Abstract

RAS蛋白是细胞信号通路的重要组成部分。在多种类型的癌症(包括胰腺癌)中经常检测到通过突变引起的RAS蛋白致癌激活。仍然需要进一步和更有效的癌症治疗。

给药
每个KRAS肽的给药剂量范围为每种肽100至5000微克(例如700微克/肽,对于7种肽的组,这将产生4900微克的肽剂量)。
如果佐剂与KRAS肽一起给药,则佐剂的剂量以体重计可以高达0.48mg/kg。示例性的给药方案如下表3所示。

实施例
实施例1:两亲分子KRas G12D引起免疫应答
确定了可溶性-KRas(KRas)或两亲分子-KRas(aKRas)(图6)突变序列和两亲分子-CpG佐剂(aCpG)引起免疫应答的功效。在这些实验中,使用的aCpG是两亲分子-KRas的五倍,因为CpG递增(escalation)实验表明该剂量可能更有效。
实验设计包括以下4组用所列化合物免疫的C57BL/6小鼠(每组n=5):
1.KRas G12D+aCpG
2.aKRas G12D+aCpG
3.KRas G12D+pIC
4.未免疫
PolyIC(pIC)被用作基准佐剂对照。
将佐剂以及肽原液溶解在水中。最终注射液用1X PBS(磷酸盐缓冲液)稀释。
所使用的aKRas肽是具有G12D取代的突变序列的18mer序列(野生型的氨基酸4-21→YKLVVVGAGGVGKSALTI(SEQ ID NO:9))。每次均使用20μg/100μl注射。
使用的aCpG序列是CpG1826序列(5’-tccatgacgttcctgacgtt-3’;SEQ ID NO:10),在5’端添加了两个鸟嘌呤(5’-ggtccatgacgttcctgacgtt-3’;SEQ ID NO:11)浓度为每100μl注射剂5nmol。CpG1826是最佳的小鼠序列,而CpG7909对人类是最佳的,并且在小鼠中的活性较弱。CpG1826和CpG7909属于同一CpG类(B类),通常在其各自的物种中具有相似的活性谱。
每100μl注射剂使用50μg的pIC。
皮下(s.c.)给予初次免疫(primer immunization)到尾巴基部(第0天),以及在2周后(第14天)一剂加强剂量。加强剂量给药后第8天(第21天),使用标准方案对脾细胞上的IFNγ进行ELISpot分析。用5μg/孔的G12D序列18mer(aa4-21)→YKLVVVGADGVGKSALTI(SEQID NO:1)激活脾细胞(106个细胞/孔)。如图1所示,aKRas与aCpG的组合活化脾细胞,而可溶性KRas与aCpG的组合,而未处理的和可溶性KRas和pIC对照组则没有。
实施例2:两亲分子KRas G12R和G12V引起免疫应答

6.aKRas G12V+aCpG

佐剂以及肽原液都溶解在水中。最终注射液用1X PBS稀释。
所使用的aKras肽是具有G12R/V取代的突变序列的18mer序列(野生型SEQ氨基酸4-21→YKLVVVGAGGVGKSALTI(SEQ ID NO:9))。每次均使用20μg/100μl注射。
使用的aCpG序列是CpG1826序列(5’-tccatgacgttcctgacgtt-3’;SEQ ID NO:10),在5’端添加了两个鸟嘌呤(5’-ggtccatgacgttcctgacgtt-3’;SEQ ID NO:11),浓度为每100μl注射剂5nmol。
每100μl注射剂使用50μg的pIC。
皮下(s.c.)给予初次免疫到尾巴基部,以及在2周后(d14)一剂加强剂量。
加强剂量给药后第7天(d21),使用标准方案对脾细胞上的IFNγ进行ELISpot分析。用5μg/孔的以下任一种激活脾细胞(106细胞/孔):G12V序列18mer(aa4-21)→YKLVVVGAVGVGKSALTI(SEQ ID NO:2)或G12R序列18mer(aa4-21)→YKLVVVGARGVGKSALTI(SEQ ID NO:3)。
如图2所示,aKRas G12R和G12V与aCpG的组合激活脾细胞,而可溶性KRasG12R和G12V与aCpG的组合,以及未处理的和可溶性KRas和pIC对照组均没有。
实施例3:两亲分子KRas G12D引起CD8+T-细胞免疫应答
在野生型B6小鼠中,很难引起Kras特异性CD8+T-细胞应答。用可溶性或两亲分子缀合形式的KRas G12D抗原以及佐剂(CpG或aCpG)免疫人类HLA基因(A2.1)转基因的B6小鼠。已知A2.1等位基因在人类中对KRas肽产生应答。
实验设计包括下列4组用所列化合物免疫的B6 HLA-A2.1 Tg小鼠(每组n=5):
1.KRas野生型+CpG
2.KRas 12D+CpG
3.aKRas 12D+aCpG
4.未免疫
将佐剂以及肽原液溶解在水中。最终注射液用1X PBS稀释。
所使用的aKRas肽是具有G12D取代的突变序列的18mer序列(野生型氨基酸4-21→YKLVVVGAGGVGKSALTI(SEQ ID NO:9))。每次均使用20μg/100μl注射。
使用的aCpG序列是CpG1826序列(5’-tccatgacgttcctgacgtt-3’;SEQ ID NO:10),在5’端添加了两个鸟嘌呤(5’-gg tccatgacgttcctgacgtt-3’;SEQ ID NO:11),浓度为每100μl注射剂5nmol。
将该疫苗皮下给药到雌性小鼠的尾巴基部两侧,每侧50μl。每两周间隔给予两次加强剂量。
分别在第二和第三次加强剂量后第7天,使用标准方案对外周血单核细胞(PBMC)上的IFNγ进行了细胞内细胞因子染色(ICS)分析,并对脾细胞上的IFNγ进行了ELISpot分析。
用5μg/孔的SEQ ID NO:1或SEQ ID NO:12-20中的一个激活PBMC/脾细胞(106细胞/孔和2x106细胞/孔),如下所示:
将特异性的突变肽仅给予用相应的18mer免疫的小鼠。
图3示出了用aKRas G12D和aCpG的组合免疫的小鼠的KRas特异性CD8+T-细胞应答,但是对于与CpG组合的可溶性KRas G12D或与CpG组合的可溶性野生型KRas没有KRas特异性CD8+T-细胞应答。
实施例4:更高抗原浓度和不同剂量间隔的分析
抗原的浓度从20μg增加到50μg,并以每两周(bw)或每周(w)的剂量间隔进行测试。
实验设计包括以下5组用所列化合物免疫的C57BL/6小鼠(每组n=5):
1.KRas 12D+CpG1826(bw)
2.KRas 12D+CpG1826(w)
3.aKRas 12D+aCpG1826(bw)
4.aKRas 12D+aCpG1826(w)
5.未免疫
将佐剂以及肽原液溶解在水中。最终注射液用1X PBS稀释。

将特异性突变肽仅给予用相应的18mer免疫的小鼠。
制备单独的平板以用于短(9mer和10mer)或长(17mer和18mer)肽刺激。用所有刺激刺激来自“未免疫”对照小鼠的细胞。
图4显示了对aKRas G12D和aCpG组合免疫的小鼠,具有KRas特异性CD8+T-细胞应答(9mer)和CD8+或CD4+T-细胞应答(18mer),但对于可溶性KRas G12D与CpG组合或未经处理的对照,却没有KRas特异性CD8+T-细胞应答(9mer)和CD8+或CD4+T-细胞应答(18mer)。
实施例5:KRAS两亲分子的进一步分析
如以上实施例中所述,已经在C57BL/6小鼠模型中证明了具有aCpG佐剂的突变KRAS肽-两亲分子疫苗的潜在免疫原性。这些研究以及进一步的研究总结在表4中,这些结果表明,由于MHC结合的物种差异,小鼠历来对突变KRAS肽的免疫应答较人类差。
·由ELISPOT和细胞因子珠阵列鉴定(图1、图2、图4和图5),皮下给药突变KRAS两亲分子-肽(G12D(图6)、G12R和G12V)可在小鼠中有效诱导KRAS特异性脾T细胞
·与突变KRAS肽与可溶性CpG或聚肌苷酸:聚胞苷酸(poly IC)的组合作为佐剂相比,突变KRAS两亲分子-肽(G12D、G12R和G12V)与aCpG组合使用作为佐剂在诱导KRAS特异性脾T细胞方面更有效(图1、图2和图5),以及
·在引起CD4和CD8 T细胞应答方面,突变KRAS两亲分子-肽(G12D)与aCpG组合比可溶性KRAS肽与可溶性CpG组合更有效,如通过长(18-mer)或最小(9-mer)肽表位再刺激时的效应细胞因子应答所证实(图4)。
