Abstract
The invention relates to the technical field of active ingredient extraction, and particularly relates to a preparation method of a paris polyphylla extractive solution, which comprises the following steps: taking a proper amount of rhizome of paris polyphylla, cleaning, slicing, crushing to 200 meshes, placing in an enzymolysis tank, adding 6 times of deionized water, placing in a constant-temperature water bath shaking table at 60-80 ℃, sequentially adding fungal protease, cellulase, pectinase and alpha-amylase, performing enzymolysis for 1h respectively, homogenizing for 20min under the pressure of 200-400 MPa, inactivating enzyme, placing in a high-pressure reaction kettle, adding a proper amount of deionized water, and performing subcritical water extraction for 20-30 min to obtain an extracting solution. According to the invention, the paris polyphylla extracting solution is prepared by adopting a method of high-pressure homogenization treatment, subcritical water extraction and enzymolysis coupling, so that the active ingredients of paris polyphylla powder can be rapidly separated out, the extraction time is shortened, the extraction efficiency is improved, and no waste residue is generated.

s3, placing the enzyme deactivation solution into a high-pressure reaction kettle, adding a proper amount of deionized water, and performing subcritical water extraction treatment for 20-30 min at 100-160 ℃ to obtain an extracting solution.
Further, in the step S2, the mass ratio of the fungal protease, the cellulase, the pectinase, the alpha-amylase and the paris polyphylla pollen is 1 to 10.
Further, in the step S2, the flow rate of the homogenization treatment is 4 to 6L/h, and the temperature of the enzymatic hydrolysate is between 10 and 20 ℃.

In the embodiment, the flow rate of homogenization treatment is 5L/h, the temperature of the zymolyte is 15 ℃, the power is 120W, the frequency is 60Hz, and the ratio of subcritical water extraction to feed liquid is 15mL/g during ultrasonic dispersion.
Proved by verification, compared with an alcohol extraction method, a water boiling method, a water ultrasonic extraction method and an alcohol ultrasonic extraction method, the extraction method can obviously reduce the extraction time and does not generate waste residues under the same target product amount.
