本发明提供了一种纺锤形氧化铁纳米材料及其制备方法和应用,属于纳米材料技术领域。本发明采用水热法,通过调控反应物的浓度并控制反应温度,能够制备出纺锤形Fe2O3纳米材料,且该纺锤形Fe2O3纳米材料的尺寸可控、形貌规则、结构均一。本发明所制备的纺锤形Fe2O3纳米材料可获得巨大表面能等材料优势,在生物分析与检测及电化学生物传感器、细胞和蛋白质分离与识别、磁共振成像、肿瘤磁热疗、药物靶向及缓释、抗菌效应等生物医学领域有着广泛的应用前景。

本发明涉及纳米材料技术领域,尤其涉及一种纺锤形氧化铁纳米材料及其制备方法和应用。
背景技术
磁性氧化铁纳米材料是指铁的氧化物及其羟基氧化物的纳米材料。以价态、晶型、结构的不同可将最为常见的氧化铁纳米材料分为Fe2O3(α、β、γ相)、Fe3O4、FeO以及FeOOH(α、β、γ、δ相)等。其中,α-Fe2O3、γ-Fe2O3和Fe3O4纳米材料因具有较高的稳定性(包括化学和热力学等)、抗腐蚀特性、催化活性、电化学特性以及传感特性等,表现出较好的实用价值,并已在磁性材料、化学颜料、活性催化剂、气敏元件以及电极材料等领域得到较为广泛的应用。近些年来,国内外针对氧化铁纳米材料的研究也主要集中于这三种。
Fe3O4为磁铁矿,反尖晶石型,其结构式可表示为Fe3+[Fe2+Fe3+]O4,其中一半Fe3+存在于四面体间隙中,Fe2+与另一半Fe3+存在于八面体间隙中。由于Fe2+与Fe3+交替排列在晶体中,在电场影响下Fe2+与Fe3+易产生电子转移,故而Fe3O4具有较高的导电性和磁性。当Fe2+完全转化为Fe3+,八面体结构中产生空位,转变为γ-Fe2O3,但仍然保持了磁铁矿的结构,具有很强的磁性,称为磁赤铁矿。γ-Fe2O3与Fe3O4具有类似的结构,加热γ-Fe2O3到一定的温度后会完全转化为α-Fe2O3,即赤铁矿。α-Fe2O3为刚玉型结构(菱形中心六面晶体结构),具有较高的化学稳定性。γ-Fe2O3为立方晶体结构四面体,Fe2+离子仅占据八面体位置。通过不同的制备方法可得到不同形貌的磁性氧化铁纳米材料,目前文献报道过的磁性氧化铁纳米材料有球形、棒状、四面体形、纺锤形、环状等诸多不规则的形貌结构。
磁性氧化铁纳米材料除具有可控的磁性能、优异的光学性能和催化性能外,还具有生物相容性好、毒副作用小、低免疫原型、表面易修饰等特点,在生物医学领域具有广阔的应用前景。目前,对磁性氧化铁纳米材料的合成与应用从基础理论、制备方法到其应用技术都有了比较深入的研究,但仍存在以下关键问题尚待解决:(1)磁性氧化铁纳米材料固有的小尺寸及表面自旋斜交效应所导致的饱和磁化强度低、磁响应缓慢等缺陷凸显,影响了它在生物医学领域的应用性能。(2)在蛋白质检测领域,目前利用磁性氧化铁纳米颗粒在NMR系统中的信号检测多局限于磁场强度较高的医学场,其弛豫性质研究一直受到由颗粒尺寸小而导致的稳定性差、易团聚等问题的困扰,还受到检出方法复杂、检测周期长、灵敏度低等问题困扰。(3)磁性氧化铁纳米材料因其过小的尺寸,不易在肿瘤组织定位和滞留,当作为被动靶向药物载体时,不利于抗癌药物在病变部位有效富集,限制了其在载药方面的应用。
针对以上问题,研究人员试图通过调节磁性氧化铁纳米材料的尺寸及几何结构,以便能够发现一种性能更为优良的新型磁性氧化铁纳米材料来弥补传统磁性氧化铁纳米材料的不足。
发明内容
本发明的目的在于提供一种纺锤形氧化铁纳米材料及其制备方法和应用,所制备的氧化铁纳米材料呈纺锤形、尺寸可控、形貌规则、结构均一。
为了实现上述发明目的,本发明提供以下技术方案:
本发明提供了一种纺锤形氧化铁纳米材料的制备方法,包括以下步骤:
将磷酸盐类化合物、硫酸盐、铁盐和水混合,将所得反应液进行水热反应,干燥后得到纺锤形氧化铁纳米材料;
所述磷酸盐类化合物、硫酸盐和铁盐的摩尔比为(0.01~0.02):(1.5~1.8)×10-4:(5.5~9.5)×10-4;
所述水热反应的温度为180~220℃,时间为32~48h。
优选的,所述磷酸盐类化合物为磷酸氢钠、磷酸氢钾、磷酸二氢钠、磷酸二氢钾、磷酸三钠或磷酸钾。
优选的,所述硫酸盐为硫酸钠、硫酸钾或硫酸镁。
优选的,所述铁盐为三氯化铁或硝酸铁。
优选的,所述磷酸盐类化合物、硫酸盐和铁盐的摩尔比为(0.012~0.018):(1.6~1.7)×10-4:(6.0~9.0)×10-4。
优选的,所述水热反应的温度为190~210℃,时间为36~42h。
优选的,所述干燥的温度为60~80℃,时间为3~6h。
本发明提供了上述技术方案所述制备方法制备得到的纺锤形氧化铁纳米材料。
优选的,所述纺锤形氧化铁纳米材料为中空结构,两端开口;所述纺锤形氧化铁纳米材料的长径为500nm~1μm。
本发明提供了上述技术方案所述纺锤形氧化铁纳米材料在吸附蛋白质中的应用。
本发明提供了一种纺锤形氧化铁纳米材料的制备方法,包括以下步骤:将磷酸盐类化合物、硫酸盐、铁盐和水混合,将所得反应液进行水热反应,干燥后得到纺锤形氧化铁纳米材料;所述磷酸盐类化合物、硫酸盐和铁盐的摩尔比为(0.01~0.02):(1.5~1.8)×10-4:(5.5~9.5)×10-4;所述水热反应的温度为180~220℃,时间为32~48h。本发明采用水热法,通过调控反应原料的浓度并控制反应温度,能够制备出纺锤形Fe2O3纳米材料,且该纺锤形Fe2O3纳米材料的尺寸可控、形貌规则、结构均一。本发明所制备的纺锤形Fe2O3纳米材料可获得巨大表面能等材料优势,在生物分析与检测及电化学生物传感器、细胞和蛋白质分离与识别、磁共振成像、肿瘤磁热疗、药物靶向及缓释、抗菌效应等生物医学领域有着广泛的应用前景。
本发明的制备工艺简便,成本低。

具体实施方式
本发明提供了一种纺锤形氧化铁纳米材料的制备方法,包括以下步骤:
将磷酸盐类化合物、硫酸盐、铁盐和水混合,将所得反应液进行水热反应,干燥后得到纺锤形氧化铁纳米材料;
所述磷酸盐类化合物、硫酸盐和铁盐的摩尔比为(0.01~0.02):(1.5~1.8)×10-4:(5.5~9.5)×10-4;
所述水热反应的温度为180~220℃,时间为32~48h。
在本发明中,若无特殊说明,所需制备原料均为本领域技术人员熟知的市售商品。
本发明将磷酸盐类化合物、硫酸盐、铁盐和水混合,将所得反应液进行水热反应。在本发明中,所述磷酸盐类化合物优选为磷酸氢钠、磷酸氢钾、磷酸二氢钠、磷酸二氢钾、磷酸三钠或磷酸钾;所述硫酸盐优选为硫酸钠、硫酸钾或硫酸镁;所述铁盐优选为三氯化铁或硝酸铁。在本发明中,所述磷酸盐类化合物、硫酸盐和铁盐独立优选为水合物盐或无水盐。
在本发明中,所述磷酸盐类化合物、硫酸盐和铁盐的摩尔比为(0.01~0.02):(1.5~1.8)×10-4:(5.5~9.5)×10-4,优选为(0.012~0.018):(1.6~1.7)×10-4:(6.0~9.0)×10-4,更优选为(0.015~0.016):(1.62~1.66)×10-4:(7.0~8.0)×10-4。
本发明对所述水的用量没有特殊的限定,能够将原料充分溶解使其满足上述摩尔比即可。
在本发明中,所述磷酸盐类化合物、硫酸盐、铁盐和水混合的过程优选在超声条件下进行,本发明对所述超声的过程没有特殊的限定,能够将物料充分溶解即可。在本发明的实施例中,所述超声的时间具体为3min。
完成所述混合后,本发明优选将所得反应液进行水热反应,所述水热反应的温度为180~220℃,优选为190~210℃,更优选为200℃,时间为32~48h,优选为36~42h,更优选为38~40h。本发明优选在反应釜中进行所述水热反应,本发明对所述反应釜没有特殊的限定,选用本领域熟知的反应釜即可。
在所述水热反应过程中,硫酸根离子和磷酸根离子发挥协同作用,即双阴离子吸附在纳米晶体(铁离子与磷酸根粒子形成的络合物)表面,不同晶面优先溶解,各向异性生长;另外双阴离子与铁离子存在配位效应,能够加速溶解过程,促进纺锤形ɑ-Fe2O3纳米晶体形成。
在本发明中,纺锤状Fe2O3纳米材料在其生长和溶解过程中,磷酸根离子可以通过替换铁氧化物表面酸性较弱的羟基(-OH)来降低铁氧化物表面的电势,其替换为磷酸根离子通过内圈络合的形式(与-OH进行配位交换)吸附在-Fe2O3的表面。在酸性条件下,磷酸根离子与-Fe2O3表面的单配位羟基交换易形成≡Fe-PO4 2-络合物,其与H+作用发生质子化后形成≡Fe-PO4H2和≡Fe-PO4H-两种络合物,而不同晶面的络合程度将对其对应晶面生长和溶解产生直接影响;随着晶体的不断生长及沿c轴的优先溶解,长度逐渐增加,最终形成纺锤状纳米材料。
完成所述水热反应后,本发明将所得物料自然冷却至室温后,倒出反应釜上清液,将釜中的下层沉淀样品进行干燥,得到纺锤形氧化铁纳米材料。在本发明中,所述干燥的温度优选为60~80℃,更优选为65~75℃,时间优选为3~6h,更优选为4~5h。
本发明提供了上述技术方案所述制备方法制备得到的纺锤形氧化铁纳米材料。在本发明中,所述纺锤形氧化铁纳米材料为中空结构,两端开口。在本发明中,所述纺锤形氧化铁纳米材料的长径为500nm~1μm。
本发明提供了上述技术方案所述纺锤形氧化铁纳米材料在吸附蛋白质中的应用。本发明对所述应用的方法没有特殊的限定,按照本领域熟知的方法应用即可。
下面将结合本发明中的实施例,对本发明中的技术方案进行清楚、完整地描述。显然,所描述的实施例仅仅是本发明一部分实施例,而不是全部的实施例。基于本发明中的实施例,本领域普通技术人员在没有做出创造性劳动前提下所获得的所有其他实施例,都属于本发明保护的范围。
实施例1
将二水磷酸二氢钠0.0562g、无水硫酸钠0.1562g和六水三氯化铁10.76g混合放入烧杯中,加入少量蒸馏水,进行超声溶解3min,将溶解好的溶液放入容量瓶中,配制成1000mL溶液,再将配好的反应液倒入反应釜中,并封装;将封装好反应釜在220℃的条件下持续反应48h;待反应釜自然冷却至室温后,倒出反应釜上清液,将釜中的下层沉淀样品放入烧杯中,在80℃下干燥4h,得到样品,记为1C。
实施例2
本实施例与实施例1的区别仅在于:二水磷酸二氢钠、无水硫酸钠和六水三氯化铁的质量为实施例1中对应各原料质量的2倍,其他条件同实施例1,所得样品记为2C。
实施例3
本实施例与实施例1的区别仅在于:二水磷酸二氢钠、无水硫酸钠和六水三氯化铁的质量为实施例1中对应各原料质量的3倍,其他条件同实施例1,所得样品记为3C。
性能测试
1)对实施例1~3制备的样品进行XRD测试,采用X-射线衍射仪分析样品的晶体结构、相组成以及晶胞参数的精确测定,实验中采用步进扫描方式收集衍射数据,每步0.02°,扫描速度为每秒一步,用多晶粉末衍射法对产物进行结构分析,来精确测量晶胞参数,2θ角扫描范围为0°到80°,结果见图1。
通过分析图1可以确定,实施例1~3制备的样品均为纯相的α-Fe2O3。对样品的特征峰的位置与标准六方Fe2O3相一致,且三种样品粉体的XRD衍射图谱的特征峰也基本一致,说明样品的特征衍射图谱不受浓度的影响,实施例1~3的不同浓度不会影响产物Fe2O3的合成。此外,图中没有杂峰,说明最终样品中没有其他物质存在,纯度较高,其中较尖锐的峰说明样品结晶度较高。
2)对实施例1~3制备的样品进行SEM测试,结果见图2~7:
图2~5为实施例2制备的样品在不同放大倍数下的扫描电镜图,其中,图2为扫描倍数MAG=5.00KX下的扫描电镜图;由图2可以看出,样品颗粒的形貌呈现出两种尺寸大小的规格,其中大颗粒部分呈现出环状和长圆柱状(米粒状),两端有开口,小颗粒未观察到环状结构,分散性差,主要呈堆积状。
图3为扫描倍数为MAG=10.00KX下的扫描电镜图,由图3可以看出,大小不同的两种颗粒的数量相差大,大颗粒数量多于小颗粒的数量,且大颗粒两端的开口明显。
图4为扫描倍数为MAG=25.00KX下的扫描电镜图;图4中只有大颗粒的形貌,其呈纺锤形,且两端开口十分明显。
图5为扫描倍数为MAG=20.00KX下的扫描电镜细节图,图5中呈现单个米粒结构,能够明显看出单个米粒呈现中空结构,且两端开口。
由图2~5综合观察,实施例2制备的样品呈现纺锤形,且颗粒的大小一致,尺寸均一,两端开口,中空结构,分散性好。
图6为实施例1制备的样品的SEM图,图7为实施例3制备的样品的SEM图,分析图2~7可知,实施例1~3在不同浓度下制备的α-Fe2O3纺锤形的形貌,随着浓度的升高,环状颗粒的长度拉长,呈纺锤形,两端有开口,中空结构;尺寸大小相差越大,分散性较好。
3)对实施例2制备的样品进行TEM表征和元素分析,结果见图8,其中,图8中的a为单一纺锤形Fe2O3颗粒的形貌特征图,图8中的b为纺锤形Fe2O3颗粒中铁元素的分布图,图8中的c为纺锤形Fe2O3颗粒中氧元素的分布图。由图8可知,Fe2O3颗粒呈现出纺锤形形貌,其单体均由Fe、O两种主要元素均匀分布构成。
应用例
实施例2制备的α-Fe2O3纳米颗粒对蛋白质吸附性能研究:
1、牛血清白蛋白(BSA)标准液的制备
(1)用分析天平准确称量牛血清白蛋白10mg,称量完毕后马上放入冰箱中,在2~8℃下保存。用去离子水使其在烧杯中溶解,移入100mL容量瓶中定容,得到浓度为0.1mg·mL-1的牛血清白蛋白溶液。
(2)取六个相同的容量瓶,依次贴上标签纸,编号分别为1、2、3、4、5、6号,用1000μg的移液枪分别按表1取样,再在六个称量瓶中分别加入5mL刚配制好的考马斯亮蓝溶液,考马斯亮蓝溶液用5mL量筒量取。
表1牛血清白蛋白标准曲线的测定
将牛血清蛋白标准溶液、去离子水和考马斯亮蓝溶液加入小称量瓶后,轻轻摇匀(若猛烈摇晃会产生大量泡沫),静置五分钟后进行紫外-可见分光光度测量。
通过以上步骤,以牛血清白蛋白的浓度为横坐标,以吸光度值为纵坐标,建立牛血清白蛋白的标准曲线,如图9所示。根据标准曲线拟合的线性方程为y=5.2377x+0.0331,相关系数R2=0.99293,说明紫外-可见分光光度测量方法在对牛血清白蛋白的测定中具有很好的线性关系,可用于测量牛血清白蛋白。
2、不同初始浓度BSA溶液的配制
为了研究牛血清白蛋白的浓度对吸附量Q的影响,进行以下实验操作:将pH=5的磷酸盐缓冲溶液作为溶剂,配制五瓶初始浓度分别为1.75mg·mL-1、2.0mg·mL-1、2.25mg·mL-1、2.5mg·mL-1、2.75mg·mL-1的牛血清白蛋白溶液。分别取100mL上述牛血清白蛋白溶液置于五个锥形瓶中并依次标好浓度,准确称取一定质量的纺锤形Fe2O3纳米颗粒依次加入五个锥形瓶中,同时制作空白对照样加入100mL、pH=5的磷酸盐缓冲溶液,测吸光度时作为参比对照组。将六个锥形瓶超声分散10min后静置3h(包括超声分散的10min)。静置完毕后,取六个称量瓶标上对应的编号,用1000μg移液枪从锥形瓶中各取1mL牛血清白蛋白溶液至对应编号的称量瓶中,再在各个称量瓶中加入5mL考马斯亮蓝溶液,用紫外-可见光分光光度计进行测量,每组读数三次并保存数据。
3、不同初始浓度BSA对吸附量的影响
根据步骤1所得标准曲线,计算上述步骤2中不同初始浓度BSA溶液下的紫外测试数据,所得浓度与初始浓度之差即为BSA的吸附量,对纺锤形Fe2O3纳米颗粒的吸附量进行比较,结果见图10,由图10可知,在一定浓度范围内,纺锤形Fe2O3纳米颗粒对牛血清白蛋白的吸附量随着牛血清白蛋白初始浓度的增加而增大,当牛血清白蛋白的初始浓度达到2.75mg·mL-1时,吸附量逐渐达到饱和状态。
以上所述仅是本发明的优选实施方式,应当指出,对于本技术领域的普通技术人员来说,在不脱离本发明原理的前提下,还可以做出若干改进和润饰,这些改进和润饰也应视为本发明的保护范围。
Claims (10)
1.一种纺锤形氧化铁纳米材料的制备方法,其特征在于,包括以下步骤:
将磷酸盐类化合物、硫酸盐、铁盐和水混合,将所得反应液进行水热反应,干燥后得到纺锤形氧化铁纳米材料;
所述磷酸盐类化合物、硫酸盐和铁盐的摩尔比为(0.01~0.02):(1.5~1.8)×10-4:(5.5~9.5)×10-4;
所述水热反应的温度为180~220℃,时间为32~48h。
2.根据权利要求1所述的制备方法,其特征在于,所述磷酸盐类化合物为磷酸氢钠、磷酸氢钾、磷酸二氢钠、磷酸二氢钾、磷酸三钠或磷酸钾。
3.根据权利要求1所述的制备方法,其特征在于,所述硫酸盐为硫酸钠、硫酸钾或硫酸镁。
4.根据权利要求1所述的制备方法,其特征在于,所述铁盐为三氯化铁或硝酸铁。
5.根据权利要求1所述的制备方法,其特征在于,所述磷酸盐类化合物、硫酸盐和铁盐的摩尔比为(0.012~0.018):(1.6~1.7)×10-4:(6.0~9.0)×10-4。
6.根据权利要求1所述的制备方法,其特征在于,所述水热反应的温度为190~210℃,时间为36~42h。
7.根据权利要求1所述的制备方法,其特征在于,所述干燥的温度为60~80℃,时间为3~6h。
8.权利要求1~7任一项所述制备方法制备得到的纺锤形氧化铁纳米材料。
9.根据权利要求8所述的纺锤形氧化铁纳米材料,其特征在于,所述纺锤形氧化铁纳米材料为中空结构,两端开口;所述纺锤形氧化铁纳米材料的长径为500nm~1μm。
10.权利要求8或9所述纺锤形氧化铁纳米材料在吸附蛋白质中的应用。
