Abstract
The invention provides a fermentation tank sampling device and a sampling method, which comprise a sampling pipeline, a sampling head, an emptying hose and a sampler, wherein the sampling pipeline is a three-way pipeline, and one end of the sampling pipeline penetrates through the top of a fermentation tank and extends to the bottom of an inner cavity of the fermentation tank for sampling samples; one end of the sampling pipeline is connected to the fermentation tank through an emptying hose and is used for balancing the air pressure in the sampling pipeline and the fermentation tank and sucking the gas in the fermentation tank to empty the sampling pipeline; one end is connected with the sampling head; the sampler is connected with the sampling head and is used for extracting a sample of fermentation liquor in the sampling pipeline; the evacuation hose is provided with a hose clamp to control the on-off of the hose. The device and the method realize the effect of emptying the liquid in the sampling pipeline, and simultaneously, external gas does not enter the fermentation tank to be treated through the sampling pipeline, namely all liquid and gas in the sampling pipeline come from the fermentation tank and flow to the direction of the outer end through the sampling pipeline, so that the fermentation tank is ensured not to be polluted to the greatest extent while the emptying sampling pipeline is realized.

Background
There are two disadvantages to the sampling of glass fermenters at present: 1. sampling in a non-emptying mode, and retaining fermentation liquor in a sampling pipeline after sampling is finished, so that the mixed bacteria pollution is easily caused; 2. the emptying sampling is to press the fermentation liquid in the sampling pipeline back to the fermentation tank through air pressure, and is also easy to cause mixed bacteria pollution.

The technical problem to be solved by the invention is as follows: in order to overcome the defects in the prior art, the invention provides a sampling device and a sampling method for a fermentation tank.
The technical scheme adopted for solving the technical problems is as follows: a sampling device of a fermentation tank comprises a sampling pipeline, a sampling head, an emptying hose and a sampler, wherein the sampling pipeline is a three-way pipeline, and one end of the sampling pipeline penetrates through the top of the fermentation tank and extends to the bottom of an inner cavity of the fermentation tank for sampling samples in the fermentation tank; one end of the sampling pipeline is connected to the top of the fermentation tank through an emptying hose, is communicated with the upper part in the fermentation tank and is used for balancing the air pressure in the sampling pipeline and the fermentation tank and sucking the gas in the fermentation tank to empty the sampling pipeline; one end is connected with the sampling head; the sampler is connected with the sampling head and is used for extracting a sample of fermentation liquor in the sampling pipeline; and the emptying hose is also provided with a hose clamp for controlling the on-off of the emptying hose. Preferably, the sampler may be a conventional disposable screw-port syringe.
Specifically, the sampler adopts a sterile injector. Disposable syringes, which are ubiquitous in the market, are readily available and low cost.
Because the emptying hose and the hose clamp are more convenient for preparation (cleaning and sterilization) before use, the emptying hose and the hose clamp are matched to control on-off instead of a valve switch.
Specifically, the fermentation tank is a small-sized glass fermentation tank.
A sampling method of a fermentation tank comprises the sampling device and further comprises the following steps:
s1: closing a hose clamp on the emptying hose to close the emptying hose, wherein the gas at the top of the fermentation tank is not communicated with the sampling pipeline;
s2: sterilizing the outer surface of the sampling head, connecting the sampler to the sampling head, and extracting fermentation liquor in the fermentation tank through a sampling pipeline;
s3: after sampling is finished, opening a hose clamp on the emptying hose, and refluxing part of fermentation liquor in the sampling pipeline into the fermentation tank under the action of pressure difference;
s4: when the backflow is stopped, the gas in the fermentation tank is slowly pumped by a sampler, enters the sampler after passing through an emptying hose, a sampling pipeline and a sampling head, and is emptied of the fermentation liquid in the sampling pipeline between the emptying hose and the sampling head;
s5: and after the sampler is taken down, disinfecting the outer surface of the sampling head to finish sampling.
Further, depending on the amount of sampling, multiple samplings may be required, and therefore, when multiple samplings are required, a plurality of samplers are required.
In order to further reduce the risk of pollution of the glass fermentation tank in the sampling link, the outer surface of the sampling head needs to be disinfected before the sampler and the sampling head are connected each time; the outer surface of the sampling head also needs to be sterilized after each disconnection of the sampler and the sampling head. Surface disinfection is usually carried out with 75% ethanol.
The invention has the beneficial effects that: the sampling device and the sampling method for the fermentation tank provided by the invention have the advantages that the effect of emptying liquid in the sampling pipeline is realized, meanwhile, external gas cannot enter the fermentation tank to be fermented through the sampling pipeline, namely all liquid and other gas in the sampling pipeline come from the fermentation tank and flow towards the outer end through the sampling pipeline, and the fermentation tank can be ensured not to be polluted to the maximum extent while the sampling pipeline is emptied.

Detailed Description
The present invention will now be described in detail with reference to the accompanying drawings. This figure is a simplified schematic diagram, and merely illustrates the basic structure of the present invention in a schematic manner, and therefore it shows only the constitution related to the present invention.
As shown in figure 1, the sampling device of the fermentation tank comprises a sampling pipeline 5, a sampling head 6, an emptying hose 4 and a sampler, wherein the fermentation tank 1 is a small-sized glass fermentation tank 1, a stirring paddle 2 is arranged in the fermentation tank 1, and the top of the stirring paddle 2 is connected with a driving motor 3; the fermentation liquid is arranged at the lower part of the inner side of the fermentation tank 1, and the gas is arranged at the upper part; the sampling pipeline is a stainless steel three-way pipeline, three ports of the sampling pipeline are respectively P1, P2 and P3 as shown in the figure, and the common end is O, wherein the P1 end penetrates through the top of the fermentation tank 1 and extends to the bottom of the inner cavity of the fermentation tank 1 for sampling a fermentation liquid sample in the fermentation tank 1; the P2 end is connected to the top of the fermentation tank 1 through an emptying hose 4, is communicated with the upper part of the inner cavity of the fermentation tank 1 and is used for balancing the air pressure of the sampling pipeline and the fermentation tank 1 and sucking the gas in the fermentation tank 1 to empty the sampling pipeline; the end P3 is bent downwards and the end part is connected with the sampling head 6; in the embodiment, an injector is used as a sampler and is connected with a sampling head 6 for extracting a sample of fermentation liquor in a sampling pipeline; and a hose clamp 5 for controlling the on-off of the emptying hose 4 is also arranged on the emptying hose 4.
A sampling method of a fermentation tank comprises the sampling device and further comprises the following steps:
s1: the emptying hose 4 is clamped by a hose clamp 5, so that the emptying hose 4 is closed, and the gas at the top of the fermentation tank 1 is not communicated with the sampling pipeline;
s2: disinfecting the outer surface of the sampling head 6 by using 75% alcohol, connecting an injector to the sampling head 6, pulling the injector, and extracting fermentation liquor in the fermentation tank 1 through a sampling pipeline;
s3: after sampling is finished, the hose clamp 5 on the emptying hose 4 is taken off, and part of fermentation liquid in the sampling pipeline flows back into the fermentation tank 1 under the action of pressure difference;
s4: when the reflux is stopped, the gas is slowly pumped by an injector, the gas in the fermentation tank 1 enters the injector through the emptying hose 4, the P2-O-P3 section of the sampling pipeline and the sampling head 6, and the fermentation liquid in the P2-O-P3 section of the sampling pipeline is emptied; the hose clamp 5 does not need to be clamped back again after the sampling pipeline is emptied, and only needs to be used in the sampling process, and the hose clamp is not clamped at other times.
S5: after the injector is taken down, the outer surface of the sampling head 6 is disinfected to finish sampling.
Further, depending on the amount of sampling, multiple samplings may be required, and therefore, when multiple samplings are required, a plurality of samplers are required.
In order to further reduce the risk of pollution of the glass fermentation tank 1 in the sampling link, the outer surface of the sampling head 6 needs to be disinfected before the sampler and the sampling head 6 are connected each time; the outer surface of the sampling head 6 also needs to be sterilized after each disconnection of the sampler and the sampling head 6.

Claims (6)
1. A sampling device of a fermentation tank is characterized in that: the device comprises a sampling pipeline, a sampling head, an emptying hose and a sampler, wherein the sampling pipeline is a three-way pipeline, and one end of the sampling pipeline penetrates through the top of the fermentation tank and extends to the bottom of an inner cavity of the fermentation tank for sampling samples in the fermentation tank; one end of the sampling pipeline is connected to the top of the fermentation tank through an emptying hose, is communicated with the upper part in the fermentation tank and is used for balancing the air pressure in the sampling pipeline and the fermentation tank and sucking the gas in the fermentation tank to empty the sampling pipeline; one end is connected with the sampling head; the sampler is connected with the sampling head and is used for extracting a sample of fermentation liquor in the sampling pipeline; and the emptying hose is also provided with a hose clamp for controlling the on-off of the emptying hose.
2. The fermenter sampling device of claim 1, wherein: the sampler adopts a sterile injector.
3. The fermenter sampling device of claim 1, wherein: the fermentation tank is a small-sized glass fermentation tank.
4. A sampling method of a fermentation tank is characterized in that: a sampling device according to any one of claims 1 to 3, comprising the further steps of:
s1: closing a hose clamp on the emptying hose to close the emptying hose, wherein the gas at the top of the fermentation tank is not communicated with the sampling pipeline;
s2: sterilizing the outer surface of the sampling head, connecting the sampler to the sampling head, and extracting fermentation liquor in the fermentation tank through a sampling pipeline;
s3: after sampling is finished, opening a hose clamp on the emptying hose to enable the sampling pipe to be communicated with the inner cavity of the fermentation tank, and enabling part of fermentation liquor in the sampling pipeline to flow back into the fermentation tank under the action of gravity;
s4: after the backflow is stopped, the gas in the fermentation tank is slowly pumped by a sampler, enters the sampler after passing through an emptying hose, a sampling pipeline and a sampling head, and is emptied of fermentation liquor in the sampling pipeline;
s5: and after the sampler is taken down, disinfecting the outer surface of the sampling head to finish sampling.
5. The method of sampling a fermenter of claim 4, wherein: when multiple sampling is needed, the number of samplers is multiple.
6. The method of sampling a fermenter of claim 5, wherein: before connecting the sampler and the sampling head each time, the outer surface of the sampling head needs to be disinfected; the outer surface of the sampling head also needs to be sterilized after each disconnection of the sampler and the sampling head.
