Abstract
本发明公开了青蒿素改善玫瑰痤疮炎症用动物和细胞模型的构建方法,涉及皮肤性疾病药剂技术领域,具体为青蒿素改善玫瑰痤疮炎症用动物和细胞模型的构建方法,包括以下步骤:(1)青蒿素改善玫瑰痤疮炎症表型的动物模型构建;(2)青蒿素改善玫瑰痤疮炎症表型的细胞模型构建。本发明的优点在于发现了一种治疗玫瑰痤疮潜在新药(青蒿素),并且对玫瑰痤疮的发病机制进行了探索,本发明发现青蒿素能明显改善LL37诱导的玫瑰痤疮样炎症小鼠皮损红斑、组织病理学改变及IL‑1β、IL‑6、TNFα、TLR2的升高,同时,青蒿素可减少皮损CD4+T细胞、巨噬细胞和中性粒细胞浸润,抑制皮损中免疫细胞相关趋化因子(CXCL10、CCL20、CCL2和CXCL2)的表达。

技术领域
本发明涉及皮肤性疾病药剂技术领域,具体为青蒿素改善玫瑰痤疮炎症用动物和细胞模型的构建方法。
背景技术
玫瑰痤疮是一种慢性面部皮肤炎症性疾病,主要表现为面部隆突部位反复发作的阵发潮红、持久性红斑、毛细血管扩张、丘疹脓疱、鼻部肥大增生等症状。玫瑰痤疮主要发生在浅肤色人种中,流行病学调查显示在不同国家玫瑰痤疮的患病率从2%到22%不等。目前玫瑰痤疮的详细发病机制尚未完全阐明,但许多因素包括紫外线、饮食、热刺激、微生物及应激状态等都被认为可诱发或加重玫瑰痤疮。虽然玫瑰痤疮并不威胁患者的生命,但是由于皮损位于面部并且往往合并反复的灼热、刺痛、瘙痒等症状,所以其对患者心理、正常社交及生活都照成明显的影响,流行病调学查也显示玫瑰痤疮患者生活质量出现明显降低。虽然玫瑰痤疮具体的病理生理机制尚不清楚,但积累的研究证据显示除了遗传因素,面部皮肤局部免疫过度激活及神经血管功能失调是玫瑰痤疮重要发病机制。
虽然玫瑰痤疮的病理生理学机制尚未完全阐明,但是目前的证据表明玫瑰痤疮多年来一直是免疫、血管和神经功能障碍的原因。在玫瑰痤疮中固有免疫和适应性免疫均明显激活,特别是Toll样受体2(TLR2)和抗菌肽(CAmp) 在局部皮肤表达升高,并且皮损中中性粒细胞、巨噬细胞、CD4T细胞均有明显浸润;另外患者皮损中血管扩张和增殖,特别是在鼻赘型患者中最为明显。同时,基因组和qPCR结果显示血管内皮生长因子(VEGF)和神经源性炎症基因 (TRP),香草醛受体1(TRPV1)在玫瑰痤疮中升高。因此,美国FDA批准的玫瑰痤疮临床药物包括多西环素和α-肾上腺素受体激动剂,这些药剂的临床功效归因于它们的抗炎或抗血管生成特性。
青蒿素(Artemisinin,ART)是从青蒿中分离出来的一种抗疟药物。目前青蒿素已治愈超过一百万的疟疾患者并且未显示出明显的副作用和不良反应。除了抗疟,因具有免疫调节作用青蒿素也用于免疫性疾病如系统性红斑狼疮,同时临床和基础研究也发现青蒿素具有抗炎和抗血管生成的作用。在上述研究中,青蒿素被用于治疗红斑狼疮、关节炎等自身免疫性疾病和肿瘤性疾病。另外,玫瑰痤疮的基础研究也证实面部皮内炎症激活如固有免疫中的中性粒细胞、巨噬细胞和肥大细胞,适应性免疫中的CD4+T细胞等均呈明显浸润,而面部毛细血管扩张和增殖更是玫瑰痤疮特征性的病理改变,在病人中导致明显的红斑表型。所以基于已有的研究发现,我们推测青蒿素对玫瑰痤疮具有良好的治疗作用。
由于青蒿素对玫瑰痤疮治疗效果尚不清楚,需要构建玫瑰痤疮小鼠模型进行药物干预明确青蒿素对玫瑰痤疮炎症表型和病理改变的影响,青蒿素虽然具有抗炎和抗血管增殖的作用,但是如何在玫瑰痤疮中发挥这些作用,具体的机制也尚不清楚,所以,需要通过各种技术手段在细胞层面来检测细胞相关的蛋白质、细胞因子,之后再研究这些分子的作用机制,靶细胞等,由于是一个逆向研究的过程,因此早期需要花费大量的人力物力进行大筛选工作的缺点。
发明内容
针对现有技术的不足,本发明提供了青蒿素改善玫瑰痤疮炎症用动物和细胞模型的构建方法,解决了上述背景技术中提出现有由于青蒿素对玫瑰痤疮治疗效果尚不清楚,需要构建玫瑰痤疮小鼠模型进行药物干预明确青蒿素对玫瑰痤疮炎症表型和病理改变的影响,青蒿素虽然具有抗炎和抗血管增殖的作用,但是如何在玫瑰痤疮中发挥这些作用,具体的机制也尚不清楚,所以,需要通过各种技术手段在细胞层面来检测细胞相关的蛋白质、细胞因子,之后再研究这些分子的作用机制,靶细胞等,由于是一个逆向研究的过程,因此早期需要花费大量的人力物力进行大筛选工作的问题。
为实现以上目的,本发明通过以下技术方案予以实现:青蒿素改善玫瑰痤疮炎症用动物和细胞模型的构建方法,包括以下步骤:
(2)青蒿素改善玫瑰痤疮炎症表型的动物模型构建
改善玫瑰痤疮症状的药物的动物模型为7周龄BALB/C小鼠,用青蒿素每天以200mg/kg灌胃,连续七天,在第5天小鼠背部皮肤剃毛,第6-7天给予背部皮肤皮下注射LL37多肽以诱导玫瑰痤疮样炎症;
(2)青蒿素改善玫瑰痤疮炎症表型的细胞模型构建
步骤一、化合物:
氨基酸序列为:LLGDFFRKSKEKIGKEFKRIVQRIKDFLRNLVPRTE,然后通过HPLC纯化,并通过质谱鉴定;
步骤二、青蒿素干预HACAT细胞炎症模型:将HACAT细胞置于30mm培养皿中用含10%血清的DMEM完全培养基在37℃、5%的二氧化碳的培养箱中培养待细胞达到90%融合度后按照实验目的,分为空白组—不做任何处理,青蒿素组—青蒿素(50μM/L)刺激12h,LL37组—LL37(4μM/L)刺激12h,青蒿素+LL37组—青蒿素(50μM/L)刺激1h后,加入LL37(4μM/L)再同时刺激12h。之后收集RNA,储存在-80℃直至分析;
步骤三、青蒿素影响HACAT细胞炎症机制模型:将HACAT细胞种板于含有细胞爬片的24孔板中用含10%血清的DMEM完全培养基在37℃、5%的二氧化碳的培养箱中培养待细胞达到90%融合度后按照实验目的,分为空白组—不做任何处理,青蒿素组—青蒿素(50μM/L)刺激5h,LL37组—LL37(4μM/L) 刺激5h,青蒿素+LL37组—青蒿素(50μM/L)刺激1h后,加入LL37(4μM/L) 再同时刺激5h,之后,取出细胞爬片,行免疫组织荧光染色检测并分析HACAT 细胞P65蛋白的核转位;
步骤四、青蒿素影响人脐静脉内皮细胞趋化模型:24孔板中放入500μL 含10%血清的1640完全培养基,根据实验需要,设置空白组、青蒿素组、LL37 组、青蒿素+LL37组;在LL37组和青蒿素+LL37组中,24孔板加入4μM/L的 LL37,将趋化小室(8mm孔径)置入24孔板中,并种入HUVECs细胞(2×103),小室中的1640培养基为200μL且不含血清;根据实验需要,向青蒿素组和青蒿素+LL37组小室中加入青蒿素(50μM/L),将24孔板置于37℃、5%CO2 细胞培养箱中培育,24小时后取出24孔板,清洗固定后用0.5%的结晶紫染色30分钟,清洗并晾干小室后于显微镜下观测粘附于小室下层膜上的细胞并拍照计数。
优选的,所述步骤(2)青蒿素改善玫瑰痤疮炎症表型的细胞模型构建还包括青蒿素影响人脐静脉内皮细胞(HUVECs)迁移模型:HUVECs(每孔5x105) 接种到6孔板中,根据实验需要,设置空白组、青蒿素组、LL37组、青蒿素 +LL37组,在LL37组和青蒿素+LL37组中;当细胞生长到100%融合时,用无菌1mlTip头在皿底做划痕,PBS冲洗每个孔,随后加入含10%血清的1640完全培养基,并在青蒿素组和青蒿素+LL37组加入青蒿素(50μM/L),LL37组和青蒿素+LL37组加入LL37(4μM/L),将HUVECs细胞在37℃、5%CO2的培养箱中培养24小时,然后用3个随机选择的区域的划痕间距评价细胞的迁移能力。
优选的,所述步骤(2)青蒿素改善玫瑰痤疮炎症表型的细胞模型构建还包括组织学:用石蜡固定玫瑰痤疮样炎症皮损的小鼠背部皮肤,切成4μm厚;切片用苏木精和伊红(H&E)染色染色,并且如前所述在标准光学显微镜 (OLYMPUS,JApAn)下观察它们的组织形态学。
优选的,所述步骤(2)青蒿素改善玫瑰痤疮炎症表型的细胞模型构建还包括皮肤组织免疫荧光:在室温下用多聚甲醛固定8mm冷冻切片15分钟,并用5%正常驴血清和0.2%TritonX在室温下封闭持续1小时;将皮肤切片用抗CD4、F4/80、LY6G和CD31抗体在4℃下处理过夜,用AlexAFluor488标记的抗山羊IgG抗体常温染色1h,使用ZeissAxioScopeA1捕获图像。
优选的,所述步骤(2)青蒿素改善玫瑰痤疮炎症表型的细胞模型构建还包括实时PCR:使用TRIzol试剂(InvitrogenLifeTechnologies,USA)从细胞或皮损组织中分离总RNA,使用PrimeScriptRT试剂盒(TAkArA,ShigA,JApAn) 将2μgRNA转录成cDNA,并在AppliedBiosystems7500机器 (LifeTechnologies)上用iTAqUniversAlSYBRGREENSupermix(Bio-RAd, CAliforniA,USA)进行qPCR。
优选的,所述步骤(2)青蒿素改善玫瑰痤疮炎症表型的细胞模型构建还包括数据统计:实验中的数据都用均数±标准误(SEM)的形式表示,使用 GrAphPAdPrism5软件作图并用其中的t检验检测两组数据的差异,取双侧检测标准、并以*P<0.05,**P<0.01和***p<0.001被认为具有显著性差异。
本发明提供了青蒿素改善玫瑰痤疮炎症用动物和细胞模型的构建方法,具备以下有益效果:
本发明的优点在于发现了一种治疗玫瑰痤疮潜在新药(青蒿素),并且对玫瑰痤疮的发病机制进行了探索,本发明发现青蒿素能明显改善LL37诱导的玫瑰痤疮样炎症小鼠皮损红斑、组织病理学改变及IL-1β、IL-6、TNFα、 TLR2的升高,同时,青蒿素可减少皮损CD4+T细胞、巨噬细胞和中性粒细胞浸润,抑制皮损中免疫细胞相关趋化因子(CXCL10、CCL20、CCL2和CXCL2) 的表达;体外实验中,青蒿素通过抑制NF-kB信号通路来抑制LL37诱导的HACAT细胞炎症因子表达,并且青蒿素还可以抑制LL37诱导的皮肤血管增殖和体外HUVECs细胞迁移,总之,实验表明青蒿素主要是通过调节免疫反应和血管生成来改善玫瑰痤疮症状。

请参阅图1至图5,本发明提供一种技术方案:青蒿素改善玫瑰痤疮炎症用动物和细胞模型的构建方法,包括以下步骤:
(1)青蒿素改善玫瑰痤疮炎症表型的动物模型构建
改善玫瑰痤疮症状的药物的动物模型为7周龄BALB/C小鼠,用青蒿素每天以200mg/kg灌胃,连续七天,在第5天小鼠背部皮肤剃毛,第6-7天给予背部皮肤皮下注射LL37多肽以诱导玫瑰痤疮样炎症;
(2)青蒿素改善玫瑰痤疮炎症表型的细胞模型构建
步骤一、化合物:

实验结果一、青蒿素缓解小鼠玫瑰痤疮样炎症表型:
过度活跃的固有免疫,特别是角质形成细胞中TLR2的过度激活,是玫瑰痤疮的重要病理机制,研究发现促炎细胞因子/趋化因子和促血管生成因子在角质形成细胞TLR2激活时释放,并与红斑、毛细血管扩张和炎症等玫瑰痤疮症状有关,如图1A、B所示,青蒿素显著减弱了LL37诱导的玫瑰痤疮样红斑,组织学分析显示,青蒿素治疗改善了玫瑰痤疮样炎症的炎症细胞浸润(图1C);大量研究表明,玫瑰痤疮中TLR2和促炎细胞因子显著上调,如图1D所示, TLR2和促炎因子在小鼠背部皮损中被青蒿素治疗所抑制,因此,这些结果表明青蒿素改善了LL37诱导的小鼠玫瑰痤疮样炎症表型,其中,图1中,(A) 脱毛后,将LL37皮下注射至背部皮肤,诱导玫瑰痤疮样红斑表型,通过灌胃给予青蒿素预处理;(B)玫瑰痤疮表型的严重程度根据红度评分;(C)玫瑰痤疮皮损处皮肤伊红染色分析;(D)qPCR法检测小鼠皮损中TLR-2、TNF α、IL-1β和IL-6的表达水平,以及结果代表了三个独立的实验,数据代表平均值±SEM,*P<0.05,**P<0.01和***P<0.001;
实验结果二、青蒿素抑制LL37诱导的玫瑰痤疮样小鼠皮损中CD4+T细胞、中性粒细胞和巨噬细胞的浸润:
玫瑰痤疮患者皮损中CD4+T细胞浸润和Th1/Th17通路激活都参与了玫瑰痤疮的病理变化,这表明适应性免疫是玫瑰痤疮的重要发病机制。通过免疫荧光,本研究检测了玫瑰痤疮样炎症小鼠皮损中的CD4+T细胞,并且观察到大量的CD4+T细胞浸润;此外,在小鼠皮损中,青蒿素可显著抑制LL37诱导的CD4+T细胞浸润(图2A、B),与CD4+T细胞浸润减少的表型一致,青蒿素还抑制了玫瑰痤疮样炎症小鼠皮肤中Th1(IFN-γ,CXCL10)和Th17(IL17A,CCL20)相关细胞因子和趋化因子的上调(图2C),其中,图2中,(A,B) 组织免疫荧光法观察皮损中CD4+T细胞的浸润,绿色表示CD4+T细胞,蓝色表示DAPI,比例尺100μm;(C)qPCR法检测小鼠皮损中Th1(CXCL10,IFN- γ)和Th17(CCL20,IL17A)趋化因子和细胞因子的表达;(D,E)组织免疫荧光法观察皮损中中性粒细胞的浸润,绿色表示Ly6G(中性粒细胞),蓝色表示DAPI,比例尺100μm;(F)qPCR法检测小鼠皮损中中性粒细胞趋化因子(CXCL2)的表达;(G,H)组织免疫荧光法观察皮损中巨噬细胞细胞的浸润,绿色表示F4/80(巨噬细胞),蓝色表示DAPI,比例尺100μm;(I) qPCR法检测小鼠皮损中巨噬细胞趋化因子(CCL2)的表达;数据代表平均值±SEM,以及*P<0.05,**P<0.01和***P<0.001;
除了CD4+T细胞外,包括中性粒细胞和巨噬细胞在内的先天性免疫细胞也在玫瑰痤疮炎症过程中聚集,如图2D、E和图2G、H所示,LL37诱导的中性粒细胞和巨噬细胞浸润被抑制,相应地,青蒿素也可抑制小鼠皮损中性粒细胞趋化因子CXCL2和单核细胞趋化蛋白-1即CCL2的表达(图2F、I);因此,这些实验证据支持青蒿素抑制玫瑰痤疮中过度激活的适应性和先天性免疫;
实验结果三、青蒿素抑制LL37诱导的HACAT细胞玫瑰痤疮相关炎症因子:
角质形成细胞中促炎因子的过度表达在玫瑰痤疮的发病机制中起着重要作用,在本研究中,青蒿素抑制了LL37诱导的HACAT细胞IL-1β、IL6、IL8、 TNFα、CCL2、CXCL10、CCL20和TLR2的上调(图3),细胞中的实验结果也与之前青蒿素在小鼠体内发现的抗炎作用一致;所以,这些结果表明青蒿素抑制LL37诱导的角质形成细胞TLR2、细胞因子和趋化因子的上调;
实验结果四、青蒿素抑制LL37诱导的HACAT细胞NF-kB信号通路的激活:
研究证明NF-kBp65蛋白核移位调节细胞因子释放。为进一步分析NF-kB 信号通路是否参与青蒿素对角质形成细胞的炎性抑制,本发明用青蒿素(50 μM)预处理HACAT细胞1小时,然后用LL37(4μM)刺激细胞30分钟通过免疫印迹法检测细胞中磷酸化p65的水平;然后检测p65的核转位,LL37刺激细5小时,然后进行免疫荧光染色;本发明发现LL37可以促进p65的磷酸化,而青蒿素可以抑制HACAT细胞的磷酸化(图4A);此外,青蒿素干预显著抑制LL37诱导的HACAT细胞NF-kB-p65核移位,如图4B所示LL37刺激可诱导细胞质中的p65蛋白转移到细胞核中而青蒿素可以抑制这一反应;所以,青蒿素可以部分通过NF-kB信号途径减轻HACAT细胞的炎症反应,其中,图4 中,(A)免疫印迹法检测HACAT细胞P65蛋白磷酸化的水平(青蒿素预处理 1小时,LL374μM刺激30分钟);(B)细胞免疫荧光法观察HACAT细胞P65 蛋白的核转位,红色表示P65蛋白,蓝色表示DAPI,比例尺40μm,以及结果代表了三个独立的实验;
实验结果五、青蒿素减少血管新生并抑制LL37诱导的HUVECs迁移:
在小鼠模型中,LL37显著诱导血管生成和促血管生成因子的表达,并且青蒿素干预可使真皮微血管数量减少,从而抑制血管生成(图5A);在小鼠皮损中,尽管青蒿素对VEGFA、VEGFb和Angpt2没有抑制作用,但它对LL37 诱导的VEGFc和Angpt2在小鼠皮损中有显著的抑制作用(图5B);在体外,不同浓度的青蒿素(25μM、50μM、75μM和100μM)对HUVECs的活性没有影响(图5C),然而,50μMART在划痕实验(图5D、E)和趋化实验(图5F、 G)中明显抑制LL37诱导的HUVECs迁移;因此,这些结果表明青蒿素可以抑制玫瑰痤疮中LL37诱导的血管生成,其中,图5中,(A)组织免疫荧光法观察皮损中血管的增殖,绿色表示CD31(血管内皮细胞),蓝色表示DAPI,比例尺100μm;(B)qPCR法检测小鼠皮损中促血管生成因子的表达。(C) CCK8实验中不同浓度青蒿素对HUVECs稳定性的影响;(D,E)体外细胞趋化实验检测青蒿素对HUVECs趋化的影响;(F,G)体外细胞划痕实验检测青蒿素对HUVECs迁移的影响,以及结果代表了三个独立的实验,数据代表平均值±SEM,*P<0.05,和**P<0.01。

Claims (6)
1.青蒿素改善玫瑰痤疮炎症用动物和细胞模型的构建方法,其特征在于,包括以下步骤:
(1)青蒿素改善玫瑰痤疮炎症表型的动物模型构建
改善玫瑰痤疮症状的药物的动物模型为7周龄BALB/C小鼠,用青蒿素每天以200mg/kg灌胃,连续七天,在第5天小鼠背部皮肤剃毛,第6-7天给予背部皮肤皮下注射LL37多肽以诱导玫瑰痤疮样炎症;
(2)青蒿素改善玫瑰痤疮炎症表型的细胞模型构建
步骤一、化合物:
氨基酸序列为:LLGDFFRKSKEKIGKEFKRIVQRIKDFLRNLVPRTE,然后通过HPLC纯化,并通过质谱鉴定;
步骤二、青蒿素干预HaCaT细胞炎症模型:将HaCaT细胞置于30mm培养皿中用含10%血清的DMEM完全培养基在37℃、5%的二氧化碳的培养箱中培养待细胞达到90%融合度后按照实验目的,分为空白组—不做任何处理,青蒿素组—青蒿素(50μM/L)刺激12h,LL37组—LL37(4μM/L)刺激12h,青蒿素+LL37组—青蒿素(50μM/L)刺激1h后,加入LL37(4μM/L)再同时刺激12h。之后收集RNA,储存在-80℃直至分析;
步骤三、青蒿素影响HaCaT细胞炎症机制模型:将HaCaT细胞种板于含有细胞爬片的24孔板中用含10%血清的DMEM完全培养基在37℃、5%的二氧化碳的培养箱中培养待细胞达到90%融合度后按照实验目的,分为空白组—不做任何处理,青蒿素组—青蒿素(50μM/L)刺激5h,LL37组—LL37(4μM/L)刺激5h,青蒿素+LL37组—青蒿素(50μM/L)刺激1h后,加入LL37(4μM/L)再同时刺激5h,之后,取出细胞爬片,行免疫组织荧光染色检测并分析HaCaT细胞P65蛋白的核转位;
步骤四、青蒿素影响人脐静脉内皮细胞(HUVECs)趋化模型:24孔板中放入500μL含10%血清的1640完全培养基,根据实验需要,设置空白组、青蒿素组、LL37组、青蒿素+LL37组;在LL37组和青蒿素+LL37组中,24孔板加入4μM/L的LL37,将趋化小室(8mm孔径)置入24孔板中,并种入HUVECs细胞(2×103),小室中的1640培养基为200μL且不含血清;根据实验需要,向青蒿素组和青蒿素+LL37组小室中加入青蒿素(50μM/L),将24孔板置于37℃、5%CO2细胞培养箱中培育,24小时后取出24孔板,清洗固定后用0.5%的结晶紫染色30分钟,清洗并晾干小室后于显微镜下观测粘附于小室下层膜上的细胞并拍照计数。
2.根据权利要求1所述的青蒿素改善玫瑰痤疮炎症用动物和细胞模型的构建方法,其特征在于,所述步骤(2)青蒿素改善玫瑰痤疮炎症表型的细胞模型构建还包括青蒿素影响人脐静脉内皮细胞(HUVECs)迁移模型:HUVECs(每孔5x105)接种到6孔板中,根据实验需要,设置空白组、青蒿素组、LL37组、青蒿素+LL37组,在LL37组和青蒿素+LL37组中;当细胞生长到100%融合时,用无菌1mlTip头在皿底做划痕,PBS冲洗每个孔,随后加入含10%血清的1640完全培养基,并在青蒿素组和青蒿素+LL37组加入青蒿素(50μM/L),LL37组和青蒿素+LL37组加入LL37(4μM/L),将HUVECs细胞在37℃、5%CO2的培养箱中培养24小时,然后用3个随机选择的区域的划痕间距评价细胞的迁移能力。
3.根据权利要求1所述的青蒿素改善玫瑰痤疮炎症用动物和细胞模型的构建方法,其特征在于,所述步骤(2)青蒿素改善玫瑰痤疮炎症表型的细胞模型构建还包括组织学:用石蜡固定玫瑰痤疮样炎症皮损的小鼠背部皮肤,切成4μm厚;切片用苏木精和伊红(H&E)染色染色,并且如前所述在标准光学显微镜(OLYMPUS,Japan)下观察它们的组织形态学。
4.根据权利要求1所述的青蒿素改善玫瑰痤疮炎症用动物和细胞模型的构建方法,其特征在于,所述步骤(2)青蒿素改善玫瑰痤疮炎症表型的细胞模型构建还包括皮肤组织免疫荧光:在室温下用多聚甲醛固定8mm冷冻切片15分钟,并用5%正常驴血清和0.2%TritonX在室温下封闭持续1小时;将皮肤切片用抗CD4、F4/80、LY6G和CD31抗体在4℃下处理过夜,用AlexaFluor488标记的抗山羊IgG抗体常温染色1h,使用ZeissAxioScopeA1捕获图像。
5.根据权利要求1所述的青蒿素改善玫瑰痤疮炎症用动物和细胞模型的构建方法,其特征在于,所述步骤(2)青蒿素改善玫瑰痤疮炎症表型的细胞模型构建还包括实时PCR:使用TRIzol试剂(InvitrogenLifeTechnologies,USA)从细胞或皮损组织中分离总RNA,使用PrimeScriptRT试剂盒(Takara,Shiga,Japan)将2μgRNA转录成cDNA,并在AppliedBiosystems7500机器(LifeTechnologies)上用iTaqUniversalSYBRGREENSupermix(Bio-Rad,California,USA)进行qPCR。
6.根据权利要求1所述的青蒿素改善玫瑰痤疮炎症用动物和细胞模型的构建方法,其特征在于,所述步骤(2)青蒿素改善玫瑰痤疮炎症表型的细胞模型构建还包括数据统计:实验中的数据都用均数±标准误(SEM)的形式表示,使用GraphPadPrism5软件作图并用其中的t检验检测两组数据的差异,取双侧检测标准、并以*P<0.05,**P<0.01和***p<0.001被认为具有显著性差异。
