As the most preferable scheme, the raw material formula of the coupling gel comprises the following components in percentage by weight: 0.15% of lysozyme, 0.5% of antibacterial peptide, 2% of cypress extract, 2% of sophora flavescens extract, 5% of propylene glycol, 0.2% of citric acid, 50.01% of vitamin B, 2% of aqueous rheology regulator and the balance of purified water.
The medical ultrasonic special sterilization coupling gel provided by the invention mainly adopts natural plant components and bioactive substances for sterilization and disinfection, is harmless to human bodies, high in safety in the using process, good in solubility, convenient to prepare and excellent in sterilization effect on various common germs. As shown in the examples, under the raw material formula of the invention, the obtained disinfection coupling gel is colorless or light yellow transparent gel, has no bubbles and insoluble foreign matters, and does not generate layering, enzyme change and peculiar smell when the product is stored for up to one month under normal storage conditions; it has a killing effect on mould and yeast less than 10CFU/mg/g, staphylococcus aureus, pseudomonas aeruginosa and beta hemolytic streptococcus; when the formula of the invention is not adopted, as shown in a comparative example, the sterilization and disinfection effects are obviously reduced.
Further, the enzyme activity of the lysozyme is not less than 20000U/mg.
Further, the antibacterial peptide is food-grade epsilon-polylysine, and the purity of the antibacterial peptide is more than 99.0%.
Furthermore, the contents of the effective components in the hinoki extracting solution and the radix sophorae flavescentis extracting solution are both more than 10%.
Further, the aqueous rheology modifier is a food grade guar gum.
The invention also aims to provide a preparation method of the medical ultrasonic special disinfection coupling gel, which comprises the following steps:
(1) adding an aqueous rheological regulator into a proper amount of purified water according to a raw material formula, and stirring for 1h to obtain transparent gel for later use;
(2) according to the formula of the raw materials, lysozyme, antimicrobial peptide, citric acid, vitamin B5 and the balance of purified water are taken, stirred for 1h at room temperature, after full dissolution, 1, 2-propylene glycol, hinoki and sophora flavescens extract PST are added into the mixture, stirred for 1h, mixed uniformly, added into the obtained substance in the step (1), continuously stirred for 30min, and after uniform dissolution, transparent colloid is obtained, namely the medical ultrasonic special disinfection coupling gel.
Further, the pH value of the transparent colloid obtained in the step (2) is 5.5-8.
Further, the mass percentage of the appropriate amount of purified water in the step (1) in the total amount of purified water is 60-80%.
The preparation method is simple, the operation is controllable, complex equipment is not needed, and the special disinfection couplant prepared by the method can well meet the standard.
Compared with the prior art, the invention has the following beneficial effects:
(1) the raw materials of the medical ultrasonic special disinfection coupling gel provided by the invention are mainly natural plant components and bioactive components, the sterilization effect is excellent, the use is safe and reliable, and the raw material components are easy to obtain;
(2) the preparation method of the medical ultrasonic special disinfection coupling gel provided by the invention has the characteristics of simple preparation process, convenience in operation and no need of complex equipment.
Detailed Description
In order to make the objects, technical solutions and advantages of the present invention more clearly understood, the present invention is described in detail below with reference to the following embodiments, and it should be noted that the following embodiments are only for explaining and illustrating the present invention and are not intended to limit the present invention. The invention is not limited to the embodiments described above, but rather, may be modified within the scope of the invention.
Example 1
A medical ultrasonic special disinfection coupling gel comprises the following raw materials in percentage by weight: 0.25% of lysozyme, 0.3% of antibacterial peptide (food grade epsilon-polylysine), 1% of hinoki extracting solution (the content of active ingredients is more than 10%), 1% of radix sophorae flavescentis extracting solution (the content of active ingredients is more than 10%), 3% of 1, 2-propylene glycol, 0.1% of citric acid, 50.005% of vitamin B, 1% of aqueous rheology modifier (guar gum) and the balance of purified water.
The preparation method comprises the following steps:
(1) according to the formula of the raw materials, taking purified water accounting for 60 percent of the total mass fraction, adding an aqueous rheological regulator into the purified water, and stirring for 1 hour to obtain transparent gel for later use;
(2) according to the formula of the raw materials, lysozyme, antibacterial peptide, citric acid, vitamin B5 and the balance of purified water are taken, stirred for 1h at room temperature, after full dissolution, 1, 2-propylene glycol, a cypress extract and a sophora flavescens extract are added into the mixture, stirred for 1h, mixed uniformly, added into the obtained substance in the step (1), continuously stirred for 30min, after uniform dissolution, a transparent colloid is obtained, and the pH value is adjusted to be 6-7, so that the medical ultrasonic special disinfection coupling gel (original disinfectant) is obtained.
Example 2
A medical ultrasonic special disinfection coupling gel comprises the following raw materials in percentage by weight: 0.12% of lysozyme, 0.4% of antibacterial peptide (food grade epsilon-polylysine), 2% of cypress extract (active ingredient content is 15%), 2% of sophora flavescens extract (active ingredient content is 20%), 4% of 1, 2-propylene glycol, 0.15% of citric acid, 50.008% of vitamin B, 3% of aqueous rheological regulator (guar gum) and the balance of purified water.
The preparation method comprises the following steps:
(1) according to the formula of the raw materials, taking purified water accounting for 80% of the total mass fraction, adding an aqueous rheological regulator into the purified water, and stirring for 1 hour to obtain transparent gel for later use;
(2) according to the formula of the raw materials, lysozyme, antibacterial peptide, citric acid, vitamin B5 and the balance of purified water are taken, stirred for 1h at room temperature, after full dissolution, 1, 2-propylene glycol, a cypress extract and a sophora flavescens extract are added into the mixture, stirred for 1h, mixed uniformly, added into the obtained substance in the step (1), continuously stirred for 30min, after uniform dissolution, a transparent colloid is obtained, and the pH value is adjusted to be 6-7, so that the medical ultrasonic special disinfection coupling gel (original disinfectant) is obtained.
Example 3
A medical ultrasonic special disinfection coupling gel comprises the following raw materials in percentage by weight: 0.15% of lysozyme, 0.5% of antibacterial peptide (food grade epsilon-polylysine), 2% of cypress extract (20% of effective component content), 2% of sophora flavescens extract (20% of effective component content), 5% of 1, 2-propylene glycol, 0.2% of citric acid, 50.01% of vitamin B, 2% of aqueous rheology modifier (guar gum) and the balance of purified water.
The preparation method comprises the following steps:
(1) according to the formula of the raw materials, taking purified water accounting for 75% of the total mass fraction, adding an aqueous rheological regulator into the purified water, and stirring for 1h to obtain transparent gel for later use;
(2) according to the formula of the raw materials, lysozyme, antibacterial peptide, citric acid, vitamin B5 and the balance of purified water are taken, stirred for 1h at room temperature, after full dissolution, 1, 2-propylene glycol, a cypress extract and a sophora flavescens extract are added into the mixture, stirred for 1h, mixed uniformly, added into the obtained substance in the step (1), continuously stirred for 30min, after uniform dissolution, a transparent colloid is obtained, and the pH value is adjusted to be 7-8, so that the medical ultrasonic special disinfection coupling gel (original disinfectant) is obtained.
Comparative example 1
Except for lysozyme which was not added, the raw material formulation and the preparation process of the reference example 1 were used to obtain a first sterile coupled gel control.
Comparative example 2
Except for the antibacterial peptide which is not added, the raw material formula and the preparation process are the same as those of the raw material formula and the preparation process of the reference example 2, and a second disinfection coupling gel control sample is obtained.
Comparative example 3
A third sterile coupled gel control was obtained according to the raw material formulation and preparation process of example 3, except that no aqueous rheology modifier was added.
Test example
The performance of the samples obtained in the examples 1-3 is detected, 100ml of gel is taken from each bottle, and 100 bottles are comprehensively tested, so that the samples well meet the industrial standard of medical ultrasonic couplant YY 0299-2016. The shape and appearance of the product are observed, the product is colorless or light yellow transparent gel, has no bubbles and insoluble foreign matters, and does not generate layering, enzyme change and peculiar smell when being stored for one month under normal storage conditions. The viscosity index at 25 ℃ is 60 pas or more, and exceeds 50 pas specified by the standard by 10 pas.
The samples obtained in examples 1 to 3 were subjected to performance testing according to the following criteria in terms of "Disinfection technical Specification" (2002 edition), "cosmetic safety technical Specification" (2015 edition), "hygienic requirements for skin disinfectants" GB 27951-2011:
1. the lead content of the disinfectant (a special disinfection coupling gel sample) is less than 1.5mg/kg, the mercury content is less than 0.001mg/kg, and the arsenic content is less than 0.01mg/kg, which all accord with the regulation of GB 27951-2011.
2. The total colony number of the disinfectant is less than 10CFU/mg/g, the mold and yeast are less than 10CFU/mg/g, staphylococcus aureus, pseudomonas aeruginosa and beta hemolytic streptococcus are not detected, and all the bacteria meet the regulation of GB 27951-2011.
3. The 0.03mol/L PBS solution containing 3% lecithin, 3% Tween-80, 1% glycine and 1% histidine is used as a neutralizer to effectively neutralize the killing effect of the disinfectant on staphylococcus aureus and candida albicans on a slide.
Respectively acting the disinfectant on Staphylococcus aureus, Pseudomonas aeruginosa, Escherichia coli and Candida albicans on the slide for 5min, wherein the average killing logarithm value is more than 3; meets the regulation of disinfection technical specification (2002 edition). The disinfectant can act on natural bacteria on skin for 5min, and has average killing log value of more than 1, and meets the specification of disinfection technical Specification (2002 edition). After the disinfectant is stored for 90 days at 37 ℃, the average killing log value of the disinfectant on various bacteria on a slide is more than 3, the disinfection performance is stable, and the disinfectant can be effectively stored for 24 months.
The acoustic characteristics of the original disinfectant obtained in the embodiment 1-3 are detected, the detection standard is according to the medical industry standard YY0299-2016 medical ultrasonic couplant, a CSC-1 type ultrasonic material acoustic performance testing device is adopted as a detection instrument, and the detection conditions are as follows: the water temperature is 35 ℃, the test frequency is 2.183MHz, the sound velocity of the original disinfectant is 1557m/s, and the acoustic characteristic impedance is 1.573 multiplied by 106Pa.s/m, sound attenuation coefficient slope 0.065 dB/(cm.MHz), and meets the requirements of YY0299-2016 medical ultrasonic couplant.
The disinfectant obtained in examples 1 to 3 was evaluated as it is according to the "Disinfection technical Specification" 2002 edition of the Ministry of health, and the results were as follows: acute oral LD of KM mice with the disinfectant as it is50Bw is more than 5000mg/kg, which belongs to actual nontoxic class; the eye cream has no irritation to the intact skin and eyes of rabbits; the result of the micronucleus test on mouse marrow pleochromocyte is negative.
The control samples obtained in the control examples 1-3 are tested for the bactericidal performance, and the bactericidal effect is far inferior to that of the original disinfectant, the bactericidal effects on staphylococcus aureus, pseudomonas aeruginosa, escherichia coli and candida albicans can not reach the standard specification, and the average killing logarithm values are all less than 1. The acoustic velocity of the disinfectant of comparative example 1-3 was measured to be 1005-1072 m/s, and the acoustic characteristic impedance (2.687-3.672) × 105Pa s/m, the slope of the acoustic attenuation coefficient is 0.13-0.28 dB/(cm MHz), and the standard requirement cannot be met.
Claims (10)
1. The medical ultrasonic special disinfection coupling gel is characterized in that the raw material formula of the coupling gel comprises the following components in percentage by weight: 0.05-0.25% of lysozyme, 0.3-0.6% of antibacterial peptide, 1-6% of cypress extract, 1-6% of sophora flavescens extract, 3-7% of 1, 2-propylene glycol, 0.1-0.4% of citric acid, 0.02% of vitamin B50.005, 1-5% of aqueous rheology regulator and the balance of purified water.
2. The medical ultrasound special sterilization coupling gel according to claim 1, wherein the raw material formula of the coupling gel comprises, by weight: 0.12-0.21% of lysozyme, 0.4-0.6% of antibacterial peptide, 2-5% of a cypress extract, 2-5% of a sophora flavescens extract, 4-6% of 1, 2-propylene glycol, 0.15-0.25% of citric acid, 50.008-0.012% of vitamin B, 1.5-3.5% of an aqueous rheology modifier, and the balance of purified water.

The medical ultrasound special sterilization coupling gel according to claim 2, wherein the raw material formula of the coupling gel comprises, by weight: 0.15% of lysozyme, 0.5% of antibacterial peptide, 2% of cypress extract, 2% of sophora flavescens extract, 5% of 1, 2-propylene glycol, 0.2% of citric acid, 50.01% of vitamin B, 2% of aqueous rheology modifier guar gum and the balance of purified water.
4. The medical ultrasound-specific disinfecting coupling gel according to any one of claims 1 to 3, characterized in that the enzymatic activity of lysozyme is not less than 20000U/mg.
5. The medical ultrasound special sterilization coupling gel according to any one of claims 1 to 3, wherein the antibacterial peptide is food grade epsilon-polylysine with a purity of 99.0% or more.
6. The disinfection coupling gel special for medical ultrasound as claimed in any one of claims 1-3, wherein the contents of effective components in the extracted solutions of Chamaecyparis obtusa and Sophora flavescens are both greater than 10%.
7. The medical ultrasound-specific disinfecting coupling gel of any one of claims 1 to 3, wherein the aqueous rheology modifier is a food grade guar.
8. A method for preparing the medical ultrasonic special sterilization coupling gel as claimed in any one of claims 1 to 7, which comprises the following steps:
(1) adding an aqueous rheological regulator into a proper amount of purified water according to a raw material formula, and stirring for 1h to obtain transparent gel for later use;
(2) according to the formula of the raw materials, lysozyme, antimicrobial peptide, citric acid, vitamin B5 and the balance of purified water are taken, stirred for 1h at room temperature, after full dissolution, 1, 2-propylene glycol, a cypress extract and a sophora flavescens extract are added into the mixture, stirred for 1h, mixed uniformly, added into the obtained substance in the step (1), continuously stirred for 30min, and after uniform dissolution, transparent colloid is obtained, namely the medical ultrasonic special disinfection coupling gel.
9. The method according to claim 8, wherein the transparent colloid obtained in the step (2) has a pH of 5.5 to 8.
10. The preparation method according to claim 8, wherein the appropriate amount of purified water in the step (1) accounts for 60-80% by mass of the total amount of purified water.
