Abstract
The invention provides a preparation method of bamboo-cypress seed oil, which is prepared by taking bamboo-cypress seeds as raw materials through the steps of crushing, steaming, extruding, standing, filtering and the like. The oil yield of the preparation method can reach 90%. The bamboo-cypress seed oil obtained by the preparation method contains abundant active substances such as oleic acid, linoleic acid, arachidic acid, arachidonic acid, alpha-linolenic acid, peanut dienoic acid, vitamin E, flavone and the like. Wherein the total content of unsaturated fatty acid is over 75%, the content of total flavone is up to 4.23%, and the content of vitamin E is up to 2.11mg/100 g. The said seed oil is edible, and has effects of lowering blood pressure, reducing blood lipid, softening, dilating blood vessel, improving microcirculation, preventing cardiovascular and cerebrovascular diseases, preventing cancer, and improving eyesight.
Description
Technical Field
The invention relates to the technical field of vegetable oil preparation, in particular to a preparation method and application of bamboo-cypress seed oil.
Background
According to the statistics provided by the national food and oil information center, since 1996, the import quantity of edible oil in China far exceeds the total production quantity in China, the edible oil becomes the first edible oil and oil import country in the world, and the self-sufficiency rate of the edible oil in China is less than four times. In recent years, chinese oil materials mainly depend on rapeseed, peanut, linseed, and the like, but it is difficult to ensure safe supply of chinese oil materials and edible oils, and a new approach is required to be developed to make a long-term development. In 2011, the consumption of domestic edible oil reaches 2515 ten thousand tons, the consumption of industry and other edible oil reaches 250 ten thousand tons, the total consumption is 2765 ten thousand tons, and the consumption of each year reaches 20.5 kilograms. In the edible oil in China, the import dependence of high-grade woody (herbaceous) edible oil reaches 95%, and the external dependence of common edible oil also reaches 68%. Therefore, the development of edible oils from other woody (herbaceous) plants is imminent.
Bamboo cypress (Podocarpus nagi), belonging to the family podocarpacaceae (Podocarpaceae), is mainly distributed in the south of the Yangtze river: such as Jiangxi, Zhejiang, Fujian, Hunan, Guangxi, Guangdong, etc. The pericarp and fruit of the bamboo cypress contain abundant volatile components and grease, and especially the oil content in the seed kernel of the bamboo cypress is higher by about 30%. The bamboo cypress contains various active ingredients, such as abundant unsaturated fatty acid, flavonoid, beta-limonene, vitamin E and the like. Therefore, in theory, the bamboo cypress oil should be a high-quality functional edible oil.
Nevertheless, the intensive research on the preparation process, physical and chemical properties, etc. of the bamboo-cypress seed oil is still lacking, and the bamboo-cypress seed oil cannot be developed into the food field.
Disclosure of Invention
In order to overcome the defects of the prior art, the invention provides a preparation method of bamboo cypress seed oil, which takes bamboo cypress seeds as raw materials and comprises the following steps:
(1) crushing the bamboo cypress seeds to obtain bamboo cypress seed powder;
(2) steaming the bamboo cypress seed powder obtained in the step (1);
(3) extruding the steamed bamboo cypress seed powder obtained in the step (2) to obtain oil;
(4) standing and filtering the extruded oil obtained in the step (3) to obtain the bamboo-cypress seed oil.
According to an embodiment of the present invention, the raw material of the bamboo cypress seeds is bamboo cypress nuts (bamboo cypress seeds are peeled).
According to an embodiment of the present invention, the weight of the seeds of bamboo cypress in step (1) is 10 to 100kg, preferably 30 to 70kg, for example 50 kg.
According to an embodiment of the present invention, the particle size of the bamboo cypress seed powder in step (1) may be 20 to 70 mesh, preferably 30 to 60 mesh, for example 40 mesh.
According to an embodiment of the present invention, the steam of step (2) may be implemented by using water vapor, and the equipment of the steam may be any one of a water bath and a steamer; the steaming time may be 5 to 30 minutes, preferably 10 to 25 minutes, more preferably 18 to 22 minutes, for example, about 20 minutes.
According to the embodiment of the invention, the extrusion in the step (3) can be realized by an extrusion device, and the pressure of the extrusion can be 0-50 MPa, for example, the pressure is increased by adopting a 0-30 MPa procedure;
the extrusion time is 5 to 60 minutes, preferably 10 to 40 minutes, more preferably 20 to 30 minutes, for example 25 minutes.
Preferably, the fine powder of steamed bamboo and cypress seeds obtained in step (2) can be artificially pressed into a fixed shape, such as a cake shape, in a fixed die before being extruded.
According to an embodiment of the present invention, the standing time in step (4) is 6 to 48 hours, preferably 8 to 24 hours, for example 12 hours.
According to an embodiment of the present invention, any one of a buchner funnel and a sand core funnel, preferably a 4 μm sand core funnel, may be used for the filtration in step (4).
According to the embodiment of the invention, the oil yield of the method for preparing the bamboo-cypress seed oil can reach 90%, wherein the oil yield is calculated by the following formula: oil yield (weight of oil actually obtained)/(weight of theoretical oil content) 100%.
The invention also provides the bamboo-cypress seed oil prepared by the method.
According to the embodiment of the invention, the content (mass percentage) of the total flavone (calculated by rutin) in the bamboo-cypress seed oil is more than or equal to 4 percent, such as 4.23 percent.
According to an embodiment of the invention, the content of unsaturated fatty acids in the bamboo cypress seed oil is 75% or more, for example 75.5%.
According to an embodiment of the invention, the content of linoleic acid in the bamboo cypress seed oil is more than or equal to 35%, for example 38.2%.
According to an embodiment of the invention, the content of arachidonic acid in the bamboo-cypress seed oil is more than or equal to 1%, for example 1.40%.
According to an embodiment of the invention, the content of arachidilic acid in the bamboo cypress seed oil is 8% or more, for example 8.04%.
According to an embodiment of the invention, the content of vitamin E in the bamboo-cypress seed oil is more than or equal to 2mg/100g, for example 2.11mg/100 g.
According to the embodiment of the invention, the bamboo-cypress seed oil has antioxidant activity, and the clearance rate of oxygen free radicals is more than or equal to 86%, preferably more than or equal to 88%.
The invention also provides application of the bamboo-cypress seed oil, which can be used as a raw material and an additive of functional foods for preventing three highs, oxidation resistance and the like, and can also be used as a raw material for preparing a blood pressure reducing medicine.
The invention has the beneficial effects that:
the invention provides a preparation method of bamboo-cypress seed oil, which can prepare the bamboo-cypress seed oil through simple and easy operation steps, and the oil yield reaches 90 percent. The prepared bamboo-cypress seed oil contains abundant vitamin E, flavonoid substances, unsaturated fatty acid including oleic acid, linoleic acid, arachidonic acid, alpha-linolenic acid, peanut dienoic acid and the like, meets the standard of edible oil, and has the effects of reducing blood pressure, reducing blood fat, softening and expanding blood vessels, improving microcirculation, preventing cardiovascular and cerebrovascular diseases, preventing cancers, improving eyesight, and the like. Wherein, the content of total flavone can reach more than 4 percent, the content of vitamin E is not less than 2mg/100g, and the content of unsaturated fatty acid is not less than 75 percent. The bamboo-cypress seed oil provided by the invention has excellent antioxidant activity, the clearance rate of oxygen free radicals reaches 88%, and meanwhile, the bamboo-cypress seed oil contains rich trace elements necessary for human bodies.
Detailed Description
The present invention will be described in further detail with reference to specific examples. It is to be understood that the following examples are only illustrative and explanatory of the present invention and should not be construed as limiting the scope of the present invention. All the technologies realized based on the above-mentioned contents of the present invention are covered in the protection scope of the present invention.
The experimental methods used in the following examples are all conventional methods unless otherwise specified; reagents, materials and the like used in the following examples are commercially available unless otherwise specified.
Unless otherwise specified, the percentages in the present invention are mass percentages.
Instruments and reagents:
the bamboo cedar seeds are collected in Shenzhou village (provided by Shibi ecological agriculture development Co., Ltd., Fujian province), a crusher, a wooden barrel, an energy-saving water bath steam stove (model: RQ-ZBL-12, produced by Shaqigong hardware kitchen ware factory in Fushan City), and a Yifeng extruder (model: 6Y1.0, produced by Changshan Yifeng oil press factory).
Example 1: preparation of bamboo cypress seed oil
Step 1: pulverizing 100 jin of dried seed of cortex Phellodendri into 40 mesh fine powder;
step 2: filling the crushed fine powder of the bamboo cypress seeds into a wooden barrel with wet gauze laid at the bottom, and putting the wooden barrel on an energy-saving water bath kettle to steam for 20 minutes by using water vapor;
and step 3: placing the steamed bamboo and cypress fine powder in a fixed die, manually pressing the powder into a fixed shape, then placing the powder into an extruder, boosting the pressure in an automatic mechanical device by adopting a program of 0-30 MPa, and extruding for 25 minutes to obtain oil;
and 4, step 4: standing the extruded oil overnight, and filtering with 4 μm sand core funnel under reduced pressure to obtain the final product with faint scent and light yellow transparent liquid (light yellow clear transparent oil).
The inventor finds that in the step 2, the steam cooking time is extremely important for the oil yield quality, the time is too short, the oil yield is not high, the cooking time is too long, the steam content is too high, the oil quality is affected, and the specific expression is that the oil is very turbid (see the following table 1). In the time controlled by the invention, the oil yield is high, and the obtained oil components most accord with the edible standard.

The obtained oil was subjected to component analysis, in which total flavonoids were measured according to "implementation manual for health food inspection and evaluation technical specifications" (2003 edition) P1082[ method for measuring total flavonoids (spectrophotometry) ]; fatty acids were determined according to the method of GB 5009.168-2016; vitamin E was measured according to the method of GB 5009.82-2016. The result shows that the oil contains a large amount of fatty acid, flavone and vitamin E which are needed by human body. The analysis results are shown in Table 2.

Example 2: examples of the use
The obtained bamboo-cypress seed oil product has good food therapy effect, such as certain antioxidant activity and blood pressure lowering effect, and in order to verify the effect, the inventor carries out the following experiments: the process that three people with hypertension eat the oil by Zhang Shibi and the like:
before edible oil, the three blood pressures are respectively: 150.8,160.5,160.0
Every person takes 5ml of the oil respectively before going to bed and fasting in the morning, and takes the oil mixed with vegetable or mixed with noodles at noon (the oil consumption is 5 ml), and every person takes 20 mg of nifedipine sustained release tablets every day in the first week, and the blood pressure of the people is detected every morning, and the blood pressure of the three people are sequentially as follows: 138, 142, 139; beginning in the second week, taking one nifedipine sustained release tablet with the dose of 20 mg every person for 2 days, and detecting the blood pressure in the morning every day, wherein the blood pressure of the three persons fluctuates by 1-2 amounts on the basis of 138, 142 and 139 respectively; beginning in the third week, each person takes one sustained-release tablet of nifedipine 20 mg every 3 days, during which the blood pressure is detected in the morning every day, and the blood pressure of the three persons fluctuates by 1-2 amounts on the basis of 138, 142 and 139 respectively and keeps stable.
Example 3 assay of antioxidant Activity of the bamboo and cypress seed oil product obtained in example 1 by DPPH method
3.1: preparation of DPPH stock solution
And (3) putting DPPH 5mg into a 25ml volumetric flask, diluting with DMSO to a constant volume, fully shaking to prepare DPPH stock solution, and storing in a dark place.
3.2: preparation of DPPH application solutions
The DPPH stock solution and DMSO are prepared into DPPH application solution (10ml) according to the proportion of 1.8:8.2, and the DPPH application solution and DMSO are fully and uniformly mixed to prepare DPPH stock solution which is stored in a dark place.
3.3: absorbance measurement
(1) 5ml of DPPH application solution was taken, and absorbance A0 was measured at 519nm and measured in triplicate.
(2) Taking 5ml of DPPH application liquid, adding 40 μ l of the bamboo-cypress seed oil product sample, mixing thoroughly, shaking, standing for 30min, measuring absorbance A at 519nm, and determining in parallel for three times.
3.4: calculation of clearance
In terms of DPPH clearance% ((A0-A)/A0)% 100%
And drawing a DPPH clearance curve by taking the concentration of the sample as an abscissa and the DPPH clearance as an ordinate. Each of the above 3.1-3.4 steps are performed in parallel five times
The results are shown in table 3 below, which demonstrates that the bamboo-cypress seed oil product of the present invention has excellent antioxidant properties.
Example 4: trace element content test of the obtained bamboo-cypress seed oil product in example 1
4.1 instruments and reagents
Reagent: nitric acid (guaranteed purity); a mixed standard solution (containing: V, Cr, Mn, Fe, Co, Ni, Cu, Zn, As, Sr, 100ug/mL), a Se unit element standard (100ug/mL), a mixed internal standard solution (containing: Sc, Ge, Rh, In, Tb, Lu, 100ug/mL) ultrapure water (resistivity of 18.2m omega cm)-1)。
The instrument comprises the following steps: inductively coupled plasma mass spectrometer (ICP-MS 78000); an MARS microwave digestion system; a Milli-Q pure water treatment system; an electronic analytical balance; all glassware was soaked in dilute nitric acid for 24 hours.
4.2 sample treatment and detection
Sample pretreatment
A sample (0.5 g) was precisely weighed and placed in a Polytetrafluoroethylene (PTFE) digestion tank. Adding 10mL of 65% nitric acid, sealing by a cover, digesting according to the following working conditions (see table 4a), cooling the digestion solution to below 60 ℃ after digestion is completed, taking out the digestion tank, cooling, transferring the digestion solution to a 50mL measuring flask, washing the digestion tank for 3 times by using a small amount of water, merging washing solutions into the measuring flask, fixing the volume by using water, and shaking up to obtain the sample test solution.
The blank control solution was prepared as above.

4.3 preparation of the solution
4.3.1 preparation of Standard working stock solution A mixed standard solution of 10 mixed elements and a nitrogen element standard substance of Se element are precisely weighed respectively and 0.1g of each solution is diluted to a mixed standard stock solution (1000ng/mL) containing 11 metal elements by using 2% nitric acid to fix the volume to 10 mL.
4.3.2 preparation of series of Standard stock solutions an appropriate amount of the above standard working stock solutions was precisely weighed and diluted with 2% nitric acid to form standard series of mixed solutions containing 0.2ng, 1ng, 5ng, 10ng, 50ng, 100ng, 500ng of each element per 1 mL.
4.3.3 preparation of internal standard working solution an appropriate amount of mixed internal standard solution is accurately weighed and diluted to 50ng/mL by 2% nitric acid, thus obtaining the internal standard solution.
4.4 Linear Range Observation: and sequentially measuring a series of standard working solutions with the concentration from small to large, drawing to obtain a standard curve by taking the ratio of the response intensity of the measuring element to the response intensity of the internal standard element as a vertical coordinate and the mass concentration as a horizontal coordinate, and solving a regression equation. The regression equation, correlation coefficient and linear range for each element are shown in table 4c below.

The test result shows that the bamboo-cypress seed oil product of the invention contains rich trace elements necessary for human body (18 kinds of the necessary trace elements which are confirmed to be related to the health and life of human body, namely iron, copper, zinc, cobalt, manganese, chromium, selenium, iodine, nickel, fluorine, molybdenum, vanadium, tin, silicon, strontium, boron, rubidium, arsenic and the like).
The embodiments of the present invention have been described above. However, the present invention is not limited to the above embodiment. Any modification, equivalent replacement, or improvement made within the spirit and principle of the present invention should be included in the protection scope of the present invention.
Claims (10)
1. A preparation method of bamboo cypress seed oil, which takes bamboo cypress seeds as raw materials, is characterized by comprising the following steps:
(1) crushing the bamboo cypress seeds to obtain bamboo cypress seed powder;
(2) steaming the bamboo cypress seed powder obtained in the step (1);
(3) extruding the steamed bamboo cypress seed powder obtained in the step (2) to obtain oil;
(4) standing and filtering the extruded oil obtained in the step (3) to obtain the bamboo-cypress seed oil.
2. The method as claimed in claim 1, wherein the raw material of the seeds of bamboo cedar is seeds of bamboo cedar.
3. The method according to claim 1 or 2, wherein the particle size of the bamboo cypress seed powder in the step (1) is 20-70 mesh.
4. The production method according to any one of claims 1 to 3, wherein the steam of step (2) is carried out by using steam, and the equipment for steam is any one of a water bath and a steamer;
the steaming time is 5-30 minutes, preferably 10-25 minutes, and more preferably 18-22 minutes.
5. The production method according to any one of claims 1 to 4, wherein the extrusion in the step (3) is carried out by an extrusion apparatus, the extrusion pressure is 0 to 50MPa, and the extrusion time is 5 to 60 minutes;
preferably, the fine powder of steamed bamboo and cypress seeds obtained in step (2) is artificially pressed into a fixed shape in a fixed die before being extruded.
6. The method according to any one of claims 1 to 5, wherein the standing time in the step (4) is 6 to 48 hours;
and (4) filtering by using any one of a Buchner funnel and a sand core funnel.
7. The method according to any one of claims 1 to 6, wherein the oil yield of the method is up to 90%.
8. Bamboo cypress seed oil prepared by the method of any one of claims 1-7.
9. The bamboo-cypress seed oil of claim 8, wherein the content of total flavonoids (in terms of rutin) in the bamboo-cypress seed oil is more than or equal to 4%;
the content of unsaturated fatty acid in the bamboo and cypress seed oil is more than or equal to 75 percent;
the content of vitamin E in the bamboo and cypress seed oil is more than or equal to 2mg/100 g.
10. The use of the bamboo cypress seed oil according to claim 8 or 9, which has antioxidant activity, and can be used as raw material and additive for functional food for preventing hypertension, hyperglycemia, hyperlipidemia, and oxidation, and can be used as raw material for preparing antihypertensive drug.
