Abstract

背景技术
国际公开号WO 2017/070578(用纳米颗粒检测并治疗龋齿和微小腔洞(microcavities))描述了用于检测和/或治疗牙齿中活动性龋齿病灶(active cariouslesions)或微小腔洞的纳米颗粒。纳米颗粒包含带有至少一个阳离子区域和/或具有净正电荷的淀粉,从而能够与牙齿上的龋齿病灶缔合。在一些示例中,纳米微粒包含防龋剂或者再矿化剂。在一些示例中,纳米颗粒是通过如美国公开号2011/0042821号所述的反应性挤出工艺由淀粉形成。在形成纳米颗粒后,其被阳离子化,并用含氟组分、含钙组分或含钙和磷酸盐的组分通过冻干强化。纳米颗粒在唾液的pH下具有正ζ电势。在一个示例中,将阴离子氟化盐填入阳离子淀粉纳米颗粒。相对于游离氟化盐的参比溶液,观察到纳米颗粒通过渗析膜在人造唾液中释放氟化物延迟了30分钟。

龋齿(蛀牙)是世界上最普遍的慢性疾病。几乎每个人都会在生活中的某个时刻生长龋齿。在任何给定的时间,有42%的儿童和25%的成人患有未经治疗的龋齿,导致并发症,所述并发症包括疼痛、感染、生活质量差,并且在极少数情况下甚至导致死亡。在全球范围内,估计每年在该疾病及其并发症的应对上花费超过2000亿美元。手术治疗导致不可逆的修复周期,随着患者年龄的增长,导致数次替代修复(replacement restoration)、套牙冠(crowns)以及最终的牙齿脱落或种植牙。当牙齿生物膜中的细菌使糖发酵并产生会使牙釉质脱矿的有机酸时,龋齿病灶开始形成。随着矿物从釉柱中浸出,该区域变得更加多孔并变脆弱。早期病灶由表层(表面区域)组成,与表面下(病灶主体)相比,该表层似乎不受龋齿侵蚀的影响。表面区域由矿物质沉淀产生,并由溶解度梯度、溶解/沉淀机理以及受唾液中存在的吸附剂保护来解释。报告表明,与孔体积为0.1%的吸音釉质(sound enamel)相比,龋齿病灶主体的孔体积(孔隙度的测量)为5-30%,并且表面区域<5%。这些表面下孔隙的区域在临床上呈现为乳白色不透明,被称为“白斑病灶”,并供临床医生确定早期龋齿。如果该过程没有逆转,牙釉质表面上微小的开放微通道将使酸继续进入表面下。因此,病灶变得越来越多孔,直到最终形成空洞,需要侵入性外科手术修复(牙齿填充)。然而,龋齿过程是动态的,早期龋齿有时可通过更好的卫生和再矿化剂(例如高氟化物治疗或牙膏)来逆转。一些龋齿自然会变得不活跃或“被遏制”,无需任何处理,因为矿物质和/或蛋白质沉积已减少了孔隙,尤其是表面的孔隙。对龋齿过程的更好理解导致了龋齿应对的范式转变(paradigm shift),强调了牙釉质的保存和微创侵入牙科,从而带来了更好的口腔健康结果。然而,需要替代和/或改进的再矿化剂。
龋齿和牙齿的其他矿化区域带负电,并且通常发现于菌斑附近,菌斑也可能带负电。当牙齿脱矿时,磷和钙会流失,而磷-钙矿物质在使牙齿再矿化时是有用的。氟化物还可用于使牙齿再矿化以及预防或治疗龋齿。
在本文所述的方法中,在制备淀粉纳米颗粒的同时,将磷化合物如SMTP用作交联剂。因此,交联剂提供了有用的元素,但是STMP交联的纳米颗粒带有负电荷,可预期其通过静电力与龋齿排斥。然而,如本文所述,通过优选添加多价阳离子和/或使淀粉阳离子化,可以至少减少负电荷,并且任选地中和或逆转负电荷,在形成纳米颗粒时,添加多价阳离子和/或使淀粉阳离子化可以任选地完成其中之一或两者。特别地,钙盐的添加用于使纳米颗粒的电荷更适合于靶向龋齿,同时还提供了可用于使牙齿再矿化的另一种元素——钙。阳离子(如钙)的添加也似乎提高了也可以在形成过程中掺入纳米颗粒中的阴离子活性剂(如氟化物或荧光素)的保留。氟化物可有效地进行反应以在嘴中形成矿物质,或可与递送至牙齿的纳米颗粒中的钙和/或磷形成矿物质。此外,氟化盐可用于在纳米颗粒形成过程中提高水相的离子强度,这可促进交联反应,同时还提供纳米颗粒中的氟化物。尽管不必添加钙盐和氟化盐中的一者或两者,但是当使用两种盐时,钙和氟化物在纳米颗粒中的保留都得以改进。或者,可以使用不添加钙和/或氟化物的纳米颗粒。还显示了具有磷酸盐化合物作为其唯一再矿化剂的纳米颗粒也可用于再矿化,其中淀粉被阳离子化,使得该纳米颗粒在pH值为5.5及以下时具有正ζ电势。鉴于活性龋齿病灶通常具有低于5.5的pH值,不受限于理论,可以预期如果纳米颗粒可选地在7以及更高的pH下具有负ζ电势,则上述纳米颗粒将有利于靶向进入龋齿病灶,或有利于pH触发的靶向。除矿化外,本文所述的各种纳米颗粒可用于治疗牙本质敏感,有助于防止或治疗龋齿,或用作定位龋齿病灶的诊断剂。

发明详述
蛀牙始于一个或多个呈白斑的脱矿区域。白斑可替代地称为龋洞前(pre-cavity)病灶、龋前病灶或白斑病灶。由于通常由释放酸的口腔细菌引起的腐蚀,白斑的牙釉质比天然牙釉质具有更大的孔隙率。牙釉质表面可保持完整,但在活动性病灶中,开放的微孔可使酸渗入牙齿。随着时间的流逝,白斑区域变得越来越多孔和脱矿。最终,白斑区域可能变得非常脆弱,以致牙釉质表面塌陷并形成龋洞。在本说明书中,除非指定了具体类型的脱矿区域,否则,当提到牙齿的脱矿区域时,其包括白斑和龋洞。然而,预期本文所述的纳米颗粒将特别用于治疗白斑,在白斑处通过常规手段接近多孔结构比较困难或具有破坏性。例如,传统的局部施用氟化物溶液可能仅密封白斑表面,从而使白斑失活,但是不能恢复表面下的主要多孔区域。在牙齿上钻孔以提供填充物或生物活性物质会去除一些天然牙釉质,该天然牙釉质随后永久失去,因为在该阶段其无法再形成。本文所述的纳米颗粒可靶向白斑本身或白斑附近的菌斑。本文所述的纳米颗粒还可用于封闭或密封牙本质中的孔,如治疗牙本质敏感。预期当白斑处于龋洞前阶段并且具有开放的微孔和表面下孔隙时,用纳米颗粒进行处理将是最有益的。然而,也可以在龋齿的更晚期阶段形成龋洞(孔)之后,用纳米颗粒进行治疗。
不受限于理论,发明人认为,随着钙从牙齿的脱矿区域浸出,在牙齿内部留下多磷酸盐负电荷。纳米颗粒可以通过活动性病灶中的微孔进入脱矿区域。加载到纳米颗粒分散体中的可用元素的浓度可能超过相同元素的对比溶液的浓度。如果纳米颗粒带正电,则由于静电吸引,其还可以与牙齿表面(包括外牙釉质表面下的表面)或菌斑缔合,由此促进纳米颗粒的元素的递送和/或保持。一旦唾液在牙齿上或牙齿内,则唾液会使生物聚合物降解,并且纳米颗粒所释放的一种或多种元素和/或矿物质可以有助于修复孔和/或表面下区域。例如,通过纳米颗粒递送的材料可以与牙齿或唾液的离子或化合物反应,为矿物质的形成提供晶种颗粒,用作可以掺入天然产生的矿物质中的生物活性化合物,或者简单填充脱矿的表面下区域。
在本文所述的方法中,生物聚合物(即淀粉)纳米颗粒是使用乳液法制备的。简而言之,将一种或多种生物聚合物分散或溶解在水中,然后将水分散(即乳化)在另一相(例如油相)中,并在生物聚合物处在分散体或乳液中的水相分散液滴中时,使生物聚合物交联。使用油作为第二相是任选的,但有助于将水溶性反应物加载到水相的液滴中。然而,可以使用淀粉的另一种非溶剂,例如乙醇或己烷,或多相水性体系。交联剂可以是磷酸盐或多磷酸盐交联剂,例如三偏磷酸钠(STMP)或三聚磷酸钠(STTP)。
任选地,该方法可以是相转化乳液法(PIE)。该相转化工艺的一个方案如图1所示。在所示的示例中,用三偏磷酸钠(STMP)交联剂制备淀粉基纳米颗粒。最初形成水包油乳液,其在温度升高后变成油包水乳液。表面活性剂可用于辅助水包油至油包水的转变以及选择发生该转变的温度。添加STMP,使得在油包水乳液的水滴内发生交联反应。通过在图1所示的三个阶段(分离的水相和油相、水包油乳液、油包水乳液)中的任一个阶段添加其他元素,可以将其添加到水相中。
参见图1,使油相与包含溶解的淀粉或分散的淀粉纳米颗粒的水相均质化。例如,油可以是石蜡油或食品级矿物油。或者,可以使用其它食品级油,例如向日葵油或橄榄油。在形成水包油乳液之后,将温度升高至高于反应条件下的相转化温度(PIT)。PIT可以根据水和油的比例、任意表面活性剂(例如吐温(Tween)85)的存在和类型、任意催化剂(例如NaCl)的存在和类型以及油的类型而变化。在一些情况中,PIT范围可以为25℃至60℃。任选地,可以通过高剪切混合器本身来提供加热,例如通过提高混合器速度来加热混合物。在对油包水乳液进行加热的同时或在相转化完成后,添加交联剂。然后,反应可以继续例如1小时。
生物聚合物在碱性条件下使用磷酸盐交联剂例如STMP进行交联。尽管可以使用其它交联剂,但在食品级制剂中可以优先使用STMP。交联剂提供了磷的来源,磷是一种用于修复脱矿牙齿的元素。部分交联剂(发明人认为该部分为约10-50%或10-30%)反应以通过单磷酸酯键在纳米颗粒内形成内部不可逆的(即,共价结合)交联。但是,除了二淀粉单磷酸酯以外,副反应还可以形成其它化合物,例如单淀粉三磷酸酯、单淀粉单磷酸酯。该反应在某种程度上是低效的,但是在反应或副反应中产生的无机或有机化合物中的悬挂磷酸根基团可用于在颗粒内或在配置在口腔中之后与钙和/或氟化物形成强缔合复合物。
在一些示例中,NaCl盐用于提供水相中的高离子强度,这有利于在后续步骤中发生STMP反应。然而,在本文所述的其他示例中,使用NaF代替NaCl或与NaCl组合使用,并且还在纳米颗粒中提供氟化物。可以将NaF添加到例如在均质化以形成图1中的O/W乳液之前所产生的水相中。或者,NaF也可以在均质化以形成O/W乳液同时或在形成O/W乳液后,或者在形成纳米颗粒后(但是可能会减少氟化物释放时间)添加。在一些示例中,在水相中加入钙盐,如氯化钙。任选地,在添加了STMP之后,或者在纳米颗粒形成之后(钙盐和/或氟化物释放时间可能有些减少),将干燥的钙盐或水溶液中的钙盐添加到图1的W/O乳液中。使水溶性组分进入水滴中并且至少部分地与纳米颗粒反应或以其它方式缔合。在另一选择中,代替NaF和NaCl、或除NaF和/或NaCl之外,可以将钙盐(如氯化钙)添加到均质化以形成图1中的O/W乳液之前所产生的水相中。STMP在所得纳米颗粒中产生负电荷。钙的添加可以阻止这些电荷中的一些电荷。然而,通常需要额外步骤,例如淀粉的阳离子化,以产生纳米颗粒,该纳米颗粒可以在龋齿病灶内或附近所存在的中性pH甚至酸性pH(即5.5或更小)下带正电。
氟化物和/或钙可以与磷和生物聚合物一起以乳液(例如油包水乳液)中的小水滴的分散体形式存在,其任选地通过表面活性剂进行稳定。含有生物聚合物的各液滴产生了交联颗粒。任选地,可以使用另一相中的水乳液。
一种或多种乳液优选使用超高剪切混合器(例如,思维森(Silverson)溶解器搅拌机)来产生。该搅拌机优选产生最小限度的空气包封,并且提供足够的剪切以产生直径平均小于700nm或小于500nm的纳米颗粒,考虑到其水凝胶特性和纳米颗粒尺寸的分布以及孔,其中大部分纳米颗粒能进入龋齿病灶的孔(尺寸为平均750至800nm)中。淀粉可以被烹煮、化学降解和/或热机械加工以帮助在水相中产生淀粉的溶液或分散体。或者,较小的淀粉纳米颗粒(20-200nm)可以用作淀粉原料,例如由EcoSynthetix公司生产的商品名为EcoSphereTM的那些。例如通过动态光散射(DLS)图中的峰、DLS测量的Z均尺寸(或ISO 13321或ISO 22412中所述的谐波强度平均粒径)、或纳米颗粒跟踪分析(NTA)测量中的平均值或D50值所测量,所得的纳米颗粒的数均尺寸或平均尺寸为小于1000nm,例如100-700nm、100-500nm、200-500nm或200-400nm。破乳后,可任选使水相离心,例如以4000rpm离心1分钟,来将上清液中的纳米颗粒与沉淀中的未缔合沉淀物分离。纳米颗粒任选地进行洗涤以去除痕量的油,但是如果使用合适的(即,食品级)的油,则不必完全除去所有痕量的油。上清液可以经冷冻干燥以获得干燥的纳米颗粒。纳米颗粒可以干燥储存或以水性分散体、凝胶或糊料形式储存更有限的时间。水性分散体凝胶或糊料可以任选地用杀生物剂、抗微生物防腐剂或生物抑制添加剂进行灭菌或稳定。
基于无水葡萄糖重复单元(AGU),所使用的交联剂的量可以是1%至50mol%的STMP。优选的样品用3%至50%的STMP、或10%至50%的STMP,例如约30%的STMP生产。粒度似乎与STMP的量没有显著关系,除了在某些示例中,极少量的STMP(即1%)产生了非常小的纳米颗粒(约100nm),少量的STMP(即1-5%)产生了大纳米颗粒(平均尺寸为约300-500nm),而大量的STMP(5%至50%)产生了中等纳米颗粒(约100-300nm)。不受限于理论,尽管3%的STMP似乎是足够的,但是用非常低的STMP(即1%)制备的样品可能没有将几乎所有可用淀粉掺入纳米颗粒中。一旦有足够的可用交联剂,由于颗粒是水凝胶并且其尺寸是在溶胀状态下测量的,因此利用较大量STMP得到的较小尺寸可能是由于较高的交联度和较小的溶胀(如通过与体积溶胀比有关的斯托克斯-爱因斯坦(Stokes-Einstein)方程预测)。所施加的剪切能的量或其它可能影响油包水乳液中液滴尺寸的因素也可能进一步影响粒度。在一些方面中,用添加的钙制成的纳米颗粒在7.0pH下的ζ电势接近中性,例如其范围为-5至+5mV。任选地,在水相中产生的未与纳米颗粒缔合的沉淀物可以通过离心分离。纳米颗粒常常保留在离心样品的上清液中。纳米颗粒呈现出溶胀性质,并且似乎是水凝胶。例如,纳米颗粒保留水,但是通过纳米颗粒所保留的水的量随着离子浓度提高而下降。
随着pH和STMP含量提高,纳米颗粒变得更为带负电(通过ζ电势测量)。在一些示例中,在未添加钙盐且未进行淀粉阳离子化的情况下,利用1-50摩尔%AGU的STMP的纳米颗粒的ζ电势范围为0至65mV(在一定范围的pH和STMP含量范围内),或在中性pH下为-10至-22mV。例如,在未添加钙盐且未进行淀粉阳离子化的情况下,由30%STMP制备的纳米颗粒在pH为3时的ζ电势测定为-15mV,在pH为8时为-45mV,并且在pH为12时进一步下降至-70mV。
添加钙(例如以CaCl2形式)、但仍然未进行淀粉阳离子化(如下文详述)降低了纳米颗粒的负ζ电势。在接近中性pH和5%的STMP含量下,添加钙的纳米颗粒的电荷范围为-5mV至0mV。由钙和30%STMP制成的纳米颗粒在中性pH值附近的ζ电势范围为-30mV至-25mV,并且未进行淀粉阳离子化。不受限于理论,所添加的钙可能使得由STMP提供的磷酸基团封端。任选地,淀粉可以被阳离子化,从而导致负ζ电势进一步降低,或在所需pH范围内产生正ζ电势。
作为STMP的替代品,可以使用三聚磷酸钠(STTP)作为交联剂。
任选地,例如,可以通过国际公开号WO 2017/070578(用纳米颗粒检测龋齿和微小腔洞(Detection and Treatment of Caries and Microcavities with Nanoparticles))中描述的方法,使纳米颗粒阳离子化。任选地,当在油包水乳液中时,淀粉可阳离子化。例如,可以在形成油包水乳液之前或之后将缩水甘油基三甲基氯化铵(GTAC)添加到水相中,GTAC任选地与水和异丙醇或2-丙醇一起添加或与它们预混合后添加。或者,淀粉可以在形成纳米颗粒之后阳离子化。或者,淀粉可以在形成纳米颗粒之前阳离子化,但是在该情况下,淀粉优选先进行烹煮或再生,以使得阳离子化不限于淀粉粒子(granule)表面。

PO4 3-、Ca2+和F-离子(视情况而定)掺入各种仅含磷酸盐的样品中、仅含磷酸盐和钙的样品中以及含有磷酸盐、钙和氟化物的样品中通过用于SEM图像区域元素分析的能量色散X射线光谱法(EDX/EDS)得到确认。
还通过电感耦合等离子体(ICP)分析测量了各种冷冻干燥样品(水分含量小于0.1%)的离子含量。纳米颗粒中的磷含量随所用STMP的量增加而增加。磷含量的增加通常随STMP的摩尔%线性增加。在一个示例中,用1-50摩尔%AGU的STMP生产的纳米颗粒的磷含量范围为约1000至约21,000ppm。在该示例中,磷含量在对样品进行6天渗析并用HCl和HNO3处理后通过ICP-MS测定。磷含量值与总磷有关,其被认为是与纳米颗粒结合的磷,但是未确定存在的磷酸盐物质的类型。任选地,STMP的范围为3至50摩尔%AGU或10-50摩尔%AGU,其在纳米颗粒中提供了4,000-21,000ppm的磷。任选地,STMP范围可以为20-40%、25-35%、或约30摩尔%AGU。
例如,用30摩尔%AGU的STMP生产但未进行阳离子化的纳米颗粒含有约15,000ppm的磷。加入基于淀粉(脱水葡萄糖重复单元)为67摩尔%的干燥CaCl2,纳米颗粒还包含约1,320,000ppm的钙。还添加了70摩尔%的NaF和67摩尔%的CaCl2(基于淀粉),纳米颗粒包含19,000ppm的氟化物以及增加至2,160,000ppm的钙含量。在不添加CaCl2(基于淀粉)的情况下添加70摩尔%NaF(基于淀粉)的情况下,纳米颗粒含有约700ppm的氟化物。
上述结果表明,当还添加钙时,氟化物保留增加,而当添加氟化物时,钙保留增加。钙被认为通过离子相互作用与磷酸盐相互作用。不受限于理论,氟化物可与磷酸盐和钙共沉淀,以在纳米颗粒中形成氟磷灰石(Ca5(PO4)3F)或其他矿物质。尚不清楚钙或氟化物是否与淀粉或附着在淀粉上的磷酸盐反应,或者钙或氟化物如何与淀粉结合。不过,在从离心样品的上清液中提取的纳米颗粒中测出本文所述的磷、钙和氟化物的含量,其中,未结合至纳米颗粒的沉淀物收集在离心机的沉淀中,并且不是所测量的钙或氟化物含量的一部分。虽然没有完美的分离方法(并且一些未结合的沉淀物可能仍存在于纳米颗粒中),但预期磷、钙和氟化物与纳米颗粒的结合具有足够的持久性,使得至少其中部分材料可以通过纳米颗粒递送至龋齿病灶。还预期添加不同的多价阳离子(例如不同的二价或三价阳离子或碱土金属离子)可以类似方式提高阴离子活性剂(如氟化物)的保留,但是添加钙没有潜在益处。
纳米颗粒可通过病灶中的孔进入龋齿病灶的病灶中或留在病灶外侧上。任选地,在施加纳米颗粒之前或同时,可以例如通过患者刷牙或牙齿卫生师的清洗来清洁牙齿以去除菌斑和/或牙菌膜(dental pellicle),从而改善龋齿病灶的可达性。随着在病灶附近或内部时纳米颗粒在唾液中分解,磷和钙和/或氟化物可能扩散到牙齿中,并可能沉积或任选地反应(不管有没有来自牙齿或唾液中的其它元素参与),从而在牙齿中形成矿物质。存在于纳米颗粒中的已经形成的矿物质(例如,磷酸钙、羟基磷灰石钙或氟磷灰石)也可以充当晶种或生物活性剂,填充部分病灶或掺入牙齿中产生的其它矿物质中。
任选地,在约7.0或更小的全部pH值或约5.5或更小的全部pH值时,纳米颗粒的ζ电势至少为+2.0mV,例如+2.0mV至+10.0mV。在一些示例中,该纳米颗粒在酸性溶液中具有正ζ电势,但是在中性或碱性条件下(即,7.0或更高的pH)具有负ζ电势。例如,纳米颗粒的ζ电势在约5.0或更小的全部pH值下可以为至少+2.0mV,但是在7.0或更高的pH值下该ζ电势为负的。由于嘴中唾液的pH值大致为中性(通常约为7.4),但活动性龋齿病灶中或附近的pH值通常为酸性(通常约为4-5),因此与即使在中性或弱碱性(即,7.0或更高的pH)条件下也具有正ζ电势的纳米颗粒相比,仅在酸性条件下具有正ζ电势的纳米颗粒更能选择性地靶向活动性龋齿。可以将纳米颗粒给予牙齿,例如作为在可以任选地包括诊断性荧光纳米颗粒的洗液或漱口水中、或在施加至牙齿的凝胶或糊料中或在牙膏中使用的分散体给予牙齿。
纳米颗粒的尺寸最高为2500nm,但是优选尺寸为1000nm或更小。如本文所用,术语“纳米颗粒”不限于如IUPAC定义中尺寸为100nm或更小的颗粒,而是还包括更大的颗粒,例如其等体积球的最大尺寸或直径例如最高2500nm或最高1000nm的颗粒。任选地,由DLS图的峰强度、DLS测量的z均值或NTA测量的数均或D50确定的纳米颗粒的数均或平均尺寸范围为约100nm至约700nm,约100nm至约600nm,或约100nm至约500nm,或约200nm至约500nm,或约100nm至约400nm。如上所述,这些尺寸范围内的颗粒将被称为纳米颗粒,这与该词在北美的常用用法一致,尤其是尺寸小于1000nm的颗粒。然而,在世界的其它地区,根据IUPAC定义,尺寸大于100nm的颗粒或者可以被称为微粒。

任选地,可以通过如美国专利6,755,915(“淀粉颗粒的制备方法(Method for thePreparation of Starch Particles)”)中所述的相转化乳液法来制备纳米颗粒。在该方法中,在两相体系中制备淀粉颗粒,该两相体系包括以下步骤:a)制备包含淀粉在水中的分散体的第一相,b)制备第一相在第二液相中的分散体或乳液,c)使存在于第一相中的淀粉交联,d)分离由此形成的淀粉颗粒。在一些示例中,第二相由疏水性液体组成,并且步骤b)包括形成水包油乳液。在一些示例中,第二相由相对于淀粉的水混溶性非溶剂组成。
纳米颗粒在干燥形式下是稳定的。如果以湿的形式储存在密闭容器中,则可以制备无菌的5%w/w含阳离子矿物质的淀粉纳米颗粒的水性分散体,或用柠檬酸/山梨酸钾或其他食品级杀菌剂稳定的未灭菌水性分散体。然而,可以不需要杀菌剂。有一些迹象表明在没有杀生物剂的情况下具有稳定性,但是尚不知道纳米颗粒是否具有抑菌或杀菌性质。包含氟化物的纳米颗粒似乎具有抑菌或杀菌性质。
纳米颗粒可以与在毒理学和功能上可接受的一种或多种补充载体(即,水、赋形剂或增量剂等)组合,以产生可给予动物或人口腔的组合物。例如,组合物可以是例如口腔洗液(mouth rinse)、洁牙剂(dentrifice)、凝胶、涂膜(varnish)、涂料(paint)、牙膏、牙粉或漱口水(mouthwash)。载体可以选自一种或多种这些配混物中的常规组分。例如,载体可以是如下物质中的一种或多种:水、醇、表面活性剂、乳化剂、发泡剂、磨料、湿润剂、粘度调节剂、增粘剂、成膜剂、增塑剂、稀释剂、pH调节剂、甜味剂、调味剂、着色剂和防腐剂。例如,纳米颗粒可以在家庭或牙科诊所中用于洗液或漱口水中,所述洗液或漱口水在嘴中漱一漱(swish),然后吸出和/或清洗掉。或者,纳米颗粒可以在牙膏中,通过在家刷釉质表面给予,或作为糊料或凝胶由卫生人员施用。在将纳米颗粒引入口腔后,其会粘附在菌斑或龋齿表面,并可通过穿过病灶的多孔表面而进入活动性龋齿病灶内部。
可以例如在没有或至少没有有效量的任何其它口腔护理活性成分(例如添加的防龋剂或再矿化剂)的情况下使用纳米颗粒。具体来说,可在不添加有效量的美国专利申请公开号US 2017/0112949 A1(也作为国际公开号WO 2017/070578(用纳米颗粒检测龋齿和微小腔洞(Detection and Treatment of Caries and Microcavities withNanoparticles)公开)中描述的任意口腔护理活性成分的情况下使用纳米颗粒。例如,可以在纳米颗粒形成期间或之后不添加以下一种或多种物质的情况下使用纳米颗粒:a)含氟化物活性成分,b)含钙成分,c)含钙和磷酸盐的化合物和d)氟化胺、酪蛋白磷酸肽、磷蛋白及其等同物和组合。在该情况下,由于存在通过添加交联剂引入或在交联剂和淀粉的反应中产生的一种或多种含磷酸盐或含淀粉和磷酸盐的配混物,因此认为纳米颗粒是有效的。不受理论的限制,尽管单独的磷酸盐不会产生矿物化合物,但纳米颗粒中的磷酸盐可能会与唾液中的钙或口腔中的其它元素、离子或化合物反应,从而在牙齿上或牙齿中产生矿物化合物,或有助于在白斑或活动性龋洞前病灶内形成核晶。
可以使用其它两相乳液,例如水和酒精或己烷。然而,油相有助于实现纳米颗粒中非油溶性活性剂的高加载量。该油可以是食品级矿物油,或其它优选的食品级油,例如向日葵油或橄榄油。还可以使用表面活性剂,例如,吐温85。转变温度可以根据水与油的比例、油的类型以及表面活性剂的类型和量而变化。
提供以下实施例对各种实施方式进行说明,并提供进一步能够公开的内容,但并不旨在对任意要求保护的发明进行限制。
实施例1——用于制备乳液中未阳离子化的淀粉纳米颗粒的方法
在1L塑料烧杯中,将30g天然蜡质玉米淀粉与16.6g NaCl一起分散到600g水中,以产生白色粒子悬浮液。使用思维森超高剪切溶解器搅拌机以6500rpm(转/分钟)混合分散体。在混合的同时添加7.5g的50%NaOH。将混合机调高至8000rpm,并且混合物的温度在约20-30分钟内从22℃升高至52℃;在41℃-42℃,混合物变为透明的;放置在载玻片上的小液滴的光学显微镜图像显示出,一小部分淀粉粒子碎片持续存在(在交叉偏振器下具有双折射)。在52℃,所有粒子碎片均在苛性碱溶液中进行烹煮(NaOH发挥了在较低温度下蒸煮淀粉以及用作随后与STMP进行交联反应的催化剂的双重作用)。pH试纸显示约13-14的pH。显微镜检查显示出最终淀粉溶液已充分烹煮。
将淀粉溶液冷却至19℃,将其分成两等份的约320g液体;预期目标为8.75g的吐温80,或目标为29g的在水中的30%糊料,但仅向两份溶液之一添加(剩下的全部)18.53g。
接下来,将300g(=360mL;密度=0.833)石蜡油添加至溶液1;在下午3:20开始混合。
用光学显微镜检查液滴尺寸。
最终产物是漂亮的白色悬浮液。将最终混合物转移至加盖的PE瓶中,用冷自来水冷却至<45℃,然后转移回1L塑料烧杯中,使用思维森混合机混合,并添加约300mL水;安装一个pH探针,并使用移液管喷射1N HCl 12次(每次约1mL),将pH值从11.6调节至8.52。中和后,悬浮液被破坏(不再稳定)。
实施例2——在乳液中未阳离子化的情况下任选地用氟化物和/或钙产生纳米颗粒
pH为13的含有0.3-0.6M NaCl或NaF的1L 4.68重量%淀粉溶液在55℃下烹煮2小时,然后冷却。通过将NaOH丸粒添加到水中,调节至pH值13。或者,将60g热机械加工的淀粉(或其它冷水可溶的淀粉)分散在400mL含10g NaCl或等摩尔的NaF的去离子水中。任一方法都会产生水相。
使35g吐温85分散在500g(600mL)石蜡油中以产生油相。
向油相中加入400g淀粉溶液或分散体(水相)。
使用思维森混合机使混合物均质化,保持跟踪温度。
一旦发生相转化(通常在温度约为50℃时),如果使用经烹煮的淀粉或STMP,则添加干燥的STMP粉;如果使用热机械加工的淀粉,则添加STMP和在3mL水中的0.95g NaOH。
使反应进行1-4小时。
加入200mL含3g 37%HCl的水进行中和(=1.5M)。
取少量样品,用水稀释3-5倍体积,然后用pH计检查pH值。目标pH为8.0+/-0.5。如需要,调节pH。
任选地,将干燥的CaCl2粉末加到剩余的样品中(在混合同时)并混合10分钟。
让油/水乳液沉降。
在不搅拌的情况下使分散体沉降(例如,在周末存放在冰箱中)。使任何游离磷酸钙沉淀物上方的上清液倾析出来。用乙醇使上清液中的纳米颗粒沉淀。使用布氏(Büchner)过滤法,用乙醇洗涤并过滤纳米颗粒三次(3x),以去除表面活性剂。
使纳米颗粒重新分散在水中,并通过离心或分液漏斗进行纯化(重复3次),以去除过量的油。
进行渗析以去除游离的磷酸盐和氯化盐(6天)和/或以4000RPM的速度进行1分钟离心以去除未结合到淀粉上的大的沉淀物(即CaPO4)团聚体。对样品进行冷冻干燥以回收产品纳米颗粒。
实施例3——在乳液中阳离子化的情况下任选地用氟化物和/或钙产生纳米颗粒
将水、淀粉、氢氧化钠、表面活性剂(吐温85)和油(矿物油或石蜡油)合并至混合容器中。用高剪切混合器开始混合,并且混合直至形成乳液约10分钟。
加入盐(NaCl)和/或氟化钠和/或荧光素钠,混合直至温度>60℃。
为使淀粉阳离子化,添加缩水甘油基三甲基氯化铵(GTAC)、去离子水和2-丙醇的混合物,并使其反应约1小时(30-90分钟)。监控温度并通过调节高速混合机的混合速率将温度维持在60℃至75℃。
将SMTP添加到混合物中,并使其反应/交联约1-2小时(30-150分钟)。监控温度并调节混合速度,以使温度保持低于75℃但高于60℃。测量这批料的pH,并确保仍为碱性(约pH 10)。
如果需要钙,则添加氯化钙(二水合物),所述氯化钙(二水合物)任选地预先溶解在水中,再混合10分钟。
如需要,通过添加HCl来中和该批料(至pH 6.5-8)(用钙制成的样品可能不需要此步骤)。
使样品冷却至室温和/或冷藏以促进乳液的相分离。
用2-丙醇以约50:50比例稀释样品并混合。以10,000RPM离心10分钟,以引起颗粒、水相/亲水相和油相/疏水相的多相分离。分离沉淀的颗粒相,以进一步纯化。用2-丙醇进行额外的颗粒冲洗可以帮助去除样品中的残留油。
任选地(对于具有钙的样品而言更优选),通过震荡使颗粒重新分散在去离子水中,然后以1000RPM离心1-3分钟以去除任何大沉淀物,将上清液用于下一步。
样品进行快速冷冻并冻干,以获得分离的载有矿物的纳米颗粒的干粉末样品。
在上文所述方法的一个示例中,具有钙的STMP交联纳米颗粒用如下物质进行制备:35g吐温85;500g石蜡油;400g水(最初);3.125g NaOH粉末;14.04g淀粉;13.8g NaCl;8.58g GTAC;7.22g 70%IPA;18g水(用于与GTAC和IPA制备混合物);3.38g STMP;和3.4gCaCl2*2H2O。
实施例4——具有各种矿物质和活性剂的纳米颗粒样品的制备和测量
根据实施例3中的方法制备阳离子化的纳米颗粒的各种样品。表1中显示了作为反应物添加的淀粉以及磷酸盐、钙和氟化物中的一种或多种的相对量。这些添加剂的量以相对于100摩尔淀粉AGU的摩尔pph计。考虑到各STMP分子的三个磷酸基团,在制备各样品时添加的STMP量为表1所示量的三分之一。在产生初始水相期间,通过添加氟化钠的方式添加氟化物。通过将氯化钙添加到油包水乳液中来添加钙。在未添加任意的磷酸盐、钙和氟化物的情况下制备的纳米颗粒提供了阴性对照(GMB 9)。通过将氟化钠溶解在去离子水中至116ppm的浓度来制备阳性对照(GMB10),其与GMB8的1小时释放数据相符,如下所述。

通过渗析方法测试了在有和没有淀粉酶的情况下样品GMB8的钙释放以及样品GMB7和GMB8的氟释放。使纳米颗粒分散在水中并置于渗析膜管中,并通过收集从渗析管外部收集的渗析液定期进行测量。
在用淀粉酶的情况下,样品GMB8在1小时后释放11ppm的钙,在72小时后释放21ppm的钙。在未用淀粉酶的情况下,样品GMB8在1小时后释放1ppm的钙,在72小时后释放11ppm的钙。这些结果表明从含钙和氟化物的纳米颗粒中时间依赖性地缓释钙。
在用淀粉酶的情况下,样品GMB7在1小时后释放1249ppm的氟化物,在72小时后释放21205ppm的氟化物。在用淀粉酶的情况下,样品GMB8在1小时后释放116ppm的氟化物,在72小时后释放7604ppm的氟化物。在未用淀粉酶的情况下,样品GMB7在1小时后释放1161ppm的氟化物,在72小时后释放21205ppm的氟化物。在未用淀粉酶的情况下,样品GMB8在1小时后释放78ppm的氟化物,在72小时后释放8287ppm的氟化物。这些结果表明,淀粉酶对释放时间没有显著影响,并且两个样品均显示出超过一小时的氟化物缓释。延迟释放氟化物是有益的,因为其可以安全地提供更大剂量的氟化物。在家庭治疗中,延迟释放也可以更好地利用附着在菌斑上且未进入龋齿病灶的纳米颗粒。快速释放的氟化物常常沉淀在病灶表面,而缓释的氟化物可以能够在沉淀之前通过病灶的孔进入。
还通过以下方法测试磷、钙和氟化物的释放:将表1中一些样品的纳米颗粒分散在去离子(DI)水的容器中,并在至多2天的不同时间测量水中磷酸盐、钙或氟化物之一的浓度。浓度代表各化合物累积释放的趋势。通过偶氮砷III比色法测量钙。通过孔雀绿比色法测量无机磷。使用TISAB II,通过离子特异性电极(ion-specific electrode)测量氟化物。
测试了GMB4、GMB6和GMB8的钙释放。两天后,在水中的GMB4和GMB8分别产生了约1mg/L的钙。GMB6在6小时后产生3.8mg/L钙,在1天后产生7.8mg/L钙,在2天后产生10.2mg/L钙。
测量了GMB4、GMB5和GMB7的氟化物释放。GMB4在1小时后产生3.6ppm氟化物,在6小时后产生40ppm氟化物,在1天后产生45ppm氟化物,在2天后产生51ppm氟化物。GMB5在2天后产生了0.1ppm的氟化物。GMB7在1小时后产生4.2ppm氟化物,在6小时后产生21ppm氟化物,在1天后产生29ppm氟化物,在2天后产生36ppm氟化物。
测试了GMB3至GMT 8的无机磷释放。GMB3在6小时后产生0.49mg/L磷,在1天后产生0.55mg/L磷,在2天后产生0.50mg/L磷。GMB4在6小时后产生0.37mg/L磷,在1天后产生0.47mg/L磷,在2天后产生0.64mg/L磷。GMB5在1小时后产生0.81mg/L磷,在6小时后产生3.78mg/L磷,在1天后产生10.0mg/L磷,在2天后产生13.6mg/L磷。GMB6在1小时后产生0.17mg/L磷,在6小时后产生3.41mg/L磷,在1天后产生7.29mg/L磷,在2天后产生9.42mg/L磷。GMB7在1小时后产生0.24mg/L磷,在6小时后产生2.14mg/L磷,在1天后产生4.62mg/L磷,在2天后产生6.29mg/L磷。GMB8在1小时后产生0.43mg/L磷,在6小时后产生4.59mg/L磷,在1天后产生5.39mg/L磷,在2天后产生6.32mg/L磷。
通过纳米颗粒跟踪分析和动态光散射测定表1中各种样品的平均尺寸。表2中提供了来自NTA分析的颗粒的数值平均(numerical average)(数均(mean))尺寸和来自DLS测量的Z均(基于强度)尺寸。由于至少一些样品的多分散性性质,认为NTA测量更准确。多分散样品难以通过基于强度的DLS测量方法进行测量,并且Z均测量结果偏向更大的粒度。

图2显示了在不同pH值下表1样品的ζ电势测量结果。具有较少磷的样品GMB1至GMB4在测试的所有pH值下均为阳离子。样品GMB6和GMB8在某些pH值下为阳离子,而在pH 7时为阴离子。由于磷含量相似,预计样品GMB5和GMB7在pH 7时也是阴离子的。该电荷变化表示样品GMB5-GMB8的“智能”(即通过pH触发)靶向潜力,其中,如果在病灶中或附近,病灶具有细菌主动产生的酸性条件,则其将仅被静电吸引至带负电荷的病灶,同时还可能在到达病灶之前减少被蛋白质吸引或非特异性细胞结合。或者,如果使用纳米颗粒而没有先清洁牙齿(如在家中而不是在牙科诊所程序中采用的治疗过程),则样品GMB5-GMB8不太可能被口腔中的不在可能带负电的龋齿病灶附近但在中性pH下的菌斑或有机物吸引。任选地,由于+2至+10mV的ζ电势足以靶向龋齿病灶或菌斑,或制备仅在酸性条件下为阳离子的样品,因此可以降低样品GMB1至GMB4的阳离子化程度。
实施例5——使用未阳离子化的纳米颗粒再矿化
总体上如实施例2所述制备纳米颗粒,使用热机械加工的淀粉作为原料以避免溶解天然淀粉粒子的步骤。纳米颗粒具有20mM的Ca2+。基于TEM图像,纳米颗粒显示出具有200-500nm的尺寸,并且通过DLS测量的Z均尺寸为约380nm。用NaCl制备一批纳米颗粒(GM 6)。用NaF替代NaCl制备另一批纳米颗粒(GM 6 F-)。
制备了六个釉质片,各片上都有脱矿区域。三个片用纳米颗粒GM 6和GM 6 F-中的每一个进行处理,并将各纳米颗粒的结果进行平均。将这些片浸入纳米颗粒分散体中38天。在第16天之前多次进行表面VMHN测量,然后在第38天再次进行表面VMHN测量。GM 6 F-的结果显示出,在开始16天里,硬度大致从约75线性增加到约165,在第38天,硬度进一步增加到超过200。GM 6 F的结果显示出,硬度大致从约75线性增加到约100,并且在第38天,硬度下降至约75。GM 6在第38天的硬度降低被认为是细菌在淀粉上生长的结果,而GM 6 F-的氟化物使这些纳米颗粒具有抗菌性。在试验过程中,用GM 6 F-处理的样品中的脱矿区域的横截面的共焦显微镜显示出病灶深度从38μm减小至3μm。
实施例6——使用阳离子化的纳米颗粒再矿化
按照Featherstone等人的描述(Featherstone,J.D.B.和Mellberg,J.R.(1981),在牛、绵羊和人类釉质中人工龋齿病灶的相对进展速度(Relative rates of progress ofartificial carious lesions in bovine,ovine and human enamel.),CariesRes,15,109-114),根据乳酸和羧甲基纤维素(CMC)凝胶方案,制备在各片上具有脱矿区域的釉质片。釉质片用不可渗透的涂膜部分保护,随后用表1中所述的各种纳米颗粒制剂进行处理,同时暴露于连续的乳酸和CMC去矿质中20天。纳米颗粒处理包括在20天的过程中使片与纳米颗粒样品之一接触4分钟,每天4次。连续的乳酸和CMC脱矿方案包括在上述乳酸和CMC脱矿溶液中进行4小时的酸循环,以及当所述片未浸入乳酸和CMC脱矿溶液中时将其浸入含淀粉酶的人工唾液中。
健康釉质的VMHN值为300或更高。软的脱矿釉质的VMHN值为低于100。表3给出了试验中用各纳米颗粒制剂处理过的釉质样品和对照物的平均起始和终止VMHN。如对照物所示,试验条件会导致VMHN随时间下降。然而,相对于两个对照物,所有样品GMB1至GMB8均提供了改进的结果。此外,用样品GMB1、GMB4、GMB5、GMB7和GMB8处理在20天后显示出VMHN升高。

有趣的是,GMB5产生了良好的结果,但其不含氟化物或钙。不受限于理论,可能是该样品的磷酸基团从唾液中获取钙,从而在牙齿上产生沉淀物。
实施例7-治疗牙本质敏感
根据上述样品GMB8,用纳米颗粒配制两种口腔凝胶。该凝胶用丙二醇进行稳定,并用食品级明胶进行增稠。制剂1具有25%w/w的纳米颗粒。制剂2具有5%w/w的纳米颗粒。两种凝胶制剂克制牙本质敏感的作用是通过将这些制剂施加至牙本质盘(dentine disc)并通过导水率测量所指示的牙本质渗透率的变化来确定的。由于认为牙本质超敏反应(DH)涉及牙本质小管内流体流动对神经的刺激,因此由导水率的变化所显示的小管阻塞表明DH的可能治疗。
从所提取的人类第三磨牙切出牙本质盘。各牙齿在牙齿最宽部分的正下方横向切割,然后切割成片,以产生厚度为0.5mm的牙本质盘。各盘具有最小为4mm的直径。所述盘用碳化物纸抛光直至平坦,然后酸蚀去除在切割和抛光过程期间产生的任何污迹层。将该盘放置在管中,并以1psi的压力将水施加至该盘上。测量在5分钟内通过该盘的水体积。在用纳米颗粒处理该盘之前和之后进行该测试,将样品在人造唾液中存储24小时之后再次进行该测试。治疗通过如下步骤完成:将该盘在人工唾液中浸泡30分钟;将凝胶制剂之一施加至各盘;将凝胶在牙本质表面上摩擦2分钟;等待10分钟;然后冲洗该盘。
在施加纳米颗粒之后立刻和24小时之后,用根据制剂1的纳米颗粒处理的样品显示出约40%的流量减少(相对于处理之前的流量)。在施加纳米颗粒之后立刻和24小时之后,用根据GMB8制剂2的纳米颗粒处理的样品显示出约75%的流量减少。
实施例9——荧光纳米颗粒
将荧光素钠和氯化钙添加到pH约10的水的小瓶中。在小瓶底部处形成了荧光素钙的沉淀物。
根据实施例3中所述的方法用如下物质制备纳米颗粒:74g吐温85;1121g石蜡油;842g水(最初);13g NaOH粉末;39g淀粉;29g NaCl;0.75g荧光素钠;25g GTAC;20g 70%IPA;50g水(用于与GTAC和IPA制备混合物);28g STMP;和预溶在200g水中的27g CaCl2*2H2O。
所得纳米颗粒具有橙黄色,显示出纳米颗粒中荧光素的存在。纳米颗粒可用于为如下淀粉纳米颗粒所述的目的,即通过EDC-NHS化学过程与偶联至淀粉纳米颗粒上的荧光素胺反应的淀粉纳米颗粒,如美国专利申请公开号US 2017/0112949A1(也作为国际公开号WO 2017/070578公开)(用纳米颗粒检测龋齿和微小腔洞(Detection and Treatment ofCaries and Microcavities with Nanoparticles))所述。然而,本文所述的制备荧光纳米颗粒的方法较便宜,并且纳米颗粒同时将有益的试剂递送至龋齿。

Claims (24)
1.一种制造纳米颗粒的方法,所述方法包括以下步骤:
制备包括淀粉在水中的溶液或分散体的第一相;
制备第一相在第二液相如油相中的分散体或乳液;
将以下物质中的一种或多种添加到第一相中:(a)一种或多种阴离子活性剂,(b)一种或多种多价阳离子,或(c)一种或多种淀粉阳离子化剂;以及
用磷酸盐交联剂使第一相中的淀粉交联。
2.如权利要求1所述的方法,其中,阴离子活性剂包含氟化物,所述氟化物任选以氟化钠添加,任选地在制备第一相在第二液相中的乳液或分散体之前添加至第一相。
3.如权利要求1或2所述的方法,其中,阴离子活性剂包含荧光素,所述荧光素任选以荧光素盐如荧光素钠添加,任选地在制备第一相在第二液相中的乳液或分散体之前添加至第一相。
4.如权利要求1至3中任一项所述的方法,其中,所述一种或多种多价阴离子包括钙,所述钙任选地以钙盐添加至第一相,任选地在制备第一相在第二液相中的乳液或分散体的同时或之后添加至第一相。
5.如权利要求1至4中任一项所述的方法,所述方法包括:以足以产生在5.5或更低的pH下具有正ζ电势的纳米颗粒的量添加一种或多种淀粉阳离子化剂。
6.如权利要求1至5中任一项所述的方法,其中,交联剂包括三偏磷酸钠。
7.如权利要求1至6中任一项所述的方法,所述方法包括:以足以产生平均尺寸或数均尺寸范围为100-700nm或100-500nm的纳米颗粒的剪切使水相在油相中的乳液混合,所述平均尺寸或数均尺寸由动态光散射(DLS)中的Z均尺寸测定或由纳米颗粒跟踪分析(NTA)中的数均尺寸测定。
8.一种通过如权利要求1至7中任一项所述的方法生产的纳米颗粒,其任选地结合到水性分散体、凝胶或糊料中。
9.含有淀粉和磷的纳米颗粒,所述磷任选地存在于淀粉-磷酸盐配混物和/或悬挂磷酸盐中,其中,纳米颗粒在5.5或更低的pH下具有正的ζ电势,其中,通过动态光散射(DLS)中的Z均尺寸测定或由纳米颗粒跟踪分析(NTA)中的数均尺寸测定,纳米颗粒的平均尺寸或数均尺寸范围为100-500nm。
10.如权利要求9所述的纳米颗粒,其包含氟化物、荧光素或钙中的一种或多种。
11.如权利要求9所述的纳米颗粒,其包含氟化物和钙。
12.如权利要求9至11中任一项所述的纳米颗粒,其在7.0或更高的pH下具有负ζ电势。
13.如权利要求9至12中任一项所述的纳米颗粒,其结合到水性分散体、凝胶或糊料中。
14.如权利要求8至13中任一项所述的纳米颗粒的用途,用于识别龋齿病灶、用于牙齿再矿化、用于治疗和预防龋齿病灶或者用于治疗牙本质敏感。
15.一种用于治疗牙齿的方法,其包括:任选地在从一个或多个牙齿去除菌斑之后,将如权利要求8至13中任一项所述的纳米颗粒施加至所述一个或多个牙齿上。
16.用钙和/或氟化物强化的多磷酸盐交联淀粉纳米颗粒。
17.用钙和/或氟化物强化的多磷酸盐交联淀粉纳米颗粒的用途,用于填充牙齿或支持牙齿再矿化。
18.一种用于填充牙齿或支持牙齿再矿化的方法,所述方法包括:将用钙和/或氟化物强化的多磷酸盐交联淀粉纳米颗粒给予牙齿。
19.一种通过使用磷酸盐交联剂的乳液法来制备生物聚合物纳米颗粒的方法,其中,氟化物和钙盐中的一种或两种存在于水在第二相中的乳液中。
20.如权利要求16至19中任一项所述,其中,纳米颗粒的尺寸为1000nm或更小。
21.如权利要求16至20中任一项所述,其中纳米颗粒是具有净正ζ电势的阳离子型或两性离子型。
22.如权利要求16至22中任一项所述,其中,纳米颗粒分散于液体、糊料或凝胶载体中,并且施加至牙齿。
23.如权利要求22所述,其中,在清除牙齿上的菌膜和/或菌斑之后或同时,将纳米颗粒施加至牙齿。
24.如权利要求16至23中任一项所述,其中,纳米颗粒包含矿物质,例如,磷酸钙、氟磷灰石或羟基磷灰石钙。
