本发明涉及一种食品中亚硝酸盐的测定前处理方法,包括如下步骤:称取适量样品,置于具塞锥形瓶中,加入饱和硼砂溶液,再加入70℃的水混匀,于沸水浴中加热15min,取出置冷水浴中冷却,并放置至室温;定量转移提取液至容量瓶中,加入亚铁氯化钾溶液,再加入乙酸锌溶液,摇匀,并置于离心机中离心过滤;继续加入亚铁氯化钾溶液及乙酸锌溶液,进行二次离心过滤,以沉淀蛋白质;加水至刻度,摇匀,加入活性炭进行物理吸附,加入氢氧化铝进行悬浮液吸附,加入正己烷提取色素,除去杂质,上清液用滤纸过滤,弃去初滤液,滤液备用。本发明针对现有技术中的缺陷,可以将食品中的各种杂质、基底颜色去除,从而大大提高了检测精度。

食品中亚硝酸盐的测定前处理方法
技术领域:
本发明涉及食品检测技术领域,尤其涉及一种食品中亚硝酸盐的测定前处理方法。
背景技术:
《GB 5009.33-2016 食品安全国家标准 食品中亚硝酸盐与硝酸盐的测定 第二法分光光度法》中12.2.4:对于其他样品的前处理方法为“定量转移提取液至200ml容量瓶中,加入5ml亚铁氯化钾溶液,摇匀,再加入5ml 220g/L乙酸锌溶液,以沉淀蛋白质,加水至刻度,摇匀,放置30min,除去上层脂肪,上清液用滤纸过滤,弃去除滤液30ml,滤液备用。”
该方法具有如下缺点:1、滤液沉淀不完全或未沉淀导致过滤十分缓慢,甚至无法过滤;2、有些样品滤液基底有颜色导致滤液无法检测比色。
发明内容:
本发明的目的是针对现有技术的缺陷,提供一种检测精度高的食品中亚硝酸盐的测定前处理方法。
本发明是通过如下技术方案实现的:
一种食品中亚硝酸盐的测定前处理方法,包括如下步骤:
(1)称取适量样品,置于具塞锥形瓶中,加入饱和硼砂溶液,再加入70℃的水混匀,于沸水浴中加热15min,取出置冷水浴中冷却,并放置至室温;
(2)定量转移提取液至容量瓶中,加入亚铁氯化钾溶液,再加入乙酸锌溶液,摇匀,并置于离心机中离心过滤;
(3)继续加入亚铁氯化钾溶液及乙酸锌溶液,进行二次离心过滤,以沉淀蛋白质;
(4)加水至刻度,摇匀,加入活性炭进行物理吸附,加入氢氧化铝进行悬浮液吸附,加入正己烷提取色素,除去杂质,上清液用滤纸过滤,弃去初滤液,滤液备用。
本发明食品中亚硝酸盐的测定前处理方法的有益效果是:本发明针对现有技术中的缺陷,可以将食品中的各种杂质、基底颜色去除,从而大大提高了检测精度。

本发明提供一种食品中亚硝酸盐的测定前处理方法,包括如下步骤:
(1)称取5g样品,置于250mL具塞锥形瓶中,加入12.5mL 50g/L饱和硼砂溶液,再加入70℃左右的水约150ml混匀,于沸水浴中加热15min,取出置冷水浴中冷却,并放置至室温;
(2)定量转移提取液至200mL容量瓶中,加入5mL 106g/L亚铁氯化钾溶液,再加入5mL220g/L乙酸锌溶液,摇匀,并置于离心机中离心30min,过滤;
(3)继续加入亚铁氯化钾溶液及乙酸锌溶液,进行二次离心过滤,以沉淀蛋白质;
(4)加水至刻度,摇匀,加入活性炭进行物理吸附,加入氢氧化铝进行悬浮液吸附,加入正己烷提取色素,除去杂质,上清液用滤纸过滤,弃去初滤液,滤液备用。

(1)称取适量样品,置于具塞锥形瓶中,加入饱和硼砂溶液,再加入70℃的水混匀,于沸水浴中加热15min,取出置冷水浴中冷却,并放置至室温;
(2)定量转移提取液至容量瓶中,加入亚铁氯化钾溶液,再加入乙酸锌溶液,摇匀,并置于离心机中离心过滤;
(3)继续加入亚铁氯化钾溶液及乙酸锌溶液,进行二次离心过滤,以沉淀蛋白质;
(4)加水至刻度,摇匀,加入活性炭进行物理吸附,加入氢氧化铝进行悬浮液吸附,加入正己烷提取色素,除去杂质,上清液用滤纸过滤,弃去初滤液,滤液备用。
