The extraction process of the centella extract comprises the following steps: reflux-extracting herba Centellae with 75% ethanol for 2 times (10 times of the amount of the extract for 1 hr each time), filtering, and mixing filtrates; recovering ethanol from the filtrate under reduced pressure, and concentrating the filtrate until no alcohol smell is obtained, which is equivalent to 1g/mL of crude drug. Purifying with HPD-100 macroporous resin column with maximum sample loading of 12 times volume of sample solution, 50% ethanol as eluent, and 6 times volume of resin column as eluent.
In comparative example 4, trollius chinensis bunge, pawpaw and centella asiatica are commercially available traditional Chinese medicinal materials.
Example 1
The preparation of the composition of this example was carried out according to the following steps:
firstly, preparing earthworm freeze-dried powder:
cutting fresh lumbricus into abdomen, removing viscera silt, cleaning with deionized water, cutting, weighing, storing in a refrigerator at-80 deg.C, freezing, crushing the frozen lumbricus, pulverizing with a low temperature pulverizer or under protection of liquid nitrogen into fine powder with particle size of 70-80 meshes, and freeze drying with a low temperature refrigerator to obtain Lumbricus lyophilized powder;
secondly, preparing an oil phase:
heating 16 parts by weight of stearic acid, 8 parts by weight of glyceryl monostearate, 8 parts by weight of No. 15 white oil, 4 parts by weight of beeswax, 12 parts by weight of liquid paraffin, 6 parts by weight of vaseline and 5 parts by weight of vitamin E to 80 ℃, and uniformly mixing to obtain an oil phase;
thirdly, preparing a water phase:
heating 15 parts by weight of earthworm freeze-dried powder, 8 parts by weight of trollius chinensis bunge extract, 8 parts by weight of pawpaw extract, 8 parts by weight of centella asiatica extract, 8 parts by weight of humectant, 8 parts by weight of emulsifier and 1 part by weight of preservative to 80 ℃, and uniformly mixing to obtain a water phase; the humectant is trehalose; the emulsifier is monostearate; the antiseptic is phenoxyethanol.
Fourthly, adding the water phase into the oil phase at the temperature of 80 ℃, continuously keeping the stirring state after the two phases are mixed, and cooling to the room temperature to obtain the earthworm protein composition emulsion.
Example 2
The preparation of the composition of this example was carried out according to the following steps:
firstly, preparing earthworm dry powder:
cutting fresh lumbricus into abdomen, removing viscera silt, cleaning, cutting into segments, sun-drying (refer to the first part of 2005 edition in Chinese pharmacopoeia), and pulverizing into fine powder with particle size of 90-100 meshes to obtain dry lumbricus powder;
secondly, preparing an oil phase:
heating 16 parts by weight of stearic acid, 8 parts by weight of glyceryl monostearate, 8 parts by weight of No. 15 white oil, 4 parts by weight of beeswax, 12 parts by weight of liquid paraffin, 6 parts by weight of vaseline and 3 parts by weight of vitamin E to 80 ℃, and uniformly mixing to obtain an oil phase;
thirdly, preparing a water phase:
heating 10 parts by weight of earthworm dry powder, 5 parts by weight of trollius chinensis bunge extract, 5 parts by weight of pawpaw extract, 5 parts by weight of centella asiatica extract, 5 parts by weight of humectant, 5 parts by weight of emulsifier and 0.5 part by weight of preservative to 80 ℃ and mixing uniformly to obtain a water phase; wherein the humectant is glycerol, the emulsifier is monostearate, and the antiseptic is phenoxyethanol.
Fourthly, adding the water phase into the oil phase at the temperature of 80 ℃, continuously keeping the stirring state after the two phases are mixed, and cooling to the room temperature to obtain the earthworm protein composition emulsion.
Example 3
The preparation of the composition of this example was carried out according to the following steps:
firstly, preparing earthworm freeze-dried powder:
cutting fresh lumbricus into abdomen, removing viscera silt, cleaning with deionized water, cutting, weighing, storing in a refrigerator at-80 deg.C, freezing, crushing the frozen lumbricus, pulverizing with a low temperature pulverizer or under protection of liquid nitrogen into fine powder with particle size of 70-80 meshes, and freeze drying with a low temperature refrigerator to obtain Lumbricus lyophilized powder;

Fourthly, adding the water phase into the oil phase at the temperature of 80 ℃, continuously keeping the stirring state after the two phases are mixed, and cooling to the room temperature to obtain the earthworm protein composition. The humectant is glycerin.
Comparative example 4
The preparation of the comparative example composition was carried out according to the following procedure:
(1) 1 part of carbomer is taken, and 200 parts of distilled water is added for full swelling.
(2) Weighing 12 parts by weight of earthworm freeze-dried powder, 6 parts by weight of trollius chinensis bunge, 6 parts by weight of pawpaw and 6 parts by weight of centella asiatica, adding 20 times of distilled water, and decocting. Then adding 4 parts by weight of vitamin E, 6 parts by weight of humectant, 6 parts by weight of emulsifier and 0.8 part by weight of preservative, uniformly stirring, concentrating into extract, and freeze-drying the extract to form freeze-dried powder. The humectant is glycerin. The emulsifier is monostearate. The preservative is phenoxyethanol.
(3) Adding 8 parts by weight of freeze-dried powder into carbomer swelling solution, adding triethanolamine into the mixed solution, and fixing the volume to obtain gel, thus obtaining the hormone face repairing cream.
100 patients with hormonal facial dermatitis were selected in dermatology department of the academy of traditional Chinese medicine of Heilongjiang province according to the standard of Table 1, wherein 60 patients were grade 3 patients, 20 patients were grade 2 patients, and 20 patients were grade 1 patients.
The earthworm protein composition emulsions prepared in examples 1, 2 and 3 and the hormone face repair cream prepared in comparative example 4 were uniformly applied to the affected parts of the face, respectively, after dividing 60 cases of 3-stage patients into four groups at random, and the grades and identification criteria of the hormone face diseases are shown in table 1, and the days and effects after application of the earthworm protein composition emulsions prepared in examples 1, 2 and 3 and the hormone face repair cream prepared in comparative example 4 are shown in table 2.
The earthworm protein composition emulsion prepared in example 1 was applied uniformly to the affected area of the face for 20 patients of grade 2 and 20 patients of grade 1, and the effect was evaluated after use.

The earthworm protein composition prepared in example 1, which is a hormone for face dermatitis, had the following effects:
for grade 1, mild hormonal facial skin (20 patients total), used for 1-5 days, the papules became smaller and the skin surface turned red. The number of papules is reduced. There was a slight improvement in inflammation. On day 5-15, most hormone faces will be withdrawn with different degrees, manifested as red, swollen, itchy, and papular. On day 15-90, the skin in withdrawal period shows red, swelling, pimple and exudation symptoms gradually disappear, the appearance of the skin mainly shows dryness and skin fading, the skin gradually approaches to normal, the skin state is unstable, and withdrawal symptoms are easy to appear repeatedly. The appearance and the function of the skin are almost recovered to be normal after days 90-180, and the skin state is greatly improved.
For grade 2, moderate hormone face skin (20 patients in total), erythema, small pimple and red swelling part tend to be reduced after 1-10 days. On day 10-20, most hormone faces will be withdrawn with different degrees, manifested as red, swollen, itchy, and papular. On the 20 th to 90 th days, the skin in the withdrawal period shows red, swollen, pimple and exudation symptoms which are gradually faded, the appearance of the skin mainly shows dry and skin-fading and gradually approaches to normal, the skin state is unstable, and the withdrawal symptoms are easy to appear repeatedly. The appearance and the function of the skin are almost recovered to be normal after days 90-180, and the skin state is greatly improved.
When the earthworm protein composition prepared in example 1 is used for patients with grade 3 hormonal facial dermatitis, the pimple is seriously swollen, the large blister is small, the red swelling part basically disappears, the skin surface is faded, the inflammation gradually disappears, the skin hyperplasia and the bulge do not appear after 1-15 days, and most hormonal faces can be in a withdrawal phenomenon with different weights, blisters and exudation after 15-30 days. On 30-120 days, the skin in the withdrawal period shows red, swelling, pimple and exudation symptoms which gradually disappear, the appearance of the skin mainly shows dryness and skin fading, the skin is gradually close to normal, the skin state is unstable, and the withdrawal symptoms are easy to appear repeatedly. The appearance and the function of the skin are almost normal after 180 days at 120, and the skin state is greatly improved.
As shown in table 2, the patients with grade 3 hormonal facial dermatitis using the earthworm protein composition prepared in example 1 can recover to grade 0 after 90-180 days of use, the skin condition is greatly improved, and the face of the patients is smooth and glossy and is close to the color of normal skin, because the earthworm freeze-dried powder in example 1 mainly contains enzymes, proteins, polypeptides, lipids and trace elements, and has the effects of anticoagulation, antithrombotic, blood pressure reduction, antitumor, antipyretic, anti-inflammatory, analgesic and wound healing acceleration. The trehalose has the functions of diminishing inflammation and inhibiting the secretion and diffusion of inflammatory factors, so that the earthworm protein composition can prevent the skin of a patient suffering from the hormonal acne from generating pigmentation and scars in the process of combining the earthworms and the trehalose.
With the earthworm protein compositions of examples 2, 3, the best condition recovered in patients with hormonal face dermatitis grade 3 can reach grade 1, and after 180 days, the patients' faces have hyperpigmentation or scarring.
With the earthworm protein composition of comparative example 4, the patients with hormonal facial dermatitis grade 3 improved, but the best recovery was achieved only in grade 2, and after 180 days, the patients had hyperpigmentation or scarring in their faces.

Claims (10)

1. A composition comprising earthworm protein, wherein the earthworm protein comprises: the active ingredients in the formula consist of the following components in parts by weight: 10-15 parts of earthworm freeze-dried powder or earthworm dry powder, 5-8 parts of trollius chinensis bunge extract, 5-8 parts of pawpaw extract, 5-8 parts of centella asiatica extract, 3-5 parts of vitamin E and 5-8 parts of humectant.
2. The earthworm protein-containing composition of claim 1, wherein: the humectant is glycerol or trehalose.
3. The earthworm protein-containing composition of claim 1, wherein: the formulation of the composition containing the earthworm protein is emulsion, ointment, cream, gel or aqua, and the formulation consists of the effective components and auxiliary material base of the corresponding formulation.

The earthworm protein-containing composition of claim 1, wherein: the formulation of the composition containing the earthworm protein is emulsion, and the formulation comprises the following components in parts by weight: 10-15 parts of earthworm freeze-dried powder, 5-8 parts of trollius chinensis bunge extract, 5-8 parts of pawpaw extract, 5-8 parts of centella asiatica extract, 3-5 parts of vitamin E, 5-8 parts of humectant, 5-8 parts of emulsifier, 0.5-1 part of preservative, 40-68 parts of oil phase matrix and 80-100 parts of purified water.
5. The earthworm protein-containing composition of claim 4, wherein: the emulsifier is one or two of sorbitan and monostearate.
6. The earthworm protein-containing composition of claim 4, wherein: the preservative is one or a combination of two of nipagin and phenoxyethanol.

The earthworm protein-containing composition of claim 4, wherein: the oil phase matrix comprises the following components in parts by weight: 12-20 parts of stearic acid, 6-10 parts of glyceryl monostearate, 6-10 parts of No. 15 white oil, 2-5 parts of beeswax, 10-15 parts of liquid paraffin and 4-8 parts of vaseline.
8. The process for the preparation of a composition containing earthworm proteins according to any one of claims 4-7, characterised in that it is carried out according to the following steps:
(1) preparing an oil phase:
taking the oil phase matrix and vitamin E according to the formula, heating to 75-85 ℃, and uniformly mixing to obtain an oil phase;
(2) preparing an aqueous phase:
adding Lumbricus lyophilized powder or Lumbricus dry powder, flos Trollii extract, fructus Chaenomelis extract, herba Centellae extract, humectant, emulsifier and antiseptic into purified water, heating to 75-85 deg.C, and mixing to obtain water phase;
(3) adding the water phase into the oil phase at 75-85 deg.C, mixing, stirring, and cooling to room temperature to obtain composition emulsion containing Lumbricus protein.
9. The method of preparing a composition comprising earthworm protein according to claim 8, wherein: the preparation method of the earthworm freeze-dried powder comprises the following steps: cutting fresh lumbricus into abdomen, removing viscera silt, cleaning with deionized water, cutting into pieces, storing in a refrigerator below-70 deg.C, freezing, crushing the frozen lumbricus, pulverizing with a low temperature pulverizer or under the protection of liquid nitrogen into fine powder with particle size of 70-120 meshes, and freeze drying with a low temperature refrigerator to obtain Lumbricus lyophilized powder.
10. Use of a composition comprising earthworm proteins according to any one of claims 1-7 for the preparation of a medicament or a skin care product for the treatment of hormonal acne.
