Description
Technical Field
The invention relates to the technical field of fish culture, in particular to a preparation for preventing and treating saprolegniasis of Tibet plateau fishes.
Background
Saprolegniasis, also called dermatomycosis, is caused by several species of the genera Saprolegnia (Saprolegnia), achlyria (achlyya), ophiocystis (Aphanomyces) and dictyosphaea (Dictyuchus) in the family saprolegniaceae (Saprolegniacae). The saprolegnia hypha is a tubular multikaryon without transverse partitions, is generally attached to the damaged part of aquatic animals like roots, has multiple branches and is fine, can penetrate into the damaged and necrotic skin and muscles to be called as inner hypha, and has the function of absorbing nutrition; the hyphae extending outside the body are coarse and have less branches, which can reach 3cm, and form a white floccule visible to the naked eye. The saprolegniasis occurs mainly because of secondary infection caused by the stress, the fish only has the influence of crowding, moving or other adverse environmental factors to cause body surface tissue injury, saprolegniasis zoospores in water are attached at random, and start to germinate on necrotic tissues to form hyphae, the hyphae can spread and invade nearby normal tissues besides parasitizing the necrotic tissues, secrete digestive ferment to decompose the covering of surrounding tissues, form sporangia at the tail end, release the zoospores into water, and spread everywhere through the water.
The ecological barrier and the safety barrier are important in China in the Tibetan region, known as the third pole of the earth. The literature data shows that the fish distributed in the area has 60 species and 13 subspecies, which belong to 3 orders, 5 families and 4 subfamilies, and 22 genera. Along with the degradation or loss of the habitat, the construction of dams and hydropower stations, the invasion of foreign species and the pollution to the environment, and the characteristics of slow growth, late sexual maturity and the like of fishes in the region, 12 threatened fishes (endangered and easily dangerous) exist, nearly 17 percent. 7 main fishes distributed in the Yalu Tibet Bujiang, and 5 fishes are in a threatened state, so that rescue and protection work for the threatened fishes in Tibet needs to be carried out urgently, and the germplasm continuity of the special fishes is ensured. The development of domestication and propagation of the Tibet plateau fishes is an effective means for preserving the germplasm resources of the Tibet plateau fishes. Water temperature for Tibetan fish culture: the water temperature is 9-20 ℃, the water temperature is in the range of the explosion of saprolegnia, and the Tibet plateau fishes are easy to cause mechanical damage in the process of fishing and transporting, so that the saprolegnia is caused to explode; in addition, wild fishes carry more parasites, and the fish body is damaged due to the breeding of the parasites, so that saprolegniasis outbreak is caused. In the breeding process of the Tibet plateau fishes, the parent is injected with an oxytocic, an agent egg and an agent sperm to cause mechanical damage to the fishes, so that the saprolegnia is bred. In the Tibetan fish culture process, as the culture fish size is increased, ponds need to be separated in time, and mechanical damage is easily caused in the transfer process, so that saprolegniasis outbreak is caused. Therefore, saprolegniasis frequently occurs in the process of domesticating and breeding the Tibet plateau fishes, and how to prevent and control the saprolegniasis of the Tibet plateau fishes becomes one of the main difficulties in the maintenance of the Tibet plateau fishes.

a preparation for preventing and treating saprolegniasis of Tibet plateau fishes comprises salt, baking soda, a sea buckthorn extract, salicylic acid and astaxanthin nanoparticles, wherein the sea buckthorn extract comprises a sea buckthorn fruit extract and a sea buckthorn leaf extract in a weight ratio of 2: 1.
Hippophae rhamnoides is a generic name of plants and fruits thereof, and is a deciduous shrub belonging to the genus Hippophae of the family Elaeagnaceae. The root, stem, leaf, flower and fruit of sea-buckthorn, especially the fruit of sea-buckthorn, contain abundant nutrient substance and biological active substance, the fruit of sea-buckthorn is rich in vitamin C, the content of fresh fruit of each hundred grams reaches 1600 milligrams, 35 times of orange, 250 times of pear, 400 times of apple, the name of king of vitamin, in addition also contain linoleic acid, protein, flavonoid substance, not only the food rich in nutrition, but also the good medicine for treating heart disease, hypertension and other diseases.
The seabuckthorn extract contains abundant active ingredients such as vitamin e, vitamin a, sod, flavone and the like, and the substances can well resist free radicals, so that the aging process of a human body becomes slower because the amount of the free radicals is small. Total Flavonoids (TFH) extracted from fructus Hippophae and folium Hippophae are a group of compounds containing 7 kinds of flavonoids, and can relieve angina pectoris and improve cardiac function and ischemic electrocardiogram. The anticancer effect of seabuckthorn fruit not only can be caused by immune mechanism or other ways, but also can directly inhibit cancer cells and block carcinogenic factors.
The effective component superoxide dismutase (SOD) of the seabuckthorn fruit has the function of removing free radicals in a human body, simultaneously can enhance the function of an immune system and regulate immunocompetent cells, is an effective immunomodulator, and has extremely obvious effects on improving the disease resistance of the human body and delaying the aging of the human body. The SOD content in the seabuckthorn fruit extract is 6 times of that of ginseng, the seabuckthorn fruit extract can block free radicals generated by peroxidation of substances in skin, can prevent premature aging of tissue and skin, can repair damaged cell tissues, can promote tissue regeneration and epithelial tissue healing, and has excellent effects on treating scald, burn, incised wound and frostbite clinically and domestically.
Folium Hippophae mainly contains nutrients and active ingredients such as flavonoids, crude fiber, polysaccharides, amino acids, vitamins, microelements, carotene, proteins, organic acids, chlorophyll, etc. The seabuckthorn leaf brass has 99 percent of flavonoid glycoside, is easier to dissolve in water than seabuckthorn fruit brass, has various pharmacological actions of increasing coronary blood flow, reducing myocardial oxygen consumption, inhibiting platelet aggregation and the like, and also has a research showing that the seabuckthorn leaf extract has the function of regulating blood fat similar to ginkgo flavonoid glycoside. The content of the water-soluble polysaccharide of the seabuckthorn leaves is 2.3-4.5%, and the water-soluble polysaccharide of the seabuckthorn can effectively remove oxygen free radicals in human bodies. The folium Hippophae contains multiple microelements such as Fe, Mg, Ca, Zn, Mo, K, etc., and has high content, magnesium and iron content higher than daily fruit and vegetable content, and carotene content of 19.1Mg/100 g.
The effective components in the seabuckthorn fruit extract and the seabuckthorn leaf extract are not completely the same, and the seabuckthorn fruit extract or the seabuckthorn leaf extract can make up the defect of singly using the seabuckthorn fruit extract or the seabuckthorn leaf extract after the seabuckthorn fruit extract and the seabuckthorn leaf extract are mixed according to a certain proportion, so that the effect of the seabuckthorn fruit extract or the seabuckthorn leaf extract is enhanced.
Salicylic acid is a plant willow bark extract, is a natural anti-inflammatory drug, and has antifungal, antipruritic and keratolytic effects in medicine. When the surface of fish body is infected with fungi, salicylic acid can also soften horny layer, when the horny layer is fallen off, hypha can also be fallen off at the same time, and in addition, salicylic acid can help other antifungal medicines to penetrate and inhibit fungi from growing.
Astaxanthin is a ketone carotenoid, is a chain-breaking antioxidant, has strong oxidation resistance, can eliminate nitrogen dioxide, sulfide, disulfide and the like, can reduce lipid peroxidation, effectively inhibit lipid peroxidation caused by free radicals, has physiological effects in various aspects such as inhibiting tumorigenesis, enhancing immunity, eliminating free radicals in vivo and the like, has good treatment effect on skin cancer caused by ultraviolet rays, has prevention and treatment effect on eye diseases caused by diabetes, and has wide application prospect in the aspects of health care products, medicines, cosmetics, food additives, aquaculture and the like. The strong antioxidant activity of astaxanthin is due to the fact that the astaxanthin can stabilize the structure of a membrane, reduce membrane permeability, limit a peroxide promoter from entering cells, and protect important molecules in the cells from oxidative damage, and meanwhile, the astaxanthin can potentially become an oxidant to induce oxidative stress. In addition, experiments show that the astaxanthin can increase the activity of antioxidant enzyme and the expression of protein, different doses of astaxanthin can obviously increase the expression of the protein of perhydrolase and superoxide dismutase in animal cells, the biological activity of the astaxanthin is obviously improved, and the substances have better antioxidant effect in vivo.
The astaxanthin nano-particles are particles with the particle size of less than 30nm developed by utilizing a nano-delivery system (NDS), so that the problems of the solubility and the stability of astaxanthin are solved, the astaxanthin nano-particles can effectively permeate skin, effective components are wrapped in nano-sized capsules, the nano-capsules are used as carriers, and the nano-capsules automatically and slowly release the effective components to skin tissues at a constant speed, so that the effective components can be maintained in effective concentration for a long time, stable effective components are obtained, and the absorption efficiency and the bioavailability are greatly increased.

TABLE 2 ingredient Table of comparative and control formulations
(2) Test of
10 groups of lasalopecuroides were raised separately at 6:00 and 7: 00 the preparation was added to 1L of water, and fed for 10 days, and the number of diseases and the mortality rate of each group were counted, and the results are shown in Table 3.
TABLE 3 comparison of results
From the above table, it can be found that the preparation prepared by the formula of the invention has an obvious cure effect on diseased rassa nojirimus nudus, probably because the hippophae rhamnoides extract contains higher SOD, the preparation has the effects of promoting tissue regeneration and epithelial tissue healing on damaged cells, and restoring the activity of diseased cells. In addition, the sea buckthorn extract and the astaxanthin can play a role in scavenging oxygen free radicals, enhance the capability of organisms in scavenging oxygen free radicals and resisting stress, enhance the resistance of fish bodies and reduce the damage of the fish bodies in stress or disease attack. In addition, the sea buckthorn extract also contains other effective components, so that the immunity of the organism is further improved. Moreover, the sea buckthorn extract is crushed into 300nm nanoparticles by a nanoscale ultrafine grinder, the astaxanthin is in the form of astaxanthin nanoparticles, and the nanoparticles are more beneficial to the effective components of the nanoparticles to penetrate through skin tissues of fish, so that the absorption efficiency and the bioavailability are greatly increased, and the saprolegniasis is more effectively prevented and treated. Salicylic acid is a natural anti-inflammatory drug, and can soften horny layer when the fish body surface is infected with fungi, so that the skin tissue can be penetrated by effective components, hypha can be simultaneously fallen off when the horny layer falls off, and in addition, the salicylic acid can help other antifungal drugs to penetrate and inhibit the growth of fungi.
The data of the preparations A-E can show that the treatment effect of simultaneously adding the seabuckthorn fruit extract and the seabuckthorn leaf extract is better than that of singly using the seabuckthorn fruit extract or the seabuckthorn leaf extract, which indicates that the seabuckthorn fruit extract and the seabuckthorn leaf extract have complementary relation and can enhance the treatment effect on saprolegniasis; and, the effect is best when the content of the seabuckthorn fruit extract is greater than that of the seabuckthorn leaf extract.
By comparing one group to four groups of data, the promotion effect exists among the sea buckthorn extract, the salicylic acid and the astaxanthin nanoparticles, and the prevention and treatment effect is reduced due to the lack of any one component; and the fact that the effect of directly using the astaxanthin and the effect of using the astaxanthin nanoparticles are also obviously different can be found by comparing the four groups, and the effect of using the astaxanthin nanoparticles is more excellent and is more beneficial to preventing and treating saprolegniasis.
The above description is only for the purpose of illustrating the preferred embodiments of the present invention and is not to be construed as limiting the invention, and any modifications, equivalents, improvements and the like that fall within the spirit and principle of the present invention are intended to be included therein.
