便秘是一种常见慢性消化道疾病,该疾病不仅会引起腹胀、腹痛、呕吐、头痛、面部粉刺和雀斑、烦躁不安、精神不振等症状,而且还与高血压、高血脂、肝脏疾病、乳腺癌、大肠癌、痔疮、肛裂等疾病有着很大的相关性,严重影响患者的情绪和生活质量。随着社会的发展、经济的提升、科技的进步及人们饮食起居等生活习惯的改变,尤其是转基因食品、深加工食品逐渐代替部分纯天然食物,使便秘的发生率逐年上升。据统计,其人群发病率高达27%。目前,西医临床改善便秘的药物可分为调节肠道的微生物制剂,泻剂、促动力药等几大类。这些药物均有较大副作用,不宜久服,需要在医生的指导下使用。中医临床采用单验方及中成药治疗便秘有着悠久的历史,中医治疗强调辨证论治,采用通里攻下药如大黄、番泻叶等,效果显著,但也不可长期使用,容易产生依赖性。由于便秘的发病机制复杂、药物治疗副作用大,市售保健品良莠不齐,普遍存在功能不确切或添加致泻成分的问题,因此,采用天然食品或药食同源的中药材为原料进行组方,开发出功能确切,毒副作用小的产品具有重要意义。
发明内容
本发明的一个目的是提供一种具有降脂和通便功效的药物组合物,本发明的另一个目的是提供一种该组合物的制备方法。
上述目的是通过以下技术方案实现的:
一种药物组合物,它由以下重量配比的原料制成:昆布30-80%、胖大海5-30%、亚麻子油10-40%。
优选地,所述原料的重量配比是:昆布60-80%、胖大海5-10%、亚麻子油10-30%。
进一步优选地,所述原料的重量配比是:昆布75%、胖大海5%、亚麻子油20%。
一种制备所述药物组合物的方法,包括以下步骤:按重量配比取昆布、胖大海、亚麻子油,将昆布粉碎成微粉,胖大海按重量加入8-12倍水煎煮提取2-3次,每次0.5-2小时,滤过,合并滤液并浓缩成稠膏,然后将稠膏、亚麻子油与昆布微粉混匀,制成药物。
优选地,在将所述昆布粉碎成微粉前还对其进行了脱腥处理,处理方法是将昆布用脱腥液进行加热浸泡,所述脱腥液由陈皮、茉莉花茶和食醋制成,制备方法如下:将陈皮与茉莉花茶按3-8:1-5的重量比混合,然后加入3-6倍重量的水煎煮0.5-2小时,过滤,向滤液中按重量加入1-5%的食醋,即得。
进一步优选地,所述脱腥液的制备方法如下:将陈皮与茉莉花茶按5:3的重量比混合,然后加入5倍重量的水煎煮1.5小时,过滤,向滤液中按重量加入2.5%的食醋,即得。
进一步优选地,所述加热浸泡的温度是60-80℃,时间是10-30min。
该药物组合物能加快小肠运动,改善小肠蠕动功能,促进排便,同时还能降低高脂动物模型的血清TC值和TG值,增加HDL-C值,因此可用于制备降脂和通便药物。
该药物组合物含有膳食纤维、多糖、α-亚麻酸等营养要素。膳食纤维具有促进肠道的推进作用,多糖成分促进肠液分泌而避免粪便干结,油脂对肠壁具有润滑作用,另外还含有具有清热通便的功能成分。该组合物还能减少脂肪与胆固醇的吸收及增加体内脂质排泄,借助其食用后的饱胀感可减少高热量食物纳入,因此具有降脂作用。
原料昆布来源为翅藻科植物,富含丰富的膳食纤维、甘露醇、多糖等成分,其所含膳食纤维既能促进胃肠蠕动、增加消化液的分泌,起到润肠通便的作用,又能整合胆固醇、减少胆固醇的吸收,对高脂血症和动脉硬化起到预防作用。原料胖大海系梧桐科植物胖大海的干燥种子,为药食两用中药材品种,性甘、寒,归肺、大肠经,具有清热润肺等作用,现代研究发现,该植物脂肪酸成分中含有亚油酸、油酸、棕榈油酸、10-十九烯酸和8-壬炔酸等,其中亚油酸是人体必需的脂肪酸,是人体组织、细胞的组成成分,亚油酸与脂类代谢和胆固醇代谢密切相关,能降低血脂和胆固醇、减少动脉粥样硬化、抑制癌细胞生长和促进大脑发育。原料亚麻子油富含α-亚麻酸,具有抗炎、抗血栓、降血脂、降低血粘度、促进智力发育、预防脑萎缩等多种保健功效,在食品、医药、化工等领域有着极为广泛的应用。
本发明的有益效果是:
体内动物实验证明,昆布、胖大海和亚麻子油三种原料经复方配伍后的疗效,优于单一原料降血脂和润肠通便的作用,说明三者经配伍组合使用后,发挥了很好的协同作用。同时这三种原料均来源于可直接食用的天然产物,具有很高的安全性,在最大可服用剂量下未发现毒副作用,也未能测出半数致死剂量LD50。因此,该组合物是一个比较温和安全的降血脂及润肠通便的药物。试验结果表明,该药物组合物能加快小肠运动,改善小肠蠕动功能,促进排便,从而缓解便秘,另外还能降低高脂动物模型的血清TC值和TG值,增加HDL-C值,说明具有降脂功效,从而能从整体上改善高脂血症及功能性便秘,且疗效确切,副作用小。
具体实施方式
下面通过具体实施例对本发明进行详细说明。
实施例1处方筛选试验
实施例1-6的原料配方见表1。

将昆布、胖大海干燥后粉碎成细粉,然后加入亚麻子油搅拌均匀,用于以下试验。
1.通便实验
造模方法:80只昆明种小白鼠(雄性,体重18-22g)随机分为8组:空白组、模型组、实施例1-6组,每组10只。小鼠分组后,各实施例组每日按2.08g/kg的剂量灌胃给予相应药物,空白组和模型组给予蒸馏水,连续灌胃14天,期间小鼠正常饮水进食和活动,最后一次给予受试物后,小鼠禁食不禁水16h,模型组和实施例组灌胃给予复方地芬诺酯(5mg/kgBW),空白组灌胃给予蒸馏水。
1.1小肠运动实验
给予复方地芬诺酯30min后,各剂量组分别灌胃给予含相应受试样品的墨汁(含5%的活性炭粉、10%的阿拉伯树胶),空白组和模型灌胃给予墨汁。25min后立即脱颈椎处死小鼠,解剖打开腹腔分离肠系膜,剪取上端自幽门、下端至回盲肠的肠管,置于托盘上,轻轻将小肠拉成直线,测量肠管长度为“小肠总长度”,从幽门至墨汁前沿为“墨汁推进长度”。按下式计算墨汁推进率:墨汁推进率(%)=墨汁推进长度(cm)/小肠总长度(cm)×100%,结果见表1。

从以上结果可以看出,(1)与空白组相比,模型组的墨汁推进率极显著降低(P<0.01),表明造模成功;(2)与模型组相比,实施例1-6组的墨汁推进率均有所增加,其中实施例3、4具有极显著性差异(P<0.01),实施例1、2、5、6具有显著性差异(P<0.05);(3)同时与空白组相比,实施例3没有明显差异(P>0.05),实施例1、4、5有显著性差异(P<0.05),实施例2、6有极显著性差异(P<0.01),以上结果表明,六个实施例1中实施例3对提高模型小鼠的小肠运动效果最好,其次是实施例4,再次是实施例1和5,实施例2和6效果最差。
1.2小鼠排便实验
给予复方地芬诺酯30min后,各实施例组分别灌胃给予含相应受试样品的墨汁(含5%的活性炭粉、10%的阿拉伯树胶),空白组和模型组灌胃给予墨汁,小鼠均单笼饲养,正常饮水进食。从灌墨汁开始,观察并记录小鼠首粒排黑便时间、6h内排黑便重量,结果见表2。

从以上结果可以看出,与空白组相比,模型组的首粒排黑便时间大幅延长,6h内排便重量大幅减少,表明造模成功;与模型组相比,实施例1-6的排便时间均有所缩短,排便量有所增加,其中,实施例3的排便时间和排便量均有极显著性差异(P<0.01)。
以上结果表明,本发明组合物能加快小肠运动,改善小肠蠕动功能,促进排便,从而缓解便秘。
2.降血脂实验
36只SD雄性大鼠(体重150-200g)按TC水平分为3组:空白组、模型组、实施例3的药物组,每组12只。大鼠分组后,空白组以基础饲料饲养,其他组均换用高脂饲料饲养,药物组每日按2.08g/kg的剂量灌胃给予相应药物,空白组和模型组给予蒸馏水,连续灌胃30天,期间小鼠正常饮水进食和活动。最后一次灌胃后,禁食不禁水12h,用10%水合氯醛(0.30mL/100gBW)经腹腔注射麻醉动物,解剖,腹主动脉取血,血样于4℃、3000r/min离心10min,分离血清,检测TC值、TG值、HDL-C值,结果见表3。

以上结果表明,本发明组合物能降低高脂动物模型的血清TC值和TG值,增加HDL-C值,说明具有降脂功效。
实施例7
昆布750g、胖大海50g、亚麻子油200g
按重量配比取昆布、胖大海、亚麻子油,将昆布干燥后粉碎成200目微粉,胖大海按重量加入10倍水煎煮提取2次,每次1.5小时,滤过,合并滤液并浓缩成稠膏,然后将稠膏、亚麻子油与昆布微粉混匀,湿法制粒,干燥,密封包装,灭菌,即得。
食用时,将颗粒用少量开水调成糊状即可食用,也可以将用大量开水融化后饮用。该食品中的主要成分是昆布,其粉末溶解性差,腥味重,让很多消费者难以接受,应用受到一定限制,申请人接着对昆布进行了脱腥处理的工艺研究。
申请人首先参考了大量文献,对应用较多的天然产物脱腥剂如山楂、食醋、食盐、陈皮、紫苏、肉桂、生姜、绿茶、茉莉花茶等进行了单一成分筛选,结果发现除食盐和绿茶外,其它物质均有一定程度的脱腥效果,但仍不能完全脱除昆布的腥味,在此基础上,申请人又将剩下的几种物质进行组合,最终从中筛选得到一种成分较少且脱腥效果更好的脱腥剂配方,它由陈皮、茉莉花茶和食醋三种成分组成,试验结果表明,使用该脱腥剂处理后的昆布其腥味评分可下降90%以上,而且处理后的昆布经粉碎后溶解性也得到改善。
以下仅列出试验中几个有代表性的实施例,由于试验样本很多,其它样本无法在实施例中全部列出。
实施例:将陈皮与茉莉花茶按5:3的重量比混合,然后加入5倍重量的水煎煮1.5小时,过滤,向滤液中按重量加入2.5%的食醋,接着将昆布原料加入到上述脱腥液中在75℃下加热浸泡15min,取出后用清水漂洗,沥去水分后干燥,最后再粉碎成200目微粉。
对比例1:将紫苏加入6倍重量的水煎煮2小时,过滤,向滤液中按重量加入3%的食盐,接着将昆布原料加入到上述脱腥液中在75℃下加热浸泡15min,取出后用清水漂洗,沥去水分后干燥,最后再粉碎成200目微粉。
对比例2:将肉桂、生姜按1:1的重量比混合,然后加入5倍重量的水煎煮2小时,过滤,向滤液中按重量加入1.5%的食醋,接着将昆布原料加入到上述脱腥液中在75℃下加热浸泡15min,取出后用清水漂洗,沥去水分后干燥,最后再粉碎成200目微粉。
对比例3:将山楂、紫苏、茉莉花茶按2:2:1的重量比混合,然后加入3倍重量的水煎煮1小时,过滤,接着将昆布原料加入到上述滤液中在75℃下加热浸泡15min,取出后用清水漂洗,沥去水分后干燥,最后再粉碎成200目微粉。
去取10g昆布微粉微粉,用50ml开水冲调后进行感官评价,将未进行脱腥处理的昆布微粉设为0分,无任何腥味设为10分,分值越高脱腥效果越好,试验人员15名,结果取平均值。
表5评分结果
结果表明,以陈皮、茉莉花茶和食醋为原料制备的脱腥液能使昆布微粉的腥味评分下降90%以上,加入到产品中后基本上闻不到腥味。同时,由于脱腥液中还含有食醋,并且进行了加热浸泡处理,使得粉碎后的昆布溶解性更好,能迅速形成均匀的糊状溶液,而未经脱腥处理的昆布溶解较慢,且溶液呈颗粒状。
本发明中制备脱腥液所使用的原料全部为天然产物,不含人工合成的化学成分,具有脱腥效果好、安全可靠等优点,而且原料数量少、来源广泛,具有成本低等优点。
Claims (1)
1.一种药物组合物的制备方法,其特征在于,所述药物组合物由以下重量配比的原料制成:昆布75%、胖大海5%、亚麻子油20%,制备方法包括以下步骤:按重量配比取昆布、胖大海、亚麻子油,将昆布粉碎成微粉,胖大海按重量加入8-12倍水煎煮提取2-3次,每次0.5-2小时,滤过,合并滤液并浓缩成稠膏,然后将稠膏、亚麻子油与昆布微粉混匀,制成药物,在将所述昆布粉碎成微粉前还对其进行了脱腥处理,处理方法是将昆布用脱腥液进行加热浸泡,所述加热浸泡的温度是60-80℃,时间是10-30min,所述脱腥液由陈皮、茉莉花茶和食醋制成,制备方法如下:将陈皮与茉莉花茶按5:3的重量比混合,然后加入5倍重量的水煎煮1.5小时,过滤,向滤液中按重量加入2.5%的食醋,即得;
该药物组合物能加快小肠运动,改善小肠蠕动功能,促进排便,从而缓解便秘;能降低高脂动物模型的血清TC值和TG值,增加HDL-C值,具有降脂功效。
