In one preferred aspect, the antibiotic of the present invention is one selected from the group consisting of tetracycline, oxytetracycline, doxycycline, minocycline, chloramphenicol, thiamphenicol, erythromycin ethylsuccinate, roxithromycin, clarithromycin, azithromycin, telithromycin, dirithromycin, kitasamycin, midecamycin, acetyl midecamycin, josamycin, milbemycin, romycin, acetyl spiramycin, lincomycin, clindamycin and sodium fusidate.
On one hand, the mass ratio of the antibiotic to the vegetable oil is preferably 0.01: 1-100: 1.
In one preferred aspect, the acne described herein is acne caused by propionibacterium acnes.
On the other hand, the composition can be prepared into external medicines.
More preferably, the external medicine of the present invention is a liquid preparation for external use; further preferably, the liquid preparation for external use is one selected from an emulsion for external use and a suspension for external use.
More preferably, the external medicine of the present invention is an external semisolid preparation; further preferably, the semisolid preparation for external use of the present invention is one selected from ointments and gels.
In a further aspect of the invention there is provided the use of a composition as hereinbefore described in the manufacture of a medicament for the treatment of acne.
In vitro test results show that the composition can generate a synergistic antibacterial effect (the combined medication index is less than 1) on the clindamycin-resistant propionibacterium acnes, and animal tests further prove that the composition can generate a synergistic therapeutic effect on a rat acne model caused by propionibacterium acnes.
Detailed Description
The present invention will be further described below by way of specific embodiments, but the present invention is not limited to only the following examples.
Test example 1 synergistic antibacterial action of the composition against clindamycin-resistant Propionibacterium acnes (CCARM 9010)
The inhibition of the test substances ① - ③ on the clindamycin-resistant propionibacterium acnes (CCARM 9010) is measured by a method disclosed by Lemna and the like (food and fermentation industry, 2017,43(02): 232-238.).
(1) The tested drugs are:
① antibiotic, ② vegetable oil, ③ antibiotic and vegetable oil in a mass ratio R (indicated as "-" connecting antibiotic and vegetable oil)
(2) Test method
① preparing test sample solution by taking a certain amount of test substance, using DMF as solvent, and adopting a specific multiple continuous dilution method to prepare test sample solution with 6 concentrations.
② bacterial suspension is prepared by inoculating activated strain to liquid culture medium (no agar), shake-culturing, calculating bacterial colony number by plate dilution method, and adjusting bacterial suspension concentration to 10 with sterile physiological saline7CFU/mL。
③ agar-hole diffusion method for determining antibacterial activity comprises cooling the sterilized culture medium to 50 deg.C, adding 6mL of bacterial suspension, mixing, pouring into a culture dish with diameter of 9cm, standing for 45min, uniformly perforating (diameter of 7mm) on the cured culture medium with an aseptic perforator, marking, adding 40 μ L of sample solution into each hole, DMF as blank control, culturing at 37 deg.C for 18h, measuring and recording the diameter (mm) of the zone of inhibition, repeating for 3 times, averaging to obtain the result, and calculating the Inhibition Ratio (IR) of the strain to be tested according to the following formula.
For antibiotics and vegetable oils, IR was plotted against the log (c)) of the test concentrations (μ g/mL) of antibiotics and vegetable oils, and the concentration of each test substance at which the inhibition of specific fa occurred was calculated from the linear regression equation and is designated as ICfa(A)And ICfa(B). For mixtures, the concentration of antibiotic in the mixture at which inhibition of a particular fa occurs is calculated from a linear regression equation using IR plotted against the logarithm of the antibiotic concentration (. mu.g/mL) and reported as ICfa(mixA)。
The Combination Index (CI) at which a specific fa inhibitory rate is produced is calculated according to the following formula.
CI=ICfa(mixA)/ICfa(A)+ICfa(mixA)/(R×ICfa(B))
When CI <1, it means that there is synergism, the smaller the CI, the stronger the synergism. The results are shown in Table 1

EXAMPLE 1 preparation of a suspension for external use containing a combination of antibiotics and vegetable oils
[ prescription ] medecamycin 5.00g, hop berry oil 2.40g, camphor spirit 250mL, sodium carboxymethyl cellulose 5g, glycerin 100g, adding purified water to 1000mL.
[ PREPARATION ] Medidycin is put into a mortar, and glycerin is added to the mortar to be ground into fine paste. And preparing sodium carboxymethylcellulose into mucilage with 200mL of water, slowly adding into mortar under stirring, transferring into a measuring vessel, adding Camphora spirit and Ceratonia strigaea fruit oil under stirring, adding purified water to full volume, and stirring.
EXAMPLE 2 preparation of emulsion for external use containing a combination of antibiotics and vegetable oils
[ prescription ] sodium fusidate 0.16g, rice germ oil 9.84g, acacia gum fine powder 125g, tragacanth gum fine powder 7g, saccharin sodium 0.1g, volatile almond oil 1mL, ethylparaben 0.5g, purified water added to 1000mL.
[ preparation ] sodium fusidate, rice germ oil and Arabic gum are ground uniformly, 250mL of purified water is added at a time, the mixture is ground in one direction forcibly to prepare primary emulsion, sodium saccharin aqueous solution, volatile almond oil and ethylparaben alcohol solution are added, then tragacanth mucilage is slowly added, the purified water is added to the full amount, and the mixture is stirred uniformly, thus obtaining the composition.
EXAMPLE 3 preparation of an ointment containing a combination of antibiotics and vegetable oils
[ prescription ] 6.13g of meleumycin, 3.87g of black grass seed oil, 12.5g of stearic acid, 17.0g of glyceryl monostearate, 5.0g of beeswax, 67.0g of white vaseline, 10.0g of aluminum distearate, 75.0g of ozokerite, 410.0mL of liquid paraffin, 1.0g of ethylparaben, 1.0g of calcium hydroxide and distilled water till 1000g.
[ preparation method ] Mebaimycin, black grass seed oil, stearic acid, glyceryl monostearate, beeswax and ceresin are heated and melted in water bath, then liquid paraffin, white vaseline and aluminum distearate are added, and the mixture is heated to 85 ℃ (oil phase); dissolving calcium hydroxide and ethylparaben in distilled water, and heating to the same temperature (water phase); slowly adding the water phase into the oil phase under stirring, and condensing to obtain the final product.
Example 4 preparation of a gel containing a combination of antibiotics and vegetable oils
[ prescription ] erythromycin ethylsuccinate 0.38g, coriander seed oil 9.62g, carbopol 94010 g, glycerin 50g, polysorbate 802g, ethylparaben 0.5g, ethanol 50g, sodium hydroxide 4g, and distilled water to 1000g.
[ preparation method ] mixing erythromycin ethylsuccinate, coriander seed oil, carbopol, glycerol and polysorbate 80 with 300mL of distilled water, dissolving sodium hydroxide in 100mL of distilled water, adding into the above liquid, stirring, dissolving ethylparaben in ethanol, gradually adding, and stirring to obtain transparent gel.
Comparative example 1 preparation of antibiotic-containing gel
[ prescription ] erythromycin ethylsuccinate 0.38g, carbopol 94010 g, glycerin 50g, polysorbate 802g, ethylparaben 0.5g, ethanol 50g, sodium hydroxide 4g, and distilled water added to 1000g.
[ preparation method ] mixing erythromycin ethylsuccinate, carbopol, glycerin and polysorbate 80 with 300mL of distilled water, dissolving sodium hydroxide in 100mL of distilled water, adding into the above liquid, stirring, dissolving ethylparaben in ethanol, and gradually adding and stirring to obtain transparent gel.
Comparative example 2 preparation of gel of vegetable oil-containing composition
[ prescription ] 9.62g of coriander seed oil, 94010 g of carbopol, 50g of glycerol, 802g of polysorbate, 0.5g of ethylparaben, 50g of ethanol, 4g of sodium hydroxide and distilled water till 1000g.
[ preparation method ] mixing coriander seed oil, carbopol, glycerin, polysorbate 80 and 300mL of distilled water, dissolving sodium hydroxide in 100mL of distilled water, adding into the above liquid, stirring, dissolving ethylparaben in ethanol, gradually adding, and stirring to obtain transparent gel.
Test example 2 therapeutic effect of a gel comprising a combination of antibiotics and vegetable oils on a model of acne in rats caused by Propionibacterium acnes
The treatment of the gels prepared in example 4, comparative example 1 and comparative example 2 on a Propionibacterium acnes-induced rat model of acne was examined using the method disclosed in Liyuan et al (Hainan medicine, 2017,28(21): 3446-3448), wherein the swelling rates of auricles after administration are shown in Table 2.

As can be seen from table 2, the rate of decrease of the propionibacterium acnes-induced swelling of the rat acne model after application of the gel of example 4 with respect to the model control group was higher than the sum of the rates of decrease of comparative example 1 and comparative example 2.

The composition of claim 1, wherein said antibiotic is selected from the group consisting of tetracycline, oxytetracycline, doxycycline, minocycline, chloramphenicol, thiamphenicol, erythromycin ethylsuccinate erythromycin, roxithromycin, clarithromycin, azithromycin, telithromycin, dirithromycin, kitasamycin, medemycin, acetylmedemycin, josamycin, meleumycin, romycin, acetylspiramycin, lincomycin, clindamycin, and sodium fusidate.
3. Composition according to claim 1 or 2, characterized in that, in one aspect, the ratio of the antibiotic according to the invention to the vegetable oil is preferably between 0.01:1 and 100:1 by mass.
4. A composition according to claim 1 or 2, characterized in that said acne is acne caused by propionibacterium acnes.
5. The composition of claim 1 or 2, wherein said composition is formulated for topical administration.
6. The composition of claim 5, wherein said topical medicament is a topical liquid formulation.
7. The composition of claim 6, wherein the liquid preparation for external use is one selected from the group consisting of an emulsion for external use and a suspension for external use.
8. The composition of claim 5, wherein said topical medicament is a topical semisolid formulation.
9. The composition of claim 8, wherein said semi-solid formulation for external use is one selected from the group consisting of an ointment and a gel.
10. Use of a composition according to claim 1 or 2 in the manufacture of a medicament for the treatment of acne.
