Abstract
The invention discloses a novel environment-friendly specimen fixing composition, a preparation method thereof and a fixing solution containing the composition, wherein the composition comprises an antiseptic and mildew-proof active ingredient, a permeation assistant and a controllable fixing regulator, wherein the antiseptic and mildew-proof active ingredient is a natural biological cross-linking agent, and the permeation assistant is a mixture of one or two or more of nitrates, sulfates or chlorides in any proportion; the controllable fixed regulator is a micromolecular organic matter. The composition is dissolved in a solvent to prepare the novel environment-friendly specimen fixative. The fixing solution disclosed by the invention does not contain formaldehyde, is safe and non-toxic, does not volatilize effective components, is harmless to human bodies and the environment, can effectively kill and inhibit various moulds on corpses and animal specimens, can be used for controllably fixing the specimens, and has the advantages of simple preparation method, easiness in obtaining raw materials, low price and good application and popularization values.

Technical Field
The invention relates to a novel environment-friendly specimen fixing composition, a preparation method thereof and a fixing solution containing the composition.
Technical Field
In the medical, biological analysis, observation and research process, animal and human body tissues are fixed firstly, and then processed, observed, detected and stored. The tissue of animal and human body is fixed by a series of treatments of fixing agent and dehydrating agent, etc. to retain the inherent form and structure of cell and tissue, so as to attain the goal of preventing bacterial corrosion and tissue autolysis, preserving cell inherent substance, hardening tissue, conveniently cutting tissue, preventing disease diffusion and enhancing dyeing, etc. The most commonly used tissue fixative is formalin, which has the advantages of uniform fixation, strong penetration, preservation of lipid substances, low cost, etc. However, the defects of high impurity content, influence on dyeing after the fixed liquid is acidified, incapability of fixing uric acid and carbohydrate, easiness in volatilization, environmental pollution, easiness in drying of specimens, poor mildew resistance and the like indicate that the fixed liquid is bound to leave a stage with fixed tissues.
In order to reduce the harm of formaldehyde to human bodies and environment, researchers have developed various materials and methods for replacing formaldehyde, such as phenolic solutions, salt solutions, benzalkonium bromide solutions, heavy metal compounds, and the like. However, the phenol solution has a damaging effect on peripheral nerve endings, phenol is easily oxidized into benzoquinone, and yellow brown mud precipitates exist in a dead body pool after a long time, so that the antiseptic effect is reduced. Common salt solution is 20% sodium chloride solution, and due to high concentration and strong dehydration effect, the corpse tissue can shrink due to dehydration, which brings certain difficulty to dissection. The benzalkonium bromide solution is a bactericide, has a common concentration of 0.1-0.2%, has no irritation, low toxicity and strong bactericidal power, but has a reduced effect because the benzalkonium bromide is easily affected by organic matters, is forbidden to be used with many inorganic salts, and aluminum instruments are avoided. The heavy metal salts are also very toxic, so their application is very limited. Aiming at the problems, the invention provides a novel environment-friendly specimen fixing composition and a fixing liquid prepared from the same. The specific technical scheme is as follows:
firstly, the invention provides a novel environment-friendly specimen fixing composition which comprises an antiseptic and mildew-proof active ingredient, a permeation assistant and a controllable fixing regulator, wherein the antiseptic and mildew-proof active ingredient is a natural biological cross-linking agent, and the permeation assistant is a mixture of one or two or more of nitrates, sulfates or chlorides in any proportion; the controllable fixed regulator is a micromolecular organic matter.
The novel environment-friendly specimen fixing composition comprises the following components in parts by weight: 0.5-5 parts of natural biological crosslinking agent, 0.5-5 parts of permeation aid and 0.5-2 parts of controllable fixed regulator.
Preferably, in the novel environment-friendly specimen fixing composition, the natural biological cross-linking agent is genipin.
Preferably, in the novel environment-friendly specimen fixing composition, the genipin is prepared from geniposide extracted from eucommia ulmoides or gardenia jasminoides.
Preferably, in the novel environment-friendly specimen fixing composition, the permeation assistant is one or a mixture of two or more of potassium nitrate, calcium nitrate, sodium nitrate, magnesium nitrate, potassium sulfate, calcium sulfate, sodium sulfate, magnesium sulfate, potassium chloride, calcium chloride, sodium chloride and magnesium chloride mixed in any proportion.
Preferably, in the novel environment-friendly specimen fixing composition, the controllable fixing regulator is urea.
The preparation method of the novel environment-friendly specimen fixing composition comprises the following steps:
A. weighing genipin and a penetration aid according to the weight parts, adding the genipin into the penetration aid, and uniformly stirring at normal temperature and normal pressure to form a mixture;
B. and D, adding the controllable fixing regulator in parts by weight into the mixture prepared in the step A, and continuously stirring uniformly at normal temperature and normal pressure to obtain the novel environment-friendly specimen fixing composition.
The method for preparing the fixing solution by using the novel environment-friendly specimen fixing composition comprises the steps of dissolving the novel environment-friendly specimen fixing composition in a solvent, and adjusting the pH of the novel environment-friendly specimen fixing composition to 5-7 by using a pH regulator.
Preferably, the solvent of the fixative solution prepared from the novel environment-friendly specimen fixative composition is water, methanol or ethanol.
Preferably, the pH regulator is one or a mixture of two of sodium hydroxide and potassium hydroxide mixed in any ratio.
The invention has the following beneficial effects:
1. the plant extract genipin is applied to corpse fixation, corrosion prevention and preservation, meanwhile, a penetration assisting agent, urea and other small molecular organic matters are added according to a certain proportion, dissolved in water, methanol and ethanol, and the pH is adjusted by using sodium hydroxide or potassium hydroxide to prepare the environment-friendly specimen fixing solution, so that various moulds of corpse and animal specimens are effectively killed and inhibited, and the specimens are controllably fixed.
2. The environment-friendly specimen fixing liquid disclosed by the invention does not contain formaldehyde, is safe and non-toxic, does not volatilize effective components, is harmless to human bodies and the environment, and tests prove that dead bodies and animal specimens are not mildewed or shrunk after being fixed, have natural colors, flexible and moist tissues and good using effect.
3. The preparation method of the environment-friendly specimen stationary liquid is simple, and the raw materials are easy to obtain and low in price, so that the environment-friendly specimen stationary liquid is convenient to popularize and apply.

Example 1
The embodiment is to prepare a novel environment-friendly specimen fixing composition and a fixing liquid thereof.
The preparation method of the composition comprises the following steps: weighing 40g of genipin, adding 20g of calcium nitrate, and uniformly stirring at normal temperature and normal pressure; and adding 10g of urea into the mixture, uniformly stirring at normal temperature and normal pressure, and standing to obtain a finished product.
The preparation method of the fixing liquid comprises the following steps: and dissolving the prepared composition in deionized water, adjusting the weight of the composition to 1000g, and adding a sodium hydroxide solution to adjust the pH value of the stationary liquid to 5.0 to obtain the novel environment-friendly specimen stationary liquid for later use.
Example 2
The embodiment is to prepare a novel environment-friendly specimen fixing composition and a fixing liquid thereof.
The preparation method of the composition comprises the following steps: weighing 50g of genipin, adding 30g of magnesium sulfate, and uniformly stirring at normal temperature and normal pressure; and adding 15g of urea into the mixture, uniformly stirring at normal temperature and normal pressure, and standing to obtain a finished product.
The preparation method of the fixing liquid comprises the following steps: dissolving the prepared composition in deionized water, adjusting the weight of the composition to 1000g, adding a potassium hydroxide solution, and adjusting the pH value of the stationary liquid to 6.0 to obtain novel environment-friendly specimen stationary liquid for later use.
Example 3
The embodiment is to prepare a novel environment-friendly specimen fixing composition and a fixing liquid thereof.
The preparation method of the composition comprises the following steps: weighing 40g of genipin, adding 20g of calcium chloride, and uniformly stirring at normal temperature and normal pressure; and adding 10g of urea into the mixture, uniformly stirring at normal temperature and normal pressure, and standing to obtain a finished product.
The preparation method of the fixing liquid comprises the following steps: dissolving the prepared composition in deionized water, adjusting the weight of the composition to 1000g, adding a potassium hydroxide solution, and adjusting the pH value of the stationary liquid to 7.0 to obtain novel environment-friendly specimen stationary liquid for later use.
Example 4
This example is to apply the novel environment-friendly specimen fixative prepared in example 1 to the fixation and preservation of pathological tissues such as fat cells, intestines, ganglia, lymph nodes, and the like. As shown in figure 1, the pathological tissue section treated by the prepared novel environment-friendly specimen stationary liquid has obvious cell morphology and background differentiation, can better reflect the original structure of the histiocyte and is beneficial to pathological analysis. Therefore, the environment-friendly specimen fixing solution has excellent fixing and preserving effects on pathological tissues such as fat cells, intestines, ganglia, lymph nodes and the like.
Example 5
This example is to apply the novel environment-friendly specimen fixative prepared in example 2 to the fixation and preservation of pathological tissues such as loose connective tissue, hyaline cartilage, mixed thrombus, intestinal adenocarcinoma, and the like. As shown in figure 2, the pathological tissue section treated by the prepared novel environment-friendly specimen stationary liquid has the same obvious cell morphology and background distinction, can better reflect the original structure of the histiocyte, and is beneficial to pathological analysis. Example 6
This example is to apply the novel environmental protection specimen fixing solution prepared in example 2 to the fixed preservation of liver, kidney, heart and muscle tissue of fresh animals. The product is soaked and stored, and the phenomena of putrefaction, shrinkage, autolysis and the like of tissues can be observed by observing the product for one year. As shown in figure 3, the porcine liver, kidney, heart and muscle tissues are preserved for one year, the fixing solution is still clear and transparent, the animal tissues do not shrink obviously, the shape is preserved completely, and the environment-friendly specimen fixing solution can better fix and preserve the fresh animal tissues.
Tests prove that the novel environment-friendly specimen fixing composition and the fixing liquid thereof can effectively kill and inhibit a plurality of moulds of corpses and animal specimens and can controllably fix the specimens. The corpse and animal specimens preserved after being fixed by the novel environment-friendly specimen fixing liquid do not go mouldy or shrink, have natural color, flexible and moist tissues and very good effect. Moreover, the novel environment-friendly specimen fixing solution does not contain formaldehyde, is safe and non-toxic, has non-volatile active ingredients, and is harmless to human bodies and the environment. In addition, the novel environment-friendly specimen stationary liquid has the advantages of simple preparation method, easily obtained raw materials, low price and very good application and popularization values.
Claims (10)
1. A novel environment-friendly specimen fixing composition comprises an antiseptic and mildew-proof active ingredient, a permeation aid and a controllable fixing regulator, and is characterized in that: the preservative and mildew-proof active ingredient is a natural biological cross-linking agent, and the permeation aid is one or a mixture of two or more of nitrates, sulfates or chlorides which are mixed in any proportion; the controllable fixed regulator is a micromolecular organic matter.
2. The novel environmentally friendly specimen fixative composition of claim 1, wherein: the novel environment-friendly specimen fixing composition comprises the following components in parts by weight: 0.5-5 parts of natural biological crosslinking agent, 0.5-5 parts of permeation aid and 0.5-2 parts of controllable fixed regulator.
3. The novel environmentally friendly specimen fixing composition according to claim 1 or 2, characterized in that: the natural biological cross-linking agent is genipin.

6. The novel environmentally friendly specimen fixing composition according to claim 1 or 2, characterized in that: the controllable fixed regulator is urea.
7. A method for preparing the novel environment-friendly specimen fixing composition as set forth in any one of claims 1 to 6, characterized in that: the method comprises the following steps:
A. weighing genipin and a penetration aid according to the weight parts, adding the genipin into the penetration aid, and uniformly stirring at normal temperature and normal pressure to form a mixture;
B. and D, adding the controllable fixing regulator in parts by weight into the mixture prepared in the step A, and continuously stirring uniformly at normal temperature and normal pressure to obtain the novel environment-friendly specimen fixing composition.

A fixative prepared using the novel environmentally friendly specimen fixative composition of any one of claims 1 to 7, characterized by: the fixing solution is prepared by dissolving the novel environment-friendly specimen fixing composition as defined in any one of claims 1 to 7 in a solvent, and adjusting the pH of the fixing solution to 5 to 7 by using a pH adjusting agent.
