The invention uses sulfonic acid functionalized tetramethylethylenediamine ionic liquid as a fermentation catalyst, has good biocompatibility and catalytic activity, can improve the fermentation environment in the fermentation process, assists and promotes microorganisms to destroy cell wall structures and extracellular polymers of biomass, reacts with a large amount of crystalline cellulose in traditional Chinese medicines, promotes the decrystallization of the cellulose, accelerates the conversion rate of cellulose sugar, and obtains the environmental condition more suitable for the growth of microorganisms. Meanwhile, the microbial inoculum is matched, and the two agents generate synergistic action, so that the fermentation efficiency is improved, the fermentation time is shortened, the gas generation is reduced, and the biological activity and the bioavailability of natural products in the fermented traditional Chinese medicine are improved. Preferably, the microbial agent comprises one or more of yeast, acetobacter xylinum, lactobacillus plantarum, lactobacillus acidophilus and lactobacillus bifidus. The microbial agent disclosed by the invention is beneficial bacteria, can adjust the clean and balanced environment in the vagina and achieves the purpose of daily maintenance of the vagina.
Lactobacillus is representative of probiotics in vagina, and Lactobacillus crispatus produces H2O2The vaginal Lactobacillus crispatus has the strongest capability, can competitively exclude the colonization of other bacteria in the vagina, and women taking the Lactobacillus crispatus as the dominant bacteria have lower proportion of the bacterial vaginosis.

Fructus Cnidii has effects of warming kidney, tonifying yang, eliminating dampness, dispelling pathogenic wind, and killing parasite. Can be used for treating sexual impotence, cold womb, leukorrhagia due to cold-dampness, lumbago due to damp arthralgia, vulval eczema, pudendal pruritus, and trichomonas vaginitis.
Centella asiatica has the effects of clearing heat, promoting diuresis, relieving swelling and removing toxicity. It is used for treating abdominal pain due to eruptive disease, summer diarrhea, dysentery, jaundice due to damp-heat pathogen, stranguria with blood, hematuria, emesis, epistaxis, hemoptysis, conjunctival congestion, larynx swelling, rubella, scabies, tinea, furuncle, carbuncle, toxic swelling, and traumatic injury.
Radix Dipsaci has antibacterial, antiinflammatory, immunity regulating, antioxidant, antiaging, bone injury healing promoting, and premature labor abortion resisting effects. Tonify liver and kidney, strengthen tendons and bones, heal fracture and stop metrorrhagia and metrostaxis. They belong to yang tonics classified under deficiency tonics.
The folium Eucommiae has tranquilizing, analgesic, blood pressure lowering, and antiinflammatory effects, and can enhance immunity and promote healing of creative fracture. Tonify liver and kidney, strengthen tendons and bones. They belong to yang tonics classified under deficiency tonics.
The guava leaf has antioxidant effect and has effect in resisting Staphylococcus aureus; has effects of eliminating dampness, invigorating spleen, clearing away heat and toxic materials, astringing, stopping bleeding, relieving inflammation, and stopping bleeding. Can be used for treating dysentery with abdominal pain, food stagnation, abdominal distention, gingival swelling and pain, rheumatalgia, eczema, ecthyma, furuncle, toxic swelling, traumatic injury, traumatic hemorrhage, and snake and insect bite.
Radix Arnebiae is dry root of arnebia euchroma (Royle) Johnst or arnebia euchroma (Royle) Johnst of Boraginaceae, and contains naphthoquinone pigment and polysaccharide compound as main ingredients, and can be released by fermentation catalysis. The lithospermum has good antibacterial, anti-tumor, antiviral, anti-inflammatory, anti-allergic, liver-protecting and enzyme-reducing effects, and is a plant with high medicinal value. The lithospermum extract has obvious bacteriostatic action on staphylococcus aureus, staphylococcus albus, pseudomonas aeruginosa, escherichia coli, typhoid bacillus, streptococcus A and streptococcus B. the pH regulator is one or more of lactic acid, sodium lactate, acetic acid, tartaric acid, disodium hydrogen phosphate, dipotassium hydrogen phosphate, sorbic acid, citric acid and malic acid; the acidity regulator used in the invention has high safety performance and low irritation, and can regulate the pH value of the product to be in a pH range beneficial to the growth of beneficial bacteria in vagina.
The nutritional humectant comprises one or more of plant collagen, yeast-beta-glucan, alpha-glucan oligosaccharide, inulin, levan and fructo-oligosaccharide, and also comprises one or more of propylene glycol, glycerol, butanediol, pentanediol, hydrogenated castor oil, betaine, polyethylene glycol-200 and polyethylene glycol-400.

s1, weighing oat, radix Sophorae Flavescentis, radix Paeoniae Rubra, Eucalyptus globulus Labill leaf, fructus Cnidii, herba Centellae, radix Dipsaci, folium Eucommiae, folium Psidii Guajavae, radix Arnebiae, Borneolum Syntheticum, radix Angelicae sinensis, and folium Nelumbinis according to formula ratio, mixing, pulverizing, and sterilizing;
s2, adding dication acidic ionic liquid and a microbial agent fermentation catalyst, adding deionized water, stirring uniformly, and fermenting;
s3, drying the fermented traditional Chinese medicine mixture, adding ethanol for extraction to obtain a traditional Chinese medicine liquid, and removing the ethanol in the traditional Chinese medicine liquid to obtain a fermented traditional Chinese medicine;
s4, mixing the obtained fermented traditional Chinese medicine with probiotics, a pH regulator, a nutrition humectant, a thickening agent, a chelating agent and deionized water to prepare the antibacterial gel.
The traditional Chinese medicine fermentation is a novel traditional Chinese medicine processing technology, changes the traditional technologies of decocting, boiling, refining, boiling and the like, and can effectively enhance the biological activity of natural products, thereby improving the medicinal health-care function of the natural products. Preferably, in step S2, the amount of the dicationic acidic ionic liquid added is 0.05-2.5% of the total mass of the traditional Chinese medicine, and the amount of the microbial agent added is 0.05-2.5% of the total mass of the traditional Chinese medicine.
Preferably, the dicationic acidic ionic liquid is added prior to or simultaneously with the microbial agent in step S2.
Preferably, step S4 specifically includes:
s41, adding the nutrient humectant of the formula amount into deionized water to prepare an aqueous solution;
s42, adding the thickening agent and the chelating agent in the formula amount into the aqueous solution, and fully swelling to prepare a matrix solution;
s43, weighing the fermented traditional Chinese medicine according to the formula amount, adding deionized water to dissolve the fermented traditional Chinese medicine, and adding the mixture into the matrix solution under the stirring condition to prepare an active solution;
s44, adding a pH regulator to regulate the pH of the active solution to 4-6, and thus obtaining the antibacterial gel.

2. Detailed Description
The technical solutions in the embodiments of the present invention will be clearly and completely described below with reference to the drawings in the embodiments of the present invention, and it is obvious that the described embodiments are only a part of the embodiments of the present invention, and not all of the embodiments. All other embodiments, which can be derived by a person skilled in the art from the embodiments given herein without making any creative effort, shall fall within the protection scope of the present invention.
The chemical reagents and laboratory equipment used in the preferred embodiment of the invention are commercially available.
The preparation process of dicationic acidic ionic liquid is shown in figure 2, and ionic liquid [ Ps2TMEDA][BF4]2Is an example.
Adding tetramethylethylenediamine and ethanol (1 mol: 1L) into a reactor respectively, and preheating at 70 ℃; 1, 3-propane sultone (2 times of the amount of the substance of tetramethylethylenediamine) was added dropwise under stirring, and after completion of the dropwise addition, stirring was continued for 8 hours to obtain a large amount of an intermediate (white solid). After the reaction is finished, the intermediate product is repeatedly washed by ether for 3-5 times to remove unreacted raw materials, and then the intermediate product is dried in a vacuum drying oven at 80 ℃ for 12 hours.
The intermediate product was added to the reactor, preheated at 70 ℃, and then a tetrafluoroboric acid solution (2 times the amount of tetramethylethylenediamine) was added dropwise to the reactor, with continued stirring for 10 h. After the reaction is finished, removing water in the reaction mixture by rotary evaporation, and finally washing the obtained product for 3-5 times by using acetone to remove the intermediate product which is possibly not reacted, thereby obtaining the ionic liquid [ Ps ]2TMEDA][BF4]2Vacuum drying at 80 deg.C for 10h gave a yield of about 92%.

s1, weighing the following traditional Chinese medicines in parts by weight according to the formula proportion: 40 parts of oat, 30 parts of radix sophorae flavescentis, 30 parts of red peony root, 25 parts of eucalyptus globulus, 25 parts of fructus cnidii, 25 parts of centella, 25 parts of teasel root, 20 parts of eucommia ulmoides leaf, 20 parts of guava leaf, 20 parts of lithospermum, 10 parts of borneol, 10 parts of angelica and 10 parts of lotus leaf, mixing, crushing and sterilizing;

The effect evaluation was performed on the bacteriostatic gels of examples 1 to 8 and comparative examples 1 to 4.

The data in Table 1 show that the formula products of the female private nursing gel have obvious antibacterial effect, the antibacterial rate to Escherichia coli, staphylococcus aureus and Candida albicans is over 93.5%, and the comparison between example 1 and example 2 shows that the dicationic acidic ionic liquid is added before the microbial agent to obtain higher antibacterial effect; the ionic liquid can destroy the crystal structure of Chinese medicine fiber to raise the dissolubility of Chinese medicine, promote the dissolution of active matter in Chinese medicine, speed the fermentation of microbial preparation and raise the available active components and bioavailability of Chinese medicine. Compared with the comparative example 1, the fermentation of the traditional Chinese medicine can greatly improve the biological activity and the utilization rate of the traditional Chinese medicine, and the bacteriostasis rate of the bacteriostasis gel is obvious; compared with the comparative example 2, the dicationic acid ionic liquid can generate a synergistic effect with the microbial agent, and the available active ingredients of the traditional Chinese medicine and the bioavailability thereof are improved, so that the bacteriostasis rate of the bacteriostasis gel is improved; compared with the comparative example 3, the addition of functional humectants such as plant collagen, yeast-beta-glucan, inulin and the like can improve the bacteriostasis rate of the bacteriostasis gel; compared with comparative example 4, the chelating agent is added to enhance the thixotropy of the gel, promote the drug effect to be exerted and improve the bacteriostasis rate of the bacteriostasis gel.
Test 2: beneficial bacteria (Lactobacillus acidophilus GDMCC1.208) growth effect test: preparation of a Lactobacillus acidophilus suspension (1.0X 10)4~9.0×104) Taking 5mL of female privacy nursing gel diluent (1:10), adding 0.1mL of acidophilic lactobacillus suspension, after acting for 2min, putting the mixture into an MSR culture medium, culturing for 12h and 24h at 37 ℃, counting viable bacterial colonies, and calculating the survival rate of lactobacillus, wherein the results are shown in Table 2. The survival rate is calculated according to the following formula:

Experiments show that the vaginal beneficial bacteria growth-promoting agent has no obvious effect on vaginal beneficial bacteria and high survival rate of vaginal beneficial bacteria, shows that the vaginal pathogenic bacteria are killed, the vaginal beneficial bacteria growth is slightly influenced, the beneficial bacteria growth can be promoted, the vaginal pathogenic bacteria growth-promoting agent is safe to use for a long time, and the vaginal beneficial bacteria growth-promoting agent has a good effect on treatment and prevention of vaginitis. Compared with comparative examples 1-4, the results show that traditional Chinese medicine fermentation, dicationic acidic ionic liquid, functional humectants such as plant collagen, yeast-beta-glucan, inulin and the like, and chelating agents all play a beneficial role in survival of beneficial bacteria, the reasons of the beneficial bacteria are probably similar to data analysis in table 1, harmful bacteria and beneficial bacteria are in a mutual competitive relationship in growth, and the survival rate of the beneficial bacteria is improved while the harmful bacteria are inhibited; wherein, the traditional Chinese medicine fermentation has the biggest influence on the survival rate of beneficial bacteria.
As can be seen from the data in Table 2, the best solution of the present invention is shown in example 2, and the contents of the components in the formula can satisfy the killing effect on harmful bacteria, promote the growth of probiotics, and maintain the normal colony balance of the vagina. The bacteriostatic gel is simple in process and beneficial to industrial production.
Finally, it should be noted that: although the present invention has been described in detail with reference to the foregoing embodiments, it will be apparent to those skilled in the art that modifications may be made to the embodiments or portions thereof without departing from the spirit and scope of the invention.

Claims (10)

1. A bacteriostatic gel, comprising: the traditional Chinese medicine fermentation product comprises a fermentation traditional Chinese medicine, a pH regulator, a nutrition humectant, a thickening agent, a chelating agent and deionized water which are subjected to fermentation catalysis by a dicationic acidic ionic liquid and a microbial agent.
2. A bacteriostatic gel according to claim 1, wherein the dicationic acidic ionic liquid is sulfonic acid-functionalized tetramethylethylenediamine ionic liquid.
3. A bacteriostatic gel according to claim 2, wherein the preparation method of the dicationic acidic ionic liquid comprises the following steps:
a. adding 1, 3-propane sultone or 1, 4-propane sultone into an ethanol solution of tetramethylethylenediamine under the condition of stirring, reacting for a period of time at 60-100 ℃, and separating the obtained solid intermediate product;
b. and (4) adding an acid solution into the solid intermediate product obtained in the step S1, stirring and reacting for a period of time at the temperature of 60-100 ℃, and removing the solvent and residual reactants in the reaction mixture to obtain the dicationic acidic ionic liquid.
4. The bacteriostatic gel according to claim 1, wherein the traditional Chinese medicines comprise: 20-50 parts of oat, 20-50 parts of radix sophorae flavescentis, 20-50 parts of red peony root, 10-40 parts of eucalyptus globulus leaf, 10-40 parts of fructus cnidii, 10-40 parts of centella, 10-40 parts of teasel root, 10-30 parts of folium cortex eucommiae, 10-30 parts of guava leaf, 10-30 parts of lithospermum, 0-20 parts of borneol, 0-20 parts of angelica sinensis and 0-20 parts of lotus leaf.
5. The bacteriostatic gel according to claim 1, wherein the microbial agent comprises one or more of yeast, acetobacter xylinum, lactobacillus plantarum, lactobacillus acidophilus and lactobacillus bifidus.
6. A bacteriostatic gel according to claim 1, characterized by further comprising at least one of the following additional technical features:
the pH regulator is one or more of lactic acid, sodium lactate, acetic acid, tartaric acid, disodium hydrogen phosphate, dipotassium hydrogen phosphate, citric acid and malic acid;
the nutritional humectant comprises one or more of plant collagen, yeast-beta-glucan, alpha-glucan oligosaccharide, inulin, levan and fructo-oligosaccharide, and also comprises one or more of propylene glycol, glycerol, butanediol, pentanediol, hydrogenated castor oil, betaine, sodium lactate, polyethylene glycol-200 and polyethylene glycol-400;
the thickening agent is one or more of methyl cellulose, carboxymethyl cellulose, hydroxyethyl cellulose, hydroxypropyl methyl cellulose, polyquaternium 1, polyquaternium 6, guar gum, xanthan gum, pectin, carrageenan, chitosan and chitosan;
the chelating agent is EDTA or EDTA disodium.
7. A bacteriostatic gel according to claim 1, wherein the pH is 4-6.
8. The preparation method of the antibacterial gel is characterized by comprising the following steps:
s1, weighing oat, radix Sophorae Flavescentis, radix Paeoniae Rubra, Eucalyptus globulus Labill leaf, fructus Cnidii, herba Centellae, radix Dipsaci, folium Eucommiae, folium Psidii Guajavae, radix Arnebiae, Borneolum Syntheticum, radix Angelicae sinensis, and folium Nelumbinis according to formula ratio, mixing, pulverizing, and sterilizing;
