Abstract
本发明涉及保健品技术领域,尤其涉及一种乌发生发组合物。本发明使用何首乌30~50份、葵花籽30~60份、胡萝卜10~40份、杏仁5~20份、无花果5~15份、和叶酸类化合物0.1~1份,进行复配,利用各组分之间的协同作用,得到一种适合孕妇食用的具有乌发生发效果的组合物,对孕妇防脱发的防治效果良好。

技术领域
本发明属于保健品技术领域,尤其涉及一种乌发生发组合物。
背景技术
脱发有病理性脱发和生理性脱发,正常人每天脱落100根以内的头发都是正常现象属于生理性脱发。病理性脱发为头发脱落速度过快或者过多。脱发是指头发异常或过度的脱落,其主要症状是头发油腻,亦或焦枯发蓬,缺乏光泽,有淡黄色鳞屑固着难脱,或灰白色鳞屑飞扬。现代医学则认为脱发与遗传因素、激素水平以及营养因素具有很密切的联系。脱发按其原因主要包含以下几类:①脂溢性脱发:目前西医治疗男性脂溢性脱发,因为男性脱发与体内双氢睾酮浓度有关。有研究指出女性脂溢性脱发与头皮睾酮水平增高有关,而与二氢睾酮水平升高无关,这说明女性的脂溢性脱发应以抗雄激素药物治疗为主。②病理性脱发:主要由于病毒、细菌、高热对毛母细胞有损伤,抑制了毛母细胞正常分裂,使毛囊处于休克状态而导致脱发,如急性传染病、长期服用某种药物等。③化学性脱发:有害化学物质对头皮组织、毛囊细胞的损害导致脱发。④物理性脱发:空气污染物堵塞毛囊、有害辐射等原因导致的脱发。⑤营养性脱发:消化吸收机能障碍造成营养不良导致脱发。⑥肥胖性脱发:大量的饱和脂肪酸在体内代谢后产生废物,堵塞毛囊导致脱发。⑦遗传性脱发:脱发也是有遗传性的,一般男性呈显性遗传,女性呈阴性遗传。
孕妇怀孕期间,由于营养不均衡、心理压力大,以及身体营养物物质需要同时滋养胎儿等因素,可能导致孕妇怀孕期间产生脱发症状,考虑到胎儿的生理发育不完全,市面上的治疗脱发的药物或者药膳等产品可能对胎儿产生影响。
中国专利申请CN111558015A公开了一种用于治疗脱发的中药组合物,包括灵芝18~22重量份、制何首乌4~6重量份、枸杞子18~22重量份、桑椹18~22重量份、人参6~10重量份、黄芪18~22重量份、淫羊藿6~8重量份、三七8~12重量份、黄精8~12重量份、当归6~8重量份、砂仁3~5重量份。所述组合物治疗脂溢性脱发480例患者的有效率为91.2%,斑秃210例患者疗效为89.3%,效果显著,但是其用药原则明确指出“孕妇禁用”。推测可能是由于灵芝、人参、枸杞子、当归等药性过强,无法排除其对胎儿的不利影响。
中国专利申请CN105412758A公开了一种有助于脱发康复的中药组合物,按照重量组分计:包括龙胆草20-25份、生地黄15-20份、高良姜15-20份、金莲花10-15份、檀香10-15份、阿胶10-15份、鸭跖草5-10份、枇杷叶4-8份、银杏叶3-6份。100例患者治愈率为72%,有效率为98%,无效率为2%。同样的,在该产品的注意事项中明月标注了“孕妇慎用”的字样。
因此,开发针对孕妇使用的治疗或预防脱发的产品是符合市场需要的。
发明内容
本发明目的在于针对现有技术的不足,提供一种乌发生发组合物。
本发明的目的通过以下方案实现。
一种乌发生发组合物,制备原料按重量份数计,包括如下组分:
何首乌30~50份、葵花籽30~60份、胡萝卜10~40份、杏仁5~20份、无花果5~15份、和叶酸类化合物0.1~1份。
进一步地,所述组合物,制备原料按重量份数计,包括以下组分:
何首乌40份、葵花籽45份、胡萝卜25份、杏仁12份、无花果10份、和叶酸类化合物0.5份。
进一步地,所述的叶酸类化合物为叶酸、甲酰四氢叶酸、L-甲基叶酸、叶酸可药用盐、叶酸或叶酸可药用盐的活性代谢产物和可在体内代谢和/或生成叶酸的物质中的一种或几种。
进一步地,所述叶酸类化合物选自叶酸、叶酸可药用盐和L-甲基叶酸中的至少一种。

本发明还提供一种所述组合物的制备方法,包括以下步骤,
S1:将所述重量份的何首乌、葵花籽、胡萝卜、杏仁和无花果混合、粉碎得混合粉料;
S2:将混合粉料在水中浸泡,过滤得浸膏;
S3:将浸膏冷冻,冷冻后回温,加入水、纤维素酶反应得到反应液;
S4:在反应液中加入乙醇,超声处理、离心收集上清液;
S5:在上清液中加入叶酸类化合物,减压移除溶剂,干燥即得所述组合物。
进一步地,S1中,所述粉碎是粉碎至10~20目;
进一步地,S2中,按重量份数计,所述混合粉料:水=1:8~10,所述浸泡时间为12~18h。

本发明利用低温冷冻技术对细胞进行破同时配合纤维素酶对细胞壁的水解作用,充分使细胞內溶物分散在提取溶剂中。
进一步地,S4中,按重量份数计,乙醇的加入量与S3中水加入量的比值为0.3~0.5:1;所述超声温度为30-40℃,功率密度为200-300W/cm2,超声时间为30-60min;所述离心转速为2000-3000rpm,离心时间为10-20min。
本发明在酶解反应液中加入乙醇,同时配合超声协助,使反应体系中的蛋白质等物质在合适乙醇和超声协同的条件下沉淀出来,减少了与本发明无关的物质在溶剂中的溶解量。此时超声能量过高,可能会使活性成分遭到破坏,而能量过低是则可能会使蛋白质等无关成分沉淀不完全而且有效成分不能完全提取。
进一步地,S5中,所述干燥是干燥至水分含量≤3%。
本发明的优势在于:
1、克服了现有促进毛发生长的中药存在针对的使用不便的缺陷,以促进和提高人的身体机能为目标,达到促进生发、乌发、防脱发的目的,并且无副作用适合孕后妇女使用;
2、本发明的方法,将何首乌、葵花籽、胡萝卜、杏仁和无花果混合后一次性提取,简化了提取步骤,节约提取时间和成本。
3、采用本方法制作的保健品具有生发、乌发、防脱发等功能,在增强体质、生发、乌发、防脱发等方面效果显著;
4、本发明的组合物既可以补充孕妇的日常营养需求,又可以防治妇女怀孕期间可能导致的脱发问题,组成简单,成本低。
5、本发明的制备方法,冷冻、超声、酶解步骤相辅相成,共同提取出符合本发明目的的有效成分来制备本发明提供的组合物。
具体实施方式
本发明所使用的“纤维素酶”是指催化降解纤维素水解而生成葡萄糖和低聚合度纤维的一组酶的总称,包括葡聚糖内切酶、葡聚糖外切酶和纤维二糖酶3个主要组分,它不是单一种酶,而是起协同作用的多组分酶系。以下实施例所使用的纤维素酶为商业化来源,CAS号:9012-54-8。
实施例1
一种乌发生发组合物
何首乌30份、葵花籽60份、胡萝卜20份、杏仁15份、无花果10份、和叶酸0.5份。
所述组合由以下方法制备得到:
S1:将所述重量份的何首乌、葵花籽、胡萝卜、杏仁和无花果混合、粉碎至10目,得混合粉料;
S2:将混合粉料在水中浸泡15h,过滤得浸膏,按重量份数计,所述混合粉料:水=1:8;
S3:将浸膏在-10℃冷冻12h,冷冻后回温至室温,加入浸膏重量10倍的水、0.01倍的纤维素酶,在35℃反应10h,得到反应液;
S4:在反应液中加入乙醇,乙醇的加入量与S3中水加入量的重量比值为0.3:1,在30℃,功率密度为300W/cm2超声处理40min,2000rpm下离心15min,收集上清液;
S5:在上清液中加入叶酸,减压移除溶剂,干燥至水分含量为3%,即得所述组合物。
实施例2
一种乌发生发组合物
何首乌50份、葵花籽30份、胡萝卜10份、杏仁20份、无花果5份、和叶酸钙1份。
所述组合由以下方法制备得到:
S1:将所述重量份的何首乌、葵花籽、胡萝卜、杏仁和无花果混合,粉碎至20目,得混合粉料;
S2:将混合粉料在水中浸泡18h,过滤得浸膏,按重量份数计,所述混合粉料:水=1:10;
S3:将浸膏在-20℃冷冻16h,冷冻后回温至室温,加入浸膏重量8倍的水、0.05倍的纤维素酶,在45℃反应8h,得到反应液;
S4:在反应液中加入乙醇,乙醇的加入量与S3中水加入量的重量比值为0.5:1,在40℃,功率密度为250W/cm2超声处理30min,3000rpm下离心10min,收集上清液;
S5:在上清液中加入叶酸钙,减压移除溶剂,干燥至水分含量为1%,即得所述组合物。
实施例3
一种乌发生发组合物
何首乌40份、葵花籽40份、胡萝卜40份、杏仁5份、无花果15份、和叶酸甲酯0.1份。
所述组合由以下方法制备得到:
S1:将所述重量份的何首乌、葵花籽、胡萝卜、杏仁和无花果混合,粉碎至16目,得混合粉料;
S2:将混合粉料在水中浸泡12h,过滤得浸膏,按重量份数计,所述混合粉料:水=1:10;
S3:将浸膏在-15℃冷冻18h,冷冻后回温至室温,加入浸膏重量8倍的水、0.03倍的纤维素酶,在40℃反应10h,得到反应液;
S4:在反应液中加入乙醇,乙醇的加入量与S3中水加入量的重量比值为0.4:1,在40℃,功率密度为200W/cm2超声处理60min,3000rpm下离心20min,收集上清液;
S5:在上清液中加入叶酸类化合物,减压移除溶剂,干燥至水分含量为3%,即得所述组合物。

一种乌发生发组合物

何首乌20份、葵花籽55份、胡萝卜22份、杏仁30份、无花果5份、和叶酸0.5份。

何首乌40份、葵花籽45份、胡萝卜25份、杏仁12份、无花果10份、和叶酸0.5份。
所述组合由以下方法制备得到:
S1:将所述重量份的何首乌、葵花籽、胡萝卜、杏仁和无花果混合,粉碎至16目,得混合粉料;
S2:将混合粉料在水中浸泡12h,按重量份数计,所述混合粉料:水=1:10;加入混合粉料0.03倍的纤维素酶,在40℃反应10h,得到反应液;
S3:在反应液中加入乙醇,乙醇的加入量与S3中水加入量的重量比值为0.4:1,在40℃,功率密度为200W/cm2超声处理60min,3000rpm下离心20min,收集上清液;
S4:在上清液中加入叶酸类化合物,减压移除溶剂,干燥至水分含量为3%,即得所述组合物。
对比例4-和实施例4相比,超声功率密度不同

何首乌40份、葵花籽45份、胡萝卜25份、杏仁12份、无花果10份、和叶酸0.5份。
所述组合由以下方法制备得到:

所述组合由以下方法制备得到:
S1:将所述重量份的何首乌、葵花籽、胡萝卜、杏仁和无花果混合,粉碎至16目,得混合粉料;
S2:将混合粉料在水中浸泡12h,过滤得浸膏,按重量份数计,所述混合粉料:水=1:10;
S3:将浸膏在-15℃冷冻18h,冷冻后回温至室温,加入浸膏重量8倍的水、0.03倍的蛋白酶,在40℃反应10h,得到反应液;
S4:在反应液中加入乙醇,乙醇的加入量与S3中水加入量的重量比值为0.4:1,在40℃,功率密度为200W/cm2超声处理60min,3000rpm下离心20min,收集上清液;
S5:在上清液中加入叶酸类化合物,减压移除溶剂,干燥至水分含量为3%,即得所述组合物。
产品毒性试验
实验动物:SPF级雌性SD大鼠,每组5只,饲喂普通饲料(符合《GB 14924.3-2001实验动物小鼠大鼠配合饲料》)。
样品剂量:每天固定时间灌胃处理一次;
所有供试品灌胃给药,剂量为2g/kg.BW。
空白对照组灌胃不含受试品的蒸馏水。
实验周期:四周。

实验前后,所有实验组和空白对照组相比,体重增长无显著性差异(P>0.05)。说明本发明提供的中药组合物对大鼠的生长没有影响。
效果例-防脱发效果
效果例-小鼠毛发生长实验
实验动物为雌性C57BL/6小鼠,体重20-22g,实验前观察三天。实验期间各组小鼠饲养于安静环境,分笼饲养,饲喂普通饲料(符合《GB 14924.3-2001实验动物小鼠大鼠配合饲料》),自由摄食饮水。每日更换饮水和饲料,保持动物生活环境通风及清洁卫生。白天(12±2)小时光照,室内温度(20±2)℃,相对湿度40%~60%。小鼠基础饲养一周后,随机分组
在轻度麻醉下,将质量比1:1混合的松香和石蜡混合物加热融化后涂于小鼠背部,待其凝固变硬后轻轻揭去,每只小鼠的脱毛区域面积约为2×2cm2,脱毛后按体重随机分组,每组5只,给药方案为每天固定时间灌胃处理一次,剂量为2g/kg.BW。连续给药21天,第22天处死,测新生毛区域鼠毛长度及鼠毛重,每天观察各组动物毛发生长情况,实验结果见下表。

由实施例4和对比例1-2的差别可以看出,本发明各组分间存在较好的协同作用。而从实施例4和对比例3-5可以看出,本发明提供的方法可以从本发明的组合物原料中更好地提取出促进生发的有效成分。

Claims (10)
1.一种乌发生发组合物,制备原料按重量份数计,包括如下组分:
何首乌30~50份、葵花籽30~60份、胡萝卜10~40份、杏仁5~20份、无花果5~15份、和叶酸类化合物0.1~1份。
2.根据权利要求1所述的组合物,其特征在于,制备原料按重量份数计,包括以下组分:
何首乌40份、葵花籽45份、胡萝卜25份、杏仁12份、无花果10份、和叶酸类化合物0.5份。
3.根据权利要求1所述的组合物,其特征在于,所述的叶酸类化合物为叶酸、甲酰四氢叶酸、L-甲基叶酸、叶酸可药用盐、叶酸或叶酸可药用盐的活性代谢产物和可在体内代谢和/或生成叶酸的物质中的一种或几种。
4.根据权利要求3所述的组合物,其特征在于,所述叶酸类化合物选自叶酸、叶酸可药用盐和叶酸甲酯中的至少一种。
5.根据权利要求1-4任一项所述的组合物,本发明的组合物还包括至少一种药学上可接受的赋形剂;所述赋形剂选自稀释剂、粘合剂、崩解剂、润滑剂、着色剂、增味剂、pH调节剂、缓冲剂、均质剂、防腐剂、抗粘合剂、助流剂、酸化剂或甜味剂。
6.一种如权利要求1-5任一项所述的组合物的制备方法,包括以下步骤,
S1:将所述重量份的何首乌、葵花籽、胡萝卜、杏仁和无花果混合、粉碎得混合粉料;
S2:将混合粉料在水中浸泡,过滤得浸膏;
S3:将浸膏冷冻,冷冻后回温,加入水、纤维素酶反应得到反应液;
S4:在反应液中加入乙醇,超声处理、离心收集上清液;
S5:在上清液中加入叶酸类化合物,减压移除溶剂,干燥即得所述组合物。
7.根据权利要求6所述的制备方法,其特征在于,S1中,所述粉碎是粉碎至10~20目。
8.根据权利要求6所述的制备方法,其特征在于,S2中,按重量份数计,所述混合粉料:水=1:8~10,所述浸泡时间为12~18h。
9.根据权利要求6所述的制备方法,其特征在于,S3中,所述冷冻温度为-10℃~20℃,冷冻时间为12~18h;所述回温是回温至室温;按重量份数计,所述浸膏:水:纤维素酶=1:8-10:0.01-0.05;所述反应的温度为35-45℃,反应时间为8-12h。
10.根据权利要求6所述的制备方法,其特征在于,S4中,按重量份数计,乙醇的加入量与S3中水加入量的比值为0.3~0.5:1;所述超声温度为30-40℃,功率密度为200-300W/cm2,超声时间为30-60min;所述离心转速为2000-3000rpm,离心时间为10-20min;
S5中,所述干燥是干燥至水分含量≤3%。
