Abstract
The present invention relates to a cosmetic composition for skin anti-aging and moisturizing containing hydrangea macrophylla extract, which comprises hydrangea macrophylla extract containing high concentration of hydrangeal phenol and specific ingredients, thereby having a greatly improved effect on skin anti-aging and moisturizing.
Description
Technical Field
The present invention relates to a cosmetic composition for skin anti-aging and moisturizing containing hydrangea macrophylla extract, and more particularly, to a cosmetic composition having a significant effect on skin anti-aging and moisturizing by containing hydrangea macrophylla extract containing hydrangeal phenol and a specific component.
Background
With the abundance of life, various methods have been studied to delay not only aging inside the body but also aging outside the body.
The area of greatest concern with external aging is skin. Skin aging is classified into chronological aging, i.e., intrinsic aging, which is manifested over time, and photoaging, which occurs in combination with degenerative changes occurring at sites exposed to sunlight and chronological aging.
Due to this aging process, the skin becomes dry, wrinkles appear, and the skin elasticity is lost. In the studies on the related skin aging, the most important parts are free radicals (free radial) and active oxygen (reactive species). It is reported that these occur naturally in the human body, but they also occur by exposure to sunlight or contact with various chemical substances and the like. The generated radicals or active oxygen exert oxidative stress on skin cells, and substances generated in this process are considered as causative substances for the production of melanin and wrinkles.
It has been found that, in addition to these aging-causing substances, active oxygen is exposed to polymeric matrices (collagen, hyaluronic acid, elastin) or the like that maintain the elasticity in the skin, whereby these matrices are sequentially cut.
In this regard, the following conclusions are drawn: in order to prevent skin aging, it is necessary to effectively remove generated active oxygen while controlling the generation of excessive active oxygen, and it is necessary to establish a system associated with inhibition of enzymes causing skin wrinkles, inhibition of enzymatic activities causing pigmentation, and the like.
Therefore, various studies on cosmetic compositions for skin anti-aging, whitening and moisturizing are conducted through such a related system, but there is a real situation that more effective and safe cosmetic compositions are still required.

Experimental example 1 MMP-1 expression inhibition experiment of hydrangea macrophylla extract
The Hs68 cell line (Human Foreskin Fibroblast cell line; Human Foreskin Fibroblast cell line) was cultured under the following conditions. DMEM medium (Lonza #12-604F) was used as a culture solution after 10% FBS (Gibco) #16000- & 044) and antibiotics (1% P/S) were put therein. At 37 deg.C, 5% CO2The cells are cultured under conditions.
Hs68 cells were added to each well (well) of a 6-well plate (well plate) at 1.8X105Cells/2mL (cells/2mL), cultured for 24 hours. After the medium was changed to Serum-free (Serum-free) medium, the medium was cultured for another 6 hours, and then UVB was applied at 20mJ/cm2After stimulating and treating hydrangeal phenol (hydrangenol) (hydrangea macrophylla extract) and a positive control (retinoic acid, 1 μ M) (n ═ 3) at the concentrations, respectively, for 48 hours, the culture solution was collected and used for ELISA (R ═ 3)&D # DY901) method to determine MMP-1 amount. To correct for differences in Cell numbers between groups, MMP-1ELISA results were normalized (normalization) to CCK-8 assay (Cell Counting Kit-8assay, described below) results.
Cell proliferation & toxicity assay (CCK-8)
At the end of the MMP-1 assay, Hs68 cells were replaced with cells containing 5% CCK-8After culturing the solution (Dojindo) # CK04), the culture was incubated at 37 ℃ with 5% CO2The incubator was further incubated for 2 hours. Finally, the presence or absence of cell proliferation or toxicity was calculated by measuring the absorbance at a wavelength of 450nm and comparing the absorbance with that of a control group.
