Abstract
The invention discloses a bacteriostatic gel for restoring female ecological balance and a preparation method thereof, and relates to the technical field of bacteriostatic drugs. A bacteriostatic gel for restoring female ecological balance is mainly prepared from bacteriostatic materials; the bacteriostatic raw materials comprise ecological balance liquid and plant extracts, wherein the ecological balance liquid comprises the following raw materials in parts by weight: 4.5-5.5 parts of acetyl heptapeptide-4, 0.3-0.8 part of recombinant human collagen, 12-25 parts of plant active polysaccharide, 15-25 parts of plant active ingredients and 10-12 parts of purified water; the antibacterial raw materials also comprise the following raw materials in parts by weight: 0.05-0.1 part of benzalkonium bromide, 0.8-1.5 parts of pH regulator, 1-3 parts of gel matrix and 30-32 parts of purified water. The invention relates to the technical field of antibacterial drugs, in particular to an antibacterial gel for restoring female ecological balance and a preparation method thereof.
Background
The gynecological antibacterial gel has effects of dispelling pathogenic wind, removing dampness, relieving itching, killing parasite, preventing putrefaction and promoting granulation, and can be used for treating symptoms caused by vulvitis, vulval ulcer, vaginitis, etc., such as vulval or vaginal congestion, swelling, burning sensation, pain, secretion increase, topical ulcer, erosion and pruritus.

The Saviae Miltiorrhizae radix extract has effects of promoting blood circulation, removing blood stasis, dredging channels, relieving pain, clearing heart fire, relieving restlessness, cooling blood, and resolving carbuncle, and can be used for treating thoracic obstruction, heart pain, abdominal mass, heat arthralgia, vexation, insomnia, menoxenia, dysmenorrhea, amenorrhea and pyocutaneous disease with swelling and pain. The Carthami flos extract has effects of promoting blood circulation, dredging channels, removing blood stasis, relieving pain, and promoting blood circulation, removing blood stasis, and eliminating dampness. The folium Artemisiae Argyi extract can be used for treating hematemesis, epistaxis, metrorrhagia, menorrhagia, vaginal bleeding due to fetal leakage, lower abdomen psychroalgia and irregular menstruation, has excellent antibacterial effect, and can be used for treating skin pruritus.
The raw materials related to the invention are all commercially available, and the types and sources of the raw materials are shown in table 1.

The plant medicinal materials used in the plant extracts are all produced in Shaanxi and can be used after being dried.
Example 1: a preparation method of bacteriostatic gel for restoring female ecological balance comprises the following steps:
s1 dissolution: 0.08kg of benzalkonium bromide is taken, 0.8kg of purified water (the conductivity is 12 mu s/cm) is added, the mixture is stirred uniformly, and the benzalkonium bromide is dissolved to prepare the antibacterial solution.
S2 plant extraction: adding water into absolute ethyl alcohol to prepare 55 percent (mass concentration) of ethanol solution. Taking 2.8kg of dried radix sophorae flavescentis, adding 2kg of cortex phellodendri, 3.5kg of fructus cnidii, 3kg of radix salviae miltiorrhizae, 2.1kg of safflower, 3.2kg of motherwort and 2.8kg of folium artemisiae argyi, uniformly mixing to obtain a plant mixture, crushing the plant mixture by using a crusher to obtain coarse powder with the particle size of not more than 2.5mm, soaking the plant mixture for 6 days at one time by using 55% ethanol solution as a solvent according to a percolation method (Chinese pharmacopoeia) under the items of fluid extract and extract, and slowly percolating the plant mixture; filtering, adding 49.8kg ethanol solution into the solid, soaking for 4 days, slowly percolating, squeezing the residue, mixing the two extractive solutions, mixing, decolorizing with active carbon, standing for 24 hr, filtering to remove active carbon, rotary evaporating the filtrate at 60 deg.C to recover ethanol, and evaporating until the material density is 1.09g/ml (60 deg.C) to obtain plant extract.
S3 swelling: dissolving 2kg of carbomer in 18kg of purified water, standing for 11 hours, swelling the gel matrix, and uniformly stirring to obtain a swelling solution.
S4 dilution: taking 7.5kg of lavender essential oil, adding 6.5kg of pterocarpus santalinus essential oil and 6kg of purified water, adding 8kg of absolute ethyl alcohol for dilution to prepare a plant active diluent, adding 11kg of collagen peptide, 0.55kg of recombinant human collagen, 8.8kg of angelica polysaccharide, 9.2kg of schizophyllan, 5kg of acetyl heptapeptide-4 and 11kg of purified water into the plant active diluent, and uniformly mixing to prepare the ecological balance liquid.
S5 gelling: adding the antibacterial liquid prepared in the step S1, the plant extract prepared in the step S2 and the ecological balance liquid prepared in the step S4 into the swelling liquid prepared in the step S4, uniformly mixing, adding the rest 12.2kg of purified water, stirring at the rotating speed of 200 revolutions per minute, slowly adding 1.2kg of triethanolamine to adjust the pH value of the system to be 5, adding 1.1kg of Tween 80 emulsifier, uniformly mixing, and gelling to prepare the antibacterial gel for reducing the ecological balance of the female.
Example 2
Example 2 differs from example 1 in that example 2 does not add collagen peptide and otherwise remains the same as example 1.
Examples 3 to 10
Examples 3 to 10 differ from example 1 in the amount of each ingredient added in examples 3 to 10, the weight ratio of purified water used for solubilizing benzalkonium bromide to benzalkonium bromide was kept the same as in example 1, the weight ratio of purified water used for solubilizing carbomer to carbomer was kept the same as in example 1, and the other examples were kept the same as in example 1, and the amounts of each ingredient added in examples 3 to 10 are shown in Table 2.

Examples 11-14 differ from example 1 in that the process parameters for each step of examples 11-14 are different, the total purified water usage of examples 11-14 is consistent with example 1, and the process parameters for each step of examples 11-14 are shown in Table 3.

Comparative example 1 is different from example 1 in that comparative example 1 does not add acetyl heptapeptide-4 and recombinant human collagen, and the others are identical to example 1.
Performance detection
1. Benzalkonium bromide stability: the stability of benzalkonium bromide needs to be tested; the stability experiments were as follows: putting the product into a constant temperature and humidity box, standing for 90 days at 38 ℃ and 75% relative humidity; and (3) measuring the content of benzalkonium bromide in the product before and after the stability experiment, and dividing the difference value of the content of benzalkonium bromide before and after the stability experiment by the content of benzalkonium bromide before the stability experiment to obtain the reduction rate of benzalkonium bromide. The method for testing the content of benzalkonium bromide comprises the following steps: taking 200g of bacteriostatic gel, adding 50ml of distilled water and 1ml of sodium hydroxide aqueous solution with the mass concentration of 4.3%, shaking up, adding 0.4ml of bromophenol blue indicator solution (prepared from 0.1g of bromophenol blue, 3ml of 0.05mol/L sodium hydroxide aqueous solution and distilled water till 200 ml) and 10ml of trichloromethane, shaking up, titrating with 0.02mol/L sodium tetraphenylborate titration solution, shaking up forcefully until the blue color of a trichloromethane layer disappears, calculating the content of benzalkonium bromide in the bacteriostatic gel, and then calculating the reduction rate of the benzalkonium bromide. The results are shown in Table 4.
2. The bacteriostatic effect is as follows: standing the product at room temperature for 40 days, and detecting the antibacterial effect; the detection method refers to GB15979-2002 hygienic Standard for Disposable sanitary articles, and detects average inhibition rate of Escherichia coli, Staphylococcus aureus and Candida albicans after 2min, 5min, 10min and 20 min. The results are shown in tables 5-7.

The lower the reduction rate of the benzalkonium bromide is, the better the stability of the benzalkonium bromide is, and the better the antibacterial effect of the product is.
Compared with the prior art, the acetyl heptapeptide-4 and the recombinant human collagen are not added in the comparative example 1, so that the benzalkonium bromide in the prepared antibacterial gel product has poor stability and low antibacterial efficiency, and is not beneficial to product market popularization.
Comparing the experimental results of the example 1 and the comparative example 1, it can be seen that, in the process of preparing the antibacterial gel, the acetyl heptapeptide-4 and the recombinant human collagen are added simultaneously, so that the stability of benzalkonium bromide in the prepared antibacterial gel product is obviously improved, the antibacterial efficiency of the product is obviously improved, and the market popularization of the product is facilitated.
Comparing the experimental results of example 1 and example 2, the collagen peptide is not added in example 2, and the stability of benzalkonium bromide in the prepared bacteriostatic gel product is slightly reduced, and the bacteriostatic efficiency is slightly reduced, which is not beneficial to the market popularization of the product.
Compared with the embodiment 1, the addition amounts of the raw materials in the embodiments 3 to 10 are different, the process parameters in the steps of the embodiments 11 to 14 are different, the stability of benzalkonium bromide in the prepared antibacterial gel product is good, the product has excellent antibacterial effects on escherichia coli, candida albicans and staphylococcus aureus, and the market popularization of the product is facilitated.
The embodiments of the present invention are preferred embodiments of the present invention, and the scope of the present invention is not limited by these embodiments, so: all equivalent changes made according to the structure, shape and principle of the invention are covered by the protection scope of the invention.
Claims (8)
1. An antibacterial gel for restoring the ecological balance of women is characterized by being mainly prepared from antibacterial raw materials; the bacteriostatic raw materials comprise ecological balance liquid and plant extracts, wherein the ecological balance liquid comprises the following raw materials in parts by weight: 4.5-5.5 parts of acetyl heptapeptide-4, 0.3-0.8 part of recombinant human collagen, 12-25 parts of plant active polysaccharide, 15-25 parts of plant active ingredients and 10-12 parts of purified water; the plant extract is prepared by extracting a plant mixture; the plant mixture comprises the following raw materials in parts by weight: 1.5-4 parts of radix sophorae flavescentis, 1-3 parts of cortex phellodendri, 2-5 parts of fructus cnidii, 1.5-4.5 parts of salvia miltiorrhiza, 1.2-3 parts of safflower, 1.8-4.5 parts of motherwort and 2-3.5 parts of folium artemisiae argyi; the antibacterial raw materials also comprise the following raw materials in parts by weight: 0.05-0.1 part of benzalkonium bromide, 0.8-1.5 parts of pH regulator, 1-3 parts of gel matrix and 30-32 parts of purified water.

The bacteriostatic gel for restoring female ecological balance according to claim 1, wherein the ecological balance liquid comprises the following raw materials in parts by weight: 4.5-5.5 parts of acetyl heptapeptide-4, 0.45-0.65 part of recombinant human collagen, 17-20 parts of plant active polysaccharide, 15-25 parts of plant active ingredients and 10-12 parts of purified water; the plant mixture comprises the following raw materials in parts by weight: 2.5-3 parts of radix sophorae flavescentis, 1-3 parts of golden cypress, 2-5 parts of fructus cnidii, 1.5-4.5 parts of salvia miltiorrhiza, 1.8-2.4 parts of safflower, 2.8-3.5 parts of motherwort and 2-3.5 parts of folium artemisiae argyi; the antibacterial raw materials also comprise the following raw materials in parts by weight: 0.05-0.1 part of benzalkonium bromide, 1-1.3 parts of pH regulator, 1.5-2.5 parts of gel matrix and 30-32 parts of purified water.

s1 dissolution: adding purified water 8-12 times the weight of benzalkonium bromide into benzalkonium bromide, and dissolving benzalkonium bromide to obtain antibacterial solution;
s2 plant extraction: pulverizing dried plant mixture into coarse powder, soaking in 55% ethanol solution (mass fraction, water) as solvent for 5-7 days, adding 3 times of ethanol solution, and percolating according to percolation method (Chinese pharmacopoeia) under the items of fluid extract and extract; filtering, adding 3 times of ethanol solution, soaking for 3-5 days, percolating, squeezing residue, mixing extractive solutions, decolorizing with active carbon, standing for 24 hr, filtering to remove active carbon, rotary evaporating filtrate to recover ethanol, and evaporating until the material density is 1.08-1.10g/ml (60 deg.C) to obtain plant extract;
s3 swelling: dissolving gel matrix in purified water 8-10 times of the weight of the gel matrix, standing for 10-12 hr to swell the gel matrix, and stirring to obtain swelling solution;
s4 dilution: diluting the plant active ingredient with 0.3-0.5 times of ethanol to obtain plant active diluent, and adding the rest raw materials to obtain ecological balance solution;
s5 gelling: and (3) adding the bacteriostatic solution prepared in the step (S1), the plant extract prepared in the step (S2) and the ecological balance liquid prepared in the step (S4) into the swelling liquid prepared in the step (S4), uniformly mixing, adding the rest purified water, stirring, adding a pH regulator, adding a Tween 80 emulsifier which is 0.01-0.02 time of the weight of the ecological balance liquid, and uniformly mixing to prepare the bacteriostatic gel for restoring the ecological balance of the female.
