Abstract
本发明公开了BBR蛋白及其编码基因和应用。本发明提供了一种蛋白质,获自水稻(Oryza sativa),命名为BBR8蛋白,为序列表中序列1所示的蛋白质。编码BBR8蛋白的核酸分子也属于本发明的保护范围。本发明还保护BBR8蛋白的应用,为如下(c1)或(c2):(c1)调控植物对白叶枯病的抗性;(c2)增加植物对白叶枯病的抗性。本发明还保护编码BBR8蛋白的核酸分子的应用,为如下(d1)或(d2):(d1)培育对白叶枯病的抗性改变的转基因植物;(d2)培育对白叶枯病的抗性增高的转基因植物。本发明对于培育抗白叶枯病植物具有重大的应用价值。
Description
技术领域
本发明属于生物技术领域,具体涉及BBR8蛋白及其编码基因和应用。
背景技术
植物在生长发育进程中,容易遭受细菌、真菌、病毒和昆虫等侵袭。植物一旦受到侵害后,其细胞或组织就会发生病理性变化,阻碍植株的正常生长和发育进程,进而影响经济效益。近年来,我国农作物病虫害总体处于重发、频发态势,各类主要病虫害年平均发生面积比1996-2005年的10年平均值增加20.82%。从病虫危害造成损失的统计结果来看,水稻、小麦、玉米为首的三大主要粮食作物占据绝大部分,实际造成的损失比例分别占到总损失量的33.67%、23.31%和35.13%。面对病原菌的侵袭,植物进化出了一套复杂的防御系统,受到抗病和抗病相关基因的调控。
由黄单胞菌Xanthomonas oryzae pv.oryzae(Xoo)引起的水稻白叶枯病(bacterial blight)是水稻生产上的主要病害之一,在世界各地的栽培区都有不同程度的发生。水稻一旦感病,叶片会枯萎,损害光合作用,从而影响籽粒的充实,造成不实粒增加、千粒重减轻等,一般减产20-30%,严重的可达50%,甚至绝收(Huang et al.,1997)。防治该病的关键在于发掘新的抗源,种植抗病品种多样化以及克隆更多的抗病基因。
发明内容
本发明的目的是提供BBR蛋白及其编码基因和应用。
本发明提供了一种蛋白质,获自水稻(Oryza sativa),命名为BBR8蛋白,为如下(a1)或(a2)或(a3)或(a4):
(a1)序列表中序列1所示的蛋白质;
(a2)在(a1)所述蛋白质的N端或/和C端连接标签得到的融合蛋白;
(a3)将(a1)经过一个或几个氨基酸残基的取代和/或缺失和/或添加得到的与植物抗白叶枯病相关的蛋白质;
(a4)来源于水稻且与(a1)具有98%以上同一性且与植物抗白叶枯病相关的蛋白质。

蛋白质可人工合成,也可先合成其编码基因,再进行生物表达得到。
编码BBR8蛋白的核酸分子也属于本发明的保护范围。
所述核酸分子具体为如下(b1)或(b2)或(b3)或(b4)或(b5):
(b1)编码区如序列表中序列2第396-1856位核苷酸所示的DNA分子;
(b2)序列表中序列2所示的DNA分子;
(b3)序列表中序列3所示的DNA分子;
(b4)来源于水稻且与(b1)或(b2)或(b3)具有95%以上同一性且编码所述蛋白质的DNA分子;
(b5)在严格条件下与(b1)或(b2)或(b3)限定的核苷酸序列杂交且编码所述蛋白质的DNA分子。
所述严格条件是在2×SSC,0.1%SDS的溶液中,在68℃下杂交并洗膜2次,每次5min,又于0.5×SSC,0.1%SDS的溶液中,在68℃下杂交并洗膜2次,每次15min。
含有所述核酸分子的表达盒、重组载体或重组微生物均属于本发明的保护范围。
可用现有的表达载体构建含有所述核酸分子的重组表达载体。使用所述核酸分子构建重组表达载体时,可在其转录起始核苷酸前加上任何一种增强型、组成型、组织特异型或诱导型启动子,它们可单独使用或与其它的植物启动子结合使用;此外,使用所述核酸分子构建重组表达载体时,还可使用增强子,包括翻译增强子或转录增强子,这些增强子区域可以是ATG起始密码子或邻接区域起始密码子等,但必需与编码序列的阅读框相同,以保证整个序列的正确翻译。所述翻译控制信号和起始密码子的来源是广泛的,可以是天然的,也可以是合成的。翻译起始区域可以来自转录起始区域或结构基因。为了便于对转基因植物或转基因微生物进行鉴定及筛选,可对所用表达载体进行加工,如加入在植物或微生物中表达可产生颜色变化的酶或发光化合物的基因、具有抗性的抗生素标记物或是抗化学试剂标记基因等。从转基因安全性考虑,可不加任何选择性标记基因,直接以表型筛选转化植物或微生物。
所述重组载体具体可为重组表达载体。所述重组表达载体具体可为如下重组质粒:以pCAMBIA2300载体为出发载体,用序列表的序列2自5’末端第396至2766位核苷酸所示的DNA分子取代了出发载体的SalI和BamH I酶切位点之间的小片段,用序列表的序列3自5’末端第1至957位核苷酸所示的DNA分子(启动子)取代了出发载体的Hind III和Pst I酶切位点之间的小片段。
本发明还保护BBR8蛋白的应用,为如下(c1)或(c2):
(c1)调控植物对白叶枯病的抗性;
(c2)增加植物对白叶枯病的抗性。
本发明还保护编码BBR8蛋白的核酸分子的应用,为如下(d1)或(d2):
(d1)培育对白叶枯病的抗性改变的转基因植物;
(d2)培育对白叶枯病的抗性增高的转基因植物。
本发明还保护一种制备转基因植物的方法,包括如下步骤:在出发植物中导入编码BBR8蛋白的核酸分子,得到对白叶枯病的抗性增高的转基因植物。所述核酸分子具体可通过以上任一所述重组表达载体导入所述出发植物。携带有所述核酸分子的重组表达载体可通过Ti质粒、Ri质粒、植物病毒载体、直接DNA转化、显微注射、电导、农杆菌介导等常规生物学方法转化到出发植物中。
本发明还保护一种植物育种方法,包括如下步骤:增加目的植物中BBR8蛋白的含量和/或活性,从而增高目的植物对白叶枯病的抗性。
本发明还保护一种制备转基因植物的方法,包括如下步骤:抑制出发植物中编码BBR8蛋白的核酸分子的表达,得到对白叶枯病的抗性降低的转基因植物。
本发明还保护一种植物育种方法,包括如下步骤:降低目的植物中BBR8蛋白的含量和/或活性,从而降低目的植物对白叶枯病的抗性。
以上任一所述植物为双子叶植物或单子叶植物。所述单子叶植物可为禾本科植物。所述禾本科植物可为水稻属植物。所述水稻具体可为水稻秋光或水稻Kasalath。
以上任一所述白叶枯病为黄单胞菌引起的白叶枯病。以上任一所述白叶枯病为白叶枯病致病菌引起的白叶枯病。以上任一所述白叶枯病为菲律宾生理小种PXO86引起的白叶枯病。
本发明对于培育抗白叶枯病植物具有重大的应用价值。

具体实施方式
以下的实施例便于更好地理解本发明,但并不限定本发明。下述实施例中的实验方法,如无特殊说明,均为常规方法。下述实施例中所用的试验材料,如无特殊说明,均为自常规生化试剂商店购买得到的。以下实施例中的定量试验,均设置三次重复实验,结果取平均值。
水稻品种“秋光”,简称水稻秋光,为白叶枯病抗病品种。水稻品种Kasalath,简称水稻Kasalath,为白叶枯病感病品种。提及pZh01载体的文献:Xiao et al.PlantMolecular Biology 52:957–966,2003.。水稻白叶枯病致病菌菲律宾生理小种PXO86,简称菲律宾生理小种PXO86。
实施例1、BBR基因的发现
水稻秋光与水稻Kasalath杂交,获得含BBR8基因的F2单株,与水稻Kasalath杂交再自交后分别产生1000个单株的F2:2群体,利用这些群体进行BBR8基因的精细遗传和物理定位,最终将BBR8基因限定在第8染色体的BAC克隆AP003914上的47.4kb物理区间内。通过分析该区间6个编码基因的序列,发现其中一个蛋白的编码基因在水稻秋光和水稻Kasalath之间有20个核苷酸的差异,特别是第15处的核苷酸差异造成了蛋白翻译提前终止,是BBR8最可能的候选基因。序列比较差异示意图见图1。
BBR8蛋白如序列表的序列1所示。BBR8基因的cDNA如序列表的序列2所示,其中第396-1856位为编码框(1461bp)。BBR8基因的基因组DNA如序列表的序列3所示。
实施例2、功能验证
一、过表达水稻的制备
(一)重组质粒PQ-BBR8的构建
1、提取水稻秋光的总RNA并反转录为cDNA。
2、以步骤1得到的cDNA为模板,用F1和R1组成的引物对进行PCR扩增,回收PCR扩增产物。
F1:5′-aaGTCGACatgaaggacctcaaggctgtcgcc-3′;
R1:5′-aaGGATCCccaatcttaatgaaattggcc-3′。
3、用限制性内切酶SalI I和BamH I双酶切步骤2得到的PCR扩增产物,回收酶切产物。
4、用限制性内切酶Sal I和BamH I双酶切pCAMBIA2300载体,回收约8.7kb的载体骨架。
5、将步骤3得到酶切产物和步骤4得到的载体骨架连接,得到重组质粒。
6、提取水稻秋光的基因组DNA。
7、以步骤6提取的基因组DNA为模板,用F2和R2组成的引物对进行PCR扩增,回收PCR扩增产物。
F2:5′-AAGCTTctagcttttgagaacctctc-3′;
R2:5′-CTGCAGccagcgcttgacggcggggttggtc-3′。
8、用限制性内切酶HindIII和Pst I双酶切步骤7的PCR扩增产物,回收酶切产物。
9、用限制性内切酶HindIII和Pst I双酶切步骤5得到的重组质粒,回收约11.1kb的载体骨架。
10、将步骤8的酶切产物和步骤9的载体骨架连接,得到重组质粒P957-BBR8。重组质粒P957-BBR8的结构示意图见图2。根据测序结果,对重组质粒P957-BBR8进行结构描述如下:以pCAMBIA2300载体为出发载体,用序列表的序列2自5’末端第396至2766位核苷酸所示的DNA分子取代了出发载体的SalI和BamH I酶切位点之间的小片段,用序列表的序列3自5’末端第1至957位核苷酸所示的DNA分子(启动子)取代了出发载体的Hind III和Pst I酶切位点之间的小片段。重组质粒P957-BBR8中,Hind III和BamH I酶切位点之间的DNA分子如序列表的序列4所示。
(二)进行功能互补试验的Kasalath转基因水稻制备
1、将重组质粒P957-BBR8导入农杆菌EHA105,得到重组农杆菌。
2、将步骤1得到的重组农杆菌与水稻Kasalath的成熟胚愈伤组织共培养,采用40-60mg/L卡那霉素筛选抗性愈伤组织,然后经过预分化、分化、生根后得到T0代植株。
3、T0代植株自交获得T1代种子,T1代种子长成的植株即为T1代植株。
4、提取T1代植株的基因组DNA,用NPT_F和NPT_R组成的引物对进行PCR鉴定,目的条带约411bp,PCR鉴定中扩增得到目的条带的植株即为转基因植株。转基因植株即为BBR8过表达植株。
NPT_F:5′-GCAGGCATCGCCATGGG-3′;
NPT_R:5′-GCCCTGAATGAACTGCA-3′。
(三)转pCAMBIA2300载体水稻的制备
用pCAMBIA2300载体代替重组质粒P957-BBR8,按照步骤(二)进行操作,得到转pCAMBIA2300载体植株。
二、抑制表达水稻的制备
(一)重组质粒pZh01-BBR8i的构建
1、提取水稻秋光的总RNA并反转录为cDNA。
2、以步骤1得到的cDNA为模板,用F3和R3组成的引物对进行PCR扩增,回收PCR扩增产物。
F3:5′-aaGTCGACGGCGACCGCCGCAAGCTGGAGCGCC-3′;
R3:5′-aaGGATCCATTGTAATTAAAACTTTATTTC-3′。
3、用限制性内切酶SalI I和BamH I双酶切步骤2得到的PCR扩增产物,回收酶切产物。
4、用限制性内切酶SalI I和BamH I双酶切pZh01载体,回收约10kb的载体骨架。
5、将步骤3的酶切产物和步骤4的载体骨架连接,得到重组质粒pZh01-BBR8i。重组质粒pZh01-BBR8i的结构示意图见图3。根据测序结果,对重组质粒pZh01-BBR8i进行结构描述如下:以pZh01载体为出发载体,用序列表的序列5所示的DNA分子取代了出发载体的SalI和BamH I酶切位点之间的小片段。
(二)抑制表达水稻的制备
1、将重组质粒pZh01-BBR8i导入农杆菌EHA105,得到重组农杆菌。
2、将步骤1得到的重组农杆菌与水稻秋光的成熟胚愈伤组织共培养,采用50mg/L潮霉素筛选抗性愈伤组织,然后经过预分化、分化、生根后得到T0代植株。
3、T0代植株自交获得T1代种子,T1代种子长成的植株即为T1代植株。
4、提取T1代植株的基因组DNA,用Hyg_F和Hyg_R组成的引物对进行PCR鉴定,目的条带约1035bp,PCR鉴定中扩增得到目的条带的植株即为转基因植株。转基因植株即为BBR8反向抑制植株。

Hyg_R:5′-TACACAGCCATCGGTCCAGA-3′。
(三)转pZh01载体水稻的制备
用pZh01载体代替重组质粒pZh01-BBR8i,按照步骤(二)进行操作,得到转pZh01载体植株。
三、植株对白叶枯病菌的抗性鉴定
供试植株为:T1代BBR8反向抑制植株8株、T1代转pZh01载体植株8株、水稻秋光8株、T1代BBR8过表达植株8株、T1代转pCAMBIA2300载体植株8株、水稻Kasalath8株。
将供试植株正常培养至分蘖盛期,然后采用人工剪叶接种法(Kauffman等,1973年)接种菲律宾生理小种PXO86(菌液是无菌水和菲律宾生理小种PXO86制备的,菌浓度为109个细胞/ml),14天后调查照相。
示例性照片见图4。水稻Kasalath的病斑长度为8.1±0.2cm。BBR8过表达植株的病斑长度为2.5±0.6cm。水稻秋光的病斑长度为1.6±0.5cm。转pZh01载体植株的病斑长度为1.5±0.3cm。BBR8反向抑制植株的病斑长度为5.5±0.6cm。结果表明,将BBR8基因导入水稻Kasalath,显著地提高了植株的抗病性。抑制水稻秋光中BBR8基因的表达,极大地降低了植株的抗病性。

3.一种制备转基因水稻属植物的方法,包括如下步骤:在水稻属植物中导入编码BBR8蛋白的核酸分子,得到对白叶枯病的抗性增高的转基因水稻属植物, 所述编码BBR8蛋白的核酸分子为编码区如序列表中序列2第396-1856位核苷酸所示的DNA分子。
