一种含有二氢杨梅素的敏感性肌肤护肤品,以二氢杨梅素作为缓解敏感性肌肤护肤品的活性成分,二氢杨梅素添加量为护肤品重量份的0.01~5份,包括面霜、乳液、面膜和精华液,本发明利用体外细胞实验证明二氢杨梅素可抑制过敏及炎症多条信号通路中的信号物质以实现缓解敏感性肌肤问题的效果,二氢杨梅素可以通过抑制NF‑κB通路中p65的表达抑制促炎性细胞因子的分泌,可以通过抑制抑制STAT5磷酸化抑制肥大细胞的激活,也可以通过抑制ERK1/2、p38和JNK磷酸化抑制过敏炎症应激通路。

背景技术:
敏感性皮肤是一种常见的皮肤类型,指的是高反应性、耐受性差或易过敏的皮肤状态。敏感性皮肤易受到外界物理化学等因素的刺激而产生不适反应,主要表现为皮肤干燥、脱屑、发红,容易产生瘙痒、烧灼、刺痛、紧绷等。敏感性皮肤易受到外源刺激,易引发刺激性接触性皮炎(irritant contact dermatitis,ICD)和变应性接触性皮炎(allergiccontact dermatitis,ACD),从而导致机体中组织胺、白三烯、血小板活化因子、缓激肽等的释放引发各种过敏反应症状。敏感性皮肤多见于女性面部,以及双手、头皮和足部,研究报道显示北京、上海、广州3个城市敏感性皮肤的发生率平均为13%,其中女性高于男性(兰宇贞,谢志强.敏感性皮肤研究进展[J].中国中西医结合皮肤性病学杂志,2013(12):199-201)。近年来敏感性皮肤的发生率也有不断增长的趋势,这将对人们的生活质量造成极大的影响。随着人们对化妆品需求的增加,大量新型化妆品原料,尤其一些化学物,不断被应用于化妆品开发中,使得人们接触更多潜在致敏物质,大大增加了皮肤过敏的风险。化妆品中含有的香精、防腐剂、乳化剂、抗氧化剂、防晒剂、激素(皮质类固醇激素、雌激素等)等均是刺激敏感性皮肤的风险物质。近年来人们对生活质量要求不断提升,对敏感性皮肤问题也越来越重视,因而各大护肤品品牌将抗敏抗刺激列入产品研发的重点项目之一。
二氢杨梅素(Dihydromyricetin,DHM)是藤茶中含量作为丰富的一类活性黄酮类化合物,除总黄酮的一般特性外,还具有保护心脏、抗糖尿病、保护肝脏、神经保护、抗肿瘤、保护皮肤等作用,医药中还可应用于细菌感染、骨质疏松、哮喘、肾损伤、肾毒性等的治疗。也有研究者通过小鼠食物过敏模型发现二氢杨梅素能够有效缓解小鼠过敏症状及相关过敏指标,具有良好的抗食物过敏活性。二氢杨梅素在偏酸性和中性条件下稳定,由于护肤品的pH为5~7,因而适应于二氢杨梅素稳定发挥作用。近年来,二氢杨梅素作为INCI中收录的藤茶提取物和显齿蛇葡萄叶提取物的活性成分被应用于化妆品中。研究显示,二氢杨梅素可以延缓皮肤黄褐斑和老年斑的出现时间、防治光损伤。同时,二氢杨梅素复配白芨多糖制备的化妆品具有一定的祛斑、美白等功效。目前,二氢杨梅素在抗敏抗刺激护肤品中的应用还未见报道,其具体作用机制尚不清楚。
发明内容:
针对现有技术的上述问题,本发明目的在于提供了一种含有二氢杨梅素的敏感性肌肤护肤品。
为达成上述目的,本发明采用如下技术方案:
一种含有二氢杨梅素的敏感性肌肤护肤品,所述护肤品以二氢杨梅素作为缓解敏感性肌肤护肤品的活性成分,二氢杨梅素添加量为护肤品重量份的0.01~5份。
进一步,所述护肤品为面霜,按照以下重量份配制:2,3-丁二醇1~8份,丙烯酰二甲基牛磺酸铵/VP共聚物0.1~1份,甘油聚醚-26 1~10份,二氢杨梅素0.01~5份,辛甘醇0.01~1份,1,2-己二醇0.1~2份,甘油硬脂酸酯0.1~3份,PEG-100硬脂酸酯0.1~3份,辛酸/癸酸甘油三酯1~8份,聚二甲基硅氧烷0.1~3份,刺阿干树仁1~6份,生育酚乙酸酯0.1~1份,余量为水。
进一步,所述护肤品为乳液,按照以下重量份配制:丁二醇1~10份,甘油0.1~8份,卡波姆0.01~1份,二氢杨梅素0.01~5份,对羟基苯乙酮0.1~3份,1,2-己二醇0.01~2份,辛酰羟肟酸0.01~2份,鲸蜡硬脂醇0.1~3份,碳酸二辛酯0.5~3份,角鲨烷0.1~6份,鲸蜡硬脂醇橄榄油酸1~5份,山梨坦橄榄油酸酯0.1~3份,生育酚乙酸酯0.1~1份,精氨酸0.01~2份,余量为水。

本发明所述的二氢杨梅素为《已使用化妆品原料目录(2015版)》中收录的藤茶提取物/显齿蛇葡萄叶提取物中的活性成分,天然来源,安全无刺激。
本发明证实二氢杨梅素可以通过抑制NF-κB、JAK/STAT5和MAPK信号通路有效抑制过敏相关炎症介质的释放,从而缓解过敏性炎症,为实现二氢杨梅素在敏感性肌肤护肤品中的应用提供理论数据支持。
临床试验表明,持续使用本发明所述的含有二氢杨梅素的面膜可以有效缓解敏感性肌肤的面部红斑情况。

下面的实施例对本发明作详细描述,但以下实施例中所公开的技术代表本发明相关人员能力范围内应用在本发明的生物学和化学领域基础技术手段;本发明实施例的描述及附图是示例性的,旨在辅助解释本发明;本发明中具体应用涉及的添加比例在有效范围内不局限于单一比例,以上均不能理解为对本发明的限制。
本发明提出《已使用化妆品原料目录(2015版)》中收录的藤茶提取物/显齿蛇葡萄叶提取物中的活性成分——二氢杨梅素在敏感性肌肤护肤品中的应用,含有该活性成分的面膜可以有效缓解敏感性肌肤的红斑情况。体外实验也进一步证实二氢杨梅素可通过抑制p65的表达抑制炎症因子的释放,可以通过抑制STAT5磷酸化抑制肥大细胞的激活,也可以通过抑制ERK1/2、p38和JNK磷酸化抑制过敏炎症应激通路。
实施例1二氢杨梅素的分离纯化及鉴定
配制浸提液:25%乙醇(含0.2%柠檬酸)。藤茶/显齿蛇葡萄叶:浸提液按照料液比=1:8浸提。温度控制为70℃,浸提两次,每次60min。混合两次浸提液,过滤,浓缩,收集结晶,干燥,纯化,利用HPLC检测如下图所示。HPLC检测条件:色谱柱——ODS-35um(4.6×250mm);柱温为40℃;检测波长为291nm;进样体积为5μL;流速为1mL/min;流动相A为甲醇,流动相B为0.1%磷酸水溶液,梯度洗脱。经过标品配制、线性关系考察、精密度考察、重复性考察、稳定性考察以及回收率测试等步骤,最终检测样品为二氢杨梅素,如图1所示。
实施例2二氢杨梅素对肥大细胞炎症介质释放的影响
实验方法:37℃,5%CO2条件下,10%胎牛血清和1%双抗的DMEM培养基培养RBL-2H3细胞。
37℃,5%CO2条件下,10%胎牛血清(热灭活)和1%双抗的RPMI-1640培养基培养KU812细胞
(1)细胞活力检测——铺板96孔板进行实验,1×104个细胞/孔,培养过夜,添加DHM(1-2000μM)孵育24h,加入WST-1试剂(10mol/L),37℃孵育1h。在450nm波长下测定吸光值,测定细胞存活率。
(2)脱颗粒检测——铺板48孔板进行实验,1×106个细胞/孔,0.1μg/ml的anti-DNP-IgE敏化细胞16h,PBS清洗两次,100ng/ml的DNP-BSA刺激细胞,利用肥大细胞脱颗粒试验试剂盒说明书测定胰蛋白酶浓度。
(3)ROS检测——铺板96孔板进行实验,1×105个细胞/孔,0.1μg/ml的anti-DNP-IgE敏化细胞16h,然后添加DHM孵育24小时。更换为PBS,添加DCFH-DA(10μM)孵育30min,PBS洗涤细胞2次后,利用荧光观察和流式细胞术检测ROS。同时,采用mito SOXTM红色探针等检测ROS。
(4)细胞因子检测——铺板48孔板进行实验,1×106个细胞/孔,0.1μg/ml的anti-DNP-IgE敏化细胞16h,PBS清洗两次,100ng/ml的DNP-BSA刺激细胞,利用ELISA试剂盒检测上清液中的炎症因子。
数据处理:所有数据均计算平均标准差(SD)。利用t检验分析组间差距显著性。*p<0.05代表组间显著性差异,**p<0.01,***p<0.001代表组间极显著差异。#p<0.05代表组间显著性差异,##p<0.01,###p<0.001代表组间极显著差异。
实验结果:(1)如图2所示,DHM在1至500μM浓度下显示对RBL-2H3细胞无明显的细胞毒性。后续实验选择DHM的浓度为100μM。
(2)类胰蛋白酶是肥大细胞激活后释放的预形成因子一种丝氨酸蛋白酶,可以用作判断肥大细胞活化(脱颗粒)指标。如图3所示,低浓度的IgE敏化后DNP-BSA刺激细胞,即可激活肥大细胞,使其释放的胰蛋白酶显著增加。添加100μM的DHM可以显著降低细胞上清中的类胰蛋白酶,说明DHM可以抑制肥大细胞脱颗粒释放类胰蛋白酶。
(3)肥大细胞受到刺激后会释放活性氧(ROS),过敏及炎症等反应发生过程中,也常常伴随着肥大细胞活性氧的升高。如图4A和4B所示,敏化刺激后的肥大细胞荧光明显增强,说明肥大细胞中的活性氧含量明显上升。添加100μM的DHM孵育24小时后,荧光反应明显减弱,说明肥大细胞中的活性氧含量下降。图4C所示,IgE和DNP刺激的肥大细胞ROS峰(红色)相比于control组(蓝色)明显右移,而添加DHM孵育后的ROS峰(紫色)相比于刺激组(红色)的峰明显左移。综上所述,说明DHM可以有效缓解肥大细胞由于过敏引起的活性氧的上升。
(4)白介素-6(IL-6)是一种多效性的促炎因子,在慢性炎症反应的发生和发展中起到重要的作用。如图5所示,低浓度的IgE敏化后DNP-BSA刺激细胞,可使细胞培养上清中的IL-6明显增加。添加100μM的DHM可以显著降低细胞上清中的IL-6,说明DHM可以抑制促炎因子的释放。
实施例3二氢杨梅素对肥大细胞信号通路的影响
实验方法:37℃,5%CO2条件下,10%胎牛血清(热灭活)和1%双抗的RPMI-1640培养基培养KU812细胞。利用胰酶将细胞消化下来,添加适量细胞裂解液,4℃,20min。利用总蛋白检测试剂盒检测裂解物的蛋白含量。利用SDS-PAGE电泳分离蛋白,将蛋白转印至PVDF膜上,经过脱脂牛奶中封闭、对应的一抗4℃孵育过夜(p53、JNK、p-JNK、p38、p-P38、ERK1/2、p-ERK1/2、p65、STAT5和pSTAT 5),二抗在室温下孵育2h,膜显色,拍照。
实验结果:(1)核转录因子kappa B(NF-κB)是细胞中重要的转录调节因子,其活化可诱导多种基因的表达,进一步产生多种细胞因子,从而参与炎症反应。p50-p65异二聚体与IκB结合使NF-κB处于非活化状态,但是p65-p65和c-Rel-p50等二聚体可以有效促进NF-κB的诱导。如图6所示的Western图,敏化刺激后的KU812细胞内p65大量表达,100μM的DHM可以抑制细胞中p65的表达,从而抑制NF-κB的诱导活化。
(2)信号转导及转录激活因子(STATs)是关键的转录因子,活化后(如JAKs磷酸化)可以将细胞外的信号传递到细胞核内,引发相应基因的转录,进而触发细胞分化、增殖、发育、凋亡以及炎症等反应。STAT5与肥大细胞活化相关。本发明中图7所示的Western图,敏化刺激前后的KU812细胞内STAT5含量变化不大,但是敏化刺激后磷酸化SATA5(p-STAT5)明显上升,100μM的DHM可以抑制肥大细胞细胞中SATA5磷酸化,从而抑制肥大细胞活化。
(3)肥大细胞激活过程伴随着一系列信号传导通路及信号分子的级联网络,包括各类酪氨酸激酶及MAPK(Mitogen-activated protein kinase,丝裂原激活的蛋白激酶/MAP激酶)信号通路。MAPK信号通路中的ERK、JNK及p38 MAPK三条信号传导通路在过敏反应中均具有重要的调控作用。同时,MAPK通路可以被ROS激活。本发明中图8所示的Western图,用IgE敏化后的KU812细胞未刺激的MAPK信号通路分子的磷酸化水平都很低,大部分处于未磷酸化。用IgE敏化后的KU812细胞受到刺激的MAPK信号通路分子被磷酸化,p-p38和p-JNK表达上升。100μM的DHM可以抑制肥大细胞细胞中p-p38和p-JNK的表达,从而抑制肥大细胞活化。
实施例4二氢杨梅素应用于面霜
面霜:2,3-丁二醇4份,丙烯酰二甲基牛磺酸铵/VP共聚物0.4份,甘油聚醚-26 5份,二氢杨梅素0.2份,辛甘醇0.2份,1,2-己二醇0.5份,甘油硬脂酸酯1.8份,PEG-100硬脂酸酯1.2份,辛酸/癸酸甘油三酯5份,聚二甲基硅氧烷2份,刺阿干树仁4份,生育酚乙酸酯0.5份,水补足100份,制备面霜。
实施例5二氢杨梅素应用于乳液
乳液:丁二醇5份,甘油5份,卡波姆0.15份,二氢杨梅素0.2份,对羟基苯乙酮0.3份,1,2-己二醇0.5份,辛酰羟肟酸0.05份,鲸蜡硬脂醇1份,碳酸二辛酯2份,角鲨烷4份,鲸蜡硬脂醇橄榄油酸2.4份,山梨坦橄榄油酸酯1.6份,生育酚乙酸酯0.5份,精氨酸0.12份,水补足100份,制备乳液。
实施例6二氢杨梅素应用于面膜
面膜:甘油3份,丁二醇5份,透明质酸钠0.2份,二氢杨梅素0.05份,黄原胶0.1份,胶原溶液2份,对羟基苯乙酮0.3份,1,2-己二醇0.5份,辛酰羟肟酸0.05份,水补足100份,制备面膜。
实施例7二氢杨梅素应用于精华液
精华液:丁二醇5份,甘油聚醚-26 5份,,透明质酸钠0.3份,二氢杨梅素0.1份,β-葡聚糖溶液3份,1,2-戊二醇3份,辛甘醇0.2份,1,2-己二醇0.5份,水补足100份,制备精华液。
实施例8面膜临床评估
试验方法:以本发明实施例6所述的面膜为例,测试其功效,进行了以下临床实验研究:
征召年龄在20至60的女性志愿者(敏感性肌肤)20名,所有受试者均被告知本次研究的详细内容,并签署由安泰医疗合作安泰天盛纪念医院(TSMH)机构审查委员会批准的知情同意书。
本测试要求试验者停止使用其他抗敏类护肤品,使用本发明中实施例6的面膜样品,每天一次,连续使用28天。利用Dual-MPA 580皮肤测试系统对皮肤进行无创检测。
试验结果:20位试验者连续使用本发明中实施例6的面膜28天后,分区检测面部红斑情况(左脸/右脸/前额)。如下图9所示,试用者整体面部红斑情况均有所改善。说明二氢杨梅素在改善面部红斑情况缓解敏感性肌肤方面有效果。

Claims (9)

1.一种含有二氢杨梅素的敏感性肌肤护肤品,其特征在于,所述护肤品以二氢杨梅素作为缓解敏感性肌肤护肤品的活性成分,二氢杨梅素添加量为护肤品重量份的0.01~5份。
2.如权利要求1所述的一种含有二氢杨梅素的敏感性肌肤护肤品,其特征在于,所述护肤品为面霜,按照以下重量份配制:2,3-丁二醇1~8份,丙烯酰二甲基牛磺酸铵/VP共聚物0.1~1份,甘油聚醚-26 1~10份,二氢杨梅素0.01~5份,辛甘醇0.01~1份,1,2-己二醇0.1~2份,甘油硬脂酸酯0.1~3份,PEG-100硬脂酸酯0.1~3份,辛酸/癸酸甘油三酯1~8份,聚二甲基硅氧烷0.1~3份,刺阿干树仁1~6份,生育酚乙酸酯0.1~1份,余量为水。
3.如权利要求1所述的一种含有二氢杨梅素的敏感性肌肤护肤品,其特征在于,所述护肤品为乳液,按照以下重量份配制:丁二醇1~10份,甘油0.1~8份,卡波姆0.01~1份,二氢杨梅素0.01~5份,对羟基苯乙酮0.1~3份,1,2-己二醇0.01~2份,辛酰羟肟酸0.01~2份,鲸蜡硬脂醇0.1~3份,碳酸二辛酯0.5~3份,角鲨烷0.1~6份,鲸蜡硬脂醇橄榄油酸1~5份,山梨坦橄榄油酸酯0.1~3份,生育酚乙酸酯0.1~1份,精氨酸0.01~2份,余量为水。
4.如权利要求1所述的一种含有二氢杨梅素的敏感性肌肤护肤品,其特征在于,所述护肤品为面膜,按照以下重量份配制:甘油1~10份,丁二醇1~8份,透明质酸钠0.01~1份,二氢杨梅素0.01~3份,黄原胶0.01~3份,胶原溶液0.1~5份,对羟基苯乙酮0.1~3份,1,2-己二醇0.1~2份,辛酰羟肟酸0.01~1份,余量为水。
5.如权利要求1所述的一种含有二氢杨梅素的敏感性肌肤护肤品,其特征在于,所述护肤品为精华液,按照以下重量份配制:丁二醇1~10份,甘油聚醚-26 0.5~5份,,透明质酸钠0.01~1份,二氢杨梅素0.01~5份,β-葡聚糖溶液0.1~5份,1,2-戊二醇0.1~5份,辛甘醇0.01~1份,1,2-己二醇0.01~2份份,余量为水。
6.如权利要求2所述的一种含有二氢杨梅素的敏感性肌肤护肤品,其特征在于,所述护肤品为面霜,按照以下重量份配制:2,3-丁二醇4份,丙烯酰二甲基牛磺酸铵/VP共聚物0.4份,甘油聚醚-26 5份,二氢杨梅素0.2份,辛甘醇0.2份,1,2-己二醇0.5份,甘油硬脂酸酯1.8份,PEG-100硬脂酸酯1.2份,辛酸/癸酸甘油三酯5份,聚二甲基硅氧烷2份,刺阿干树仁4份,生育酚乙酸酯0.5份,水补足100份以常规方式制备面霜。
7.如权利要求3所述的一种含有二氢杨梅素的敏感性肌肤护肤品,其特征在于,所述护肤品为乳液,按照以下重量份配制:丁二醇5份,甘油5份,卡波姆0.15份,二氢杨梅素0.2份,对羟基苯乙酮0.3份,1,2-己二醇0.5份,辛酰羟肟酸0.05份,鲸蜡硬脂醇1份,碳酸二辛酯2份,角鲨烷4份,鲸蜡硬脂醇橄榄油酸2.4份,山梨坦橄榄油酸酯1.6份,生育酚乙酸酯0.5份,精氨酸0.12份,水补足100份以常规方式制备乳液。
8.如权利要求4所述的一种含有二氢杨梅素的敏感性肌肤护肤品,其特征在于,所述护肤品为面膜,按照以下重量份配制:甘油3份,丁二醇5份,透明质酸钠0.2份,二氢杨梅素0.05份,黄原胶0.1份,胶原溶液2份,对羟基苯乙酮0.3份,1,2-己二醇0.5份,辛酰羟肟酸0.05份,水补足100份以常规方式制备面膜。
9.如权利要求5所述的一种含有二氢杨梅素的敏感性肌肤护肤品,其特征在于,所述护肤品为精华液,按照以下重量份配制:丁二醇5份,甘油聚醚-26 5份,,透明质酸钠0.3份,二氢杨梅素0.1份,β-葡聚糖溶液3份,1,2-戊二醇3份,辛甘醇0.2份,1,2-己二醇0.5份,水补足100份以常规方式制备精华液。
