本发明公开一种检测氟唑环菌胺药物残留的试剂盒,沿样本液流动方向依次设置有加样区、反应膜设置区和吸水垫设置区,所述加样区内设有样本垫,所述反应膜设置区内设有反应膜,所述吸水垫设置区内设有吸水垫,所述样本垫和所述反应膜之间依次设有结合垫和加速垫,所述样本垫上设有样本孔,所述反应膜上沿样本液流动方向依次设有包被有氟唑环菌胺抗原的T线和包被有载体蛋白抗体的C线。本发明专利样本前处理快速、简单,操作简便,检测快速、准确、灵敏、稳定,操作人员技术含量要求不高,减少了检验样本所需要的时间,成本较低。

技术领域
本发明涉及化合物检测技术领域,特别涉及一种检测农产品中氟唑环菌胺药物残留的试剂盒,具体的说是应用胶体金间接竞争法的原理来达到检测农产品中氟唑环菌胺药物残留的目的,为氟唑菌酰胺环境残留监测及安全使用提供技术支撑。
背景技术
氟唑环菌胺是一种吡唑酰胺类杀菌剂,也可以作为种子处理杀菌剂, 化学名称:2-顺式异构体与2-反式异构体的混合物:2'-[(1RS,2RS)-1,1'-双向环丙-2-基]-3-(二氟甲基)-1-甲基吡唑-4-碳酰苯胺;2'-[(1RS,2SR)-1,1'-双向环丙-2-基]-3-(二氟甲基)-1-甲基吡唑-4-碳酰苯胺,分子结构C72H76F8N12O4,英文通用名称: Sedaxane。氟唑环菌胺高效、广谱、内吸,具有保护和治疗作用,以保护作用为主。它能预防土壤中的多种病害,尤其是对病原菌丝核菌,这些病菌难以被检测到又具有一定的侵袭性,氟唑环菌胺对其都有特效。适用于谷物、大豆、玉米、蔬菜、甘蔗、葡萄、马铃薯、棉花、梨果和油菜等众多作物,防治多种土传和种传病害,也可防治早期叶面病害,对丝核菌引起的病害和丝黑穗病防效优异。氟唑环菌胺低毒,用作种衣剂时,对多种种传、土传病害有较好的防治效果,能促进作物根系生长,降低非光化学淬灭,使作物增产,还可促进作物根系的生长。但是近些年来,由于农业工作者的不规范使用导致氟唑环菌胺在农产品中的残留量超标严重,对人体造成了巨大的危害,为了加强农药残留管理工作,保证农产品质量安全,西方国家分别制定了氟唑环菌胺药物残留的标准,其中美国规定残留限量范围为0.01-0.075ppm,因此加强对农产品中的氟唑环菌胺药物残留检测是非常必要的。
目前关于氟唑环菌胺药物残留的检测主要应用的是液相色谱-串联质谱法、气相色谱分析方法、高效液相色谱法等,应该说色谱技术对头氟唑环菌胺药物检测是非常有效、准确、敏感的。但待检测样本需要经过一系列的处理,繁琐费时,从样本前处理到得出检测结果需要较长的时间;同时色谱法需要价格昂贵的仪器设备,且需专业人员操作,严重阻碍了这种检测方法的普及推广,更无法实现现场检测。
目前免疫分析领域缺乏能够快速检测氟唑环菌胺药物残留的技术及其应用。因此,在实践中有必要建立一种敏感度高、操作简单、成本低、时间短,适合大规模的检测方法。
发明内容
本发明的目的是提供一种检测农产品中氟唑环菌胺药物残留的试剂盒,用于氟唑环菌胺药物的检测,试剂盒特异性好、敏感性高,检测成本低廉,适于推广。
为解决上述技术问题,本发明的技术方案为:
本发明提供一种检测农产品中氟唑环菌胺药物残留的试剂盒,沿样本液流动方向依次设置有加样区、反应膜设置区和吸水垫设置区;所述加样区内设有样本垫,所述反应膜设置区内设有反应膜,所述吸水垫设置区内设有吸水垫,所述样本垫和所述反应膜之间依次设有结合垫和加速垫,所述样本垫上设有样本孔,所述反应膜上沿样本液流动方向依次设有包被有氟唑环菌胺抗原的T线和包被有载体蛋白抗体的C线。
其中,优选地,所述T线与所述C线平行,并与样本液的流动方向垂直。
其中,优选地,所述吸水垫上覆盖有封盖。
其中,优选地,所述样本垫的长度为16mm,所述结合垫和所述加速垫的长度均为4mm,所述反应膜的长度为25mm,所述吸水垫的长度为16mm。
其中,优选地,所述T线距所述加速垫6-8mm,所述C线距所述说吸水垫4-6mm。其中,优选地,所述反应膜、所述样本垫、所述吸水垫通过双面胶固定在底板上。
本发明的有益效果: (1)操作、快速,检测过程中无需其他试剂,在5-8分钟内即可判定结果。
(2)样本前处理快速、简单,可同时批量处理样本。
(3)结果判定简单明了。检测卡以肉眼可见的红色检测印迹作为判定标准。一条C线印迹表示被检样本中含有氟唑环菌胺药物残留, C、T线两条印迹表示被检样本中不含氟唑环菌胺药物残留。
(4)检测成本低,应用价值高,人人可以操作;具有较高的社会应用价值和经济效益,可在食品安全检测中发挥重大作用,适用于现场快速检测,是国标方法强有力的辅助手段。

下面将结合本发明具体实施例,对本发明的技术方案进行清楚、完整地描述,显然,所描述的实施例仅仅是本发明一部分实施例,而不是全部的实施例。基于本发明中的实施例,本领域普通技术人员在没有做出创造性劳动前提下所获得的所有其他实施例,都属于本发明保护的范围。
如图1所示,本实施例提供检测氟唑环菌胺药物残留的试剂盒,沿样本液流动方向依次设置有加样区、反应膜设置区和吸水垫设置区,加样区内设有样本垫3,反应膜设置区内设有反应膜2,吸水垫设置区内设有吸水垫6,样本垫3和反应膜2之间依次设有结合垫4和加速垫5,样本垫3上设有样本孔10,反应膜2上沿样本液流动方向依次设有包被有氟唑环菌胺抗原的T线8和包被有载体蛋白抗体的C线9,并且具有氟唑环菌胺的特异性抗体为单克隆抗体。载体蛋白抗体为羊抗鼠多克隆抗体。
其中,T线8与C线9平行,并与样本液的流动方向垂直。T线8靠近加速垫5,C线9可靠近吸水垫6。
其中,吸水垫6上覆盖有封盖7。封盖7为防水材质的,用于固定检测卡的各个组件,避免样本试剂与外界环境中的物质发生反应,影响检测卡的检测效果。
其中,样本垫3的长度为16mm,结合垫4和加速垫5的长度均为4mm,反应膜2的长度为25mm,吸水垫6的长度为16mm。
其中,T线8距加速垫7mm,C线9距说吸水垫5mm。
其中,反应膜2、样本垫3、吸水垫6通过双面胶固定在底板1上。
本发明的检测原理:
当样本溶液中含有氟唑环菌胺时,在扩散过程中,样本中的氟唑环菌胺通过竞争结合抑制了胶体金标记的特异性抗体同T线上的具有氟唑环菌胺抗原分别结合,T线无法显色,而C 线的载体蛋白抗体可与胶体金标记的特异性抗体分别结合,C线即显示红色印迹。当样本中不含氟唑环菌胺或其含量低于检测卡检测限时,胶体金标记的特异性抗体与T线上的具有氟唑环菌胺抗原分别结合,T线显示红色印迹,液体渗透到C线时,C线上的抗载体蛋白抗体可与胶体金标记的特异性抗体结合,C线显示红色。

1.一种检测农产品中氟唑环菌胺药物残留的试剂盒,其特征在于:沿样本液流动方向依次设置有加样区、反应膜设置区和吸水垫设置区,所述加样区内设有样本垫,所述反应膜设置区内设有反应膜,所述吸水垫设置区内设有吸水垫,所述样本垫和所述反应膜之间依次设有结合垫和加速垫,所述样本垫上设有样本孔,所述反应膜上沿样本液流动方向依次设有包被有氟唑环菌胺抗原的T线和包被有载体蛋白抗体的C线。
2.根据权利要求1所述的一种检测农产品中氟唑环菌胺药物残留的试剂盒,其特征在于:所述T线与所述C线平行,并与样本液的流动方向垂直。
