Abstract

技术领域
本发明涉及均借助于微流体装置的微生物菌株开发、抗体产生、抗体分析领域。本申请涉及微流体装置和系统中及通过微流体装置和系统进行的细胞培养和细胞分析领域。本申请还涉及微流体领域,具体是微流体分析和装置领域以及细胞分选、细胞排序和/或聚集的开发。提供了用于将悬浮在移动流体中的颗粒聚集到一个或多个局部流线中的各种系统、方法和装置。
背景技术
颗粒分离和过滤已经应用于工业、医学和研究中的许多技术方案。已经开发了各种宏观尺度技术用于颗粒分离以解决这些应用。微尺度技术提供的优点在于,按比例缩小允许使用独特的流体动力学效应,以及增强了电磁分离力。介电泳力已经用于基于粒度或一些介电标签来区分颗粒。用于连续分离的其它技术依赖于用于在壁处排布的不同粒度颗粒的流的层流剖面和不同交叉横截面。进一步的微尺度技术涉及精确设计的过滤器或柱阵列,这些过滤器或柱阵列基于粒度沿颗粒方向形成分叉。这些技术可以根据颗粒的粒度或介电性质产生非常精确的分离。例如,对于通过不对称排布的障碍物产生的确定性位移,报告直径为1μm的颗粒的分辨率小于20nm。另外,这些系统的复杂程度可以很低。
目前的微尺度分离系统的缺点在于,按比例缩放通常限制了这些技术的吞吐量。在大多数情况下,颗粒体积分数保持在远低于1%,因为颗粒-颗粒相互作用可能会显著影响性能。另外,小容积流速会导致微通道中的大平均流体速度,从而导致分离力作用在颗粒上的时间不足。这些系统的通常流速范围为1-50μL/min,这对于许多制备性应用(例如,大容量血液中稀有细胞的浓缩、超声造影剂的过滤或大量胶体/乳剂的制备)来说是不够的。在微流体系统中使用微滴或液滴(乳剂)作为有限空间的一个基本问题在于,用期望样本填充大比例的液滴。例如,人们想要i)在一个液滴中携带两个不同的细胞,ii)在一个液滴中携带一个细胞和一个探针,iii)在一个液滴中携带抗原和抗体,iv)在一个液滴中携带抗原和B或T细胞,或v)在一个液滴中携带化学物质和细菌细胞等。然而,在泊松分布之后,大多数液滴将不包含所期望的组合。具有实现并利用悬浮在行进通过微流体通道的流体中的颗粒的自排序的多个系统、装置、设备和方法,将是有帮助的。这将有助于将悬浮在移动流体中的颗粒聚集到一个或多个局部流线中。在该系统中,具有基底和设置在基底上的至少一个具有用于聚集细胞的入口和出口的通道是有帮助的。同样有利的是具有包括悬浮颗粒和驱动流体层流的泵送元件的层流中的通道,使得细胞在通道中被分选进入一个或多个流线。此外,具有提供用于评估微流体通道内的颗粒分布和/或定位的确定性途径的方法将是有帮助的。本发明人已经解决了这些问题。
发明内容

具体实施方式

本发明人惊奇地发现,通过“混合”不同粘度的流体,可以在通道中稳定地组织颗粒流。当然,混合并不意味着通常意义上的混合,而是相反地,在这里意味着将例如两个不同粘度的流体混合在一起,使其尽可能长时间地以层流方式并行流动而不彼此混合。两种流体的这种并行流的效果是,这两个流之一中的颗粒被精确地限制在该流中。这种流或流体流线可以用于将颗粒带入期望空间或位置。在本发明的优选方面,这种流被用于具体地将两个不同颗粒或细胞带入一个液滴中。
通道可具有液力直径,并且聚集颗粒粒度与液力直径之比大于或等于约0.07。粒度与液力直径之比可以小于或等于约0.5。
一个或多个聚集流线的宽度可以小于或等于聚集颗粒粒度的约五倍、四倍、三倍、两倍和/或1.5倍。该系统的实施例可以增加溶液中的颗粒浓度。
可以使用检测器对在通道中的颗粒的局部流通量中行进的颗粒进行计数。这些方法可进一步包括用能够由检测器检测到的标签标记选定颗粒的系统,从而检测器检测选定颗粒并对选定颗粒进行计数。
与本领域已知的其它方法不同,由第一流体和第二流体构成的流体系统内的一个或多个颗粒的定位基本上由粘弹性力或粘性力或其组合控制。因此,在一个实施例中,表征本文公开的流体系统的雷诺数小于1。
在任何及所有方面,这些方法可以包括其中可以排他性地由粘弹性力或粘性力引起聚集的系统。其它方法实施例可以包括其中可由粘弹性力或粘性力及其它力引起聚集的系统。在上述任意方面,实施例可以包括一种设备,其中,通道从入口到出口的横截面形状和面积可以是一致的。在其它实施例中,通道从入口到出口的横截面形状和面积可以变化。一个或多个局部流线的宽度可以小于或等于预定颗粒粒度的约五倍、四倍、三倍、两倍和/或1.5倍。将具有预定粒度的颗粒的流体悬浮液从入口移动至出口将预定粒度的颗粒聚集成四个局部流、两个局部流和/或单个局部流。
在列举的这些方面或在其实施例的任一个中,可以提供用于从颗粒群中分离目标颗粒的方法,其中,划分是被动地进行的。目标颗粒可以具有不同于群体中其它颗粒的粒度,并且目标颗粒可以在通道内的预定位置形成局部流通量。在一些实施例中,可以定位第一输出分支的入口以包围目标颗粒的局部流通量在通道内的预定位置。该方法的实施例还可以包括用标签选择性地标记颗粒,该标签由可操作地连接至通道的划分系统使用。标签可以增加选择性标记的颗粒的粒度,并且标签可以是磁性标签。
在本发明中,颗粒优选包含在第二流体中,并且选择第一流体的粘度使得第二流体中的颗粒被第一流体限制在由第二流体占据的空间。如图4所示,两个入口使两种流体在入口接合点处相遇。一旦流体汇集在一起,就会因粘度不同而分开行进。结果,每种流体中的颗粒被限制流体在通道内所具有的空间。因此,颗粒以有序方式一个接一个地行进。这意味着该排序可以再次用于其它应用。
在本发明中,优选地,第一微流体通道的高度选自由2μm-60μm、5μm-50μm、10μm-45μm、15μm-40μm、25μm-35μm组成的组。
在本发明中,优选地,第一微流体通道的宽度选自由2μm-150μm、5μm-125μm、25μm-100μm、40μm-85μm、50μm-70μm组成的组。更优选地,第一微流体通道的宽度为约60μm。
在另一实施例中,第一和/或第二微流体通道的宽度与被输送流体的粘度成反比。
在另一实施例中,第一和/或第二微流体通道的宽度与颗粒的粒度成反比。
理想地,第一入口和第二入口以Y形方式在基本相同的位置进入微流体通道。这种几何形状确保了两种流体不会快速混合,而是并行地行进。因此,理想地,第一入口和第二入口之间的角度小于或等于180°。
优选地,在根据本发明的方法中,排序通道基本上是直的,并且具有从1mm-40mm、2mm-35mm、5mm-25mm、8mm-20mm、10mm-20mm和12mm-18mm组成的组中选择的长度。
优选地,第一流体和第二流体是含水流体,并且第一流体的粘度在100cP-2000cP之间,第二流体的粘度在0.001cP-2cP之间。
更优选地,第一流体的粘度选自由100cP-2000cP、200cP-1500cP、300cP-1200cP和500cP-1000cP组成的组。
更优选地,第二流体的粘度选自由0.001cP-100cP、0.010cP-50cP、0.250cP-10.0cP和0.5cP-1.0cP组成的组。
在进一步的实施例中,第一流体和第二流体是含水流体,并且第一流体和第二流体的粘度在0.001cP-100cP之间。
在另一实施例中,第二流体的粘度为约1cP。在进一步的实施例中,第一流体的粘度是第二流体的粘度的两倍。
在一个实施例中,本文公开的流体系统的滞留时间足以确保第一流体和第二流体之间的粘度梯度。滞留时间是由不同参数确定的测量值,例如第一流体和第二流体的组成、流速和通道长度。
优选的是,含水第一流体包含从以下物质组成的组中选择的物质:有机聚合物、天然聚合物、纤维素、葡萄糖、果糖或任何其它糖、DNA、RNA、聚(乙二醇)、聚(乙二醇)二丙烯酸酯、双丙烯酰胺、聚丙烯酰胺、链霉亲和素-丙烯酰胺、聚-N-丙烯酰胺、聚N-异丙基聚丙烯酰胺、琼脂糖、藻酸或其混合物。如本文所用的,术语“有机聚合物”是指通过化学合成获得、主要由包含碳和氢元素的单体组成的聚合物,其可以与不限于氧、氮和硫的任何其它元素进行组合。在本发明的上下文中,有机聚合物的一个示例可以是合成肽或蛋白质。如本文所用的,术语“天然聚合物”是指天然出现的、主要由包含碳和氢元素的单体组成的聚合物,其可以与不限于氧、氮和硫的任何其它元素进行组合。在本发明的上下文中,天然聚合物的一个示例可以是透明质酸。天然或合成聚合物也可以包括油及其衍生物。
在一个实施例中,粘性含水第一流体包括从包含非聚合物油及其衍生物,例如氟化油(参见例如图9),的组中选择的物质。
在一个实施例中,第一出口是乳剂出口,并且至少一个颗粒在离开第一出口时被封装在液滴中。
在一个实施例中,微流体系统包括第三入口。在该实施例中,高粘性流体可以用于分离低粘度流体;例如参见图1。
在该实施例中,第三流体在第三入口注入,并且选择这三种流体的粘度使得第一流体将第二流体和第三流体分离,并且所有三种流体至少在排序通道的长度上以基本上未混合的层流方式流动。
颗粒可以选自包括细胞、真核细胞、原核细胞、珠、抗体或其片段、抗原等的组。如上所述,优选存在两个不同的细胞类型并且这两个细胞类型以有序方式集合在在一个液滴中。
优选地,在此类方法中,超过20%、30%、40%、50%、60%、70%、80%、90%或95%的含有两个细胞的液滴具有两个期望的细胞。
本发明还涉及一种颗粒的排序、分选和/或聚集方法,其中,将根据本发明的第一微流体通道系统与根据本发明的另一微流体通道系统进行组合,并且两个系统的出口在一个共用通道中接合。该实施例在图2中示出。因此,本发明还涉及一种2个或更多个微流体通道在接合点处汇集的系统,其中,2个或更多个通道中的至少两个通道装载有并行流动的两种流体,其中一流体具有比另一流体更高的粘度。

本发明还涉及根据本发明的方法或根据本发明的系统用于将不同颗粒分选至一个液滴的用途。如本文所用的,术语“不同颗粒”可以指多个相同或不同颗粒。
与本领域已知的其它方法不同,本文所公开的方法和系统的另一个优点在于,通过混合两个不同粘度的流体,可以在将颗粒封装在液滴中之前控制颗粒在微流体系统中的定位。这种对流体内颗粒的优良控制允许将至少一个颗粒精确封装在单个液滴中,而不采用会不利地影响封装步骤的高流速。
因此,在另一实施例中,根据本发明的方法和/或系统允许每个液滴封装至少一个颗粒。更优选地,根据本发明的方法和/或系统允许每个液滴封装仅一个颗粒。
本发明还涉及一种包括在接合点处汇集的两个或更多个微流体通道的系统,其中,两个或更多个通道中的至少两个通道装载有并行流动的两种流体,其中一流体具有比另一流体更高的粘度。这里,一个通道可以输送颗粒(参见以上对颗粒可以是什么的定义),而另一通道可以输送不同颗粒。在两个通道汇合的接合点处形成液滴。可以重复该过程以引入第三个或其它颗粒。

示例
微流体芯片装置设计、制造和制备
所有微流体装置均使用2D计算机辅助设计(CAD)软件(AutoCAD20,Autodesk)设计。在涂覆SU-8之前,将4″硅晶片置于热板上,在200℃下烘干至少5分钟。
通过以2000-4000RPM作为最终速度30秒旋涂SU-8树脂(SU-8-2025或SU-2035,MicroChem),在晶片上涂覆SU-8层,然后首先在65℃下预烘1-3分钟,再在95℃下预烘3-10分钟,并在UV曝光之前冷却至室温。使用光刻机(MJB4接触式光刻机,SUSS MicroTec)通过高分辨率透明掩模(JD Photo Tools,UK)对SU-8涂覆晶片进行365nm UV曝光,以构造微通道。以WEC接触模式使用光刻机,并且通过以10-16mW/cm2的剂量曝光树脂10-40秒。曝光后的SU-8涂覆晶片首先在65℃下预烘1-3分钟,再在95℃下预烘1-10分钟,并在显影之前冷却至室温。使用PGMEA(Microchem,Y020100)在定轨振荡器上的玻璃容器中以100RPM的速度显影干烘后的SU-8晶片1-10分钟,随后用氮气干燥晶片并在200℃下硬烘至少5分钟。SU-8模具上的微通道高度用触针轮廓仪(Dektak 6M,Veeco)或白光干涉测量法(NT9100,Veeco)测量。将聚二甲基硅氧烷(PDMS)(Sylgard184,DowCorning)以1:10(固化剂:碱助剂)的比例在盘中混合,并倒在SU-8模具上。随后将PDMS在真空干燥器中脱气几分钟,以从模具中除去气泡。将PDMS在烘箱(VWR,法国)中以70℃固化2小时。将固化的PDMS从模具上剥离,并且用0.75mm的活检穿孔器(Harris,USA)刺穿出入口孔。用Scotch胶带从穿孔的PDMS板坯中除去颗粒和灰尘,随后用异丙醇和去离子水漂洗。使用加压氮气干燥清洁后的PDMS板坯。通过将PDMS板坯和玻璃暴露于氧等离子体(Pico,Diener等离子体)1分钟并使它们在暴露后接触,将PDMS板坯上的微流体通道网粘合到载玻片上(50mm×75mm,Dow Corning)。最后一步,通过用含有1%(v/v)硅烷(Alfa Aesar,L16584)的HFE-7500(Novec,3M)溶液冲洗所述网,然后用HFE-7500冲洗通道并用加压氮气吹扫剩余的HFE-7500,将亲氟涂层施加到微流体通道壁上。
仪表设备设置
对于液滴产生和再注入,在倒置的落射荧光显微镜(TiE,Nikon,法国)上进行实验,该显微镜已经被改进并与488nm、561nm和638nm激光器(Omicron,德国)和4个具有相应带通滤波器(PMT1:440/40、PMT2:525/40、PMT3:593/46、PMT4:708/75、PMT5:809/81,Semrock)的光电倍增管(PMT)(H10723,Hamamatsu)连接。来自PMT的信号被馈送至FPGA卡(NI-USB7856R,National Instruments)和专有软件程序(uDrop 3.5-3.9,HiFiBiO)记录的液滴数据中。
将高速照相机(Phantom系列,Vision Research)附接至显微镜的左端口,以监测液滴产生和细胞封装。使用5通道注射泵(Nemesys,Cetoni)驱动微流体芯片中的所有流体。
对于静态液滴阵列,激发光由LED源(SOLA光引擎,Lumencor Inc.)提供。使用适当的带通滤光器(GFP和TRITC滤光器组,Nikon,以及Cy5滤光器组,Semrock)和照相机设置(Orca R2,Hamamatsu)在室温(25℃)和环境氧浓度下记录特定通道的荧光信号。使用10x物镜(NA 0.45)获取图像。
细胞培养和细胞标记
在标准细胞培养箱(37℃、5%的CO2)中速度为125rpm的定轨振荡器上的无菌Erlenmayer烧瓶中培养CHO-S(Freestyle,ThermoFisher)细胞系。在添加青霉素/链霉素、0.5%的Pluronic F-68(ThermoFisher)和L-谷氨酰胺的CHO-S Freestyle培养基中培养细胞。
根据制造商提供的标准方案,细胞悬浮液通常用Calcein AM绿色、CellTracker橙色、CellTracker红色或Nucred(均为ThermoFisher)标记。
纤维素制备
在室温下,在Erlenmayer烧瓶中使用磁搅拌器和搅拌棒通过搅拌(500RPM-2000RPM)将2%(w/w)的甲基纤维素(Sigma-Aldrich)溶解在去离子水中。
细胞封装程序和监测
收获细胞并通过15μm、10μm过滤器过滤。用流式细胞仪(Guava EasyCyte,Millipore)计数,并将细胞浓度调节至10m-80m细胞/mL。然后,将细胞抽吸至与微流体装置相容的定制储存器中。连续相由Novec HFE7500氟化油中2%(w/w)的008-氟表面活性剂(RAN生物技术)组成。水相在芯片上同流。流速(油为约1000-6000μl/h,各个水溶液为100-800μl/h)调节为通常生成80-400pl的单分散液滴以测量负载效果。在液滴形成期间,使用自制的附件将细胞悬浮液冷却至~5℃,以减缓抗体分泌并保持细胞生存力。使用激光/PMT系统或使用速率为10'000至100'000帧/秒的高速成像收集数据。在使用激光/PMT系统进行测量的情况下,数据处理和分析由uDrop执行。在使用静态液滴阵列对液滴成像的情况下,将乳剂直接注射到2D室系统中,或收集在1.5ml的Eppendorf管中,该Eppendorf管含有0.1%(w/w)008-氟表面活性剂的氟化油,不管是在冰上(细胞实验)还是在室温下(无细胞实验)。如果收集在管中,用含有0.1%(w/w)008-氟表面活性剂的氟化油1:1地稀释液滴,然后使用定制PDMS阀将液滴引入观察室。然后以750μl/h的流速将液滴重注入该室中。在室的填充完成后,轻轻关闭室并将其安装在荧光显微镜(Ti Eclipse,Nikon)上。
图像分析
使用专用的ImageJ和Matlab程序处理高速图像。使用ImageJ,随着时间推移提取感兴趣区域的平均灰度级,从而产生灰度级曲线。在Matlab中,通过去除背景(整个曲线的平均灰度级)和信号反转来进行进一步的图像处理,产生具有表示细胞的峰的曲线。
该程序将使用内置函数来确定峰之间的间距,然后将数据汇集成直方图。
Claims (15)
1.一种用于在第一微流体通道系统中对颗粒进行排序、分选和/或聚集的方法,所述方法包括以下步骤:
i.提供第一微流体通道,所述第一微流体通道至少包括第一入口和第二入口以及第一出口,
ii.通过所述第一入口将第一流体注入所述通道中,
iii.通过所述第二入口将第二流体注入所述通道中,其中,所述第一流体的粘度高于所述第二流体的粘度,从而使这两种流体以不混合的层流方式并行流动,并且所述两种流体之一包含待排序、分选和/或聚集的颗粒。
2.根据权利要求1所述的方法,其中,所述颗粒优选包含在所述第二流体中,并且选择所述第一流体的粘度使得所述第二流体中的颗粒被所述第一流体限制在由所述第二流体占据的空间。

6.根据权利要求1-6中任意一项所述的方法,其中,所述第一流体和所述第二流体是含水流体,并且所述第一流体的粘度在100cP-2000cP之间,第二流体的粘度在0.001cP-100cP之间,或者其中,所述第一流体和所述第二流体是含水流体,并且所述第一流体和所述第二流体的粘度在0.001cP-100cP之间。
7.根据权利要求1-7中任意一项所述的方法,其中,含水的第一流体包括从包括以下物质的组中选择的物质:有机聚合物、天然聚合物、纤维素、葡萄糖、果糖或任何其它糖、DNA、RNA、聚(乙二醇)、聚(乙二醇)二丙烯酸酯、双丙烯酰胺、聚丙烯酰胺、链霉亲和素-丙烯酰胺、聚-N-丙烯酰胺、聚N-异丙基聚丙烯酰胺、琼脂糖、藻酸或其混合物。
8.根据权利要求1-8中任意一项所述的方法,其中,所述第一出口是乳剂出口,并且至少一个颗粒在离开所述第一出口时被封装在液滴中。
9.根据权利要求1-9中任意一项所述的方法,其中,所述微流体系统包括第三入口。
10.根据权利要求1-10中任意一项所述的方法,其中,第三流体在所述第三入口注入,并且选择这三种流体的粘度使得所述第一流体将所述第二流体和所述第三流体分离,并且所有三种流体至少在所述排序通道的长度上以基本上未混合的层流方式流动。
11.根据权利要求1-11中任意一项所述的方法,其中,颗粒能选自包括单个细胞或聚合细胞、真核细胞、原核细胞、珠、抗体或其片段、抗原和水凝胶珠的组。
12.一种颗粒的排序、分选和/或聚集方法,其中,将根据权利要求1-12中任意一项的第一微流体通道系统与根据权利要求1-12中任意一项的另一微流体通道系统进行组合,并且两个系统的所述出口在一个共用通道中接合。
13.一种微流体通道系统,包括
i.第一微流体通道,所述第一微流体通道至少包括第一入口和第二入口以及第一出口,
ii.第一流体,
iii.第二流体,其中,所述第一流体的粘度高于所述第二流体的粘度,从而使这两种流体能够以不混合的层流方式并行流动,并且所述两种流体之一包含待排序、分选和/或聚集的颗粒。
14.根据权利要求1-13中任意一项的方法或根据权利要求14的系统用于将不同颗粒分选进入一个液滴的用途。
15.一种包括在一个接合点处汇集的两个或更多个微流体通道的系统,其中,所述两个或更多个通道中的至少两个通道装载有并行流动的两种流体,其中一流体具有比另一流体高的粘度。
