The anti-wrinkle and moisturizing compositions prepared in examples 1 to 6 were subjected to acute oral toxicity verification and efficacy verification, and the verification results were as follows.
(1) Acute oral toxicity verification
The test samples were the wrinkle resistant moisturizing compositions prepared in examples 1 to 6. The test method refers to the operation method of a one-time limit method in an acute toxicity test of health food inspection and evaluation technical specification (2003 edition), and the sample to be tested acts on the test mouse in a stomach-perfusion mode in batches with a normal dose of 4.5mL/kg and a high dose of 20 mL/kg.
The test object was Kunming white mouse.
The specific test operation is as follows:
screening 60 female and male mice respectively, dividing the mice into 12 groups, and 5 female and male mice in each group, wherein 6 groups are tested in a normal dosage, and numbering is carried out according to corresponding test samples; high dose test 6 groups, also numbered according to the corresponding test samples.
The test mice were fasted for 8 hours before gavage, observed for 7 days after gavage, and the general status, toxicity performance and death number of the mice were recorded. The test results were averaged by panel to give tables 1 and 2.
TABLE 1 test mouse mortality and abnormal Activity rates
TABLE 2 weight change before and after gastric lavage of test mice
From the results of tables 1 and 2, it can be seen that the mice in the normal group and the high dose group had normal activities, a mortality rate of 0, and a normal weight gain after the gavage. It is demonstrated that the anti-wrinkle moisturizing compositions prepared in examples 1 to 6 of the present invention are safe and non-toxic.
(2) Efficacy verification
60 volunteers between 28-45 years of age were selected and divided into 6 groups.
Firstly, detecting the moisture, grease and elasticity of the face of a volunteer by using a Real Bube RBX-916 detector to obtain initial data; then, each group of volunteers used the anti-wrinkle moisturizing composition prepared in examples 1 to 6, 50mL per day, and continuously took for 8 weeks; the water, grease and elasticity of the facial skin of the volunteers are detected by a Real Bubee RBX-916 detector at a fixed time every week, and data are recorded. The average values were obtained to compare the moisture, oil and skin elasticity data of the skin of the volunteers before and after using the anti-wrinkle moisturizing compositions prepared in examples 1 to 6.
The results are shown in fig. 1, fig. 2 and fig. 3, which are obtained by recording and comparing the facial skin moisture, oil and elasticity data of volunteers before, after 1 week, 2 weeks, 3 weeks, 4 weeks, 5 weeks, 6 weeks, 7 weeks and 8 weeks after the administration of the anti-wrinkle moisturizing composition, and averaging the data of each group. The results showed that the moisture content of facial skin of volunteers was significantly increased (see fig. 1), the lipid content was significantly decreased (see fig. 2), and the elasticity of facial skin was increased (see fig. 3) after the collagen products prepared in examples 1 to 6 were administered.
Secondly, detecting the facial wrinkles and spots of the volunteers by using a Visia tester, and taking a picture for recording; then, each group of volunteers used the anti-wrinkle moisturizing composition prepared in examples 1 to 6, 50mL per day, and continuously took for 3 months; after 3 months, the facial wrinkles and spots of the volunteers are detected by a Visia tester and photographed and recorded. Note: the volunteers do not take the anti-wrinkle moisturizing composition prepared by the invention at the same time when taking the anti-wrinkle moisturizing composition.
By comparing skin conditions of the volunteers before and after using the anti-wrinkle moisturizing compositions prepared in examples 1 to 6, it can be seen that wrinkles and spots on the facial skin of the volunteers are remarkably reduced. As shown in fig. 4 a-9 b, after the volunteers used the collagen drinks prepared in examples 1, 2 and 4 of the present invention for 3 months, the facial wrinkles and spots were significantly improved, which proves that the technical solution of the present invention has the ability of collagen supplementation and oxidation resistance.
