Abstract

本发明公开了一株金针菇菌种‘农万金8号’,属于金针菇属(Flammulina velutipes),已保藏于中国微生物菌种保藏管理委员会普通微生物中心。由于本发明所提供的‘农万金8号’菌种具有菇体洁白、菌盖略大、菌柄略粗,菇柄坚挺,基部绒毛较少等优良金针菇菌种的品质特点。改良了其亲本F0321菌盖较大、较薄,菌柄较软,商品感观较差等缺点,以及另一亲本F0323栽培管理难度大,容易出现菌被、顶芽等缺点,比对照菌株‘农金3号’(Flv0303)增产28.6%,商品外观良好,适合于市场需求,综合性状优良并适合进行工厂化生产,具有良好的推广前景。

一种适合工厂化栽培的金针菇及其分子鉴定方法
技术领域
本发明涉及一株金针菇菌种。具体涉及一种适合工厂化栽培的金针菇农万金8号及其分子鉴定方法。
背景技术
金针菇[Flammulina velutiper (Fr.) Sing] 隶属于担子菌亚门、层菌纲、伞菌目、白蘑科、金针菇属,其菌盖滑嫩、柄脆、营养丰富、味美适口而著称于世。福建省是中国南方金针菇的主栽地区。随着金针菇栽培规模的扩大,市场上对金针菇在栽培,颜色和产量上也产生了越来越高的评判标准,如,菌柄粗细和均匀与否、剖切面空隙、菌柄脆度、保鲜持久度、菌盖大小、菇体颜色等评价标准。目前菌柄均匀、菇体密集、菇体白成为金针菇市场的主要趋势。现有的金针菇主要栽培品种中,‘农金3号’(Flv0303)菌盖白色、半球形、圆整、边缘内卷,相对湿度低于80%时,菌盖中央略显淡黄色。菌柄白色、圆柱形、中空,基部有绒毛,2012年福建省作物品种审定委员会认定,该品种适宜福建省工厂化袋式栽培。但随着市场的发展,市场对金针菇品质要求越来越高,要求白色金针菇不仅要产量高,色白还要口感脆嫩,致使生产企业后备品种不足的问题日益凸显,选育品质优、产量高、商品性佳、市场前景好的优良品种显得尤为重要。
发明内容
本发明要解决现有生产中使用的金针菇品种单产低,出菇不整齐,色偏黄等的技术问题,从而提供一株芽数多,菇体整齐,菇体白,菇柄口感脆度好,品质优、产量高的金针菇菌种。
本发明的金针菇菌种‘农万金8号’,属于金针菇属 (Flammulina velutipes ),该菌株的分类命名为:金针菇(Flammulina velutipes ) 农万金8号保藏于中国微生物菌种保藏管理委员会普通微生物中心,保藏地址为北京市朝阳区北辰西路1号院3号,保藏日期为 2020 年6月15日,保藏编号为CGMCC NO.20203。
‘农万金8号’菌种在20℃、PDA 培养基上菌丝生长最快,菌丝最强壮、均匀,尖端菌丝较整齐、粗壮,菌落边缘较圆整。子实体菌盖略大、菌柄略粗 ,菇体坚挺,基部绒毛少,菇体颜色比对照品种农金3号更稳定,商品外观良好,适合于市场需求。
‘农万金8号’由金针菇‘F0321’和‘F0323’通过杂交育成,杂交菌株‘农万金8号’编号为‘Flv0012’,筛选后进入栽培试验。F0321具有产量较高、菇根白、周期较短等特点,但是菌盖较大、较薄,菌柄较软,商品感观较差。国内工厂化栽培(瓶栽)主要品种,源于日本。F0323的优点是菌盖较小、厚实,产量高。但该品种栽培管理难度大,容易出现菌被、顶芽等问题,一旦出现,大幅度减产或无收成。该品种的栽培周期比F0321长2-3天。国内工厂化栽培(瓶栽)主要品种,源于日本。对照品种F0303,经2012年福建省作物品种审定委员会认定并推广的福建省工厂化袋式栽培品种,该品种菌盖白色、半球形、圆整、边缘内卷,相对湿度低于80%时,菌盖中央略显淡黄色。菌柄白色、圆柱形、中空,基部有绒毛。多次栽培试验表明,杂交菌株‘Flv0012’比对照菌株‘农金3号’(Flv0303)增产28.6%,其菌盖大小、菌盖厚度和菌柄直径的数据均优于对照菌株‘农金3号’(Flv0303)。对于商品外观,(Flv0012)菇体洁白、菌盖略大、菌柄略粗,菇柄坚挺,基部绒毛较少,商品外观良好,适合于市场需求。定名为‘农万金8号’(图1)。
从图2的拮抗结果可看出,杂交子农万金8号与亲本F321、F0323以及对照菌株F0303无明显的拮抗线,但F0012的菌丝形态明显区别于F0321和F0303,但与F0323无明显区别,F0012与亲本F0321、F0323以及对照菌株F0303是否为不同菌株需通过分子手段进一步验证。
本发明的金针菇菌种根据基因组测序数据采用Kmer差减组装技术,获得差异片段(SV)(图4),选取10个SV序列设计特异性引物10对(P1~P10),对F0321、F0012和F0323进行扩增,除了P3没有扩增成功外,其它引物均获得PCR产物,扩增结果见图5。各特异性片段的PCR扩增结果分析如图3所示。F0321有7个特异片段(P1、P2、P4、P5、P8、P9和P10),F0323有2个特异片段(P6和P7),而F0012继承了两亲本的特异片段,进一步证明它是杂交子。根据省食用菌品种鉴定技术规程的要求,我们把省认定的2个金针菇品种也进行了DNA指纹检测。结合‘F0306’的基因组数据,进一步开发了9对特异性扩增引物(P11~P19),对五个品种进行扩增和产物测序,扩增结果见图5。各特异性片段的PCR扩增结果分析如图3所示。从图3可以看出,P12、P14和P15是‘农金3号’和‘农金6号’的特异序列;P16、P17、P18、P19是‘F0321’、‘F0323’和‘农万金8号’的特有序列;P11和P13是‘农万金8号’区别于其它品种的序列。综合上述检测结果,可以确定F0012是‘F0321’和‘F0323’的杂交子,它与亲本及省认定品种的DNA指纹有显著的差异,是不同的品种。
本发明的优点在于:本发明所提供的‘农万金8号’(Flv0012)平均产量365.35g/瓶,对照品种‘农金3号’(Flv0303)平均产量为284.02g/瓶,“农万金8号”(Flv0012)比对照菌株增产28.6%,其菌盖大小、菌盖厚度和菌柄直径的数据均优于对照菌株’农金3号’(Flv0303)。对于商品外观,‘农万金8号’(Flv0012)菇体洁白、菌盖略大、菌柄略粗,菇柄坚挺,基部绒毛较少,符合市场需求。

具体实施方式
实施例1:选用主栽的金针菇‘F0321’和‘F0323’做杂交亲本,选取亲本‘F0321’单孢菌株20个,‘F0323’单孢菌株20个生长速度较快、气生菌丝较发达的单孢菌株,按照全组合方式配对,共配制获得60个组合,经镜检含有锁状联合的杂交子数量47份,进行初筛出菇实验后,选择能够出菇的杂交子20个进入复筛。进过复筛后有14个杂交子正常出菇,产量高于对照菌株’农金3号’的杂交子数量有13个,但产量高于400g/瓶的菌种仅有3个,其中Flv0012综合性状较好,达到预期育种目标,该杂交菌株定名为“农万金8号”,用于开展后续的批次试验。
以拮抗反应法对‘农万金8号’菌株与其亲本进行差异对比。
检测依据:依据中华人民共和国农业行业标准《NY/T 1845-2010 食用菌菌种区别性鉴定 拮抗反应》进行待检样品的接种、培养和观察。
检测材料:金针菇菌株4个,其中待检菌株为‘农万金8号’,亲本F0321、F0323以及对照菌株F0303。
检验方法:严格按照无菌操作,用Φ8 mm 打孔器分别打取活化后适龄菌种菌落边缘作接种块,两接种块间隔10 mm,菌丝朝上。在20℃的温度下,通风、避光培养至接种块菌丝接触后,再在自然光下培养7d。
结果与分析:在灯下或自然光下,观察培养物表面两菌株交界处菌丝反应,结果如图所示(图2)。
检测结论:从图2的拮抗结果可看出,‘农万金8号’,亲本F0321、F0323以及对照菌株F0303之间的菌丝均存在较明显差异,菌丝形态完全不同,F0321、F0323菌丝偏稀疏,F0303菌丝较浓密,‘农万金8号’菌丝最浓密。但不同菌株间的拮抗线不太明显。
实施例2:用SV检测技术对本菌株进行与其亲本的DNA差异对比。
检测材料:金针菇菌株5个,其中待检菌株为 ‘农万金8号’,对照菌株 F0321、和F0323、F0303(农金3号)、农金六号。
检验方法:根据基因组测序数据,采用Kmer差减组装技术,获得差异片段(SV),相应选取若干个SV序列设计特异性引物对待测样品进行扩增。(参考文献见 Alkan, C.,Coe, B. P. & Eichler, E. E. Genome structural variation discovery andgenotyping. Nature reviews. Genetics, 2011 (12):363–376;朱风丽等, 韩国赤芝全基因组重测序分析,世界科学技术-中医药现代化,21(4):764-774.)。SV试验引物序列由擎科生物技术有限公司合成。
待检菌株菌丝体活化,培养,基因组DNA提取,电泳检测均参照中华人民共和国农业行业标准进行。

实施例3:‘农万金8号’共进行了多次稳产性试验,结果表明,‘农万金8号’具备工厂化栽培的要求,产量显著高于亲本F0303(农金3号)。
稳产性试验参试菌株:‘农金3号’(Flv0303)。
试验地点:福建万辰生物科技股份有限公司。
试验安排:菌种统一制作,试验点按照当地生产管理方式进行工厂化栽培,瓶栽,采用多区制移动床架模式栽培(塑料瓶,0.43kg(干料)/瓶)。
试验结果:试验制种时期‘农万金8号’菌丝在 PDA 中菌丝较浓密,最适生长温度为 20 ℃。‘农万金8号’栽培结果如图1所示。
栽培发现:(1)接种到采收的时间,‘农万金8号’与对照菌株‘农金3号’相同均为45天;(2) ‘农万金8号’(Flv0012)平均产量365.35g/瓶,对照品种‘农金3号’(Flv0303)平均产量为284.02g/瓶,“农万金8号”(Flv0012)比对照菌株增产28.6%,(3)‘农万金8号’(Flv0012)其菌盖大小、菌盖厚度和菌柄直径的数据均优于对照菌株‘农金3号’(Flv0303)。(4) 对于商品外观,‘农万金8号’(Flv0012)菇体洁白、菇柄坚挺,基部绒毛较少,符合市场需求的。

菌株发菌速度统计:两个品种菌丝生长速度相近,出菇时间一样,从接种到采收均为45天,两个品种没有差异。
‘农万金8号’(Flv0012):带根鲜菇平均产量为400.5g/瓶,‘农金3号’(Flv0303):带根鲜菇平均产量为278.9g/瓶,‘农万金8号’(Flv0012)产量比对照菌株‘农金3号’(Flv0303)高43.6%。

1.一种金针菇(Flammulina velutipes)菌株农万金8号,其特征在于,该菌株的分类命名为:金针菇(Flammulina velutipes)农万金8号,已于2020年6月15日保藏于中国微生物菌种保藏管理委员会普通微生物中心,保藏编号为CGMCC NO.20203;
所述金针菇农万金8号的亲本为金针菇F0321和F0323。
2.一种如权利要求1所述金针菇菌株农万金8号的真实性鉴定方法,其特征在于,根据基因组测序数据,采用Kmer差减组装技术,获得差异片段SV,相应选取SV序列设计特异性引物对待测样品进行扩增,判断与亲本DNA指纹的差异性;
