背景技术
随着社会经济发展，人们生活品质逐年提升，犬猫的家庭饲养角色已逐渐从家畜向家庭成员转变，宠物健康福利也日益受到重视。肾衰竭是指肾脏部分或全部功能丢失，剩余肾单位不足以维持机体正常代谢活动，机体很快出现脱水、贫血、呕吐、腹泻等症状，进而昏迷死亡。犬猫肾衰竭是临床常见疾病之一，发病原因复杂，如误食有毒物质、细菌感染、高血压、先天性肾发育不良等，加上药物滥用、营养过剩、加上品种宠物自身免疫力缺陷和宠物主人对于养宠的不专业，导致肾衰竭的发病率越来越高。由于犬猫对于疼痛的耐受力较强，宠物主人往往不能及时发现病情，机体出现明显症状时往往会错过最佳治疗期，这也是本病治愈率不高的原因。对于宠物肾衰竭的治疗，国内目前多采用西医方法，应用输液、血液透析、腹膜透析，联合使用抗生素进行治疗。西医治疗疗效快，效果显著，但只是单纯起到快速排毒，代替肾功能的作用，不能对已经受损的肾脏起到修复作用，长期使用会使尚有功能的肾单位逐渐丧失功能，直至病情恶化，并不能修复受损肾单位，并且治疗成本高，患宠痛苦大，一般家庭难以承担高昂的治疗费用。

为克服西医治疗的种种弊端与不足，本发明以中兽医理论为基础，结合临床和实验室研究，提供一种治疗犬猫肾衰竭的中兽药组合物。
为实现上述目的，发明以人中兽医药理论为基础，经过长期实践研究，筛选出本发明配方，其特征如下：
一种治疗犬猫急慢性肾脏衰竭的中兽药组合物，其特征在于，按照以下重量对原料药进行配比：黄芪5-50份，人参5-30份，生地黄5-30份，山茱萸5-30份，山药5-50份，茯苓5-30份，黄柏5-30份，丹参10-50份，鹿衔草10-50份，六月雪10-50份，大黄10-50份，淫羊藿10-50份，仙茅5-30份，女贞子5-30份，桑寄生5-30份，天花粉5-30份，菟丝子5-30份，仙鹤草5-30份，牡蛎10-50份，竹茹5-30份，半夏5-30份，车前子10-50份，泽泻10-50份。
一种治疗犬猫急慢性肾脏衰竭的中兽药组合物，其特征在于，所述各原料药重量配比为：黄芪20-40份，人参10-20份，生地黄10-20份，山茱萸10-20份，山药20-40份，茯苓10-20份，黄柏10-20份，丹参20-40份，鹿衔草20-40份，六月雪20-40份，大黄20-40份，淫羊藿10-30份，仙茅10-20份，女贞子10-20份，桑寄生10-20份，天花粉10-20份，菟丝子10-20份，仙鹤草10-20份，牡蛎20-40份，竹茹10-20份，半夏5-20份，车前子10-20份，泽泻10-20份。
一种治疗犬猫急慢性肾脏衰竭的中兽药组合物，其特征在于，所述各原料药优选配比为：黄芪30份，人参12份，生地黄10份，山茱萸10份，山药33份，茯苓12份，黄柏10份，丹参30份，鹿衔草30份，六月雪30份，大黄30份，淫羊藿15份，仙茅10份，女贞子10份，桑寄生10份，天花粉10份，菟丝子15份，仙鹤草15份，牡蛎30份，竹茹12份，半夏8份，车前子15份，泽泻15份。
本发明采用行业通用方法水提法制得：按所需重量份数称取各药材，加入药材总重量10～12倍量的水，煎煮两次，每次煎煮1.0小时，滤过，合并两次滤液，滤液减压浓缩至1g/ml，按所需剂型的制备工艺制成相应制剂，即得。
本发明组合物的在制药领域的应用方式为：将上述组合物为原料制备成口服液、片剂、胶囊剂、膏剂等内服剂型。
本发明的组方原理如下：
中医学认为，肾衰竭属于“溺毒”、“关格”、“癃闭”、“肾风”、“水肿”、“虚劳”、“肾劳”的范畴，治疗上多采用温补脾肾、祛湿降浊、活血化瘀之法。
本发明方中黄芪、丹参、大黄等，健脾补中，破积行瘀，利水消肿，为君药；茯苓，泽泻，六月雪等，健脾宁心，渗湿利水，为臣药；人参，半夏，山茱萸等，补益肝肾，降逆止呕，生津养血，为佐药；仙茅为使药，温补肾阳，引药入肾、脾经。多药合用，共奏温补脾肾、祛湿降浊、活血化瘀的功效。
发明相对于现有技术具有如下的优势及效果：
本发明是一种来源天然植物用于治疗犬猫急慢性肾脏衰竭的中兽药组合物，通过内服给药的方式，发挥温补脾肾、祛湿降浊、活血化瘀、益气固本的功效，多途径调节患病犬猫脾、肾等脏腑功能，安全、省时省力。有效缓解犬猫急慢性肾脏衰竭导致的氮质血症，代偿性肥大，修复肾功能，延缓肾组织纤维化，从而治疗犬猫急慢性肾脏衰竭。本发明着眼于调节机体全身状态，扶正固本，可增强动物机体免疫力和抗病能力。本方的药源丰富，价格低廉，方剂大，功效涵盖面广，可用于多种原因引起的肾衰竭。此外，本发明还可避免患宠由于血液透析，腹膜透析，输液治疗带来的痛苦，减轻西医治疗给主人带来的经济压力和心理负担。

下面结合实施例对本发明做进一步的说明，以下所述，仅是对本发明的较佳实施例而已，并非对本发明做其他形式的限制。凡是未脱离本发明方案内容，依据本发明的技术实质对以下实施例所做的任何简单修改或等同变化，均落在本发明的保护范围内。
实施例1：一种治疗犬猫急慢性肾脏衰竭的中兽药组合物，由下列药物组成：
取黄芪27份，人参10份，生地黄9份，山茱萸9份，山药30份，茯苓14份，黄柏10份，丹参28份，鹿衔草28份，六月雪25份，大黄28份，淫羊藿14份，仙茅8份，女贞子8份，桑寄生12份，天花粉12份，菟丝子14份，仙鹤草16份，牡蛎25份，竹茹14份，半夏8份，车前子14份，泽泻15份
所述中药的制备方法如下：
按所需重量份数称取各药材，加入药材总重量10～12倍量的水，煎煮两次，每次煎煮1.0小时，滤过，合并两次滤液，滤液减压浓缩至1g/ml，即得治疗犬猫急慢性肾脏衰竭的中药水煎剂。
实施例2：一种治疗犬猫急慢性肾脏衰竭的中兽药组合物，由下列药物组成：
黄芪30份，人参12份，生地黄10份，山茱萸10份，山药33份，茯苓12份，黄柏10份，丹参30份，鹿衔草30份，六月雪30份，大黄30份，淫羊藿15份，仙茅10份，女贞子10份，桑寄生10份，天花粉10份，菟丝子15份，仙鹤草15份，牡蛎30份，竹茹12份，半夏8份，车前子15份，泽泻15份。
所述中药的制备方法如下：
按所需重量份数称取各药材，加入药材总重量10～12倍量的水，煎煮两次，每次煎煮1.0小时，滤过，合并两次滤液，滤液减压浓缩至1g/ml，即得治疗犬猫急慢性肾脏衰竭的中药水煎剂。
实施例3：一种治疗犬猫急慢性肾脏衰竭的中兽药组合物，由下列药物组成：
黄芪25份，人参12份，生地黄10份，淫羊藿15份，仙茅10份，女贞子13份，桑寄生17份，天花粉14份，菟丝子17份，仙鹤草17份，山茱萸16份，山药38份，茯苓14份，黄柏12份，丹参25份，鹿衔草27份，六月雪21份，大黄27份，牡蛎33份，竹茹12份，半夏16份，车前子16份，泽泻14份。
所述中药的制备方法如下：
按所需重量份数称取各药材，加入药材总重量10～12倍量的水，煎煮两次，每次煎煮1.0小时，滤过，合并两次滤液，滤液减压浓缩至1g/ml，即得治疗犬猫急慢性肾脏衰竭的中药水煎剂。
一、治疗犬猫急慢性肾脏衰竭实验室试验
将上述中药在实验室进行效果试验。
(一)急性肾衰竭效果试验：
1.试验材料：
动物：选用32只SD雄性大鼠，体重200±50g，购自北京维通利华实验动物技术有限公司。
药物：选取实施例1、实施例2制得的中药水煎剂对应作为药液1、药液2。
2.试验方法：
取健康SD雄性大鼠32只，分成四组，分别为空白组、模型组、药液1组，药液2组。
大鼠禁水24h后，取50％甘油溶液按照10mL/kg体重，对除空白组外其余三组大鼠两侧后肢肌肉注射，制备急性肾脏衰竭模型，2天后正式进行试验。

肾脏衰竭后，代谢废物和毒素不能及时排除，血液中CREA和UREA会显著升高，引发氮质血症。
表2可以看出，与空白组相比，模型组血清UREA和CREA含量显著升高，且差异明显(P<0.05)，说明造膜成功；使用了药液1和药液2后血清UREA和CREA含量有所下降，说明实施例1和实施例2均可以治疗急性肾脏衰竭引起的氮质血症，且药液2效果更佳。
(二)慢性肾衰竭实验室试验
1.试验材料：
动物：选用32只Wistar雄性大鼠，体重200±20g。购自北京维通利华实验动物技术有限公司。
药物：选取实施例1和2制得的中药水煎剂对应作为药液1和2
2.试验方法：
取健康Wistar雄性大鼠32只，分成四组，分别为空白组、模型组、药液1组，药液2组。除空白组外，其余三组用2.5％腺嘌呤混悬液按照200mg/kg灌胃，连续21天，制备慢性肾脏衰竭模型。

表4可以看出，与空白组相比，模型组血清UREA和CREA含量显著升高，且差异明显(P<0.05)，说明造模成功。使用了药液1和药液2后血清UREA和CREA含量有所下降，药液1与药液2对比差异不明显(P>0.05)，药液1与模型组对比差异明显(P<0.05)，药液2与模型组对比差异明显(P<0.05)，说明实施例1，实施例2对于慢性肾脏衰竭引起的氮质血症，均有良好的治疗效果。

表5可以看出，与空白组相比，模型组肾脏重量和脏器均显著升高，且差异明显(P<0.05)，说明造模成功；使用了药液1和药液2后脏器系数均降低，药液1与药液2对比模型组均差异明显(P<0.05)，说明实施例1和2能够抑制慢性肾脏衰竭引起的器官肿大，维持肾脏正常生理结构。
二、治疗犬猫急慢性肾脏衰竭临床试验
将上述实施例1-3进行临床效果对比试验。
1.供试地点：河北廊坊，涿州，张家口某宠物连锁机构
2.试验动物：70只宠物犬，年龄5-13岁；39只猫，年龄6-12岁。
3.临床症状：患病犬猫食欲不振，畏寒，可视粘膜发白，口臭，腹泻，经血液生化，血常规，尿液检测判断为肾衰竭，急性肾衰竭犬37只；慢性肾衰竭犬33只，猫39只。
4.试验剂型：按照实施例1-3制备方法，将实施例1制成药液1；实施例2制成药液2，实施例3制成药液3。
5.疗效判定标准：
(1)痊愈：食欲恢复正常，贫血症状消失，无口臭；经生化，血常规，尿液检测判定各项指标恢复正常
(2)有效：食欲有所恢复，贫血症状减轻，口臭现象减轻；生化，血常规，尿液检测判定各项指标有恢复，但不如正常值。
(3)无效：食欲不振，贫血状态未改善甚至加重，口臭现象未改善甚至加重；生化，血常规，尿液检测判定各项指标无恢复甚至更加严重。
6.急性肾衰竭试验方法：
将急性肾衰竭犬分为4组，分别为急性犬1，急性犬2，急性犬3组，对照组；每组分别为9只，10只，10只，8只。

由表7可以看出，在治疗犬急性肾衰竭的效果上，本发明具有极高的有效率。
7.慢性肾衰竭试验方法：
将慢性肾衰竭犬分为3组，分别为慢性犬1，慢性犬2，慢性犬3，犬对照组；每组分别为8只，9只，9只，7只。慢性肾衰竭猫分为3组，分别为慢性猫1，慢性猫2，慢性猫3，猫对照组；每组分别为10只，10只，10只，9只。

由表9可以看出，在治疗犬猫慢性肾衰竭的效果上，本发明拥有很高的有效率。
以上试验结果表明，本发明中的中兽药组合物对于犬猫急慢性肾衰竭具有较好的治疗效果。

1.一种治疗犬猫急慢性肾脏衰竭的中兽药组合物，其特征在于，按照以下重量对原料药进行配比：黄芪5-50份，人参5-30份，生地黄5-30份，山茱萸5-30份，山药5-50份，茯苓5-30份，黄柏5-30份，丹参10-50份，鹿衔草10-50份，六月雪10-50份，大黄10-50份，淫羊藿10-50份，仙茅5-30份，女贞子5-30份，桑寄生5-30份，天花粉5-30份，菟丝子5-30份，仙鹤草5-30份，牡蛎10-50份，竹茹5-30份，半夏5-30份，车前子10-50份，泽泻10-50份。
2.根据权利要求1所述的治疗犬猫急慢性肾脏衰竭的中兽药组合物，其特征在于，所述各原料药重量配比为：黄芪20-40份，人参10-20份，生地黄10-20份，山茱萸10-20份，山药20-40份，茯苓10-20份，黄柏10-20份，丹参20-40份，鹿衔草20-40份，六月雪20-40份，大黄20-40份，淫羊藿10-30份，仙茅10-20份，女贞子10-20份，桑寄生10-20份，天花粉10-20份，菟丝子10-20份，仙鹤草10-20份，牡蛎20-40份，竹茹10-20份，半夏5-20份，车前子10-20份，泽泻10-20份。

4.根据权利要求1～3中任一所述的治疗犬猫急慢性肾脏衰竭的中兽药组合物，其特征在于，所述的原料药制备方法如下：按所需重量份数称取各药材，加入药材总重量10～12倍量的水，煎煮两次，每次煎煮1.0小时，滤过，合并两次滤液，滤液减压浓缩至相对密度1.10～1.20(65℃)，按所需剂型的制备工艺制成相应制剂，即得。
5.根据权利要求1～4中任一所述的中兽药组合物在制备治疗犬猫急慢性肾脏衰竭的药物中的应用。
