Abstract
本发明公开了一种保湿抗菌皂及其制备方法。所述的保湿抗菌皂包含以下重量百分比计的组分:皂基75~85%、甘油1~5%、燕麦β‑葡聚糖0.5~1%、固定化酶4~9%、枯草菌脂肽钠0.5~1.5%、螯合剂0.1~1%、香精0.1~0.3%,余量为去离子水;所述的固定化酶是将酶包被于壳聚糖‑虫胶酸复合的笼形结构中,所述的酶固载量为0.5~10wt%。本发明提供的保湿抗菌皂具有高效的抑菌作用,抑菌谱广,去污能力强,保湿效果好,安全无毒,刺激性低,在有效清洁的同时保持肌肤健康、水嫩柔滑,适合长期使用。

技术领域
本发明属于日化洗涤用品技术领域,具体涉及一种保湿抗菌皂及其制备方法。
背景技术
香皂是日常生活中必不可少的洗涤护理用品,以脂肪酸钠和其他表面活性剂为主要原料,添加品质改良剂和外观改良剂,经过加工成型后制成的产品。随着日用化工的快速发展,香皂的品种不再单一,除了具有基本的去污、洗涤性能外,还具有养颜、润肤,外观美观等特征。尽管洗面奶、沐浴露、香皂等在近年来得到充足的发展,但由于香皂使用方便,去污效果好,价格便宜,刺激性低,在国内外仍然是重要的皮肤清洁调理用品。
传统的香皂多成碱性,使用后冲洗不净会对人体皮肤造成刺激,且长期使用使人皮肤变得干涩、紧绷和粗糙;个别使用者对香皂可能会出现过敏现象;使用后水沥不净,香皂表面就会溶解损失、外观变形,存在遇水易糊烂的缺陷;有些香皂添加了植物源精油,但储存期间精油会慢慢挥发损失,且洗手后精油的芳香味只能维持很短的时间;有些香皂的抗硬水能力差,使用生活用水时因钙、镁等离子对一价离子的置换,去污能力变差。
此外,香皂作为日常生活中必不可少的洗涤护理用品,人们对其需求不再仅仅满足去污效果,具有杀菌效果的香皂更受青睐,市面上的抑菌香皂是利用三氯卡班、对氯间二甲苯酚、DMDM乙内酰脲等化学物质抑菌,其安全性需要探究,并且长期使用某种化学消毒剂易导致微生物产生耐药性。某些中药成分有较强的抗菌作用和美容功效,它们是由天然植物加工制得,对人体不会产生不良反应和副作用,但是往往添加量较大,成本较高。
发明内容
本发明的目的是提供一种保湿抗菌皂及其制备方法。所述的保湿抗菌皂具有高效的抑菌作用,抑菌谱广,去污能力强,保湿效果好,安全无毒,刺激性低,在有效清洁的同时保持肌肤健康、水嫩柔滑,适合长期使用。
本发明是通过以下技术方案予以实现的:
一种保湿抗菌皂,包含以下重量百分比计的组分:皂基75~85%、甘油1~5%、燕麦β-葡聚糖0.5~1%、固定化酶4~9%、枯草菌脂肽钠0.5~1.5%、螯合剂0.1~1%、香精0.1~0.3%,余量为去离子水。
优选,所述的固定化酶是将酶包被于壳聚糖-虫胶酸复合的笼形结构中,所述的酶固载量为0.5~10wt%。
所述的酶选自溶菌酶、肽聚糖水解酶中的至少一种。进步一步优选,所述的肽聚糖水解酶选自糖苷酶、糖基转移酶、酰胺酶、肽链内切酶中的至少一种。更优选,所述的酶为溶菌酶。
本发明的固定化酶,其制备步骤为:
1)将壳聚糖溶解于0.1~10wt%的乙酸溶液中,使其浓度为1~2mg/mL,得到壳聚糖溶液;
2)将虫胶酸溶于乙醇,制成浓度为0.4~1mg/mL的虫胶酸溶液;
3)将酶溶于去离子水,得到总蛋白浓度为0.1~2.5mg/mL的酶溶液;
4)将壳聚糖溶液和虫胶酸溶液混合,室温下磁力搅拌10~20min,然后加入酶液,继续搅拌5~10min,按终浓度为1~3wt%的比例加入京尼平,继续搅拌至体系呈凝胶状,滴加适量碳酸钠溶液,调节pH值至5.5~7,常温下固化25~30min,真空低温干燥,得到固定化酶。
所述的固定化酶制备步骤中,壳聚糖和虫胶酸的质量比为1:0.1~0.25;所述的固定化酶中酶的固载量为0.5~10wt%。
优选,所述的皂基选自棕榈酸钠/钾、肉豆蔻酸钠/钾、月桂酸钠/钾、硬脂酸钠/钾、椰油酸钠/钾、油酸钠/钾中的至少一种。
更优选,所述的皂基为月桂酸钠、肉豆蔻酸钠和椰油酸钠以质量比1:1:1组成。
优选,所述的螯合剂选自次氮基三乙酸钠、乙二胺四乙酸二钠、柠檬酸钠、二乙烯三胺五羧酸、酒石酸钠、三聚磷酸钠、偏磷酸钠、焦磷酸钠、乙二胺四甲叉磷酸钠中的至少一种。
本发明还提供一种所述的保湿抗菌皂的制备方法,其包括以下步骤:
1)往皂基中加入去离子水,加热至60~65℃,搅拌使其完全融化;
2)降温至40~45℃,加入甘油、燕麦β-葡聚糖、固定化酶、枯草菌脂肽钠、螯合剂和香精,搅拌均匀,得到混合料;
3)将混合料进行均化处理,经压条成型、切块后得到成品保湿抗菌皂。
本发明所述的皂基优选为月桂酸钠、肉豆蔻酸钠和椰油酸钠三者的混合物,选用清洁力强,水溶性好的月桂酸钠、肉豆蔻酸钠作为皂基主成分,添加适量的椰油酸钠,一方面降低月桂酸钠、肉豆蔻酸钠刺激性比较大的不足,另一方面,弥补椰油酸钠清洁能力较弱的缺陷,使制得的保湿抗菌皂保持高效清洁能力的同时,降低对皮肤的刺激。
本发明中的溶菌酶和肽聚糖水解酶均能催化病原菌细胞壁降解,发挥抑菌作用,其中,溶酶菌主要通过破坏细胞壁中的N-乙酰胞壁酸和N-乙酰氨基葡糖之间的β-1,4糖苷键,使细胞壁不溶性黏多糖分解成可溶性糖肽,导致细胞壁破裂内容物逸出而使细菌溶解,对革兰阴性菌和阳性菌均具有较好的抑菌作用。肽聚糖水解酶,通过作用于N-乙酰葡萄糖胺和N-乙酰胞壁酸之间的糖苷键,或作用于N-乙酰胞壁酸和肽桥之间的酰胺键,或作用于氨基酸侧链的肽键或肽桥上氨基酸之间的肽键,促使细菌自溶,对革兰阳性菌的抑菌作用佳,对革兰阴性菌的作用稍弱。优选的,本发明以溶菌酶作为抑菌酶,将其包被于壳聚糖-虫胶酸复合的笼形结构中,一方面可避免皂基体系碱性降低溶菌酶的活性,削弱溶菌酶的抑菌效果,从而促使溶菌酶稳定发挥长效的抑菌作用,另一方面,载体壳聚糖-虫胶酸复合物亦具有一定的抑菌效果,固定化溶菌酶后,抑菌效果显著提升,具有明显的协同增益作用。本发明人在前期试验中发现,仅以壳聚糖作为载体,溶菌酶的包被率较低,固定化效果稍弱,抑菌效果降低,并且不能改善抗菌皂使用后水沥不净,香皂表面就会溶解损失、外观变形,存在遇水易糊烂的缺陷。出乎意料之外,本发明人发现,利用壳聚糖分子中含有-OH、-NH-等基团与虫胶酸的-COOH相互作用,在京尼平交联下,形成容纳量较大的笼形结构,能显著提升对酶的包被效果,提高酶的固载量,同时发现虫胶酸的加入,可改善抗菌皂的沥水性,使抗菌皂在水中不易糊烂,延长了香皂的使用寿命。
本发明中添加的枯草菌脂肽钠(CAS号:302933-83-1)是由纳豆菌发酵得到的一款表面活性剂,安全无刺激,可生物降解,含有由7个氨基酸组成的环状肽亲水部分和长链烷烃的疏水部分形成的独特分子结构,具有良好的乳化及助乳化性。本发明人发现,在抗菌皂中添加枯草菌脂肽钠对革兰阴性菌的抑菌效果明显提升,推测这是由于枯草菌脂肽钠能有效溶解革兰阴性菌厚厚的外膜结构,促使病原菌暴露细胞壁,进一步被溶菌酶溶解。
此外,由于细菌细胞外膜的脂多糖是通过钙离子和镁离子的相互作用而连接的,本发明加入的螯合剂与枯草菌脂肽钠相互作用,能进一步提升对细菌外膜结构的溶解作用,利用螯合剂处理,能有效将二价阳离子除去,进而破坏细胞外膜的结构,从而提高菌体的通透性。
甘油和燕麦β-葡聚糖的加入,能有效降低抗菌皂对皮肤的刺激性,提升皮肤的保湿锁水功效,使用抗菌皂后,不会产生干涩感。
与现有技术相比,本发明具有以下有益效果:
本发明提供的保湿抗菌皂具有高效的抑菌作用,抑菌谱广,对革兰阳性菌和革兰阴性菌均具有较好的抑菌效果,去污能力强,保湿效果好,安全无毒,刺激性低,在有效清洁的同时保持肌肤健康、水嫩柔滑,适合长期使用。

以下实施例是对本发明的进一步说明,而不是对本发明的限制。下述实施例中配方的组分,若无特别说明,均为常规市售产品。
实施例1
本实施例固定化酶的制备包括以下步骤:
(1)称取100mg壳聚糖溶解于2wt%的乙酸溶液中,使其浓度为1mg/mL,得到壳聚糖溶液;
(2)称取10mg虫胶酸溶于乙醇,制成浓度为0.4mg/mL的虫胶酸溶液;
(3)称取5mg溶菌酶溶于去离子水,得到总蛋白浓度为0.5mg/mL的酶溶液;
(4)将壳聚糖溶液和虫胶酸溶液混合,室温下磁力搅拌10min,然后加入酶液,继续搅拌10min,按终浓度为1wt%的比例加入京尼平,继续搅拌至体系呈凝胶状,滴加适量碳酸钠溶液,调节pH值至6,常温下固化30min,真空低温干燥,得到固定化酶。
实施例2
本实施例固定化酶的制备包括以下步骤:
(1)称取100mg壳聚糖溶解于10wt%的乙酸溶液中,使其浓度为2mg/mL,得到壳聚糖溶液;
(2)称取25mg虫胶酸溶于乙醇,制成浓度为1mg/mL的虫胶酸溶液;
(3)称取10mg溶菌酶溶于去离子水,得到总蛋白浓度为0.1mg/mL的酶溶液;
(4)将壳聚糖溶液和虫胶酸溶液混合,室温下磁力搅拌20min,然后加入酶液,继续搅拌5min,按终浓度为2wt%的比例加入京尼平,继续搅拌至体系呈凝胶状,滴加适量碳酸钠溶液,调节pH值至7,常温下固化25min,真空低温干燥,得到固定化酶。
实施例3
本实施例固定化酶的制备包括以下步骤:
(1)称取100mg壳聚糖溶解于0.5wt%的乙酸溶液中,使其浓度为1mg/mL,得到壳聚糖溶液;
(2)称取15mg虫胶酸溶于乙醇,制成浓度为0.5mg/mL的虫胶酸溶液;
(3)称取1mg溶菌酶溶于去离子水,得到总蛋白浓度为0.1mg/mL的酶溶液;
(4)将壳聚糖溶液和虫胶酸溶液混合,室温下磁力搅拌15min,然后加入酶液,继续搅拌10min,按终浓度为3wt%的比例加入京尼平,继续搅拌至体系呈凝胶状,滴加适量碳酸钠溶液,调节pH值至5.5,常温下固化30min,真空低温干燥,得到固定化酶。
比较例1
本比较例1固定化酶的制备包括以下步骤:
(1)称取100mg壳聚糖溶解于2wt%的乙酸溶液中,使其浓度为1mg/mL,得到壳聚糖溶液;
(2)称取5mg溶菌酶溶于去离子水,得到总蛋白浓度为0.5mg/mL的酶溶液;
(4)往壳聚糖溶液中加入酶液,搅拌10min,按终浓度为1wt%的比例加入京尼平,继续搅拌至体系呈凝胶状,滴加适量碳酸钠溶液,调节pH值至6,常温下固化30min,真空低温干燥,得到固定化酶。
本比较例1与实施例1比较,区别在于,以壳聚糖作为载体,不含虫胶酸。
实施例4-9
实施例4-9保湿抗菌皂的制备包含下表所示重量百分比计的组分:
制备步骤:
(1)往皂基中加入去离子水,加热至60℃,搅拌使其完全融化;
(2)降温至40℃,加入甘油、燕麦β-葡聚糖、固定化酶、枯草菌脂肽钠、螯合剂和香精,搅拌均匀,得到混合料;
(3)将混合料进行均化处理,经压条成型、切块后得到成品保湿抗菌皂。

对比例1保湿抗菌皂的制备与实施例4比较,区别在于,所述的固定化酶为比较例1制得。其制备步骤参考上述实施例。

试验例一、泡沫测试和糊烂程度分析
(1)按照行业标准QB/T 2486-2008测定抗菌皂的发泡力,结果见表1。
(2)糊烂程度分析:
仪器:1.尖头的铅丝挂钩;2.钢尺(卡尺)刻度0.5mm;3.两端装柄的薄刀片;4.广口保温瓶2300mL。
测定步骤:1.取抗菌皂,将尖头铅丝插入其一端,悬挂于广口保温瓶中,瓶中盛有温度为20±1℃的蒸馏水1800mL,香皂不与瓶壁接触。使香皂的2/3浸泡在水中,浸泡12小时后取出。
2.待香皂水滴流完,将香皂侧立于光滑平整的桌面,用两端装柄的薄刀片从垂直于桌面的方向剖开香皂。被浸的3个完整剖面有明显的糊烂白色镶边。
计算:用钢尺量出剖面的3个糊烂层的厚度,取其平均数,单位mm,结果见表1。

结果显示,本发明实施例4-9制得的保湿抗菌皂具有较佳的发泡能力,(5min)/mL≥400,满足《中华人民共和国轻工行业标准洗衣皂》中I型洗衣皂的要求,具有较好的清洗力、起泡性及易冲洗性,并且香皂在水中不易糊烂,延长了香皂的使用寿命,效果优于对比例1-4制得的保湿抗菌皂。
试验例二、抑菌效果检测
按照QB/T 2738《日化产品抗抑菌效果评价办法》悬液定量法对本发明制得的保湿抗菌皂进行抑菌效果检测,其中,测试浓度为1:100,作用时间为20min,试验菌种为金黄色葡萄球菌ATCC 6538、大肠杆菌8099、绿脓杆菌ATCC 15442和白色念珠菌ATCC 14053,结果见下表2。

结果显示,本发明实施例4-9制得的保湿抗菌皂具有较佳的抑菌效果,对金黄色葡萄球菌的抑菌率高达99.65%,对大肠杆菌的抑菌率高达99.04%,对绿脓杆菌的抑菌率高达98.76%,对白色念珠菌的抑菌率高达96.42%,效果优于对比例1-4制得的保湿抗菌皂。
试验例三、皮肤刺激性和保湿效果检测
分别对实施例4-9和对比例1-4制得的保湿抗菌皂进行皮肤刺激性测试,具体为:实验用皂液浓度为5wt%,用量为0.3mL时,由HET VAM测试测定皮肤反应分级(相对刺激值分级),强烈刺激>2.0,刺激1.2-2.0,中等刺激0.8-1.2,轻微刺激0.0-0.8,结果见下表3。
用水分测试仪分别测试洗涤前皮肤的含水量、以及洗涤2h后的皮肤含水量,计算出皮肤含水量增加的百分率,结果见下表3。

结果显示,本发明实施例4-9制得的保湿抗菌皂具有较佳的保湿效果,使用后可增加皮肤的含水量,显著降低皮肤的刺激性,效果优于对比例1-4制得的保湿抗菌皂。

Claims (10)
1.一种保湿抗菌皂,其特征在于,包含以下重量百分比计的组分:皂基75~85%、甘油1~5%、燕麦β-葡聚糖0.5~1%、固定化酶4~9%、枯草菌脂肽钠0.5~1.5%、螯合剂0.1~1%、香精0.1~0.3%,余量为去离子水。
2.根据权利要求1所述的保湿抗菌皂,其特征在于,所述的固定化酶是将酶包被于壳聚糖-虫胶酸复合的笼形结构中,所述的酶固载量为0.5~10wt%。
3.根据权利要求2所述的保湿抗菌皂,其特征在于,所述的酶选自溶菌酶、肽聚糖水解酶中的至少一种。
4.根据权利要求3所述的保湿抗菌皂,其特征在于,所述的肽聚糖水解酶选自糖苷酶、糖基转移酶、酰胺酶、肽链内切酶中的至少一种。
5.根据权利要求4所述的保湿抗菌皂,其特征在于,所述的酶为溶菌酶。
6.根据权利要求1-4任一所述的保湿抗菌皂,其特征在于,所述的固定化酶的制备步骤为:
1)将壳聚糖溶解于0.1~10wt%的乙酸溶液中,使其浓度为1~2mg/mL,得到壳聚糖溶液;
2)将虫胶酸溶于乙醇,制成浓度为0.4~1mg/mL的虫胶酸溶液;
3)将酶溶于去离子水,得到总蛋白浓度为0.1~2.5mg/mL的酶溶液;
4)将壳聚糖溶液和虫胶酸溶液混合,室温下磁力搅拌10~20min,然后加入酶液,继续搅拌5~10min,按终浓度为1~3wt%的比例加入京尼平,继续搅拌至体系呈凝胶状,滴加适量碳酸钠溶液,调节pH值至5.5~7,常温下固化25~30min,真空低温干燥,得到固定化酶。
7.根据权利要求6所述的保湿抗菌皂,其特征在于,所述的固定化酶制备步骤中,壳聚糖和虫胶酸的质量比为1:0.1~0.25;所述的固定化酶中酶的固载量为0.5~10wt%。
8.根据权利要求1所述的保湿抗菌皂,其特征在于,所述的皂基选自棕榈酸钠/钾、肉豆蔻酸钠/钾、月桂酸钠/钾、硬脂酸钠/钾、椰油酸钠/钾、油酸钠/钾中的至少一种。
9.根据权利要求1所述的保湿抗菌皂,其特征在于,所述的螯合剂选自次氮基三乙酸钠、乙二胺四乙酸二钠、柠檬酸钠、二乙烯三胺五羧酸、酒石酸钠、三聚磷酸钠、偏磷酸钠、焦磷酸钠、乙二胺四甲叉磷酸钠中的至少一种。
10.一种制备权利要求1-9任一所述的保湿抗菌皂的方法,其特征在于,包括以下步骤:
1)往皂基中加入去离子水,加热至60~65℃,搅拌使其完全融化;
2)降温至40~45℃,加入甘油、燕麦β-葡聚糖、固定化酶、枯草菌脂肽钠、螯合剂和香精,搅拌均匀,得到混合料;
3)将混合料进行均化处理,经压条成型、切块后得到成品保湿抗菌皂。
