Description

Millettia speciosa champ extract, preparation method thereof and application thereof in medicines for reducing blood fat and losing weight
Technical Field
The invention belongs to the technical field of medical health-care products, and particularly relates to a beautiful millettia root extract, a preparation method thereof and application thereof in medicines for reducing blood fat and losing weight.
Background
Obesity and dyslipidemia are important risk factors for cardiovascular and cerebrovascular diseases. With the increase of living standard of people, the obesity and overweight people rapidly rise, the obesity becomes one of the great factors causing human death, and at least 280 ten thousand adults die every year in the world due to obesity-related diseases. Hyperlipidemia is one of the most typical complications of obesity, and 1200-1600 million people die of cardiovascular diseases caused by hyperlipidemia every year in the world. Therefore, the research and development of lipid-lowering and weight-losing medicines have great significance in both scientific and civil aspects, and the development potential is great. At present, weight-loss drugs such as sibutramine on the market have side effects such as abnormal liver function although they have some effects.
Radix Millettiae Speciosae is root of Millettia speciosa of Millettia of Leguminosae, and is named as radix Chimaphilae, radix Stephaniae Tetrandrae, rhizoma Nelumbinis, radix seu caulis Kadsurae Heteroclitae, and DALI. Produced in southern areas of China such as Fujian, Hunan, Guangdong, Guangxi, Hainan, Yunnan and Guizhou. The record of the materia medica records beautiful millettia root, strong bones and muscles, detoxication, internal injury regulation, traumatic injury treatment, wine infusion and kidney nourishing. The Chinese herbal medicine compilation records that the beautiful millettia root has sweet nature and flavor, is mild, tonifies deficiency and moistens lung, strengthens tendons and activates collaterals, and is used for lumbar muscle strain, rheumatoid arthritis, phthisis, chronic bronchitis, chronic hepatitis, spermatorrhea and leucorrhea. The Chinese plant record states that the beautiful millettia root has rich starch, can be used for brewing wine and can be used as a medicine, and has the functions of dredging the meridian passage, tonifying deficiency, moistening lung and strengthening spleen. Radix Millettiae Speciosae contains protein, polysaccharide, flavonoid, terpenoids, alkaloid, etc., and has effects of enhancing immunity, eliminating phlegm, relieving cough, resisting inflammation and relieving fatigue. The beautiful millettia root is commonly used for cooking soup, making medicated diet, medicinal liquor and the like in southern areas of China, such as Guangdong, Guangxi, Hainan and other areas, and has the remarkable effects of tonifying kidney and waist and strengthening tendons and bones. The health care and edible value of the beautiful millettia root is very high, and the beautiful millettia root is also a raw material medicine of Chinese patent medicines such as waist strengthening and kidney invigorating pills, strong body building capsules, and radix astragali tendon relaxing and collateral dredging tablets. However, the application of beautiful millettia root medicinal materials has not been developed completely, and the pharmacopoeia does not contain beautiful millettia root medicinal materials.

Example 5
Effect of Millettia speciosa samples on proliferation of precursor adipocytes
Complete medium (DMEM medium supplemented with 10% by volume fetal calf serum, 1% by volume penicillin and streptomycin) was used at 37 deg.C and 5% CO2Culturing 3T3-L1 precursor adipocytes in a saturated humidity incubator, and changing the culture solution every 2-3d for 1 time according to the cell growth state.
Taking 3T3-L1 precursor adipocytes in logarithmic growth phase, after trypsinization, suspending the culture medium into single cells, and diluting the cell concentration to 5 × 104And each/ml is inoculated to a cell proliferation detection culture plate special for RTCA, 100 mu L of cell suspension is added into each hole, after the cells adhere to the wall, culture media containing beautiful millettia root extracts with different concentrations (wherein the concentrations of the beautiful millettia root aqueous extracts are 0.25mg/ml and 0.5mg/ml respectively, the concentrations of the beautiful millettia root polysaccharides are 0.25mg/ml and 0.5mg/ml respectively, the concentrations of the beautiful millettia root polysaccharide filtrates are 0.25mg/ml, 0.5mg/ml and 1mg/ml respectively, and the concentrations of the beautiful millettia root alcohol extracts are 0.25mg/ml, 0.5mg/ml and 1mg/ml respectively) are added, a blank control group is arranged (the culture media do not contain beautiful millettia root extracts), real-time dynamic monitoring is carried out by adopting RTCA software, and instrument software observes a 3T3-L1 precursor adipocyte proliferation curve.
As can be seen from fig. 2, 3, 4 and 5, the beautiful millettia root aqueous extract, the beautiful millettia root polysaccharide filtrate and the beautiful millettia root alcohol extract at different concentrations all inhibited the proliferation of 3T3-L1 precursor adipocytes, and the inhibition ability was enhanced with the increase of the beautiful millettia root extract concentration compared with the blank control group.
Example 6
Inhibiting differentiation of precursor adipocytes into adipocytes
3T3-L1 cells in logarithmic growth phase described in example 5 were cultured at 1 × 105The density of each/ml was seeded in 96-well plates at 100. mu.l per well. After 48 hours, using culture media containing 0.5mmol/L IBMX, 1 mu mol/L dexamethasone, 10mg/L insulin and millettia speciosa champ extracts with different concentrations (0.1mg/ml millettia speciosa champ aqueous extract, 0.1mg/ml millettia speciosa champ polysaccharide, 1mg/ml millettia speciosa champ polysaccharide filtrate and 1mg/ml millettia speciosa champ ethanol extract) to induce and differentiate for 48 hours, then using the culture media containing 10mg/L insulin to culture for 48 hours, finally using complete culture media (10% by volume fetal calf serum and 1% by volume penicillin and streptomycin in total are added into the DMEM culture media to continue culture, changing the culture media 1 time every 2 days, and after culturing day 4, differentiating the cells into mature fat cells, using oil red O to carry out staining. Lipid droplet formation was observed under an inverted microscope and photographed.
As can be seen from fig. 6, compared with the blank control group, the beautiful millettia root aqueous extract, the beautiful millettia root polysaccharide filtrate and the beautiful millettia root alcohol extract can inhibit the 3T3-L1 precursor fat cells from being converted into fat cells, can reduce oil red O staining fat droplets, and have an inhibition effect on the formation of the fat droplets and an anti-obesity effect.

During the test period, all volunteers live according to the respective past living habits such as diet, work and rest, exercise and the like, and the volunteers are weighed and recorded at the beginning of the test; thereafter, the body weight was weighed at the same time every week by the same scale, recorded for eight weeks, and the results were subjected to statistical analysis.
As can be seen from fig. 7 and 8, the weight average of the subjects in 8 weeks showed a downward trend, and the corresponding BMI index also showed a downward trend, and most of the subjects gradually showed a normal range, while the 12# volunteers failed to decrease to a normal level after 8 weeks due to the excessive initial weight and initial BMI index, but the weight gradually decreased and the BMI gradually decreased in the whole 8 weeks.
In addition, during the period of taking the beautiful millettia root polysaccharide, symptoms such as malaise, allergy and the like do not appear in each volunteer.
The foregoing is only a preferred embodiment of the present invention, and it should be noted that, for those skilled in the art, various modifications and decorations can be made without departing from the principle of the present invention, and these modifications and decorations should also be regarded as the protection scope of the present invention.
