Abstract

药用凹凸棒石的制备方法属药品制造技术。凹凸棒石的主成分是一种具有特殊晶体结构的富镁粘土矿物,具有吸收和吸附性能,能吸收水分、吸附病菌和毒素,现已成为一种中药材。但原矿的粒度、纯度和酸碱度常不符合药用要求,以致影响制剂的质量。本发明旨在提高其细度、纯度和吸附性,降低碱性,提高药材和制剂的质量和稳定性。采用本发明提供的方法,可用低品位的原料制备成符合质量标准、性能稳定的药材,减轻药物对皮肤和粘膜的刺激。

Description

药用凹凸棒石的制备方法
本发明涉及供药用的天然矿物凹凸棒石的提纯和活化,属制药工艺领域。
凹凸棒石(Attapulgite)是1935年在美国佐治亚州Attapulgus地方发现的一种灰白色或青灰色粘土,主要成分是短纤维状(棒状)的坡缕石,并含有石英、蛋白石、白云石、磁铁矿、蒙皂石、海泡石等比重和硬度不同的多种杂质。坡缕石为含水镁铝硅酸盐,常含有铁,其结构式为:
(Mg,Al,Fe)5(Si,Al)8O20(OH)2(OH2)4·4H2O其晶体结构属层状硅酸盐(如滑石、云母、蒙皂石等),但带有链状硅酸盐(如闪石、辉石等)的特点,可称链-层状结构。与典型层状硅酸盐结构不同处在于其上下两片Si-O四面体只沿a轴延伸(成链状),而在b轴方向上则每隔两条Si-O四面体链就上下颠倒一次,使结构单元一上一下排列,形成实体与孔道相间的结构(如果四面体不上下颠倒,就会沿ab面延伸成无孔道间隔的层状结构)。实体之间靠Si-O-Si联结,容易沿c轴裂开,所以,坡缕石的晶体呈细长的纤维状或棒状,而不是片状。结构式中的4个(OH2)充盈在孔道中间,不易散失,不同于平常的水分,而与沸石中的水分相似,故称沸石水。晶体的粗细和长短即结晶程度与生成环境和发育好坏有关。典型的坡缕石是热液作用形成的,生成条件比较稳定,结晶体呈长纤维状。凹凸棒石中的坡缕石为地表多变环境下形成的,纤维甚短,一般长1微米左右,最长可达4-5微米,宽度约0.01-0.025微米,如果放大100万倍,一根纤维就变成1-2.5厘米粗、1米长的棒。坡缕石晶体从头到尾贯穿着数十或上百个孔道,孔道截面积大约3.8×6.3,表面凹凸相间,布满小孔。
凹凸棒石在工农业方面有广泛的用途,其中最主要的用途是供作吸收-吸附剂和抗盐钻井泥浆原料[1,2],最近在国内已开发成为一种新药材,研制成功一种外用新药肤疡散[3],主要利用其吸收和吸附活性。但目前药用凹凸棒石在制备方法上存在两大缺点:
1)由于采用未经提纯的原矿,以致白云石等碳酸盐太多,碱性偏高,且含有颗粒较粗、硬度较高的石英、蛋白石等杂质,容易刺激皮肤和粘膜。药品的性能不稳定,质量不易控制。
2)手选原矿用干法粉碎、过筛后,将矿粉装入玻璃瓶内按常规进行高压灭菌(如新肤疡散外用药的制备方法,专利ZL86105579.9),无法逐尽沸石水,不得发挥其应有的吸收和吸附性能,活性不高。
针对以上缺点,本发明根据凹凸棒石的矿物组成和特性,提出以下工艺加以改进:
1、提纯
1)凭肉眼观察,手选矿石,剔除杂石,晾晒干燥;
2)取少量矿样用1∶10稀盐酸测试分解其中碳酸盐所需的稀盐酸量;
3)在耐酸容器中将凹凸棒石与适量水一起搅拌,逐渐加入足够的稀盐酸,使凹凸棒石中的白云石等碳酸盐分解;
4)将泥浆通过60目筛,用大量水冲洗,转移到池中使坡缕石悬浮,依颗粒沉降原理,采用分选法,弃去石英、蛋白石、磁铁矿等比重较大的杂质和颗粒较细的蒙皂石等杂质;
5)用水漂洗泥浆至无氯离子反应,检测pH值;
6)取样作X线衍射分析和电子显微镜照相,检查其中坡缕石的晶体和结构有无损坏,纯度是否合格,如不合格,再次淘洗及至合格。
2、脱水
1)让泥浆在沉淀池中静置沉淀,排除大量水分;
2)用离心法或挤压法进一步脱水,于100℃以下干燥、粉碎,过200目筛,或采用喷雾干燥法直接用泥浆制粉。
3、活化-灭菌
1)将通过200目筛网的纯化土粉铺于不锈钢方盘中,厚度在2厘米左右为宜;
2)放入温度可调控并能透气的电烘箱中,逐渐升温至180-200℃(实验表明,超过400℃会导致结构改变,降低表面活性),待水汽基本排出后再保温2-4小时,活化与灭菌同步进行;
3)待凉透后,取出,随即移至密封的洁净容器中,抽真空以防受潮。
通过上述三个步骤,不仅可以克服原矿不纯所带来的药品质量问题,而且可以发挥其应有的吸附活性,改善药品的性能,甚至用品位稍次的原矿也可制备成符合质量标准的药材。整个工艺过程简便易行。

  1. 一种药用凹凸棒石的制备方法,其特征是用1∶10的稀盐酸分解凹凸棒石原矿中的碳酸盐,通过湿筛法在水中使原矿分散、悬浮、分选,取得坡缕石富集的浆料,直接用喷雾干燥法制粉后,在180°-200℃驱除沸石水,提高吸附活性,同时完成灭菌。
