本发明属于生物领域,尤其涉及一种慢病毒载体、构建方法及其应用。本发明公开了一种慢病毒载体的制备方法,包括:以基于LTR1.27‑GEW质粒为骨架,在5’端添加R和U5区,同时删除3’端的PBS序列,并且将其中的5’CMV启动子更换为猕猴免疫缺陷病毒的U3启动子,得到慢病毒载体Obio‑02。本发明制备得到的新型的第四代慢病毒载体在保障安全性的前提下,还能够具有高产量的病毒滴度,可应用于基因治疗等领域。

一种慢病毒载体、构建方法及其应用
技术领域
本发明属于生物领域,尤其涉及一种慢病毒载体、构建方法及其应用。
背景技术
慢病毒载体是以 HIV-1 (人类免疫缺陷 I 型病毒)为基础发展起来的基因治疗载体。区别一般的逆转录病毒载体,它对分裂细胞和非分裂细胞均具有感染能力。慢病毒载体的研究发展得很快,研究的也非常深入,慢病毒载体可以将外源基因有效地整合到宿主染色体上,从而达到持久性表达。在感染能力方面可有效地感染神经元细胞、肝细胞、心肌细胞、肿瘤细胞、内皮细胞、干细胞等多种类型的细胞,从而达到良好的的基因治疗效果,在美国已经开展了临床研究,效果非常理想,因此具有广阔的应用前景。
慢病毒属于逆转录病毒科,但其基因组结构复杂,除基因gag、pol 和env这3个和单纯逆转录病毒相似的结构基因外,还包括4个辅助基因vif、vpr、nef、vpμ和2个调节基因tat 和 rev。HIV-1是慢病毒中最具特征性的病毒,第一个慢病毒载体系统即以此病毒为基础进行构建的。慢病毒载体的构建原理就是将 HIV-1 基因组中的顺式作用元件(如包装信号、长末端重复序列)和编码反式作用蛋白的序列进行分离。载体系统包括包装成分和载体成分:包装成分由HIV-1基因组去除了包装、逆转录和整合所需的顺式作用序列而构建,能反式提供产生病毒颗粒所需的蛋白;载体成分与包装成分互补,含有包装、逆转录和整合所需的HIV-1顺式作用序列。同时具有异源启动子控制下的多克隆位点及在此位点插入的目的基因。HIV-1的LTR区是由U3区,R区和U5区组成,为降低两种成分同源重组产生有复制能力的病毒(RCV)的可能性,第三代载体将包装成分的 5’LTR的U3区被换成巨细胞病毒(CMV)立即早期启动子或者RSV启动子,同时将3’LTR 的U3区进行缺失突变, 并添加 SV40 polyA位点等。将包装成分分别构建在两个质粒上,即一个表达基因gag和 pol、另一个表达基因env。根据这个原理构建了三质粒表达系统。
三质粒表达系统包括包装质粒、包膜蛋白质粒和穿梭质粒。其中包装质粒在CMV启动子的控制下,表达 HIV-1复制所需的全部反式激活蛋白,但不产生病毒包膜蛋白及辅助蛋白vpμ;包膜蛋白质粒编码水泡性口炎病毒 G蛋白(VSV-G),应用VSV-G包膜的假构型慢病毒载体扩大了载体的靶细胞嗜性范围,而且增加了载体的稳定性,允许通过高速离心对载体进行浓缩,提高了滴度;穿梭质粒中除含有包装、逆转录及整合所需的顺式序列,还保留约350 bp的gag和 RRE,并在其中插入目的基因或标志基因(绿色荧光蛋白 GFP)。将载体系统分成三个质粒最大的益处是使序列重叠的机会大大减少,减少载体重组过程中产生 RCV的可能性。通过三质粒共转染 293T 细胞,超速离心后病毒滴度可达 10^9 IU /ml。
为了减少 HIV-1 包装结构的序列同源性,进一步减少重组成RCV的可能性,Dull等人将辅助基因去除。但由于基因gag-pol的转运需要基因rev,因此,在上述的三质粒系统的基础上,构建成四质粒表达系统,该系统加上含基因rev的质粒减少了产生RCV的可能性,而且对非分裂期细胞转导效率无影响。tRNA 作为反转录引物与5’端 PBS结合,反转录产物,跳到3’端继续反转录最终获得的反转录产物是带有包装信号(ψ)、RRE等HIV基因组序列。
最近报道的第四代慢病毒,将5’端的R和U5区删除,包装信号(ψ)和RRE等序列放到了3’端相对三代病毒载体,反转录时候直接从3’端开始反转录,最终得到的反转录产物没有包装信号(ψ),RRE等HIV基因组序列,使整合到基因组中的序列只包含HIV-1基因组的序列的4.8%,进一步增强了安全性,但是随之带来的问题是滴度和产量的严重下降,根据报道仅能达到第三代慢病毒载体的35%,也正是因为这个原因,第四代慢病毒载体并没有在实践中得以应用。tRNA 作为反转录引物与3’端 PBS结合并反转录到5’端,最终获得的反转录产物不带有包装信号(ψ)、RRE等HIV基因组序列,安全性得到进一步提升。现有的已知的第四代慢病毒报道的滴度如图1所示,从图1可以看出,现有的第四代慢病毒滴度较低。
发明内容
本发明的目的在于,发明一种新型的慢病毒载体,其在不改变第四代慢病毒载体安全性的前提下,能大幅度提升病毒产量,可以达到三代慢病毒载体病毒产量的水平。
具体的,本发明的技术方案如下:
本发明第一个方面公开了一种慢病毒载体的制备方法,包括:
以基于LTR1.27-GEW质粒为骨架,在5’端添加R和U5区,同时删除3’端的PBS序列,得到慢病毒载体Obio-01。将慢病毒载体Obio-01中的5’CMV启动子更换为猕猴免疫缺陷病毒的U3启动子,得到慢病毒载体Obio-02。
所述基于LTR1.27-GEW质粒的核苷酸序列如SEQ ID NO:1所示,在本发明的一些优选实施例中,所述慢病毒载体Obio-01的核苷酸序列如SEQ ID NO:2所示。
在本发明的一些优选实施例中,所述慢病毒载体Obio-02的核苷酸序列如SEQ IDNO:3所示。
优选的,将慢病毒载体Obio-01中的5’CMV启动子更换为人免疫缺陷病毒-1的U3启动子,得到慢病毒载体Obio-03。
在本发明的一些优选实施例中,所述慢病毒载体Obio-03的核苷酸序列如SEQ IDNO:4所示。

优选的,所述制备病毒产品的方法包括:
S1、将包含上述的慢病毒载体共转染细胞;S2、收获病毒;S3、将上清液分离得到病毒产品。
更优选的,所述步骤S2包括:转染45-50小时后,第一次收获病毒,收集培养基并给细胞更换新鲜的完全培养基;转染70-75小时后,第二次收获病毒,收集培养基,丢弃细胞。
在本发明的一些具体实施例中,所述步骤S2包括:转染48小时后,第一次收获病毒,收集培养基并给细胞更换新鲜的完全培养基;转染72小时后,第二次收获病毒,收集培养基,丢弃细胞。
优选的,使用上述慢病毒载体进行病毒包装的方法,包括:
转染前一天,将293T细胞接种到培养皿中;转染前一小时取出细胞培养皿,去除原有细胞培养基,加入Opti-MEM培养基,将细胞放回培养箱;制备转染试剂和质粒的复合物,包括以下步骤:
a. 将待转染的病毒载体质粒(骨架质粒pCAG-gagpol-Tat、包膜蛋白质粒pHCMV-VSVG和穿梭质粒)溶于Opti-MEM培养基,轻轻混匀,静置后得到质粒稀释液;所述穿梭质粒为基于LTR1.27-GEW的质粒或Obio-01~Obio-04;
b. 将转染试剂溶于Opti-MEM培养基,轻轻混匀,静置后得到转染试剂稀释液;
c. 将转染试剂稀释液滴加到质粒稀释液中,边加边轻轻混匀后在室温放置15-25min,使DNA和转染试剂充分结合形成稳定的转染复合体;取出细胞培养皿,将准备好的DNA-转染试剂复合体加入到细胞培养皿中,放回培养箱;5-8h后吸去培养基,用PBS溶液洗涤后加入新鲜完全培养基培养。在本发明的一些具体实施例中,所述骨架质粒pCAG-gagpol-Tat、包膜蛋白质粒pHCMV-VSVG和穿梭质粒的质量比为2:1:3。
本发明第五个方面公开了上述的方法、上述的慢病毒载体、上述的细胞或上述的方法在基因治疗领域中的应用。
在符合本领域常识的基础上,上述各优选条件,可任意组合,而不超出本发明的构思与保护范围。

本发明发明人通过大量的创新劳动,发明了一种新型的第四代慢病毒载体,该慢病毒载体在保障安全性的前提下,还能够具有高产量的病毒滴度,可应用于基因治疗等领域,具有广阔的应用前景。

具体实施方式
下面结合附图和实施例对本发明的技术方案进行详细描述,但并不因此将本发明限制在所述的实施例范围之中。
下列实施例中未注明具体条件的实验方法,按照常规方法和条件,或按照商品说明书选择。本发明所用试剂和原料均市售可得。
实施例1
本发明以LTR1.27-GEW为骨架,构建了一系列载体,如图2所示。其中推测Obio-01的反转录过程分如图3所示,图3中:RNA genome指“RNA 基因组”,Minus strand strongstop synthesis指“负链强终止合成”,Minus strand transfer指“负链跳转”,Minusstrand extension指“负链延伸”,Plus strand strong stop synthesis指“正链强终止合成”,Plus strand transfer指“正链跳转”,dsDNA synthesis指“DNA 合成”。LTR1.27-GEW的质粒图谱如图4所示。Obio-01、Obio-02、Obio-03、Obio-04的质粒图谱分别如图5-8所示。所述Obio-01、Obio-02、Obio-03、Obio-04的核苷酸序列分别如SEQ ID NO:2-5所示。基于LTR1.27-GEW的核苷酸序列如SEQ ID NO:1所示。本实施公开了使用上述构建的质粒进行病毒包装的方法,具体如下:
转染前一天,将293T细胞接种到100mm培养皿中;转染前一小时取出细胞培养皿,去除原有细胞培养基,加入10ml的Opti-MEM培养基,将细胞放回培养箱;制备转染试剂和质粒的复合物,包括以下步骤:
a. 将待转染的病毒载体质粒32μg(骨架质粒pCAG-gagpol-Tat:包膜蛋白质粒pHCMV-VSVG:穿梭质粒的质量比为2:1:3)溶于Opti-MEM培养基,总体积为500μl,轻轻混匀,静置5分钟;所述穿梭质粒为基于LTR1.27-GEW的质粒或Obio-01~Obio-04;
b. 将转染试剂溶于Opti-MEM培养基,总体积为500μl,轻轻混匀,静置5分钟;
c. 将转染试剂稀释液滴加到质粒稀释液中,边加边轻轻混匀后在室温放置20min,使DNA和转染试剂充分结合形成稳定的转染复合体。取出细胞培养皿,将准备好的DNA-转染试剂复合体加入到细胞培养皿中,放回培养箱;6h后吸去培养基,PBS洗一次,加入10mL新鲜完全培养基培养。
实施例2
本实施例公开了病毒纯化的方法,具体包括:
转染48小时后,第一次收毒,收集培养基至50ml离心管中,并给细胞更换新鲜的完全培养基。转染72小时后,第二次收毒,收集培养基至50ml离心管中,丢弃细胞;
将收集的培养上清,用离心机3500rpm,10min,室温离心后,上清倒入新的50ml离心管;用超速离心机30,000rpm,4℃离心2h后,小心地弃去上清,离心管倒扣在灭菌过的吸水纸上,加入DPBS重悬沉淀,收集到1.5ml EP管中,-80℃冰箱保存。
实施例3
本实施例公开了一种滴度检测方法,具体包括:
采用Real time定量PCR法测定慢病毒滴度,具体步骤如下:
制备样品:按1×105细胞每孔接种293T细胞于24孔板;第二天加入病毒,12~20h后更换新鲜培养基;感染后72h拍照记录荧光,后收取细胞抽取基因组DNA并做定量PCR实验测定滴度。
Real-time PCR 在ABI7500仪器上完成。使用试剂SYBR Master Mixture来自TAKARA;
1.按下列比例配置反应体系:
SYBR premix ex taq:10 μl;
ROX:0.4μl;
上游引物(25μM):0.5μl;
下游引物(25μM):0.5μl;
Genomic DNA:2.0μl;
水6.6μl;
2.设定程序为两步法Real-Time定量。预变性95℃,15S,之后每一步变性95℃,5S,退火延伸60℃,34S,共进行40个循环。每次在延伸阶段读取吸光值;
PCR程序为:
Cycle 1:(1X)
Step 1:95.0 ℃ for 00:15;
Cycle 2:(40X)
Step 1:95.0 ℃ for 00:05
Step 2:60.0 ℃ for 00:34;
启用数据收集和实时分析;
3.制作熔解曲线。PCR 结束后,在95℃变性1min。然后冷却至55℃,使DNA 双链充分结合。从55℃开始到95℃,每一步增加0.5℃,保持30S,同时读取吸光值;
制作溶解曲线程序为:
Cycle 3:(1X)
Step 1:95.0 ℃ for 01:00;
Cycle 4:(1X)
Step 1:55.0 ℃ for 01:00;
Cycle 5:(81X)
Step 1:55.0 ℃-95.0℃ for 00:30
循环2次后将设定点温度提高0.5℃。
将pCLenti-CMV-EGFP-WPRE(第三代慢病毒载体)、基于LTR1.27-GEW、Obio-01、Obio-02、Obio-03和Obio-04包装好的慢病毒按照0.1ul、1ul和10ul体积分别感染293T细胞,感染72h后记录荧光图片,荧光结果如图9所示,慢病毒载体Obio-02的荧光亮度优于基于LTR1.27-GEW和第三代慢病毒载体,表明滴度更优。同时收取感染后的细胞抽取细胞基因组DNA并做定量PCR实验测定慢病毒滴度,滴度统计结果如图10所示,病毒滴度柱状图显示,慢病毒载体Obio-02的滴度优于基于LTR1.27-GEW和第三代慢病毒载体。
实施例4
本实施例公开了一种慢病毒安全性的检测方法,具体包括:
将pCLenti-CMV-EGFP-WPRE(第三代慢病毒载体)、基于LTR1.27-GEW、Obio-01、Obio-02、Obio-03和Obio-04包装好的慢病毒分别感染293T细胞,48小时后抽提细胞基因组DNA,利用PCR方法分别检测质粒和细胞基因组DNA相关元件:土拨鼠肝炎病毒转录后调控序列(WPRE)、包装信号(Psi,Ψ)、Rev反应元件(RRE)及各自载体U5端的启动子(CMV、猕猴免疫缺陷病毒(SIV)和人免疫缺陷病毒(HIV1、HIV2))的情况。具体步骤如下:
1、细胞感染
将293T细胞铺于24孔板;第二天分别加入pCLenti-CMV-EGFP-WPRE(第三代慢病毒载体)、基于LTR1.27-GEW、Obio-01、Obio-02、Obio-03和Obio-04包装好的慢病毒,放入37℃、5% CO2培养箱中培养;感染48h后收集细胞;
2、抽提细胞基因组DNA
将步骤1收集的细胞,使用基因组 DNA 小量抽提试剂盒(Axygen,AP-MN-MS-GDNA-250G)提取基因组DNA;
3、PCR检测
将pCLenti-CMV-EGFP-WPRE(第三代慢病毒载体)、基于LTR1.27-GEW、Obio-01、Obio-02、Obio-03和Obio-04质粒及细胞基因组DNA采用表1中引物,进行PCR。PCR具体反应体系(50μl)为DNA模板1μl、正反向扩增引物(F/R)各1μl,2xHieFF PCR mix(上海翊圣生物)25μl,ddH2O 22μl;PCR条件为95℃,5min,1cycle;95℃,30s、55℃,30s、72℃ 40s35cycles;72℃ 5min,1cycle;4℃ 5min,1cycle。

Claims (6)

1.一种慢病毒载体的制备方法,其特征在于,包括:
以基于LTR1.27-GEW质粒为骨架,在5’端添加R和U5区,同时删除3’端的PBS序列,并且将其中的5’CMV启动子更换为猕猴免疫缺陷病毒的U3启动子,得到慢病毒载体Obio-02。
2.根据权利要求1所述的方法得到的慢病毒载体。
3.一种细胞,其特征在于,所述细胞通过权利要求2所述的慢病毒载体感染得到。
4.一种制备病毒产品的方法,其特征在于,将细胞与有效量的权利要求2所述的慢病毒载体接触,以产生病毒产品。
5.根据权利要求4所述的方法,其特征在于,包括:
S1、将包含权利要求2所述的慢病毒载体共转染细胞;S2、收获病毒;S3、将上清液分离得到病毒产品。
6.根据权利要求5所述的方法,其特征在于,所述步骤S2包括:转染45-50小时后,第一次收获病毒,收集培养基并给细胞更换新鲜的完全培养基;转染70-75小时后,第二次收获病毒,收集培养基,丢弃细胞。
