Abstract
本公开提供一种全人源LAG‑3结合分子,该结合分子从天然噬菌体库筛淘获得。其具有显著优于LAG3.5的亲和力,在疾病治疗领域尤其是癌症治疗领域有巨大的应用潜力。

技术领域
本发明属于生物医药技术领域,具体的,本发明提供一种LAG-3结合分子及其应用。
背景技术
淋巴细胞活化基因-3分子(lymphocyte activation gene3,LAG-3,LAG3,CD223)主要分布于活化的T淋巴细胞、NK细胞和树突状细胞中。LAG3作为表达于淋巴细胞表面的抑制性分子,参与调节T淋巴细胞、效应T淋巴细胞等的增殖与活化,并对维持生物体内环境稳定发挥一定的作用,与肿瘤的发生发展密切相关。
淋巴细胞活化基因-3,即LAG-3,是免疫球蛋白超家族的一员,定位于人12号染色体(12p13.3)。成熟的LAG3分子由498个氨基酸组成,分子量为70KDa。LAG-3分子由胞外区、跨膜区和胞浆区组成,其中,胞外区由4个免疫球蛋白超家族样结构域(D1~D4)组成。D1属于V系免疫球蛋白超家族,而D2,D3和D4属于C系免疫球蛋白超家族。D1结构域中包括一个由富含脯氨酸的30个氨基酸组成的环状结构,与主要组织相容性复合体Ⅱ类分子,MHCⅡ特异性结合。大部分LAG-3通过D1区形成的二聚体形式表达于细胞膜。尽管LAG-3二聚化相对较弱,但二聚化是LAG-3结合MHCⅡ分子的必要形式。
至今,已发现LAG-3的配体有5种。第一种配体为主要组织相容性复合体Ⅱ类分子,MHCⅡ类分子。LAG3与CD4分子结构的高度同源性使得MHCⅡ类分子同样也是LAG-3分子的配体。LAG-3与MHCⅡ类分子的结合位置位于D1结构域中富含脯氨酸的环状结构中。而且LAG-3与MHCⅡ类分子的亲和力(KD:60nmol/L)是CD4分子的100倍,表明LAG3分子能够有效竞争CD4与MHC类分子的结合,有效抑制T细胞活化。LAG3的另外4种配体分别为肝窦内皮细胞凝集素(liver sinusoidal endothelial celllectin,LSECtin)、半乳糖凝集素-3(galectin-3)、α-突触核蛋白原纤维(α-synucleinfibrils,α-Syn fibrils)和纤维蛋白样蛋白1(fibrinogen-like protein 1,FGL1)。2019年陈列平团队发现FGL1是LAG3的T细胞抑制功能的配体,提出一条新的肿瘤免疫逃逸通路FGL1-LAG-3,阻断FGL1与LAG-3的相互作用可以增强T淋巴细胞的抗肿瘤作用,对肿瘤免疫疗法的研究具有重要指导意义。
LAG-3负调节CD4+T细胞,CD8+T细胞以及Treg细胞。参与调控记忆性T细胞池,最大化调节T细胞功能与增殖,调控树突状细胞,参与淋巴细胞与肿瘤的相互作用,对维持内环境的稳定起着非常重要的作用。LAG-3活化第一信号轴“CD3-TCR-MHCⅡ”中MHCⅡ分子特异性结合,一方面阻断T细胞活化的信号转导通路,另一方面LAG-3分子胞内段产生免疫抑制信号下调CD4+T细胞活性。同样,LAG-3分子能够促进Treg细胞分化,参与信号转导及转录激活因子5下游信号,从而增强Treg细胞的抑制作用。2003年HUARD等研究表明,在人CD4+T细胞中抑制LAG-3会使白介素2(interleukin-2,IL-2)、IL-4、IFN-γ和肿瘤坏死因子α(tumornecrosis factoralpha,TNF-α)的分泌增加。在小鼠实验中,抑制LAG-3分子可刺激CD8+T细胞的增殖,并能增强其细胞毒活性。抑制LAG-3分子可以明显地抑制Treg细胞的功能,同时,抑制LAG-3分子还可以通过抑制Treg细胞功能,使效应性T细胞扩增。因此,抑制LAG-3分子功能可以增强T细胞的抗肿瘤作用,该分子有可能是一个潜在的肿瘤免疫治疗靶点。
目前,至少12种LAG-3抑制剂作为抗癌治疗药物处于临床试验中或者正在招募参与者。Bristol-Myers Squibb公司开发的一种抗LAG-3的全人源IgG4单克隆抗体BMS-986016,Novartis公司开发的一种抗LAG-3的全人源IgG4单克隆抗体LAG525,主要与抗PD-1/PDL1联合使用治疗多种实体瘤和血液系统恶性肿瘤,并取得了良好效果。综上,LAG-3作为一种新型免疫治疗靶点,在肿瘤的免疫治疗中发挥着重要的临床意义。目前虽然已有多家公司在研发抗LAG-3抗体,但均未获得批准上市。目前尚有很多恶性肿瘤并未获得明确的免疫治疗获益证据,抗LAG-3抗体免疫治疗的应用范围和治疗有效性仍有很大研究空间。
发明内容
本发明提供了一种LAG-3结合分子,所述LAG-3结合分子包括重链可变区和轻链可变区,所述重链可变区序列包含如SEQ ID NO 2所述的序列;所述轻链可变区包含如SEQ IDNO 4所述的序列。
本发明提供一种LAG-3结合分子,所述LAG-3结合分子包括重链可变区和轻链可变区,所述重链可变区含有分别如SEQ ID NO 7、8、9所述的HCDR1、HCDR2、HCDR3;所述轻链可变区含有分别如SEQ ID NO 10、11、12所述的LCDR1、LCDR2、LCDR3。
本发明提供一种LAG-3结合分子,所述LAG-3结合分子包括重链可变区和轻链可变区,所述重链可变区含有分别如SEQ ID NO 7、8、9所述的HCDR1、HCDR2、HCDR3;所述轻链可变区含有分别如SEQ ID NO 10、11、12所述的LCDR1、LCDR2、LCDR3;所述重链CDR序列选自含SEQ ID NO 2所述序列,所述轻链CDR序列选自SEQ ID NO 4所述序列。
进一步的,还包括重链恒定区,所述重链恒定区为人IgG1、IgG2、IgG3、IgG4中的一种;

有益效果
本发明提供的LAG-3结合分子9G1具有显著优于LAG3.5的亲和力。

具体实施方式
下述实施例中所述实验方法,如无特殊说明,均为常规方法:所述试剂和生物材料,如无特殊说明,均可从商业途径获得。
(一)术语说明
本发明所述CDR为“互补决定区”,是抗体可变结构域中在序列上高变并且形成在结构上确定的“超变环”和/或含有抗原接触残基“抗原接触点”的区域。CDR主要负责与抗原表位结合。重链和轻链的CDR通常被称作CDR1、CDR2和CDR3,从N-端开始顺序编号。位于抗体重链可变结构域内的CDR被称作HCDR1、HCDR2和HCDR3,而位于抗体轻链可变结构域内的CDR被称作LCDR1、LCDR2和LCDR3。在一个给定的轻链可变区或重链可变区氨基酸序列中,各CDR的精确氨基酸序列边界可以使用许多公知的抗体CDR编号系统的任一种或其组合确定,所述编号系统包括例如:基于抗体的三维结构和CDR环的拓扑学的Chothia,基于抗体序列可变性的Kabat,AbM(University of Bath),IMGT,Contact,根据不同的CDR编号系统,9G1分子CDR的残基确定略有差异,具体如表1:

除非另有说明,否则在本发明中,当提及抗体可变区中的残基位置时,是指根据Kabat编号系统确定的编号位置。
所述CHO细胞为中国仓鼠卵巢细胞(chinese hamster ovary cell);ExpiCHO S细胞(Thermo Fisher)为悬浮生长的CHO细胞;CHO-K1为贴壁生长的CHO细胞。
适用于本发明的“抗体及其抗原结合片段”包括但不限于多克隆、单克隆、单价、双特异性、异缀合物、多特异性、重组、异源、异源杂合、嵌合、人源化(特别是嫁接有CDR的)、去免疫的、或人的抗体,Fab片段、Fab’片段、F(ab’)2片段、由Fab表达库产生的片段、Fv、二硫化物连接的Fv(dsFv)、单链抗体(例如scFv)、双抗体或四抗体、纳米抗体(nanobody,也称为单结构域抗体)、抗独特型(抗Id)抗体(包括例如针对本发明抗体的抗Id抗体)和上述任一种的表位结合片段。
Fab片段包括重链可变结构域和轻链可变结构域,并且还包括轻链的恒定结构域以及重链的第一恒定结构域(CH1)。Fab’片段因在重链CH1结构域的羧基末端增加了一些残基(包括来自抗体铰链区的一个或多个半胱氨酸)而与Fab片段不同。
Fv是包含完整抗原结合位点的最小抗体片段。双链Fv种类由一个重链可变结构域和一个轻链可变结构域以紧密的,非共价缔合的二聚体组成。在单链Fv(scFv)种类中,一个重链可变结构域和一个轻链可变结构域可以通过柔性肽接头共价连接从而使轻链和重链可以以类似于双链Fv种类的“二聚体”结构缔合。在这种构型中,正是每个可变结构域的三个CDR作用来限定了VH-VL二聚体的表面上的抗原结合位点。六个CDR将抗原结合特异性赋予抗体,然而即使是单个可变结构域(或只包含对抗原特异的三个CDR的一半Fv)也具有识别和结合抗原的能力,尽管亲和性低于完整结合位点。
Fab’-SH是对其中恒定结构域的半胱氨酸残基携带一个游离硫醇基的Fab’的称谓。F(ab’)2抗体片段最初是作为成对Fab’片段生成的,在Fab’片段之间具有铰链半胱氨酸。抗体片段的其它化学偶联也是已知的。
术语“全长抗体”、“完整的抗体”和“完整抗体”在本文被可交换地用于指结构与天然抗体结构基本相似或具有包含如本文所定义的Fc区的重链的抗体。
术语“Fc区”在本文中用于定义免疫球蛋白重链的C端区域,所述区域包含至少一部分的恒定区。该术语包括天然序列Fc区和变体Fc区。
“分离的”抗体是指,其已经与其天然环境的组分分离。在一些实施方案中,将抗体纯化至超过95%纯度,如通过例如电泳(例如,SDS-PAGE,等电聚焦(IEF),毛细管电泳)或层析(例如,离子交换或反相HPLC))确定的。
“分离的”核酸是指这样的核酸分子,其已经与其天然环境的组分分离。分离的核酸包括包含在通常包含该核酸分子的细胞中的核酸分子,但是该核酸分子存在于染色体外或在不同于其天然染色体位置的染色体位置处。
“分离的编码抗LAG-3抗体或其片段的核酸”是指一个或多个核酸分子,其编码抗体重链或轻链(或其片段),包括在单一载体或分开的载体中的这样的核酸分子,以及存在于宿主细胞中的一个或多个位置处的这样的核酸分子。
术语“核酸”、“核酸序列”、“核苷酸序列”或“多核苷酸序列”和“多核苷酸”互换使用。它们指聚合物形式的任何长度的核苷酸(脱氧核糖核苷酸或核糖核苷酸)或其类似物。多核苷酸可以是单链或双链,并且如果为单链,可以是编码链或非编码(反义)链。多核苷酸可以包含修饰的核苷酸,如甲基化核苷酸及核苷酸类似物。核苷酸的序列可以被非核苷酸组分打断。可以在聚合后进一步修饰多核苷酸,如通过与标记组分缀合。核酸可以是重组多核苷酸或在自然界中不存在或与另一个多核苷酸以非自然布局连接的基因组来源、cDNA来源、半合成来源或合成来源的多核苷酸。
术语“多肽”、“肽”和“蛋白质”(如果为单链)在本文中互换地使用并且为任意长度的氨基酸聚合物。该聚合物可以是线形或分支的,它可以包含修饰的氨基酸,并且它可以由非氨基酸隔断。该术语也包括已经被修饰(例如,二硫键形成、糖基化、脂质化、乙酰化、磷酸化或任何其他操作,如以标记组分缀合)的氨基酸聚合物。多肽可以从天然来源分离,可以通过重组技术从真核或原核宿主产生并且可以是合成方法的产物。
(二)实施例
实施例1、富集筛选Fab噬菌体库抗体库
1)准备工作:先将LAG3抗原进行生物素化标记(LAG3-biotin),链霉亲和素包被的磁珠2%脱脂牛奶封闭,以链亲和素与Fab噬菌体抗体库孵育,去除非特异吸附的Fab噬菌体抗体。,
2)淘选:取抗体库0.5ml(约1×1012-13个Fab噬菌体颗粒),与0.5、1或3μg LAG3-biotin室温下孵育1小时;加入链霉亲和素包被的磁珠以俘获LAG3-biotin和抗LAG3噬菌体抗体复合物。经充分淘洗,去除非特异和亲和力较低的噬菌体抗体,以1mg/ml的胰蛋白酶洗脱噬菌体抗体。以洗脱的噬菌体侵染TG1大肠杆菌即可恢复成为特异性针对LAG3靶标的亚库(sub-library)。重复此淘选过程,可富集具有较强亲和力的噬菌体抗体。
计算富集效率,进行噬菌体扩增。富集效率结果如表2所示。

实施例2、单克隆噬菌体ELISA鉴定
将第二轮、第三轮洗脱下来的噬菌体感染TG1细胞,涂ZXYTAG平板获得单菌落。挑取600个单菌落培养在96孔深孔板中,加入辅助噬菌体M13进行噬菌体扩增。吸取扩增后的噬菌体进行ELISA鉴定。结果显示15个单克隆噬菌体显示阳性结果,如图1所示。
实施例3、原核表达蛋白ELISA阻断实验鉴定
将实施例2中所述15个噬菌体ELISA阳性单克隆菌摇菌至OD600=0.6,加入IPTG进行诱导,30℃诱导表达过夜,次日,将菌液离心,PBS洗涤两遍后裂解菌液提取总蛋白。
以配体FGL1包被ELISA板,TG1原核表达蛋白、抗LAG3抗体(anti-LAG3)分别为阴性和阳性对照。将原核表达总蛋白与LAG3蛋白(1.2μg/ml)等体积混匀后,37℃孵育1h,加入FGL1包被的ELISA板上,37℃孵育1h。PBST洗涤4遍,加入抗mFc-HRP,37℃孵育1h。PBST洗涤4遍。TMB显色,酶标仪检测OD450值。其中,有4个单克隆原核表达蛋白对LAG3和配体FGL1的结合阻断活性优于LAG3.5(CN104411723A),如图2所示。
实施例4、全长抗体构建和表达
将结合和阻断均为阳性的噬菌体单克隆送样测序,依据获得的可变区序列构建全长抗体,其中重链可变区VH均嫁接到人IgG4(氨基酸序列如SEQ ID NO 5)恒定区,而轻链可变区VL的C-末端与人κ(氨基酸序列如SEQ ID NO 6)恒定区连接。轻重链克隆至表达载体pHr,依据说明书瞬转入ExpiCHO-S(life technology)细胞进行表达,并采用Protein A层析柱进行亲和纯化。
实施例5:Elisa检测LAG3-FGL1阻断活性
用PBS将FGL1稀释到1μg/ml,按50μl/孔加入酶标板进行板孔的包被。孔内以200μl封闭液(0.05%PBST配制的5%牛奶)封闭后,加入LAG3-biotin和不同浓度全长LAG3重组抗体的混合液孵育,继之以HRP-链亲和素检测LAG3-biotin的结合,结合信号以TMB显色为准。试验结果如图3所示。
实施例6:FACS检测LAG3结合活性
胰酶消化CHO-K1-LAG3细胞,收集细胞离心去上清,重悬计数,将细胞数调整到每孔1.5x105个细胞。1000rpm离心2min,以1%BSA(溶于PBS)溶液洗一次,去上清。将待测抗体稀释(1%BSA,同上)到20μg/ml作为起始浓度,5倍倍比稀释,共得到7种不同浓度,取每个浓度的抗体反应液取50μl,分别加到细胞沉淀中,重悬细胞,混匀,4℃孵育30min,以稀释液作为对照,做同样的孵育反应。孵育完成后离心弃去液体,将50μlPE标记的抗人IgG二抗加入到细胞悬液中,4℃孵育30min后离心,以1%BSA缓冲液洗一次,然后用稀释液200μl重悬细胞。用流式细胞仪检测PE荧光强度。结果如图4所示。
实施例7:FACS检测LAG3-MHCⅡ结合阻断活性
收集Raji细胞并计数,将细胞数调整到每孔3x105个细胞。1000rpm离心2min,稀释缓冲液(或称稀释液,1%BSA溶于PBS)洗一次,弃去上清。将LAG3-biotin稀释到20μg/ml,同时将待测抗体稀释到40μg/ml作为起始浓度,5倍倍比稀释,共得8个浓度的待测抗体溶液(包含0点)。将LAG3-biotin与各浓度抗体等体积混合,4℃孵育30min。取50μl混合液,分别加到细胞沉淀中,重悬细胞,混匀,冰上孵育30min。反应结束后弃去上清,以100μl稀释液重悬细胞。将二抗PE-Streptavidin加人50μl到细胞悬液中,在冰上孵育30min后离心,用稀释液洗一次,然后用约200μl稀释液重悬。用流式细胞仪检测PE荧光强度。结果如图5所示。
实施例8:基于表面等离子共振(SPR)技术检测9G1与9G1抗原的亲和力
使用BIACORE X100 PLUS对9G1对LAG-3抗原的亲和力活性进行测定。实验方法如下:溶液配制:从冰箱取出10×运行缓冲液HBS-EP+,按照HBS-EP+:超纯水=1:9的比例配制一定体积的运行缓冲液,0.22μm膜过滤后备用。偶联配体:通过计算公式:Rmax=(分析物MW/配体MW)×RL×SmRL=配体偶联水平Rmax描述了芯片表面的最大结合容量,对于动力学低偶联需要Rmax≤100Sm=化学计量比(分析物:配体,未知时选择Sm=1)本实验中配体为9G1,分子量为145KD,分析物为9G1抗原,分子量为47KD,Sm=1根据公式计算得出RL≤308RU.通过检测9G1浓度为0.5μg/ml时RU值为270,故抗体浓度设定为0.5μg/ml,上样时间为180s。运行分析物:分析物浓度:26.6nM,13.3nM,6.65nM,3.325nM,1.6625nM,0.83125nM,0nM contact time 180s;stability time 0s,dissociation time:600s,regenerationtime:60s,再生buffer为10mM Gly-HCl pH1.5,根据前面步骤优化的条件设好参数,用运行Buffer稀释好样品,按照仪器提示放好样品,再生Buffer,新鲜的去离子水,开始实验。实验结果:用Biacore X100 Evaluation software对获得的数据进行动力学分析(1:1拟合),获得数据如表3所示,其亲和力显著高于LAG3.5的亲和力KD(M)0.20E-9(CN 104411723 A)。结果如图6。

实施例9:抗体转录激活的活性
通过人工构建报告基因细胞系,如以Jurkat细胞为宿主细胞,进行细胞遗传工程操作,转入荧光素酶(luciferase),且处于NFAT启动子控制之下,同时使之稳定表达LAG3,在有外部TCR激活信号,同时有LAG3结合MHCII的情况下,荧光素酶仅有较低水平表达,而在培养体系内加入不同浓度的LAG3抗体,解除MHCII/FGL1通过结合LAG3引起的抑制信号传导后,荧光素酶的转录增强,有较高水平表达,细胞裂解后,加底物,就可以检测荧光信号。本发明采用了Cobioer公司开发的两种报告基因系统分别检测了9G1和LAG3.5-similar转录激活的活性次,结果如图7A/图7B/图7C。
实施例10 LAG3抗体T淋巴细胞激活实验
1)CD4+T细胞、CD8+T细胞分选扩增
从人全血中提取PBMC,将细胞密度调整到5E7/mL,取0.25-2mLPBMC于分离管,向管中加入Isolation Cocktail50μL/mL,室温静置5分钟后,将Rapid Spheres 50μL/mL加入分离管中,用缓冲液补足2.5mL,混匀。将分离管(开盖)放入磁极中室温静置3分钟,将管中液体倒入新的试管。将分离出的CD4+T细胞、CD8+T细胞转移至T细胞扩增培养基,调整细胞密度支1E6/mL,加入激活剂和IL-2,第3天,第5天,第7天分别扩大培养体积8倍,4倍,4倍,第10天收集细胞冻存或开展实验。
2)SEB激活CD4+T细胞、CD8+T细胞分泌IL-2和INFγ
实验前一天将2μg/ml FGL-1 50uL/孔包被到96孔细胞培养板。第二天去除孔板溶液,将扩增后的CD4+T细胞、CD8+T细胞密度调整到1x106/ml,每孔100μL铺到96孔板中。将待测抗体稀释到400nM作为起始浓度,10倍梯度稀释4个浓度点,加入50μL到孔板细胞中,设置3个复孔。稀释SEB到400ng/mL,加入50μL到孔板细胞中。将孔板置于37℃,5%CO2培养箱孵育72h。SEB终浓度为100ng/ml,抗体起始终浓度为100nM。72h后收集上清,按说明书,用ELISA试剂盒(RD,DY202,DY285B)检测上清中IL-2和INFγ分泌水平,结果如图8A/图8B/图8C。

Claims (13)
1.一种LAG-3结合分子,包括重链可变区和轻链可变区,其特征在于:所述重链可变区含有分别如SEQ ID NO 7、8、9所述的HCDR1、HCDR2、HCDR3;和/或,所述轻链可变区含有分别如SEQ ID NO 10、11、12所述的LCDR1、LCDR2、LCDR3。
2.如权利要求1所述的LAG-3结合分子,其特征在于:所述HCDR序列选自如SEQ ID NO 2所述的序列;所述LCDR序列选自如SEQ ID NO 4所述的序列。
3.如权利要求1-2任一项所述的LAG-3结合分子,其特征在于:还包括重链恒定区,所述重链恒定区为人IgG1、IgG2、IgG3、IgG4中的一种,优选为人的IgG4。
4.如权利要求1-2任一项所述的LAG-3结合分子,其特征在于:还包括轻链恒定区,所述轻链恒定区为人κ、λ链或其变体,优选为人κ链或其变体。
5.如权利要求1-2任一项所述的LAG-3结合分子,其特征在于:所述LAG-3结合分子为LAG-3全长抗体或LAG-3抗体片段。
6.如权利要求5所述的LAG-3结合分子,其特征在于:所述LAG-3抗体片段包括Fab、Fab’、F(ab)2、F(ab’)2、Fv或ScFv中的一种或几种组合。
7.一种分离的LAG-3结合分子,其特征在于:该分离的LAG-3结合分子与前述任一项所述的LAG-3结合分子竞争性结合人LAG-3抗原;或与前述任一项所述的LAG-3结合分子结合相同的人LAG-3抗原表位。
8.一种多核苷酸序列或组合,其特征在于:所述多核苷酸序列或组合编码所述的LAG-3结合分子的氨基酸序列。
9.一种重组DNA表达载体,其特征在于:所述重组DNA表达载体包含如权利要求8所述的多核苷酸序列或组合。
10.一种转染如权利要求9所述载体的宿主细胞,其特征在于:所述宿主细胞包括原核细胞、酵母细胞、昆虫细胞、哺乳动物细胞、ExpiCHO S细胞或CHO-K1细胞。
11.一种药物或药物组合物,其特征在于:其含有如前所述的抗LAG-3结合分子以及一种或多种药学上可接受的载体、稀释剂或赋形剂。
12.一种包含如前所述的抗LAG-3结合分子的分离多肽或蛋白,或包含如前所述的药物组合物,或如前所述的核苷酸序列或组合在制备治疗癌症的药物中的用途.
13.一种所述的LAG-3结合分子在制备治疗癌症疾病药物中的应用,所述癌症疾病包括黑色素瘤、非小细胞肺癌、软组织细胞癌、头颈癌、头颈部鳞状细胞癌、胃癌、食管癌、MSS结直肠癌、脊索瘤、血液肿瘤、非霍奇金淋巴瘤、霍奇金淋巴瘤、宫颈癌、子宫内膜癌、胰腺癌、乳腺癌、腹膜癌、肾细胞癌等。
