1.4.2 mouse carbon clearance test
The method comprises injecting diluted India ink (10m L/kg) into mouse tail vein according to body weight, and immediately timing the injection of the ink, respectively, collecting blood 20 μ L from angular venous plexus at 2 and 10 min after the injection of the ink, and immediately adding the obtained blood to 2m L0.1.1% Na2CO3In solution, 0.1m L of each was pipetted into a 96-well microplate, and the Optical Density (OD) was measured at 600nm wavelength with a microplate reader and Na2CO3The solution was used as a negative control. 1.4.3 measurement of serum hemolysin
The method comprises the steps of hemagglutination, sheep blood taking, washing for 3 times by using normal saline, centrifuging for 10 minutes (2000 r/min) each time, preparing 2% (v/v) cell suspension by using packed SRBC by using the normal saline, injecting 0.2m L into the abdominal cavity of each mouse for immunization, removing eyeballs, taking blood from the eyeballs, placing the blood in a centrifugal tube for about 1 hour, stripping coagulated blood from the tube wall, fully separating out serum, centrifuging for 10 minutes at 2000 r/min, collecting the serum, carrying out agglutination reaction, diluting the serum by using the normal saline in a multiple ratio, respectively placing the serum with different dilution degrees in a trace hemagglutination experimental plate with 100 mu L per hole, adding 100 mu L0.5% (v/v) SRBC suspension into the sample, mixing the mixture, placing the sample in a wet flat disc, covering the flat disc, incubating the sample in a 37 ℃ incubator for 3 hours, observing the hemagglutination degree, and calculating the number of antibodies according to the level of the hemaggl.
1.4.4 detection of antibody-producing cells
Washing defibered sheep blood with physiological saline for 3 times, centrifuging (2000 r/min) for 10 minutes each time, injecting 2% (v/v) SRBC 0.2m L into the abdominal cavity of each mouse, dislocating and killing the cervical vertebra of the mouse immunized by the SRBC for 4-5 days, taking out the spleen, placing the spleen in a small plate containing hank's solution, slightly grinding the spleen to prepare cell suspension, filtering the cell suspension by a 200-mesh screen, centrifuging (1000/min) for 10 minutes, washing the cell suspension for 2 times by the hank's solution, finally suspending the cell in 5m L RPMI1640 culture solution, counting the cells, and adjusting the cell concentration to 5 × 106/m L.
The plaque is determined by heating and dissolving surface layer culture medium (1g agarose added with double distilled water to 100m L), placing in 45-50 deg.C water bath, keeping temperature, mixing with equal amount of hank's solution with pH of 7.2-7.4 and 2 times concentration, subpackaging into small tubes with 0.5m L per tube, adding 50 μ L10% SRBC (v/v, prepared with SA buffer solution), and 20 μ L spleen cell suspension (5 × 10)6Pieces/m L), quickly mixing, pouring onto a slide with a brushed agarose thin layer, making parallel pieces, after the agar is solidified, horizontally buckling the slide on a slide rack, putting the slide rack into a carbon dioxide incubator to incubate for 1.5h, adding a complement (1: 8) diluted by SA buffer solution into a groove of the slide rack, continuously incubating for 1.5h, and counting the number of hemolytic plaques.
2. Results
2.1 Effect of this product on mouse body weight
The weight of the mice in each experimental group in the table 1, the table 2 and the table 3 before and after the test is compared with the weight of the solvent control group and the weight of the negative control group, and the difference is not significant, which indicates that the sea buckthorn solid beverage has no obvious influence on the weight of the mice.

2.2 Effect of solid Hippophae rhamnoides drink on animal lymph organ/body weight ratio
Table 4 the thymus/body weight ratio and spleen/body weight ratio of each group of animals were compared with the solvent control group and the negative control group, and the differences were not significant (p > 0.05).

As can be seen from Table 5, the high-dose group of fructus Hippophae solid beverage significantly increased the phagocytic index of mouse carbon clearance (P < 0.05) compared with the solvent control group and the negative control group.

2.4 influence of solid beverage of seabuckthorn on phagocytosis of chicken red blood cells by macrophages in abdominal cavity
As can be seen from Table 6, the high dose group of the solid beverage of seabuckthorn significantly improved the phagocytosis rate and phagocytosis index (P < 0.05) compared with the solvent control group and the negative control group.

2.5 Effect of sea-buckthorn solid beverage on mouse serum hemolysin titer level
As can be seen from Table 7, the content of serum hemolysin (P < 0.05) in the high dose group of seabuckthorn solid beverage can be significantly increased compared with the solvent control group and the negative control group.

2.6 Effect of solid beverage of Hippophae rhamnoides on antibody-producing cell function
As can be seen from Table 8, the high dose group of solid beverage of Hippophae rhamnoides significantly enhanced the antibody-producing cell capacity (P < 0.01, P < 0.05) compared to the solvent control group and the negative control group.

3. Discussion of the related Art
(1) The sea-buckthorn solid beverage can obviously increase the content of the mouse serum hemolysin;
(2) the sea-buckthorn solid beverage can obviously enhance the function of macrophages in abdominal cavity of a mouse for phagocytosing chicken red blood cells;
(3) the sea-buckthorn solid beverage can obviously enhance the carbon clearance capability of mice;
(4) the sea-buckthorn solid beverage can obviously enhance the antibody cell-forming capacity;
(5) the sea-buckthorn solid beverage has no negative effect on improving the immunity of mice.
And (3) clinical trials:
1) general data: the inventor carries out clinical observation on 90 cases, wherein 47 cases are male, and 43 cases are female; age 18 years minimum, 60 years maximum; 37 cases of skin diseases, 30 cases of hepatitis, 23 cases of qi-invigorating in intestines and stomach, and other 4 cases (including qi-kick, physical weakness, aversion to cold, chronic pharyngitis, tonsillitis, common cold and the like).
