(II) preparing nutritious goat milk powder
The raw materials in examples 1-6 were weighed according to table 2, the specific process parameters are shown in table 3, and the preparation was carried out according to the following steps:
firstly, taking goat milk, adding compound bacteria powder and nutrition supplementing powder with corresponding dosage, dissolving, and fermenting to obtain a mixed material I;
dissolving an application amount of choline chloride, composite mineral substances and calcium carbonate into water to obtain a mixed material II;
secondly, uniformly mixing the whey protein powder, the phospholipid, the mixture I and the mixture II in the application amount, homogenizing, spray-drying, cooling, adding the compound vitamin and the compound bacteria powder in the corresponding amount, and uniformly mixing by a dry method to obtain the nutritional goat milk powder (correspondingly marked as M1-M6) suitable for the diarrhea children.
The goat milk is standardized, subjected to impurity separation, pasteurized and homogenized, and the obtained nutritional goat milk powder suitable for diarrhea children is suitable for diarrhea children, and is used for adjuvant treatment and diarrhea symptom relief.
Table 2 examples 1-6 table of raw materials of nutritious goat milk powder
Table 3 table of process parameters for examples 1-6
Example 7 Effect of nutritional goat milk powder suitable for diarrhea Children on diarrhea model mice
After a diarrhea model is successfully constructed by taking 1 month old C57BL/6 mice and adopting a method of continuously feeding salmonella for 3 days by 0.3 ml/mouse, 21 mice with diarrhea symptoms are selected, half of the mice are female and male, the mice are randomly divided into 7 groups, each group comprises 3 mice,
the contrast group is perfused with 0.2mL of purified water every morning;
experimental group MX1-MX6 stomach-drenching 0.2mL each day of the nutritional goat milk powder M1-M6 prepared in examples 1-6 suitable for diarrhea children (brewing manner: 1g milk powder dissolved in 10mL purified water, 50 deg.C warm water); the experiment lasts for 5 days, the diarrhea condition of the mice is observed every day, the stool dilution rate is calculated (the result is shown in table 4), the feces of the mice are collected in a metabolism cage, and the content of lactobacillus and bifidobacterium in the feces of each group of mice is measured (the result is shown in table 5);
wherein, the calculation formula of the rare excrement rate is as follows:
the stool dilution rate is the number of stools discharged/total stool discharge in a certain period
TABLE 4 stool dilution Rate statistics
Group of | 1 day | 2 days | 3 days | 4 days | 5 days |
Control group | 87.2% | 89.3% | 77.6% | 65.5% | 45.1% |
MX1 | 63.2% | 33.8% | 0 | 0 | 0 |
MX2 | 60.2% | 34.5% | 12.1% | 0 | 0 |
MX3 | 60.4% | 32.1% | 0 | 0 | 0 |
MX4 | 60.5% | 35.4% | 5.1% | 0 | 0 |
MX5 | 66.7% | 33.3% | 0 | 0 | 0 |
MX6 | 63.5% | 36.7% | 6.4% | 0 | 0 |
And (3) observation and statistical result display: although diarrhea is slightly relieved after 3 days, the mice in the control group still have continuous diarrhea, defecation is performed for many times every day, water-like feces are formed at the beginning, and the feces are changed into particles after 5 days; in the mice of the experimental group, diarrhea stops after 1 day, the defecation frequency is reduced, and the defecation liquid is thick; after 2 days, no diarrhea exists, and the excrement is granular; after 3 days, the normal state is recovered. The experiments show that the mice in the control group can be self-healed slowly, and the nutritional goat milk powder suitable for diarrhea children has an obvious effect of relieving diarrhea symptoms of diarrhea model mice.
Due to the difficulty in collecting water samples and extracting nucleic acid of mice during diarrhea, except salmonella, lactobacillus and bifidobacterium are hardly detected in excrement;
from day 7 after the experiment, after the mice had healed and formed their stools, the stools were taken for the determination of lactic acid bacteria and bifidobacteria, and the results are shown in table 5:
TABLE 5 mean probiotic content after self-healing of mice
Group of | Lactic acid bacteria content (Lg CFU/g) | Bifidobacterium content (Lg CFU/g) |
Control group | 6.19 | 5.22 |
MX1 | 8.79 | 6.76 |
MX2 | 8.32 | 6.74 |
MX3 | 8.22 | 6.27 |
MX4 | 8.43 | 6.35 |
MX5 | 8.92 | 6.83 |
MX6 | 7.56 | 6.14 |
As can be seen from the table 5, the numbers of the lactic acid bacteria and the bifidobacteria in the experimental group are higher than those in the control group, which shows that the nutritional goat milk powder suitable for the children with diarrhea can inhibit the growth of pathogenic bacteria, assist in curing the diarrhea, promote the colonization of intestinal probiotics, regulate the intestinal microenvironment steady state, and increase the contents of the two probiotics to improve the diarrhea symptoms.
