Abstract
The invention discloses a haematococcus pluvialis micelle extract for removing wrinkles, repairing and whitening skin and a preparation method thereof. The haematococcus pluvialis micelle extract for removing wrinkles, repairing and whitening is prepared from the following raw materials: tween 80 accounting for 10% of the total volume of the extract, 1% astaxanthin concentrated solution accounting for 5-10% of the total volume of the extract, medium-chain triglyceride accounting for 2% of the total volume of the extract, ascorbic acid accounting for 0.05% of the total volume of the extract, and ethylenediaminetetraacetic acid accounting for 0.1% of the total volume of the extract. The haematococcus pluvialis extract prepared by the method micellizes astaxanthin, so that the astaxanthin can form a good dispersion system in oil and water, the application performance of the astaxanthin in a water-soluble system is greatly improved, and the formed astaxanthin-based micelle system has excellent stability.
Description
Technical Field
The invention relates to the technical field of haematococcus pluvialis extracts, in particular to a haematococcus pluvialis micelle extract for wrinkle removal, repair and whitening and a preparation method thereof.
Background
Natural antioxidants are widely available in nature, and ascorbic acid (vitamin C), tocopherol (vitamin E), tea polyphenol, carotene and the like are common, wherein the most powerful antioxidant is astaxanthin of carotene class. Astaxanthin obtained commercially on a large scale is mainly derived from haematococcus pluvialis, and is fermented, extracted and purified to obtain commercial products with different purities. The astaxanthin has the function of resisting oxidation, contains active ingredients, can eliminate free radicals in cells, enhances the regeneration capability of the cells, can also maintain the balance of the organism, reduces the accumulation of aged cells and enables people to keep young; astaxanthin also has a function of ultraviolet radiation protection, so astaxanthin is widely used in cosmetics and can repair and whiten skin.
The astaxanthin molecule contains a conjugated olefinic chain in the middle and contains hydroxyl ketone groups on two side rings. Astaxanthin has a higher bioavailable activity than other antioxidants because it binds to the cell membrane from the inside and outside. The ring structures on both sides eliminate free radicals inside and outside the cell membrane, while the lipophilic alkene chain in the middle can eliminate free radicals in the cell membrane. After astaxanthin scavenges oxygen radicals, the carotenoid structure is reduced back to the original state after energy transfer is dispersed by the solvent. According to the report of the literature, the antioxidant activity of the astaxanthin is 10 times that of lutein, zeaxanthin and canthaxanthin and 100 times that of alpha tocopherol.
The molecular formula of astaxanthin is as follows:
natural antioxidants can be mainly classified into two major groups according to their solubility in water, water-soluble antioxidants such as ascorbic acid, tea polyphenols; oil-soluble antioxidants, such as vitamin E, carotene, astaxanthin. Water-soluble antioxidants are difficult to exert antioxidant effects in oil-based systems, and oil-soluble antioxidants are also difficult to exert antioxidant effects in oil-based systems.

The invention also relates to a preparation method of the haematococcus pluvialis micelle extract for wrinkle removal, repair and whitening, which is characterized by comprising the following specific steps of:
1) adding tween 80, 1% astaxanthin concentrated solution, medium chain triglyceride, ascorbic acid, and ethylenediamine tetraacetic acid into deionized water at a certain proportion, and stirring thoroughly;
2) and (3) shearing the solution obtained in the step (1) at a high speed of 3000rpm, standing, filtering, and centrifuging to form stable and transparent micellar haematococcus pluvialis extract liquid.
The invention has the beneficial effects that: the astaxanthin is micellized, so that a good dispersion system can be formed in oil and water, the application performance of the astaxanthin in a water-soluble system is greatly improved, and the formed astaxanthin micelle system has excellent stability.

The technical solutions in the embodiments of the present invention will be clearly and completely described below with reference to the drawings in the embodiments of the present invention, and it is obvious that the described embodiments are only a part of the embodiments of the present invention, and not all of the embodiments. All other embodiments, which can be derived by a person skilled in the art from the embodiments given herein without making any creative effort, shall fall within the protection scope of the present invention.
Example 1
The haematococcus pluvialis micelle extract for removing wrinkles, repairing and whitening skin is prepared from the following raw materials: tween 80 accounting for 10% of the total volume of the extract, 1% astaxanthin concentrated solution accounting for 5-10% of the total volume of the extract, medium-chain triglyceride accounting for 2% of the total volume of the extract, ascorbic acid accounting for 0.05% of the total volume of the extract, and ethylenediaminetetraacetic acid accounting for 0.1% of the total volume of the extract.
The preparation method of the haematococcus pluvialis micelle extract for removing wrinkles, repairing and whitening is characterized by comprising the following specific steps of:
1) adding tween 80, 1% astaxanthin concentrated solution, medium chain triglyceride, ascorbic acid, and ethylenediamine tetraacetic acid into deionized water at a certain proportion, and stirring thoroughly;
2) and (3) shearing the solution obtained in the step (1) at a high speed of 3000rpm, standing, filtering, and centrifuging to form stable and transparent micellar haematococcus pluvialis extract liquid.
The mixture of the extracts of Haematococcus pulvialis obtained in this example was compared with a commercially available astaxanthin solution (containing 500ppm ascorbic acid) for the stability of fluorescence irradiation. The samples obtained in this way have the best photostability every 12 hours, as the disappearance of the colour is observed visually.
Example 2
Adding 12% Tween 80, 10% astaxanthin oil concentrate 1%, and 2% MCT (medium chain triglyceride) into deionized water, stirring, shearing at 3000rpm, standing, centrifuging to obtain stable transparent liquid.
The light transmittance was measured at 500nm to check the transparency. Experiments found that the transmittance remained constant after more than 10% tween and more than 2% MCT (medium chain triglyceride) in the above system. The particle size was 10-20nm as measured by dynamic light scattering.
Example 3
The haematococcus pluvialis micellar extract sample prepared in the example 1 for removing wrinkles, repairing and whitening skin is diluted into 0.05% diluent by water together with commercially available nano-astaxanthin, micro-emulsified astaxanthin and 1% astaxanthin stock solution, and 500ppm ascorbic acid is added.
The samples were subjected to fluorescent irradiation and the astaxanthin content of the samples was measured for different irradiation times to compare the stability. The results show that the micellization treated astaxanthin retained the highest astaxanthin content.
The astaxanthin is micellized, so that a good dispersion system can be formed in oil and water, the application performance of the astaxanthin in a water-soluble system is greatly improved, and the formed astaxanthin micelle system has excellent stability; the haematococcus pluvialis micelle extract prepared by the invention is applied to cosmetic products, and can achieve better effects of repairing and whitening skin.
It will be apparent to those skilled in the art that various changes and modifications may be made in the present invention without departing from the spirit and scope of the invention. Thus, if such modifications and variations of the present invention fall within the scope of the claims of the present invention and their equivalents, the present invention is also intended to include such modifications and variations.
Claims (2)
1. The haematococcus pluvialis micelle extract capable of removing wrinkles, repairing and whitening is characterized by being prepared from the following raw materials: tween 80 accounting for 10% of the total volume of the extract, 1% astaxanthin concentrated solution accounting for 5-10% of the total volume of the extract, medium-chain triglyceride accounting for 2% of the total volume of the extract, ascorbic acid accounting for 0.05% of the total volume of the extract, and ethylenediaminetetraacetic acid accounting for 0.1% of the total volume of the extract.
2. The preparation method of the haematococcus pluvialis micelle extract with the functions of wrinkle removal, repair and whitening is characterized by comprising the following specific steps:
1) adding tween 80, 1% astaxanthin concentrated solution, medium chain triglyceride, ascorbic acid, and ethylenediamine tetraacetic acid into deionized water at a certain proportion, and stirring thoroughly;
2) and (3) shearing the solution obtained in the step (1) at a high speed of 3000rpm, standing, filtering, and centrifuging to form stable and transparent micellar haematococcus pluvialis extract liquid.
