(7) preparing the folium artemisiae argyi-honeysuckle traditional Chinese medicine extract, namely adding the folium artemisiae argyi extract and the honeysuckle extract prepared in the step (1) into propylene glycol according to the mass ratio of 9: 2, and controlling the total concentration of the traditional Chinese medicine extract in the formula to be 20 mug/m L, which is recorded as 1 f.
The results of the measurements are shown in table 1, using the inhibition of Propionibacterium acnes, the inhibition of Propionibacterium acnes outer membrane vesicle release, and the anti-inflammatory effect as indicators.

The table 1 shows that the sophora flavescens extract, the artemisia leaf extract and the honeysuckle extract have a good effect of inhibiting propionibacterium acnes, the artemisia leaf extract has an effect of inhibiting the release of outer membrane vesicles of propionibacterium acnes, and the honeysuckle extract has an effect of enhancing an anti-inflammatory effect and reducing cytotoxicity, each extract has no cytotoxicity when acting alone, but has a poor effect of reducing I L-8, namely, the final anti-inflammatory effect is poor.
Example 2
The embodiment provides a traditional Chinese medicine extract with multi-target acne removing efficacy and a preparation method thereof:
(1) the preparation method of the sophora flavescens extract comprises the steps of drying sophora flavescens to constant weight, crushing, sieving with a 60-mesh sieve, carrying out reflux extraction for 1: 8 by using 80% ethanol solution according to the material-liquid ratio of g: m L for 1 hour, continuously stirring, filtering after the first extraction is finished, extracting residues once by using 80% ethanol, filtering, combining filtrates obtained in two times, carrying out rotary evaporation and concentration on the filtrate to obtain extract, and carrying out freeze drying to obtain the sophora flavescens extract;
the preparation method of the folium artemisiae argyi extract comprises the steps of drying folium artemisiae argyi to constant weight, crushing, sieving with a 60-mesh sieve, performing reflux extraction for 1h by using a 60% ethanol solution according to a material-liquid ratio of g: m L, continuously stirring, filtering after the first extraction is finished, extracting residues once by using 60% ethanol, filtering, combining filtrates obtained in the two times, performing rotary evaporation and concentration on the filtrate to obtain an extract, and performing freeze drying to obtain the folium artemisiae argyi extract;

(3) preparing radix Sophorae Flavescentis Chinese medicinal extract by adding radix Sophorae Flavescentis extract in (1) into propylene glycol, and controlling concentration of radix Sophorae Flavescentis extract to be 40 μ g/m L, as 2 b;
(4) preparing folium Artemisiae Argyi traditional Chinese medicine extract by adding folium Artemisiae Argyi extract in (1) into propylene glycol, controlling folium Artemisiae Argyi extract concentration to 40 μ g/m L, and recording as 2 c;
(5) preparing a honeysuckle traditional Chinese medicine extract, namely adding the honeysuckle extract in the step (1) into propylene glycol, and controlling the concentration of the honeysuckle extract to be 40 mug/m L, which is recorded as 2 d;
(6) preparing a radix sophorae flavescentis-honeysuckle flower traditional Chinese medicine extract, namely adding the radix sophorae flavescentis extract and the honeysuckle flower extract prepared in the step (1) into propylene glycol according to the mass ratio of 9: 2, controlling the total concentration of the traditional Chinese medicine extract in the formula to be 40 mug/m L, and recording as 2 e;
(7) preparing the folium artemisiae argyi-honeysuckle traditional Chinese medicine extract, namely adding the folium artemisiae argyi extract and the honeysuckle extract prepared in the step (1) into propylene glycol according to the mass ratio of 9: 2, and controlling the total concentration of 40 mug/m L in the traditional Chinese medicine extract in the formula as 1 f.
The results of the measurements are shown in Table 2, using Propionibacterium acnes-inhibiting, Propionibacterium acnes-inhibiting outer membrane vesicle release-inhibiting, and anti-inflammatory effects as indices.

As can be seen from Table 2, when radix Sophorae Flavescentis and folium Artemisiae Argyi were used alone, the cytotoxicity was too high to be practical. The cytotoxicity of the traditional Chinese medicine extract is reduced after combination, and example 2 has the effects of inhibiting the reproduction of propionibacterium acnes and inhibiting bacteria from releasing outer membrane vesicles, and can play a better role in inhibiting acne inflammation than a single extract under the condition of no cytotoxicity.
Example 3
The embodiment provides a traditional Chinese medicine extract with multi-target acne removing efficacy and a preparation method thereof:
(1) the preparation method of the sophora flavescens extract comprises the steps of drying sophora flavescens to constant weight, crushing, sieving with a 60-mesh sieve, carrying out reflux extraction for 1h by using 80% ethanol solution according to the material-liquid ratio of g: m L, continuously stirring, filtering after the first extraction is finished, extracting residues once by using 80% ethanol, filtering, combining filtrates obtained in two times, carrying out rotary evaporation and concentration on the filtrate to obtain extract, and carrying out freeze drying to obtain the sophora flavescens extract;
the preparation method of the folium artemisiae argyi extract comprises the steps of drying folium artemisiae argyi to constant weight, crushing, sieving with a 60-mesh sieve, performing reflux extraction for 1h by using a 60% ethanol solution according to the material-liquid ratio of g: m L, continuously stirring, filtering after the first extraction is finished, extracting residues once by using 60% ethanol, filtering, combining filtrates obtained in the two times, performing rotary evaporation and concentration on the filtrate to obtain an extract, and performing freeze drying to obtain the folium artemisiae argyi extract;

(2) Adding the sophora flavescens extract, the artemisia leaf extract and the honeysuckle extract prepared in the step (1) into propylene glycol according to the mass ratio of 7: 7: 6, and controlling the total concentration of the traditional Chinese medicine extract in the formula to be 40 mu g/m L, which is recorded as 3 a.
(3) Preparing a sophora flavescens traditional Chinese medicine extract, namely adding the sophora flavescens extract prepared in the step (1) into propylene glycol, and controlling the concentration of the sophora flavescens extract to be 40 mu g/m L, which is recorded as 3 b;
(4) preparing folium Artemisiae Argyi traditional Chinese medicine extract by adding folium Artemisiae Argyi extract in (1) into propylene glycol, controlling folium Artemisiae Argyi extract concentration to 40 μ g/m L, and recording as 3 c;
(5) preparing a honeysuckle traditional Chinese medicine extract, namely adding the honeysuckle extract in the step (1) into propylene glycol, and controlling the concentration of the honeysuckle extract to be 40 mug/m L, which is recorded as 4 d;
(6) preparing a radix sophorae flavescentis-honeysuckle flower traditional Chinese medicine extract, namely adding the radix sophorae flavescentis extract and the honeysuckle flower extract prepared in the step (1) into propylene glycol according to the mass ratio of 7: 6, controlling the total concentration of the traditional Chinese medicine extract in the formula to be 40 mug/m L, and recording as 4 e;
(7) preparing folium artemisiae argyi-honeysuckle traditional Chinese medicine extract, namely adding the folium artemisiae argyi extract and the honeysuckle extract prepared in the step (1) into propylene glycol according to the mass ratio of 7: 6, and controlling the total concentration of the traditional Chinese medicine extract in the formula to be 40 mug/m L, which is recorded as 4 f.

As can be seen from Table 3, when radix Sophorae Flavescentis and folium Artemisiae Argyi were used alone, the cytotoxicity was too high to be practical. The traditional Chinese medicine extract has reduced cytotoxicity after combination, has the effects of inhibiting the reproduction of propionibacterium acnes and inhibiting bacteria from releasing outer membrane vesicles, and can play a better role in inhibiting acne inflammation than a single extract under the condition of no cytotoxicity. Fig. 1 is a comparison graph of cytotoxicity of extracts and combinations thereof, and it can be seen from fig. 1 that cytotoxicity is generated at a lower concentration when a certain herb extract is used alone, and irritation may be generated to the skin, which makes practical application difficult. The extract combination can improve the application concentration of the extract, and can achieve better effect on relieving acne inflammation. The traditional Chinese medicine extract has no additive effect on cytotoxicity, and the formula not only reduces the cytotoxicity, but also has the effects of inhibiting the reproduction of propionibacterium acnes and inhibiting bacteria from releasing outer membrane vesicles, and can effectively inhibit acne inflammation.
Example 4
The embodiment provides a traditional Chinese medicine extract with multi-target acne removing efficacy and a preparation method thereof:
(1) the preparation method of the sophora flavescens extract comprises the steps of drying sophora flavescens to constant weight, crushing, sieving with a 60-mesh sieve, carrying out reflux extraction for 1h by using 80% ethanol solution according to the material-liquid ratio of g: m L, continuously stirring, filtering after the first extraction is finished, extracting residues once by using 80% ethanol, filtering, combining filtrates obtained in two times, carrying out rotary evaporation and concentration on the filtrate to obtain extract, and carrying out freeze drying to obtain the sophora flavescens extract;
the preparation method of the folium artemisiae argyi extract comprises the steps of drying folium artemisiae argyi to constant weight, crushing, sieving with a 60-mesh sieve, performing reflux extraction for 1h by using a 60% ethanol solution according to the material-liquid ratio of g: m L, continuously stirring, filtering after the first extraction is finished, extracting residues once by using 60% ethanol, filtering, combining filtrates obtained in the two times, performing rotary evaporation and concentration on the filtrate to obtain an extract, and performing freeze drying to obtain the folium artemisiae argyi extract;

(4) Preparing folium Artemisiae Argyi traditional Chinese medicine extract by adding folium Artemisiae Argyi extract in (1) into propylene glycol, controlling folium Artemisiae Argyi extract concentration to 80 μ g/m L, and recording as 4 c.
(5) And (2) preparing a honeysuckle traditional Chinese medicine extract, namely adding the honeysuckle extract in the step (1) into propylene glycol, and controlling the concentration of the honeysuckle extract to be 80 mu g/m L, which is recorded as 4 d.

It can be seen that the traditional Chinese medicine extract formula has a good antibacterial effect and also plays a certain role in inhibiting the release of vesicles and I L-8, but cell death is caused by excessively high addition amount, and the addition amount may damage human skin, so that practical application is difficult.

The embodiment provides a traditional Chinese medicine extract formula and a preparation method thereof:
(1) the preparation method of the angelica sinensis extract comprises the steps of drying angelica sinensis to constant weight, crushing, sieving by a 60-mesh sieve, carrying out reflux extraction for 1h by using 80% ethanol solution according to the material-liquid ratio of g: m L for 1: 6, stirring continuously, filtering after the first extraction is finished, extracting residues by using 80% ethanol for one time, filtering, combining filtrates obtained in the two times, carrying out rotary evaporation and concentration on the filtrate to obtain extract, and carrying out freeze drying to obtain the angelica sinensis extract;
the preparation method of the lotus leaf extract comprises the steps of drying lotus leaves to constant weight, crushing, sieving by a 60-mesh sieve, carrying out reflux extraction for 1h by using 80% ethanol solution according to the material-liquid ratio of g: m L, continuously stirring, filtering after the first extraction is finished, extracting residues by using 80% ethanol once, filtering, combining filtrates obtained in the two times, carrying out rotary evaporation and concentration on the filtrate to obtain extract, and carrying out freeze drying to obtain the lotus leaf extract;
the preparation method of the mulberry leaf extract comprises the steps of drying mulberry leaves to constant weight, crushing, sieving with a 60-mesh sieve, carrying out reflux extraction for 3 hours by using 80% ethanol solution according to the material-liquid ratio of 1: 8 in terms of g: m L, continuously stirring, filtering after the first extraction is finished, extracting residues once by using 80% ethanol, filtering, combining filtrates obtained in the two times, carrying out rotary evaporation and concentration on the filtrate to obtain extract, and carrying out freeze drying to obtain the mulberry leaf extract.

(2) Preparing Chinese angelica extract, namely adding the Chinese angelica extract prepared in the step (1) into propylene glycol, and controlling the concentration of the Chinese angelica extract to be 20 mu g/m L, which is recorded as 5 b.
(3) And (2) preparing a lotus leaf traditional Chinese medicine extract, namely adding the lotus leaf extract prepared in the step (1) into propylene glycol, and controlling the concentration of the lotus leaf extract to be 20 mu g/m L, which is recorded as 5 c.
(4) And (3) preparing a mulberry leaf traditional Chinese medicine extract, namely adding the mulberry leaf extract prepared in the step (1) into propylene glycol, and controlling the concentration of the mulberry leaf extract to be 20 mu g/m L, wherein the concentration is recorded as 5 d.
The results of the measurements are shown in Table 5, using Propionibacterium acnes-inhibiting, Propionibacterium acnes-inhibiting outer membrane vesicle release-inhibiting, and anti-inflammatory effects as indices.

From table 5, it can be seen that the formula of the traditional Chinese medicine extract, angelica sinensis has a weak vesicle inhibition effect, lotus leaves have a weak bacteriostatic effect, and folium mori has two effects but has a weak effect. 5a extract has high cytotoxicity and weak anti-acne inflammation effect, and is difficult to have practical application.
Example 6
On the basis of example 3 (the total concentration of the traditional Chinese medicine extract in the formula is controlled to be 40 mu g/m L), the influence of the compounding ratio of the sophora flavescens extract, the artemisia leaf extract and the honeysuckle extract on the inhibition of propionibacterium acnes, the inhibition of the release of outer membrane vesicles of propionibacterium acnes and the anti-inflammatory effect is discussed, and the conditions and the results are shown in table 6.

As can be seen from table 6, in the same case of experiment 2 in which the total concentration was 40 μ g/m L, the cytotoxicity was the least and the inhibition ratio of I L-8 was the best, i.e., the anti-inflammatory effect was the best, when the radix angelicae sinensis extract or the folium mori extract was added to the formulation, the cytotoxicity was greatly increased and the anti-inflammatory effect was deteriorated, compared to the case where the weight ratio of the radix sophorae flavescentis extract to the folium artemisiae argyi extract to the flos lonicerae extract was 1: 2: 1.

The preparation method of the cosmetic comprises the following steps: mixing and heating the phase A to 75-85 ℃ to be completely dissolved, mixing and heating the phase B to 40-50 ℃, adding the completely dissolved phase B into the phase A at 75-85 ℃ and uniformly stirring, cooling to below 40 ℃ after uniformly mixing the A, B phases, adding the phase C and uniformly stirring, and preparing the cosmetic.
The formula is verified to meet the pear flower index evaluation of cosmetics in cold resistance, heat resistance, pH value, viscosity, centrifugal test and the like; and the stability tests such as 7-cycle hot-cold circulation, appearance, physical and chemical indexes, active ingredients, microstructure and the like are performed, and the sanitation index evaluation such as corrosion resistance challenge, mercury, arsenic and lead content test and the like is simultaneously met.
As can be seen from Table 1, the composition (the traditional Chinese medicine extract with the multi-target acne removing effect prepared in example 3) is used in the formula at a concentration of 20-50 μ g/m L, and the addition amount of the composition accounts for 3% of the weight of the cosmetic, if the concentration of the composition in the formula exceeds 50 μ g/m L, certain toxicity is shown in a cell experiment, and certain safety risk exists, and when the concentration of the composition in the formula is lower than 20 μ g/m L, the acne inflammation inhibition effect is poor.
The invention discloses a composition with multi-target-point acne removing effect and a preparation method and application thereof. The inhibition of the formula on the propionibacterium acnes reproduction is embodied in a multi-target form, and the formula can ensure the efficacy of inhibiting acne inflammation and the safety. The sophora flavescens, the folium artemisiae argyi and the honeysuckle extracts in the formula can inhibit the reproduction of propionibacterium acnes and can also inhibit propionibacterium acnes from releasing outer membrane vesicles, and the effect of inhibiting acne inflammation is achieved.
Meanwhile, the inventor finds that when one traditional Chinese medicine extract is singly used in the sophora flavescens extract, the artemisia leaf extract and the honeysuckle extract, cytotoxicity is generated at a lower concentration, skin irritation is generated, and practical application is difficult. The traditional Chinese medicine extract has no additive effect on cytotoxicity, and the compound of the radix sophorae flavescentis extract, the folium artemisiae argyi extract and the honeysuckle extract can reduce cytotoxicity, has the effects of inhibiting propionibacterium acnes reproduction and inhibiting bacteria from releasing outer membrane vesicles, and can effectively inhibit acne inflammation.
It should be noted that the above-mentioned embodiments are only for illustrating the technical solutions of the present invention and not for limiting, and although the present invention has been described in detail with reference to the preferred embodiments, it should be understood by those skilled in the art that modifications or equivalent substitutions may be made on the technical solutions of the present invention without departing from the spirit and scope of the technical solutions of the present invention, which should be covered by the claims of the present invention.

Claims (10)

1. A composition with multi-target acne removing efficacy is characterized in that: comprises the steps of (a) preparing a mixture of a plurality of raw materials,
the traditional Chinese medicine composition comprises 25-50% of radix sophorae flavescentis extract, 25-50% of folium artemisiae argyi extract and 10-25% of honeysuckle extract in percentage by mass of the raw materials.
2. The composition with the multi-target acne removing effect according to claim 1, wherein the preparation method of the sophora flavescens extract comprises the steps of drying sophora flavescens to constant weight, crushing, sieving by a 60-mesh sieve, carrying out reflux extraction for 1-2 hours by using an 80-100% ethanol solution according to a material-liquid ratio of g: m L to obtain a powder, continuously stirring, filtering after the first extraction is finished, extracting residues by using 80-100% ethanol for one time, filtering, combining two filtrates, carrying out rotary evaporation and concentration on the filtrates to obtain an extract, and carrying out freeze drying to obtain the sophora flavescens extract.

The composition with the multi-target acne removing effect according to claim 1, wherein the preparation method of the folium artemisiae argyi extract comprises the steps of drying folium artemisiae argyi to constant weight, crushing, sieving by a 60-mesh sieve, performing reflux extraction for 1-2 hours by using 60-80% ethanol solution according to the material-liquid ratio of g: m L to be 1: 6-8, performing continuous stirring, filtering after the first extraction is finished, extracting residues by using 60-80% ethanol for one time, filtering, combining the two filtrates, performing rotary evaporation and concentration on the filtrates to obtain an extract, and performing freeze drying to obtain the folium artemisiae argyi extract.

The composition with the multi-target acne removing effect according to claim 1, wherein the honeysuckle extract is prepared by drying honeysuckle to constant weight, crushing, sieving with a 60-mesh sieve, performing reflux extraction for 2-3 hours by using a 60-80% ethanol solution according to a material-liquid ratio of 1: 5-8 in terms of g: m L, continuously stirring, filtering after the first extraction is finished, extracting residues once by using 60-80% ethanol, filtering, combining two filtrates, performing rotary evaporation and concentration on the filtrates to obtain an extract, and performing freeze drying to obtain the honeysuckle extract.
5. The composition with multi-target acne removing efficacy of claim 1, wherein: the composition with the multi-target acne removing effect comprises 25% of the sophora flavescens extract, 50% of the folium artemisiae argyi extract and 25% of the honeysuckle extract in percentage by mass of the raw materials.
6. A method for preparing the composition with multi-target acne removing efficacy according to any one of claims 1 to 5, wherein the composition comprises the following components in percentage by weight: comprises the steps of (a) preparing a mixture of a plurality of raw materials,
drying radix sophorae flavescentis to constant weight, crushing, sieving with a 60-mesh sieve, performing reflux extraction for 1: 8-10 in terms of g: m L according to a material-liquid ratio, performing reflux extraction for 1-2 h by using 80-100% ethanol solution, stirring continuously, filtering after the first extraction is finished, extracting residues once by using 80-100% ethanol, filtering, combining filtrates obtained in two times, performing rotary evaporation and concentration on the filtrate to obtain an extract, and performing freeze drying to obtain the radix sophorae flavescentis extract;
