NSAID在急性和慢性病中的疼痛以及术后疼痛的处置中起主要作用,且更广泛地,NSAID在所有其中疼痛与炎症相关的那些病症中均起主要作用。
在NSAID中,酮洛芬、酮洛芬盐、典型地为细胞溶素盐(lysin salt)和右酮洛芬(dexketoprofen,酮洛芬的右旋立体异构体)、典型地为氨丁三醇盐(trometamol salt)等属于最具活性的NSAID,它们涉及丙酸衍生物类别,广泛被开据处方并且在一些国家作为非处方药得到。它是非常有潜力的,非常有效地缓解因急性和慢性情况下的创伤、骨科和风湿病导致的疼痛,以及处置儿童和成年人的发热。
除对环加氧酶的作用外,酮洛芬还可逆地抑制脂氧合酶,而脂氧合酶介导花生四烯酸转化成白三烯,类二十烷酸(eicosanoid)炎症介质家族。酮洛芬还显示出抑制缓激肽(一种炎症和疼痛化学介质),并防止溶酶体酶的释放,而溶酶体酶负责炎症反应中组织破坏的介导。
口服施用的酮洛芬很容易被胃肠道吸收,其峰值浓度为0.5-2小时;其特征在于半衰期短(1-4小时),在肝脏中迅速代谢,并且其代谢物在尿中排泄,实际上没有生物蓄积性(口服施用后24h内排泄率约为80%)。
有意义地,已经证实,酮洛芬和其他NSAID一样具有周围和中枢作用部位,由于其脂溶性而迅速通过血脑屏障。
所有这些特征均有助于快速起效,灵活的剂量和可靠的耐受特性。
然而,酮洛芬的特征还在于在水性介质中的溶解性和稳定性差,其中水溶性在22℃为0.051mg/mL,且pKa为4.45。大部分NSAID共有的这些特征使得难以在药物组合物中、特别是在液体剂型中配制酮洛芬。实际上,如大部分NSAID一样,酮洛芬对口腔、咽喉和咽部具有化学感觉(刺激剂)作用,并具有苦味。此外,苦味也被描述为源自用于溶解NSAID和/或降低其刺激作用的成分。
这些问题已在本领域得到解决,并提出了几种不同的解决方案,例如US 5895789、WO 99/52528、US 2012/0208887、WO 2004/05454、US 5183829和WO 2007/112274中所述。
申请人已经在WO 2005/058276中面对了这些问题,其中公开了包含NSAID并且具有良好适口性的药物口服剂型。该组合物利用氨丁三醇来溶解药物并消除化学感觉作用,而甘氨酸、维生素B6或其混合物则用来克服苦味。
EP1974751公开了包含NSAID的药物组合物,其中使用β-环糊精和氨丁三醇实现增溶并抑制化学感觉作用和苦味。WO 97/18245中公开了一种类似的组合物,其专门用于萘普生。这些申请解决了掩蔽NSAID的苦味和化学感觉作用的问题,但是没有解决在其他成分例如防腐剂存在下这些溶液的稳定性的技术问题。
如所述的,即在上述举出的专利和专利申请中,环糊精广泛应用于增强疏水性药物的水溶性和稳定性以及掩味剂。
环糊精为通过成环的α-(1,4)键连接的α-D-吡喃葡萄糖苷单元构成的环状寡糖,且最常见的由6(α-环糊精)、7(β-环糊精)或8(γ-环糊精)个单元构成。其特征在于疏水腔和亲水表面,由此能够通过置换存在于腔中的水分子形成包合物而截留客体分子。
不受特定理论的束缚,将环糊精描述为,能够嵌入其中的分子提供空腔。然而,所谓“复合物”,申请人预期包含这样一种复合物,其中一种成分(主体)形成空腔,该空腔包含长隧道或通道形状的空间,其中第二种化学种类(客体)的分子实体位于其中(包合物),或掩盖苦味和化学感觉作用必不可少的不同成分的简单组合,它们以特定的摩尔比存在,并且能够满足与NSAID施用相关的技术问题,且能够保证化学物理和微生物学稳定性。
本发明的复合物未通过共价键连接,不同分子之间的吸引力通常归因于范德华力以及疏水和偶极-偶极相互作用。就包合物而言,它们是环糊精与客体分子之间的尺寸、几何形状有限的配合,驱动力是疏水客体分子对腔的亲和力,而该复合物的稳定性依赖于主体与客体之间的分子间相互作用的次数。
尽管环糊精广泛用作溶解度增强剂,但它们显示出有限的水溶性,β-环糊精是最不可溶的物质之一,其在水中的溶解度为18mg/mL。这种差的溶解性主要归因于分子中存在的各种羟基之间形成的分子间和分子内氢键。由于这一原因,已经合成了几种具有不同OH取代度的衍生的环糊精,以便在水溶性和与客体分子相互作用的能力方面定制其性能。
在β-环糊精衍生物羟丙基-β-环糊精中,无定形亲水衍生物显示出改善的水溶性(600mg/mL)、低毒性和令人满意的配合能力。磺丁基醚-β-环糊精(SBECD)也为具有改善溶解度的β-环糊精衍生物。
如上所述,EP1974751和WO 97/18245中已经公开了氨丁三醇、羟基烷基胺与环糊精一起在溶解NSAID中的用途。氨丁三醇能够稳定环糊精与药物之间的包合物,并形成三元复合物,其中氨丁三醇与环糊精和药物之间显示出很强的分子间相互作用,从而不仅增强药物溶解度,而且特别地增强该复合物的掩味作用。
尽管如此,但是环糊精与氨丁三醇之间的相互作用这样的能力(尽管已经与该药物形成复合物)也表明环糊精与最终存在于溶液中的其他适合物质相互作用的能力。
实际上,已经证实,环糊精在药物或其他客体分子存在下也趋向于与水溶性聚合物形成牢固的复合物(R.S.Hirlekar等人Studies on the Effect of Water-SolublePolymers on Drug–Cyclodextrin Complex Solubility,AAPS PharmSciTech 2009,10(3),858-863;T.Loftsson等人The effect of water-soluble polymers on the aqueoussolubility and complexing abilities ofβ-cyclodextrin,International Journal ofPharmaceutics 1998,163(1–2))。所得的复合物改变了药物与环糊精之间的结合常数,同时降低了溶液中游离聚合物的浓度。
当试图获得一种液体组合物时,例如当作为本发明的那些口服施用时,或者添加额外的成分时,例如当制备具有更高粘度的凝胶组合物时,这变得尤为重要。用于口服或以其他方式施用的液体药物组合物通常包含几种药学上可接受的赋形剂,以获得期望的制剂并确保长期贮存的稳定性以及微生物学稳定性。
分子与大分子、例如环糊精之间的相互作用是高度不可预测的,并且就所用分子的性质和量而言难以控制。
因此,获得具有良好的适口性、在延长存储期间显示出良好的物理-化学和微生物学稳定性的的组合物是非显而易见的。
发明内容
申请人面临的问题在于得到包含酮洛芬的用于口服施用的液体组合物,该液体组合物具有良好的适口性并具有改善的化学-物理和微生物学稳定性。
特别地,申请人面临获得用于口服施用的液体组合物的问题,该液体组合物包含酮洛芬、β-环糊精和烷基胺的复合物,具有良好适口性并具有改善的化学-物理和微生物学稳定性。
实际上,申请人注意到,酮洛芬、羟基烷基胺和β-环糊精的复合物产生具有良好适口性的水溶性组合物,但其化学-物理和微生物学稳定性并不令人满意。实际上,已经发现,β-环糊精的存在可能降低常用于制药领域的一些防腐剂的活性,从而可能导致欧洲药典(European Pharmacopoeia)要求的挑战测试失败。
复合物的絮凝和/或沉淀也可能损害低温下的稳定性。
申请人目前令人惊讶地发现,除所述酮洛芬复合物外,由对羟基苯甲酸甲酯和对羟基苯甲酸丙酯组成的防腐剂系统还能够确保得到的液体组合物在长期储存后的物理学、化学和微生物学稳定性。
因此,本发明的第一个方面涉及液体组合物,其包含:(i)a)酮洛芬、右酮洛芬及其盐、b)羟基烷基胺和c)β-环糊精或衍生物,例如羟丙基-β-环糊精或磺丁基醚-β-环糊精(SBECD),组成的复合物,其中所述羟基烷基胺选自:氨丁三醇、乙醇胺、二乙醇胺、三乙醇胺、葡甲胺(meglumina)、2-氨基-2-甲基-1,3-丙二醇和2-氨基-1,2,3,-丙三醇,最优选氨丁三醇、二乙醇胺和三乙醇胺;和(ii)对羟基苯甲酸甲酯和对羟基苯甲酸丙酯组成的防腐剂系统,其中酮洛芬或其衍生物在所述溶液中的量等于或低于2%w/V。更优选地,酮洛芬以等于或低于1.5%的w/V浓度使用,更优选0.01%-1%的浓度,甚至更优选0.2%–0.8%的浓度,包含约0.5%的优选浓度。因此,基于分子量差异调整酮洛芬细胞溶素盐和右酮洛芬氨丁三醇(dexketoprofen trometamol)的w/V浓度。例如,0.5%酮洛芬酸的浓度相当于约0.8%w/V酮洛芬细胞溶素盐和约0.74%w/V右酮洛芬氨丁三醇。
以摩尔计,由a)酮洛芬、右酮洛芬及其盐,b)羟基烷基胺和c)β-环糊精组成的复合物i)包含相对于活性成分至少为3倍摩尔比的羟基烷基胺和0.05–1摩尔比的β-环糊精。
发明详述
本发明涉及液体组合物,其包含(i)复合物,优选由如下成分组成:a)酮洛芬、右酮洛芬及其盐、b)羟基烷基胺和c)β-环糊精或衍生物,例如羟丙基-β-环糊精(HP-β-CD)或磺丁基醚-β-环糊精(SBECD),其中所述羟基烷基胺优选选自:氨丁三醇、乙醇胺、二乙醇胺、三乙醇胺、葡甲胺、2-氨基-2-甲基-1,3-丙二醇和2-氨基-1,2,3,-丙三醇,最优选氨丁三醇、二乙醇胺和三乙醇胺;和(ii)防腐剂系统,其由对羟基苯甲酸甲酯和对羟基苯甲酸丙酯组成,其中酮洛芬或其衍生物在所述溶液中的量等于或低于2%w/V。
可以预期,在下文中,术语β-环糊精是指未修饰的和上述举出的β-环糊精衍生物。
以摩尔计,优选由a)酮洛芬、右酮洛芬及其盐、b)羟基烷基胺和c)β-环糊精组成的复合物i)包含相对于活性成分至少为3摩尔过量(其中甚至更优选选择3.5-6.5摩尔比)的羟基烷基胺,以及0.05–1摩尔比的β-环糊精。
下文中的术语酮洛芬预期包含酮洛芬酸和酮洛芬的盐,优选细胞溶素盐和右酮洛芬(酮洛芬的右旋立体异构体),典型地为氨丁三醇盐。
更优选地,酮洛芬以等于或低于2%的w/V浓度、更优选0.01%-1.5%的浓度、甚至更优选0.2%–1%的浓度使用,包含约0.5%w/V的优选浓度。酮洛芬细胞溶素盐和右酮洛芬氨丁三醇的以w/V计的浓度由此可以基于分子量差异调整。例如,0.5%酮洛芬酸浓度相当于约0.8%w/V酮洛芬细胞溶素盐和约0.74%w/V右酮洛芬氨丁三醇,其中所谓高于%w/V预期为相对于组合物总体积的值。因此,2%w/V酮洛芬酸浓度相当于3.6%w/V酮洛芬细胞溶素盐和约3%w/V右酮洛芬氨丁三醇。
在本发明的液体组合物中,羟基烷基胺以相对于活性成分至少3-倍摩尔过量,或更优选相对于活性成分至少4-倍摩尔过量,乃至更优选至少5-倍摩尔过量存在,其优选自:氨丁三醇、乙醇胺、二乙醇胺、三乙醇胺、葡甲胺、2-氨基-2-甲基-1,3-丙二醇和2-氨基-1,2,3,-丙三醇,最优选氨丁三醇、二乙醇胺和三乙醇胺,最优选氨丁三醇、三乙醇胺和二乙醇胺。
更优选地,羟基烷基胺以相对于活性成分至少为约6倍的摩尔过量存在,其中所谓约6我们是指包含相对于活性成分5.5-6.5摩尔过量的范围。甚至更优选地,羟基烷基胺相对于活性成分的摩尔过量包含从上述下限值到约10的上限值的范围。
特别优选约6-倍的羟基烷基胺摩尔过量,其中羟基烷基胺优选为氨丁三醇。
术语“约”是指这样的数值,其包含关注的值和+/-同一所述值的1%-2%的变化形式。
如上所述,本发明的液体组合物优选包含β-环糊精或衍生物,其相对于活性成分的摩尔比为0.05-1。相对于活性成分,更优选β-环糊精或衍生物的摩尔比由0.1-7或更优选0.3-0.6构成。甚至更优选β-环糊精或衍生物的摩尔比由0.35-0.55构成。
特别优选这样的组合物,其中复合物i)包含约0.5%w/V酮洛芬,羟基烷基胺为氨丁三醇,β-环糊精为HP-β-环糊精,它们分别以近似于如下的摩尔比存在:1/6/0.5。
该复合物中的上述和摩尔比允许良好的适口性,并且在长期储存后在4℃在溶液中保持稳定而没有沉淀和絮凝。
然而,可能由于与复合物中的β-环糊精的相互作用,仅有很少的防腐剂使本发明的组合物具有良好的微生物学稳定性。实际上,申请人已经确定了一对防腐剂,它们在不改变化学-物理性质的情况下为液体组合物提供长期的微生物学稳定性。
因此,根据这些观察结果,本发明的液体组合物包含相对于组合物的总体积为0.005-1%w/V的量的对羟基苯甲酸甲酯,并且优选还包含相对于组合物的总体积量为0.001-0.5%w/V的对羟基苯甲酸丙酯。
更优选地,本发明的液体组合物包含相对于组合物的总体积量为0.01-0.5%w/V的对羟基苯甲酸甲酯。
甚至更优选地,本发明的液体组合物包含相对于组合物的总体积量为0.1-0.3%w/V的对羟基苯甲酸甲酯。
本发明的液体组合物优选包含相对于组合物的总体积量为0.001-0.5%w/V的对羟基苯甲酸丙酯。
更优选地,本发明的液体组合物包含相对于组合物的总体积量为0.005-0.25%w/V的对羟基苯甲酸丙酯。
甚至更优选地,本发明的液体组合物包含相对于组合物的总体积量为0.01-0.1%w/V的对羟基苯甲酸丙酯。
此外,根据本发明的一个优选实施方案,所述液体组合物包含重量比约10:1-约1:1、更优选约8:1-约2:1的对羟基苯甲酸甲酯:对羟基苯甲酸丙酯。
最优选地,所述液体组合物包含重量比约6:1-约4:1的对羟基苯甲酸甲酯:对羟基苯甲酸丙酯。
优选地,本发明的液体药物组合物为用于口服施用的水性制剂。
优选地,将本发明的液体药物组合物制备成适合的剂型,例如溶液、混悬液、糖浆剂、凝胶和喷雾剂。更优选地,所述剂型为溶液或凝胶。甚至更优选地,所述剂型为粘性溶液或凝胶。
水优选用作本发明液体药物组合物的主要溶剂,特别是去离子水、纯水或蒸馏水等。
本发明的药物组合物可以包含另外的药学上可接受的成分和/或赋形剂。
术语药学上可接受的赋形剂应理解为包含、但不限于任何适合于制备待施用于生物体的液体药物组合物的材料,例如助溶剂,稳定剂,抗氧化剂,pH校正剂,缓冲剂,表面活性剂,螯合剂,着色剂,增香剂,糖,甜味剂和/或香料。
有利地,本发明的液体药物组合物包含一种或多种矫味剂,例如葡萄柚香精、覆盆子香精、柠檬香精、橙子香精、焦糖香精、香草香精、奶油香精等。
有利地,本发明的液体药物组合物包含一种或多种甜味剂,例如阿司帕坦、糖精、乙酰舒泛、三氯蔗糖等。
有利地,本发明的液体药物组合物包含一种或多种糖,例如乳糖、葡萄糖、蔗糖等。
有利地,本发明的液体药物组合物包含一种或多种螯合剂,例如二乙烯三胺五乙酸(DTPA)、乙二胺四乙酸(EDTA)、次氮基三乙酸(NTA)等。
优选地,本发明的液体药物组合物包含一种或多种共溶剂,其选自二元醇和多元醇,例如甘油、丙二醇、1,3-丁二醇等。
必须口服施用的水性组合物的pH优选接近中性,即为5-8,优选5.2-7.5,更优选5.5-6.5。
在一个优选的实施方案中,本发明的液体药物组合物为包含粘度改进剂的水凝胶。
优选地,粘度改进剂为亲水性聚合物,选自藻酸盐、卡波姆、聚丙烯酸酯、纤维素衍生物(例如羟乙基纤维素、羟丙基纤维素和羧甲基纤维素)、树胶(例如黄原胶、瓜尔胶)、蛋白质(例如明胶)和果胶、和高分子量多糖(例如卡拉胶)。
在如此获得的水凝胶组合物中,相对于药物组合物的总体积,粘度改进剂的存在量为0.01-1.0%w/V。相对于组合物的总体积,甚至更优选粘度改进剂的量为0.01-1.0%w/V,且甚至更优选由0.30-0.50%w/V构成。
在凝胶组合物中,额外存在的粘度改进剂(典型地为聚合物)需要进一步评估稳定性,如实验部分详细描述的。因此,根据该优选的实施方案,复合物i)由如下组成:a)酮洛芬、右酮洛芬或其盐;b)羟基烷基胺;和c)β-环糊精,相对于活性成分酮洛芬或其衍生物,羟基烷基胺至少为3–倍摩尔比,优选3-7摩尔比,甚至更优选3.5-6.5摩尔比;以及β-环糊精,其摩尔比通常低于在液体组合物中摩尔比,即0.08-0.4,更优选0.1-0.4,甚至更优选约0.35摩尔比。
活性成分等于或低于2%w/V。更优选地,酮洛芬是酮洛芬酸,并且以等于或低于1.5%w/V浓度使用,更优选以0.01%-1%的浓度使用,甚至更优选以0.2%–0.8%w/V的浓度使用,包含约0.5%的优选浓度,其中上述%w/V浓度旨在包含该范围的上限和下限,并涉及最终组合物的总体积。基于分子量差异,将相应调整以w/V计的酮洛芬细胞溶素盐和右酮洛芬氨丁三醇浓度。例如,浓度为0.5%w/V的酮洛芬酸相当于约0.8%w/V的酮洛芬细胞溶素盐和0.74%w/V的右酮洛芬氨丁三醇。
所述凝胶组合物还包含以上定义的具有相同量和优选量的防腐剂系统。
根据一个优选的实施方案,该组合物包含粘度改进剂和复合物i),其包含下列成分或由其组成:约0.5%w/V的酮洛芬,羟基烷基胺为氨丁三醇,β-环糊精为2-HP-β-环糊精,其中活性成分/羟基烷基胺和β-环糊精分别以如下近似的摩尔比存在:1/6/0.35。
优选地,本发明的液体药物组合物的特征在于等于或高于1mPa*s,且优选等于或低于2000mPa*s的粘度。甚至更优选地,本发明的液体药物组合物的特征在于500mPa*s-1500mPa*s的粘度。最优选地,本发明的液体药物组合物的特征在于约1000mPa*s的粘度。
实验例
材料
实施例1-适口性测试
制备酮洛芬(0.5w/V%)和递增量的氨丁三醇的五种水溶液,并进行了适口性测试,以评估氨丁三醇掩蔽酮洛芬的化学感觉作用和苦味的能力。
水溶液1-5中包含的氨丁三醇的量如下表1中所述。

NSAID对口腔粘膜的刺激性显示出显著的个体差异性,因此必须适当选择用于适口性测试的个体组。实际上,对于某些来体而言,刺激性可能是“略微微明显的”,其他人则将其定义为“强”或“非常强(Breslin等人"Ibuprofen as a chemesthetic stimulus:evidence of a novel mechanism of throat irritation",Chem.Sens.26:55-65,2001)。为了选择仅对NSAID的刺激作用明显敏感的那些个体,进行了初步测试,施用包含0.5%w/V酮洛芬酸的水溶液。
要求40位20-40岁之间的个体服在用该溶液时遵循以下标准程序:-啜10ml去离子水,在口腔中保持10秒钟,且然后吞咽,-啜10ml溶液,在口腔中保持10秒钟,且然后吞咽。
给出了正确定义可感知刺激性刺激的指征,如下所示:

然后,要求这40位个体在施用后的0、30秒,1分钟和5分钟的时间,在考虑上述每种刺激的情况下,评估口腔中的刺激强度,并给他们评分,将感觉定义为“强”的为3分。将感觉定义为“中度”的为2分,将感觉定义为“温和”的为1分,将感觉定义为未引起刺激的为0分。
因此,仅选择对酮洛芬产生的不适感表现出更大敏感性(总计超过40分)的那些个体。
然后按照相同的步骤并如上所述给予分数,将溶液1-5施用于20位选择的个体。
在这种情况下,使用了更多的评估时间点,因为要求20位个体评估在施用后0、30秒、1分钟、2分钟、3分钟、5分钟、10分钟和15分钟时的口腔刺激强度和感知的味觉。
分别计算每位个体的灼烧感、刺痛感、针刺感和麻木感的评估总和(0-15分钟),以及所有感觉的评估总和(0-15分钟)。
还要求个体描述所感知到的苦味,将苦味描述为“强”的人员定为3分,将苦味描述为“中度”的人员定为2分,描述为“适度”的人员定为1分。
通过Wilcoxon“符号秩检验”方法分析这些参数以比较溶液。最终评分如下表2所示。

实施例2–稳定性测试
对溶液2-5(在前述实施例1中证实没有化学感觉作用)进行化学-物理稳定性测试,以通过将样品维持在低温(4℃)下3个月来验证不存在沉淀和/或絮凝。

实施例3–适口性测试
制备4种包含0.5%的酮洛芬、1.5%的氨丁三醇和增加量的2-羟丙基-β-环糊精(2HP-β-CD)的水溶液(溶液6-8)并进行适口性测试以评估2HP-β-CD从溶液中消除苦味的能力。
如上述实施例1中所述进行测试,将结果与水溶液6-9中所含的2HP-β-CD的量一起概括在下表4中。

首先评估样品在制备过程中可能导入的任何微生物或病原体的存在。然后,将给每种样品接种200μL不同的微生物,并在30-35℃下温育多达28天,以便给样品接种细菌,并且在20-25℃下温育多达28天,以便给样品接种真菌。
接种后(时间0)以及温育14和28天后立即分析全部样品,以评估每mL样品中的活细菌或真菌细胞数(CFU/mL)。
对于测试的4种细菌的任一种,防腐剂挑战测试需要截止到第14天距初始计数不小于3.0的微生物浓度对数减少,且在第28天的微生物浓度水平相对于第14天时测定的那些无增加。对于3种真菌的任一种,需要截止到第14天距初始计数不少于1.0的微生物浓度对数减少,且在第28天时微生物浓度水平无增加。仅当对于所有测试微生物表现出阳性结果时,才将组合物视为符合。
下表7显示对于液体组合物10-15各自得到的结果。

实施例5–测试在4℃下的物理稳定性
然后将前述实施例4中所述的液体组合物13-18进行化学-物理稳定性测试,以通过将样品在低温(4℃)下维持3个月来验证不存在沉淀和/或絮凝。

仅包含对羟基苯甲酸甲酯/对羟基苯甲酸丙酯作为防腐剂系统的组合物13和16显示出足够的稳定性。
因此,由对羟基苯甲酸甲酯/对羟基苯甲酸丙酯组成的防腐剂系统是经证实唯一能够同时确保包含0.5%的酮洛芬、1.5%的氨丁三醇和1-1.5%的2HP-β-CD的液体组合物的微生物学和物理-化学稳定性的防腐剂系统。
实施例6–微生物稳定性测试
制备分别包含1和1.5%2HP-β-CD的水凝胶组合物22和23,且其组成在下表11中所述。

按照上述实施例4中描述的相同方法进行挑战测试,且将结果概括在下表12中。

从表12可以明显看出的,添加获得粘性组合物所需的亲水聚合物(例如黄原胶)会影响所得组合物的微生物学稳定性。在上述实施例中,浓度为1.5%w/V的2HP-β-CD显示与聚合物发生相互作用,可能还与防腐剂系统发生相互作用,使得该组合物不符合要求。
为确保微生物学稳定性,与在水凝胶中选择的防腐剂系统一起使用的2HP-β-CD的最大量应低于1.5%.%w/V,相对于环糊精的不同分子量进行调整在本领域中是众所周知的。
实施例7–制备例–液体组合物
本发明的液体组合物可以由下表13中所述的组合物表示,其中使用不同的β-CD,例如磺丁基醚-β-环糊精(SBECD)、2羟丙基-β-环糊精(2HP-β-CD)和β-环糊精(β-CD)以及酮洛芬的不同盐或对映体形式。

实施例9–长期物理、化学和微生物学稳定性.
根据ICH Q1A“新药物物质和产品的稳定性测试”指南、在如下ICH条件下测试从预试验中得到的稳定的组合物:
-25℃/60%RH
-30℃/65%RH
-40℃/75%RH
6个月时的结果如表15中所示:

对于所有测试条件和所有参数,本试验均呈正向,表明活性成分在溶液中在6个月时是稳定的,并且在表15所指向的凝胶组合物中也是稳定的,防腐剂得以保持,且溶液中没有任何污染,如上所述。
还在ICH条件下在12和18个月时评估组合物。在30℃和65%湿度(30℃/65%RH)的条件下在12个月时并且在25℃和60%湿度(25℃/60%RH)的条件下在18个月时它们均保持稳定。
概括而言,评估了根据本发明的组合物的物理-化学和微生物学特性的稳定性,直至18个月。
初步评估了API浓度为1%的凝胶制剂的稳定性,评价如下:
-预期的物理稳定性应力(4℃ 1个月):结果符合产品规格;
-预期的化学稳定性的应力(50℃ 1个月):结果符合产品规格,此外,如果与ICH稳定性下的制剂比较,则在相同条件下(50℃ 1个月)施加应力;和
-微生物学测试(Ph.Eur)以确保制剂的微生物学质量和防腐。
在上述测定条件下,证实凝胶制剂是符合标准和稳定的。
Claims (20)
1.水性组合物,包含:(i)由a)酮洛芬或选自其盐和对映体的衍生物、b)羟基烷基胺和c)β-环糊精组成的复合物;和(ii)由对羟基苯甲酸甲酯和对羟基苯甲酸丙酯组成的防腐剂系统,其中在复合物i)中,相对于活性成分,羟基烷基胺以至少为3的摩尔比存在,而β-环糊精以0.05-1的摩尔比存在,且活性成分的量等于或低于2%w/V。
2.权利要求1的水性组合物,其中所述酮洛芬衍生物选自:右酮洛芬、右酮洛芬氨丁三醇和酮洛芬细胞溶素盐。
3.权利要求1-2任一项的水性组合物,其中羟基烷基胺选自:氨丁三醇、乙醇胺、二乙醇胺、三乙醇胺、葡甲胺、2-氨基-2-甲基-1,3-丙二醇和2-氨基-1,2,3,-丙三醇。
4.权利要求3的水性组合物,其中羟基烷基胺选自:氨丁三醇、二乙醇胺和三乙醇胺。
5.权利要求3-4任一项的水性组合物,其中羟基烷基胺以相对于活性成分至少为4的摩尔比存在于复合物i)中。
6.权利要求3-4任一项的水性组合物,其中羟基烷基胺以相对于活性成分为3-7的摩尔比存在于复合物i)中。
7.权利要求1-6任一项的水性组合物,其中β-环糊精选自:2-HP-β-环糊精和磺丁基醚-β-环糊精。
8.权利要求1-6任一项的水性组合物,其中β-环糊精为2-HP-β-环糊精。
9.权利要求1-8任一项的水性组合物,其中β-环糊精相对于活性成分的摩尔比为0.1-0.7。
10.权利要求1-8任一项的水性组合物,其中复合物i)中β-环糊精相对于活性成分的摩尔比为0.08-0.4。
11.权利要求10的水性组合物,还包含粘度改进剂。
12.权利要求11的水性组合物,其中所述粘度改进剂为亲水性聚合物,其选自藻酸盐;卡波姆;聚丙烯酸酯;纤维素衍生物,例如羟乙基纤维素、羟丙基纤维素和羧甲基纤维素;树胶,例如黄原胶、瓜尔胶;蛋白质和高分子量多糖。
13.权利要求12的水性组合物,其中所述蛋白质选自明胶和果胶。
14.权利要求12-13任一项的水性组合物,其中所述高分子量多糖为卡拉胶。
15.权利要求11-14任一项的水性组合物,其中所述粘度改进剂的存在量相对于该药物组合物的总体积为0.01-1.0%w/V。
16.权利要求1-15任一项的水性组合物,其中对羟基苯甲酸甲酯的存在量相对于该组合物的总体积为0.005-1%w/V。
17.权利要求1-16任一项的水性组合物,其中对羟基苯甲酸甲酯的存在量相对于该组合物的总体积为0.01-0.5%w/V。
18.权利要求16-17任一项的水性组合物,其中对羟基苯甲酸丙酯的量相对于该组合物的总体积为0.001-0.5%w/V。
19.权利要求18的水性组合物,其中对羟基苯甲酸丙酯的量相对于该组合物的总体积为0.005-0.25%w/V。
20.权利要求1-19任一项的水性组合物,其中pH为5-8。
