The preparation method of the umbilical cord mesenchymal stem cell exosome extract comprises the following steps:
step 1, culturing umbilical cord mesenchymal stem cells
Culturing the umbilical cord mesenchymal stem cells with an alpha-MEM culture medium until the umbilical cord mesenchymal stem cells grow to a fusion degree of more than 80%, collecting a culture solution, washing with PBS for three times, digesting with 0.25% pancreatin for passage, continuing to subculture, collecting a culture supernatant each time, replacing a fresh culture medium, and collecting the culture supernatant in the continuous culture process.
Step 2, extracting exosomes
a. Centrifuging the cell culture supernatant at 400g of 300-;
b. taking the supernatant collected in the step a, centrifuging for 20-30min at 3000g of 2000-3000g, continuously collecting the supernatant, and discarding the precipitate;
c. filtering the supernatant collected in the step b by using a filter with the pore diameter of 0.22 mu m, and collecting filtrate;
d. ultrafiltering the supernatant with a Stired Cell 8050 type ultrafiltration device, wherein the pore diameter of the filter membrane is 30-60nm, adding PBS and ultrafiltering again after ultrafiltration is completed, and repeating for 3 times;
e. washing exosomes on the filter membrane by PBS to prepare a concentrated culture solution, measuring the protein concentration by using a BCA kit, and refrigerating and storing.
Thickening agent
Thickeners can have a significant effect on the appearance of the product. Emulsifier

The amount of the surfactant used can be selected by those skilled in the art depending on the kind of the surfactant, and for example, 0.1 to 1 part by weight can be used when polyglycerin-3-methylglucidistearate is used, and 1 to 3 parts by weight can be used when cetearyl alcohol and cetearyl glucoside are used.

step 2, adding 0.4 part of xanthan gum, 6 parts of glycerol, 6 parts of butanediol, 1.5 parts of sodium hyaluronate and 1 part of methylparaben into 43.6 parts of water, heating to 80 ℃, fully stirring and dissolving, and cooling to below 40 ℃ to obtain a first reagent;
step 3, adding 4 parts of tranexamic acid, 4 parts of ascorbic acid and 1 part of dipotassium glycyrrhizinate into 9 parts of water, and dissolving at the temperature of below 40 ℃ to obtain a second reagent;
and 4, emulsifying the fourth reagent and the first reagent, cooling to below 40 ℃, adding the second reagent, 1 part of the umbilical cord mesenchymal stem cell exosome extract and 2 parts of phenoxyethanol, and uniformly stirring to obtain the cream.

To further illustrate the beneficial effects of the present invention, comparative example 2 is provided, which comparative example 2 differs from example 3 in that: the butanediol of the present invention is replaced with xylitol.

Experiment 1: wrinkle removal test
A subject: 130 volunteers with 20-60 years old and severe raised lines and canthus lines are divided into 13 groups, and each group contains 10 persons;
the test method comprises the following steps: samples of examples 1-10 and comparative examples 1-3 are respectively taken and smeared on the face, the samples are respectively used once in the morning and at the evening for 60 days continuously, and a skin diagnostician-Visia is used for wrinkle detection; the results are shown in Table 1.
Experiment 2: antiallergic test
A subject: 130 volunteers with skin of 20-60 years old in allergic phase, divided into 13 groups of 10 persons each;
the test method comprises the following steps: samples of examples 1-10 and comparative examples 1-3 are respectively taken and smeared on the face, the samples are respectively used once in the morning and at the evening for 60 days continuously, and a skin diagnostician-Visia is used for detecting allergic regions; the results are shown in Table 1.

Remarking: the number of people with reduced wrinkles is 50% or more;
the allergy-reducing population is a population having an allergy reduction rate of 50 or more.
From the above results, it can be seen that:
the umbilical cord mesenchymal stem cell exosome used in the reagent combination can simulate partial functions of umbilical cord mesenchymal stem cells to play roles of promoting tissue repair and immunoregulation, has small molecular weight, and enables the skin to have better absorbability, and the specific tranexamic acid, ascorbic acid and dipotassium glycyrrhizinate matched with the reagent combination can better improve dry or damaged skin, and can better resist allergy, diminish inflammation and relieve the skin; specifically, by using the skin care reagent combination provided by the invention, the wrinkle reduction ratio can reach more than 70%, and the allergy reduction ratio can reach as high as 80%, which indicates that the skin care reagent combination has the technical effects of better wrinkle removal, better anti-allergy property and better absorbability by screening the types of the components in the skin care reagent combination or adjusting the weight parts of the components in the skin care reagent combination. The existing skin care products have certain wrinkle removing effect, but the anti-allergic effect is poor, and the skin condition of people with skin allergy cannot be well improved.
Similarly, the umbilical cord mesenchymal stem cell exosome used in the toning lotion can simulate partial functions of umbilical cord mesenchymal stem cells to play roles of promoting tissue repair and immunoregulation, and has small molecular weight, so that the skin absorbability is better, and the dried or damaged skin can be better improved by matching the specific tranexamic acid, the ascorbic acid and the dipotassium glycyrrhizinate, so that the skin can be better antiallergic, anti-inflammatory and soothing; specifically, by using the toning lotion provided by the invention, the wrinkle reduction ratio can reach more than 80%, and the allergy reduction ratio can reach 80%, which shows that the toning lotion has the technical effects of better wrinkle removal, better antianaphylaxis and better absorbability by screening the types of the components in the toning lotion or adjusting the weight parts of the components in the toning lotion.
Similarly, the umbilical cord mesenchymal stem cell exosome used in the facial mask can simulate partial functions of umbilical cord mesenchymal stem cells to play roles of promoting tissue repair and immunoregulation, and has small molecular weight, so that the skin absorbability is better, and the dried or damaged skin can be better improved by matching the specific tranexamic acid, the ascorbic acid and the dipotassium glycyrrhizinate, so that the skin can be better antiallergic, anti-inflammatory and soothing; specifically, by using the mask provided by the invention, the wrinkle reduction ratio can be up to more than 80%, and the allergy reduction ratio can be up to 80%, which shows that the mask has the technical effects of better wrinkle removal, better antianaphylaxis and better absorbability by screening the types of the components in the mask or adjusting the weight parts of the components in the mask.
Similarly, the umbilical cord mesenchymal stem cell exosome used in the emulsion can simulate partial functions of umbilical cord mesenchymal stem cells to play roles of promoting tissue repair and immunoregulation, and has small molecular weight, so that the skin absorbability is better, and the dried or damaged skin can be better improved by matching the specific tranexamic acid, the ascorbic acid and the dipotassium glycyrrhizinate, so that the skin can be better antiallergic, anti-inflammatory and soothing; specifically, by using the emulsion provided by the invention, the wrinkle reduction ratio can reach more than 90%, and the allergy reduction ratio can reach 90%, which shows that the emulsion has the technical effects of better wrinkle removal, better antianaphylaxis and better absorbability by screening the types of the components in the emulsion or adjusting the weight parts of the components in the emulsion.
Similarly, the umbilical cord mesenchymal stem cell exosome used in the cream can simulate partial functions of umbilical cord mesenchymal stem cells to play roles of promoting tissue repair and immunoregulation, and has small molecular weight, so that the skin absorbability is better, and the dried or damaged skin can be better improved by matching the specific tranexamic acid, the ascorbic acid and the dipotassium glycyrrhizinate, so that the skin can be better antiallergic, anti-inflammatory and soothing; specifically, by using the cream provided by the invention, the wrinkle reduction ratio can reach more than 80%, and the allergy reduction ratio can reach 90%, which shows that the cream has the technical effects of better wrinkle removal, better antianaphylaxis and better absorbability by screening the types of the components in the cream or adjusting the weight parts of the components in the cream.

the first agent comprises glycerol, butylene glycol, and sodium hyaluronate;
the second agent comprises tranexamic acid, ascorbic acid and dipotassium glycyrrhizinate;
the third agent comprises an extract of exosomes of umbilical cord mesenchymal stem cells.
2. The reagent combination according to claim 1, wherein the reagent combination comprises, in parts by weight,
the first agent comprises 1-6 parts of glycerol, 1-6 parts of butanediol and 0.1-1.5 parts of sodium hyaluronate.
3. The reagent combination according to claim 1, wherein the reagent combination comprises, in parts by weight,
the second reagent comprises 1-4 parts of tranexamic acid, 1-4 parts of ascorbic acid and 0.1-1 part of dipotassium glycyrrhizinate.
4. The reagent combination according to claim 1, wherein the reagent combination comprises, in parts by weight,
the third reagent comprises 0.1-1 part of umbilical cord mesenchymal stem cell exosome extract.
5. An agent combination for skin care comprising a first agent, a second agent and a third agent, wherein the first agent, the second agent and the third agent are, in parts by weight,
the first reagent comprises 1-6 parts of glycerol, 1-6 parts of butanediol and 0.1-1.5 parts of sodium hyaluronate;
the second reagent comprises 1-4 parts of tranexamic acid, 1-4 parts of ascorbic acid and 0.1-1 part of dipotassium glycyrrhizinate;
the third reagent comprises 0.1-1 part of umbilical cord mesenchymal stem cell exosome extract.
6. The combination of agents of claim 1, the first agent further comprising one or more of a thickening agent, an emulsifier, a surfactant, a skin conditioner, a grease, a fragrance, and a preservative, wherein,
the thickening agent is one or more selected from carbomer, xanthan gum, sodium carrageenan, Arabic gum, polyethylene glycol-14M, polyethylene glycol-90M, succinoglycan, SEPIPLUS 265, hydroxyethyl cellulose, hydroxypropyl cellulose and hydroxypropyl methyl cellulose;
