1H−ピラゾール(A−83−01)、2−(5−クロロ−2−フルオロフェニル)プテリジン−4−イル)ピリジン−4−イルアミン(SD−208)、3−(ピリジン−2−イル)−4−(4−キノニル)]−1H−ピラゾール、2−(3−(6−メチルピリジン−2−イル)−1H−ピラゾール−4−イル)−1,5−ナフチリジン(以上、メルク社)、SB431542(Sigma Aldrich社)などが挙げられる。好ましくはA−83−01が挙げられる。TGFβ受容体阻害薬には、TGFβ受容体アンタゴニストも含まれる。
これらのTGFβ受容体阻害薬は、1種の化合物を用いてもよいし、2種以上の化合物を組み合わせて用いてもよい。
これらのGSK3阻害薬は、1種の化合物を用いてもよいし、2種以上の化合物を組み合わせて用いてもよい。
これらのROCK阻害薬は、1種の化合物を用いてもよいし、2種以上の化合物を組み合わせて用いてもよい。
(a)TGFβ受容体阻害薬としてA−83−01(A)と、GSK3阻害薬としてCHIR99021(C)との組み合わせ(AC)。
(b)TGFβ受容体阻害薬としてA−83−01(A)と、ROCK阻害薬としてY−27632(Y)との組み合わせ(YA)。
(c)TGFβ受容体阻害薬としてA−83−01(A)と、GSK3阻害薬としてCHIR99021(C)及びROCK阻害薬としてY−27632(Y)との組み合わせ(YAC)。
また、培地には、5−20%の血清(FBS等)を添加することもできるが、無血清培地であってもよい。無血清培地の場合、血清代替物(BSA、HAS、KSR等)を添加してもよい。さらに、通常、増殖因子、サイトカイン、ホルモン等の因子が添加される。これらの因子としては、例えば上皮増殖因子(EGF)、インスリン、トランスフェリン、ヒドロコルチゾン21−ヘミコハク酸又はその塩、デキサメタゾン(Dex)等が挙げられるが、それらに限定されない。
GSK3阻害薬の培地への添加濃度は、例えば、0.01−100μM、好ましくは1−10μMの範囲から適宜選択され得る。
ROCK阻害薬の培地への添加濃度は、例えば、0.0001−500μM、好ましくは1−50μMの範囲から適宜選択され得る。
これらの阻害薬が水不溶性又は水難溶性の化合物の場合、少量の低毒性の有機溶媒(例えば、DMSO等)に溶解した後、上記の最終濃度となるよう培地に添加することができる。
膵外分泌細胞の場合も、102〜106細胞/cm2、好ましくは103〜105細胞/cm2の細胞密度で培養容器上に播種することができる。培養は、CO2インキュベータ中、1−10%、好ましくは2−5%、より好ましくは約5%のCO2濃度の雰囲気下において、30−40℃、好ましくは35−37.5℃、より好ましくは約37℃で行うことができる。培養期間としては、例えば1−4週間、好ましくは1−3週間が挙げられる。1−3日ごとに新鮮な培地に交換する。
胎児膵臓の膵芽細胞も膵幹/前駆細胞(PSC)に包含される。
好ましい一実施態様において、本発明のリプログラミング方法により得られるPSCは、上記(a)及び(b)の性質に加えて、(c)マスター因子であるPdx1及びNkx6.1を発現するほか、Gata4、Hes1、Sox9、Foxa2、CK19、CD133などを発現する。但し、本発明のリプログラミング方法により得られるPSCは、他の既知PSCで発現しているLgr5を発現していない。従って、本発明のPSCは新規なPSCであるといえる。
(d)少なくとも10継代、好ましくは20継代以上、みかけ上の増殖速度が低下しない。
(e)少なくとも10継代、好ましくは20継代以上、膵内分泌細胞への分化能を保持する。
(f)膵外分泌細胞と比較して核/細胞質(N/C)比が高い。
(g)Pdx1、Nkx6.1より選択される1以上のPSCマーカー遺伝子の発現が、膵外分泌細胞と比較して上昇している
(h)Pdx1、Nkx6.1より選択される1以上のタンパク質の発現が、膵外分泌細胞と比較して上昇している
好ましい実施態様においては、本発明のPSCは、上記(d)〜(h)の特徴をすべて有するものである。
従って、本発明は、哺乳動物の内胚葉組織又は器官(肝臓を除く)由来の細胞から、当該細胞の幹/前駆細胞を誘導する方法であって、インビトロで前記内胚葉組織又は器官由来の細胞に、TGFβ受容体阻害薬、又はTGFβ受容体阻害薬とGSK3阻害薬及び/又はROCK阻害薬との組み合わせを接触させることを含む方法が提供される。
TGFβ受容体阻害薬、GSK3阻害薬及びROCK阻害薬はそのままPSC誘導剤として使用できるが、適当な溶媒に溶解して液剤とすることもできる。あるいは、これらの阻害薬は、上記した膵外分泌細胞からPSCを誘導用培地と組み合わせてキット化することもできる。
上記のようにして得られた本発明の幹/前駆細胞は、コラーゲン又はマトリゲルでコーティングした培養容器上で、それぞれTGFβ受容体阻害薬、GSK3阻害薬及びROCK阻害薬の存在下で継代培養することにより、効率よく維持・増幅することができる。
培養容器としては、成熟細胞から幹/前駆細胞を誘導するために用いた培養容器と同様の培養容器を用いることができる。
培養容器としては、膵外分泌細胞からのPSCの誘導に用いたのと同様の培養容器を用いることができる。
幹/前駆細胞から成熟細胞への分化誘導は、例えばYAC添加SHM(Small hepatocyte medium)培地を用いて超低吸着培養皿上で7日間浮遊培養させることにより行うことができる。あるいは、Phorbol 12,13−dibutyrate、LDN193189、Keratinocyte Growth Factor、SANT1、Retinoic Acid、XXI、Betacellulin等を添加した培養液で培養する方法(Cell.2014 Oct 9;159(2):428−39.)等を用いてもよい。
PSCから膵内分泌細胞への分化誘導についても、上記と同様の方法を適用することができる。あるいは、Phorbol 12,13−dibutyrate、LDN193189、Keratinocyte Growth Factor、SANT1、Retinoic Acid、XXI、Betacellulin等を添加した培養液で培養する方法(Cell.2014 Oct 9;159(2):428−39.)等を用いてもよい。
上記4.に記載されるようにして本発明の幹/前駆細胞から再分化した成熟細胞は、例えば、被検化合物の代謝や細胞又は組織毒性の評価等に利用できる。
被検化合物の代謝や毒性の評価には、従来、動物モデル等が用いられていたが、一度に評価できる被検化合物の数に制限があり、また動物モデル等で得られた評価を、そのままヒトに適用できないという問題があった。そのため、ヒト癌細胞株や初代正常ヒト培養細胞を用いる評価方法が採用されている。しかしながら、ヒト癌細胞株は、ヒト癌細胞株で得られた評価が、ヒト正常細胞に適用できないという可能性が残る。また、初代正常ヒト培養細胞は安定供給やコストの面での問題がある。さらに、初代正常ヒト培養細胞を不死化した細胞株は、不死化していない場合と比較して、その活性が低下していることが示されている。本発明の方法により製造された細胞を利用することで、このような問題を解決し得る。
従って、本発明の方法において、膵外分泌細胞を用いる場合は、本発明の方法により製造された膵外分泌細胞に被検化合物を接触させる。次いで、膵外分泌細胞に接触させた被検化合物の代謝を測定する。
生体異物としては、例えば薬剤や食品の候補化合物、既存の薬剤や食品が挙げられるが、これらに限定されるものではなく、生体にとって異物である限り、本発明の生体異物に含まれる。より具体的には、Rifampin、Dexamethasone、Phenobarbital、Ciglirazone、Phenytoin、Efavirenz、Simvastatin、β−Naphthoflavone、Omeprazole、Clotrimazole、3−Methylcholanthrene等が例示できる。
例えば、被検化合物の接触により、インスリン等の酵素遺伝子の発現が誘導された場合や、これら酵素の活性が上昇した場合に、被検化合物が代謝されたと判定される。
また本発明は、細胞が分泌する酵素の分泌誘導物質のスクリーニング方法であって、本発明の方法により得られた幹/前駆細胞、本発明の膵幹/前駆細胞、又は本発明の誘導方法により誘導された細胞に被検化合物を接触させる工程、及び前記細胞において分泌された物質を測定する工程を含む、前記方法が提供される。
例えば、被検物質を、膵幹/前駆細胞又は誘導された膵細胞に接触させた後、細胞から分泌される各酵素が分泌誘導されるか否かを指標として、分泌調整物質をスクリーニングすることができる。
本発明において、被検化合物の膵毒性を評価する場合は、本発明の方法により製造された膵外分泌細胞に被検化合物を接触させる。次いで、被検化合物を接触させた膵外分泌細胞の障害の程度を測定する。障害の程度は、例えば膵外分泌細胞の生存率や膵障害マーカーを指標に測定できる。
なお、すでに膵毒性の有無が判明している化合物を対照として用いることで、より正確に、被検化合物が膵毒性を有するか否かを評価することができる。
膵前駆細胞の樹立
マウスまたはラットの膵外分泌細胞を既知の手法に従ってコラゲナーゼ処理および単離した(Reichert et al.Cold Spring Harb Protoc.2015 Jun 1;2015(6):558−61)。単離した細胞内にインスリン分泌細胞が含まれていないことを、ジチゾン(DTZ)染色により確認した。単離した膵外分泌細胞を、3種類の低分子化合物(Y−27632[10μM],A−83−01[0.5μM],CHIR99021[3μM];以下YAC)を添加したSmall hepatocyte medium(SHM)培地を用いてI型コラーゲンコート培養皿上で平面培養した。

初代培養の14日目に、YAC存在下で培養した細胞をトリプシン処理して回収し、YACを添加したSHM中に9x103細胞/cm2で播種した。マウスの場合、I型コラーゲンコート培養皿上で、ラットの場合、マトリゲルコーティングした培養皿上で細胞を培養した。CELLBANKER(登録商標)1(宝酒造、大津)を用いて凍結ストックを調製した。少なくとも10継代後、BD FACSAria IIセルソーターを用いてPSCのクローニングを実施した。
初代膵外分泌細胞をコラーゲンでコーティングした35mmプレート(IWAKI)上に、YACの存在下又は非存在下で1x102細胞/cm2で播種した。1日目に培地を交換した。2回目の培地交換後、BZ9000オールインワン蛍光顕微鏡(キーエンス、大阪)を用いて低速度撮影を行った。位相差像は2日目から6日目まで30分間隔で300回撮影し、動画は解析野ごとに作成した。次に、個々の細胞を撮影期間を通じて追跡し、目的の細胞に由来する最終の細胞数を計測した。また、個々の細胞に由来するアポトーシス細胞の総計も計数し、アポトーシス頻度を、総アポトーシス細胞/最初の総細胞数(低速度撮影の開始時に計数)として定量した。
膵外分泌細胞およびPSC細胞から、miRNeasy Mini Kit(QIAGEN)を用いて全RNAを単離した。逆転写反応は、High−Capacity cDNA Reverse Transcription Kit(Lifetechnologies)を用い、製造者のガイドラインに従って実施した。得られたcDNAを鋳型として、Platinum SYBR Green qPCR SuperMix UDG(Invitrogen)を用いてPCRを行った。標的遺伝子の発現レベルは内在性コントロールであるβ−アクチンで標準化した。
細胞を4%パラホルムアルデヒドにより15分間固定した。ブロッキング液(Blocking One)(ナカライテスク、京都)で、4℃、30分インキュベートした後、細胞を一次抗体と室温で1時間又は4℃で一晩インキュベートした。次に、Alexa Fluor 488又はAlexa Fluor 594−結合二次抗体(Lifetechnologies)を用いて、一次抗体を検出した。核をHoechst 33342(Dojindo)で共染色した。
細胞を、5%FBSを添加したSHM+YAC中に懸濁し(細胞濃度:1x104〜1x106細胞/cm2)、超低吸着培養皿上で7日間培養した。培地交換は4日目に行った。
1)膵前駆細胞の樹立
膵外分泌細胞含有培地にYACを添加することにより、図1に示す様に高い増殖能を有する小型上皮細胞を樹立した。この細胞は20回以上の継代が可能であり、一細胞培養も可能であった。またインスリン合成能を有する内分泌系細胞へと分化しうることから、膵前駆細胞としての特徴を有していると言える。
樹立した細胞は、図2,3に示されるように膵前駆細胞におけるマスター転写因子であるPdx1とNkx6.1を含む幹細胞マーカーを発現していた。
樹立した膵前駆細胞は、YAC添加SHM培地を用いて超低吸着培養皿上で7日間浮遊培養させることにより、図4に示されるように細胞がクラスター状に集積しながら増殖し、このクラスターはDTZ染色陽性、また免疫染色によってインスリンの染色性が検出された。このことから、膵前駆細胞はインスリン産生細胞へと分化したことがわかる。実際、インスリンに加えて膵内分泌細胞のマスター転写因子であるNgn3やグルコーストランスポーター(GLUT2)のmRNA上昇がみられた(図5)。またインスリン分泌を反映するCペプチド分泌も検出された(図5)。
さらに、膵内分泌細胞への分化誘導後、培地のグルコース濃度を低濃度(3mM)と高濃度(20mM)の条件で比較すると、高濃度グルコースによるInsulin mRNAや培地中Cペプチドの上昇がみられ、グルコース濃度応答性を獲得していることが示された(図7)。
ストレプトゾトシン投与糖尿病モデルマウスを用い、インスリン産生細胞への分化誘導処理を行った膵前駆細胞をMatrigelに封入し膵被膜下へ移植した。図8に示されるように、移植3日後において血糖値の改善が得られた。さらに移植部位において移植細胞のクラスターの多くは腺管様の細胞に分化している一方、一部の細胞はInsulin,Pdx1,Nkx6.1が陽性であり、β細胞へと分化している様子が観察された(図9)。
Claims (26)
- 哺乳動物の内胚葉組織又は器官(肝臓を除く)由来の細胞から、当該細胞の幹/前駆細胞を作製する方法であって、インビトロで前記内胚葉組織又は器官由来の細胞にTGFβ受容体阻害薬を接触させることを含む、前記方法。
- さらに、GSK3阻害薬及び/又はROCK阻害薬を、インビトロで前記内胚葉組織又は器官由来の細胞に接触させることを含む、請求項1に記載の方法。
- さらに、GSK3阻害薬及びROCK阻害薬を、インビトロで前記内胚葉組織又は器官由来の細胞に接触させることを含む、請求項1に記載の方法。
- 内胚葉組織又は器官が、消化管、肺、甲状腺、膵臓、分泌腺、腹膜、胸膜、喉頭、耳管、気管、気管支、膀胱、尿道又は尿管である、請求項1〜3のいずれか1項に記載の方法。
- 内胚葉組織又は器官が膵臓である請求項1〜4のいずれか1項に記載の方法。
- 膵臓由来の細胞が膵外分泌細胞である請求項5に記載の方法。
- 前記膵外分泌細胞と前記TGFβ受容体阻害薬との接触が、該阻害薬の存在下で該膵外分泌細胞を培養することにより実施される、請求項6に記載の方法。
- 前記膵外分泌細胞と前記GSK3阻害薬及び/又は前記ROCK阻害薬との接触が、該阻害薬の存在下で該膵外分泌細胞を培養することにより実施される、請求項6又は7に記載の方法。
- 哺乳動物がヒト、ラット又はマウスである、請求項1〜8のいずれか1項に記載の方法。
- 以下の特徴を有する哺乳動物の膵外分泌細胞由来の膵幹/前駆細胞。
(a)自己再生能を有する
(b)膵内分泌細胞に分化し得る
(c)Pdx1及びNkx6.1を発現するが、インスリンを発現しない - TGFβ受容体阻害薬を含む、哺乳動物の内胚葉組織又は器官(肝臓を除く)由来の細胞から当該細胞の幹/前駆細胞への誘導剤。
- GSK3阻害薬及び/又はROCK阻害薬の組み合わせをさらに含む、請求項11に記載の誘導剤。
- GSK3阻害薬及びROCK阻害薬の組み合わせをさらに含む、請求項11に記載の誘導剤。
- 内胚葉組織又は器官が、消化管、肺、甲状腺、膵臓、分泌腺、腹膜、胸膜、喉頭、耳管、気管、気管支、膀胱、尿道又は尿管である、請求項11〜13のいずれか1項に記載の誘導剤。
- 内胚葉組織又は器官が膵臓である請求項11〜13のいずれか1項に記載の誘導剤。
- 膵臓由来の細胞が膵外分泌細胞である請求項15に記載の誘導剤。
- 哺乳動物がヒト、ラット又はマウス由来である、請求項11〜16のいずれか1項に記載の誘導剤。
- 請求項1〜9のいずれか1項に記載の方法により得られた幹/前駆細胞、又は請求項10に記載の膵幹/前駆細胞の維持又は増幅剤として使用される、請求項11〜17のいずれか1項に記載の誘導剤。
- 請求項1〜9のいずれか1項に記載の方法により得られた幹/前駆細胞、又は請求項10に記載の膵幹/前駆細胞の維持又は増幅方法であって、コラーゲン又はマトリゲルでコーティングした培養容器上で、TGFβ受容体阻害薬、GSK3阻害薬及びROCK阻害薬の存在下で、当該幹/前駆細胞又は膵幹/前駆細胞を継代培養することを特徴とする、前記方法。
- 請求項1〜9のいずれか1項に記載の方法により得られた幹/前駆細胞、又は請求項10に記載の膵幹/前駆細胞から、哺乳動物の内胚葉組織又は器官由来の細胞、又は膵外分泌細胞に誘導する方法であって、TGFβ受容体阻害薬、GSK3阻害薬及びROCK阻害薬の存在下で前記幹/前駆細胞、又は膵幹/前駆細胞を培養する工程を含む、前記方法。
- 被検化合物の哺乳動物体内での代謝を評価する方法であって、
(i)請求項1〜9のいずれか1項に記載の方法により得られた幹/前駆細胞、請求項10に記載の膵幹/前駆細胞、又は請求項20に記載の方法により得られた細胞に被検化合物を接触させる工程、及び
(ii)前記細胞における被検化合物の代謝を測定する工程
を含む、方法。 - 細胞が分泌する酵素の分泌誘導物質のスクリーニング方法であって、
(i)請求項1〜9のいずれか1項に記載の方法により得られた幹/前駆細胞、請求項10に記載の膵幹/前駆細胞、又は請求項20に記載の方法により誘導された細胞に被検化合物を接触させる工程、及び
(ii)前記細胞において分泌された物質を測定する工程、
を含む、前記方法。 - 哺乳動物に対する被検化合物の内胚葉組織又は器官毒性を評価する方法であって、
(i)請求項1〜9のいずれか1項に記載の方法により得られた幹/前駆細胞、請求項10に記載の膵幹/前駆細胞、又は請求項20に記載の方法により得られた細胞に被検化合物を接触させる工程、及び
(ii)被検化合物と接触した細胞における障害の有無又はその程度を測定する工程
を含む、前記方法。 - 請求項1〜9のいずれか1項に記載の方法により得られた幹/前駆細胞、請求項10に記載の膵幹/前駆細胞、又は請求項20に記載の方法により得られた細胞を含む、内胚葉組織又は器官障害改善剤。
- 哺乳動物における内胚葉組織又は器官障害の改善方法であって、当該組織又は器官障害を有する哺乳動物に、請求項1〜9のいずれか1項に記載の方法により得られた幹/前駆細胞、請求項10に記載の膵幹/前駆細胞、又は請求項20に記載の方法により誘導された細胞の有効量を投与することを含む、前記方法。
- 内胚葉組織又は器官障害改善剤として使用するための、請求項1〜9のいずれか1項に記載の方法により得られた幹/前駆細胞、請求項10に記載の膵幹/前駆細胞、又は請求項20に記載の方法により得られた細胞。
