Abstract
The invention provides a composition with an immunity improving function, which is characterized by comprising 5-50 parts by weight of poria cocos; 3-30 parts of lentinus edodes; 2-20 parts of wolfberry fruit; 1.5-20 parts of agaricus blazei; 1.5-20 parts of grifola frondosa; 1.5-20 parts of lily; 1.5-10 parts of ginseng. The composition and the preparation of the invention have the functions of promoting the proliferation of thymus and spleen of mice, improving the capability of phagocytizing inert carbon granules by a reticuloendothelial system of the mice and having obvious in-vivo anti-tumor effect on S180 tumor mice.

Technical Field
The invention relates to the field of medicines, foods or health-care products, in particular to a composition with an immunity improving function and a preparation method and application thereof.
Background

The invention also provides application of the composition in preparing a medicine, a health-care product or food with the function of improving immunity or resisting tumors.
The invention also provides a medicine, a health-care product or a food containing the composition with the function of improving the immunity.
Specifically, the medicine, health product or food is selected from oral, injection or external preparation. The medicine, food or health care product is prepared from the composition and auxiliary materials or additives acceptable on the medicine, food or health care product.
The adjuvants can be white sugar, lactose, mannitol, sorbitol, erythritol, stevioside, citric acid, malic acid, potassium sorbate, sodium benzoate, sodium carboxymethylcellulose, gelatin, tragacanth, xanthan gum, carrageenan or konjac gum.

taking poria cocos, shiitake mushroom, wolfberry fruit, agaricus blazei, grifola frondosa, lily and ginseng, removing impurities, cleaning, adding water which is 8-12 times of the total weight of the medicinal materials, boiling and extracting for 2-4 times, wherein the extraction time is 0.5-1.5 h each time, filtering, combining filtrates, concentrating the filtrate at 60-70 ℃ under reduced pressure until the relative density is 1.05-1.10 to obtain a water extraction concentrated solution, refrigerating and centrifuging the concentrated solution.
Further, the preparation of the composition comprises the following steps:
taking poria cocos, shiitake mushroom, wolfberry fruits, agaricus blazei, grifola frondosa, lily and ginseng, removing impurities, cleaning, adding water with the weight being 10 times of the total weight of the raw materials, boiling and extracting for 2 times, wherein the time for boiling and extracting each time is 1 hour, filtering through 120 meshes, combining two filtrates, concentrating the filtrate at 60-70 ℃ under reduced pressure until the relative density is 1.05-1.10 to prepare a water extract concentrated solution, refrigerating the concentrated solution at 0-10 ℃ for 48 hours, and centrifuging at 6000-7000 r/m to obtain the composition.
The invention also provides a preparation method of the medicine, health-care product or food with the function of improving immunity, which is characterized in that when the medicine, health-care product or food is selected from oral preparations, the preparation method comprises the following steps:
taking poria cocos, shiitake mushroom, wolfberry fruit, agaricus blazei, grifola frondosa, lily and ginseng, removing impurities, cleaning, adding water which is 8-12 times of the total weight of the raw materials, boiling and extracting for 2-4 times, wherein the extraction time is 0.5-1.5 h each time, filtering, combining filtrates, concentrating the filtrate at 60-70 ℃ under reduced pressure until the relative density is 1.05-1.10 to obtain a water extraction concentrated solution, refrigerating and centrifuging to obtain the composition; mixing water with the weight 5-10 times that of the composition, the sorbitol and the citric acid with the composition, fully stirring for dissolving, boiling for 30-60 min, cooling, filtering, fixing the volume, refrigerating and centrifuging to obtain a preparation solution; homogenizing the mixed solution, packaging, and sterilizing.
Specifically, the oral preparation comprises the following steps: (1) extraction: respectively taking cleaned poria cocos, shiitake mushroom, wolfberry fruit, agaricus blazei, grifola frondosa, lily and ginseng, adding water which is 8-12 times of the total weight of the medicinal materials, boiling and extracting for 2 times, wherein the extraction time is 1 hour each time, filtering by 120 meshes, and combining the filtrates of the two times; concentrating the filtrate at 65 deg.C under reduced pressure to obtain concentrated solution with relative density of 1.07, refrigerating at 0-10 deg.C for 48 hr, and centrifuging at 7000r/m to obtain composition; (2) blending: mixing water with the weight 5-10 times that of the composition, the sorbitol and the citric acid with the composition, fully stirring and dissolving, boiling for 30-60 min, cooling to room temperature, fine filtering with 200 meshes, fixing the volume, refrigerating the constant volume liquid at 0-10 ℃ for 48h, and centrifuging at 14000r/m to obtain a prepared liquid, wherein the mass percentage concentration of the sorbitol in the prepared liquid is 10-20%, and the mass percentage concentration of the citric acid is 0.01-0.2%; (3) homogenizing: heating the prepared liquid to 60-70 ℃, homogenizing twice in a high-pressure homogenizer at the homogenizing pressure of 30-60 Mpa to obtain a homogeneous liquid; (4) filling: filling and sealing the homogeneous liquid by an automatic filling and sealing machine, and rolling and covering; (5) and (3) sterilization: and (4) filling and sealing the mixture, and then placing the mixture in a sterilizer for sterilization at the temperature of 115-121 ℃ for 20-30 min to obtain the product.
Preferably, when the medicine, health product or food is selected from oral preparation, the preparation method comprises the following steps:
(1) extraction: adding water which is 10 times of the total weight of the medicinal materials into the medicinal materials, boiling and extracting for 2 times, wherein the time for boiling and extracting each time is 1 hour, filtering through a 120-mesh filter, combining filtrates of the two times, concentrating the filtrate at 65 ℃ under reduced pressure until the relative density is 1.07 to prepare a water extraction concentrated solution, refrigerating the concentrated solution at 0-10 ℃ for 48 hours, and centrifuging at 7000r/m to obtain the composition;
(2) blending: mixing 8 times of the weight of the composition of water, sorbitol and citric acid with the composition, stirring for dissolving, boiling for 40min, cooling to room temperature, fine filtering with 200 meshes, fixing the volume, refrigerating the constant volume liquid at 0-10 ℃ for 48h, and centrifuging at 14000r/m to obtain a prepared liquid, wherein the mass percentage concentration of the sorbitol in the prepared liquid is 14%, and the mass percentage concentration of the citric acid is 0.1%;
(3) homogenizing: heating the prepared liquid to 60-70 ℃, homogenizing twice in a high-pressure homogenizer at the homogenizing pressure of 30-60 Mpa to obtain a homogeneous liquid;
(4) filling: filling and sealing the homogeneous liquid by an automatic filling and sealing machine, and rolling and covering;
(5) and (3) sterilization: filling, sterilizing in a sterilizer at 115 deg.C for 30 min.
The composition provided by the invention has the functions of maintaining the balance of functions in vivo, strengthening the body resistance to consolidate the constitution, improving the immunity of the human body, resisting tumors, protecting the health of the human body, preventing invasion of exogenous pathogenic factors and reducing the probability of infection diseases of people, and the composition has reasonable compatibility of the components, can effectively improve the cell repair function of sub-health patients, old people, chronic patients, seriously ill people and the like, promotes the improvement of the immune function of the organism, delays aging and promotes the rapid rehabilitation of the diseases. Wherein the content of the first and second substances,
poria is dry sclerotium of Poria cocos (a fungus in Polyporaceae), and traditional Chinese medicinal materials used as both medicine and food have high compatibility in common clinical formula of traditional Chinese medicine, and the principle is 'ten prescriptions and nine poria cocos'. It has sweet and light taste, mild property, and no damage to vital qi, and has effects of calming heart and tranquilizing mind.
Lentinus edodes is the fruiting body of Lentinus edodes of Pleurotaceae, also known as flower mushroom and Lentinus edodes, and is called "the king of mountain delicacies" and is also a well-known edible and medicinal fungus. It has sweet taste and mild property, and has effects of strengthening body resistance, tonifying deficiency, invigorating spleen, promoting appetite, dispelling pathogenic wind, promoting eruption, eliminating phlegm, regulating qi-flowing, and removing toxic substance.
The medlar is a dry mature fruit of the solanaceae plant medlar, commonly called as the medlar, the red eardrop, the blood medlar and the like, and is a rare plant with homology of medicine and food. Example 2
A composition for improving immunity and resisting tumor is prepared from the following raw materials in parts by weight: 26 parts of poria cocos; 12 parts of mushroom; 10 parts of wolfberry fruit; 6 parts of agaricus blazei; 6 parts of grifola frondosa; 6 parts of lily; 3.5 parts of ginseng. The preparation method comprises the following steps: (1) extraction: taking the medicinal materials according to the weight parts, removing impurities, cleaning, adding water which is 10 times of the total weight of the medicinal materials, boiling and extracting for 2 times, wherein the time of each boiling and extracting is 1 hour, filtering through a 120-mesh sieve, combining filtrates obtained in two times, concentrating the filtrate at 65 ℃ under reduced pressure until the relative density is 1.07 to prepare a water extraction concentrated solution, refrigerating the concentrated solution at 0-10 ℃ for 48 hours, and centrifuging at 7000r/m to obtain the composition; (2) blending: mixing 8 times of water, a proper amount of sorbitol and citric acid and the composition, fully stirring and dissolving, boiling for 40min, cooling to room temperature, fine filtering with 200 meshes, fixing the volume, refrigerating the constant volume liquid at 0-10 ℃ for 48h, and centrifuging at 14000r/m to obtain a prepared liquid, wherein the mass percentage concentration of the sorbitol in the prepared liquid is 14%, and the mass percentage concentration of the citric acid is 0.1%; (3) homogenizing: heating the prepared liquid to 60-70 ℃, homogenizing twice in a high-pressure homogenizer at the homogenizing pressure of 30-60 Mpa to obtain a homogeneous liquid; (4) filling: filling and sealing the homogeneous liquid by an automatic filling and sealing machine, and rolling and covering; (5) and (3) sterilization: filling, sterilizing at 115 deg.C for 30min to obtain oral preparation (1ml equivalent to 0.058g crude drug).

The efficacy of the experiment for improving the immunity of the organism and resisting the tumor function of the invention is evaluated as follows.
1 materials of the experiment
1.1 Experimental animals and tumor strains
ICR mice, clean grade, 18-22g, male; s180 tumor strain.
1.2 materials and reagents
The composition or oral dosage form prepared in examples 1-3, wherein the concentration of the composition is 0.155 g/mL; 0.9% physiological saline; india ink; 0.1% Na2CO3And (3) solution.

2 method of experiment
2.1 Effect of example samples on mouse thymus index and spleen index

On day 12, the water supply and food supply were stopped, the weight was weighed, the cells were sacrificed by the conventional method, the tumors were taken and weighed, and the tumor inhibition rate was calculated.

As can be seen, the thymus index and spleen index of the mice in the experimental group are significantly different (P <0.05) compared with those in the blank group, which indicates that the samples in the examples can obviously increase the thymus and spleen mass of the adult mice.

Therefore, the carbon clearance phagocytosis index of the mice in the experimental group is obviously different from that of the blank group (P <0.05), which indicates that the samples in the examples can improve the phagocytosis index α of the mice and enhance the function of a phagocytosis system.

Therefore, the tumor inhibition rates of the experimental groups are all over 30 percent, and compared with the blank group, the experimental groups have significant difference (P <0.05), which indicates that the samples in the examples have obvious in-vivo anti-tumor effect on S180 tumor mice.
The experimental results show that the composition and the preparation have the effects of promoting the proliferation of thymus and spleen of mice, improving the ability of phagocytizing inert carbon granules by a reticuloendothelial system of the mice and having obvious in-vivo anti-tumor effect on S180 tumor mice, and the composition has the effects of improving immunity and resisting tumor.

7. The medicament, health product or food of claim 6, wherein the medicament, health product or food is selected from oral, injectable or topical dosage forms.
8. The drug, health product or food of claim 7, wherein the drug, health product or food comprises an oral tablet, a buccal tablet, a chewable tablet, an effervescent tablet, a hard capsule, a soft capsule, a granule, a syrup, a medicated wine, an oral liquid.
9. A process for the preparation of a composition according to any one of claims 1 to 4, comprising the steps of:
adding water which is 8-12 times of the total weight of the medicinal materials into poria cocos, shiitake mushrooms, wolfberry fruits, agaricus blazei murill, grifola frondosa, lily and ginseng, boiling and extracting for 2-4 times, wherein the extraction time is 0.5-1.5 h each time, filtering, combining filtrates, concentrating the filtrate at 60-70 ℃ under reduced pressure until the relative density is 1.05-1.10, obtaining a water extraction concentrated solution, refrigerating and centrifuging.
10. The preparation method of the medicine, health-care product or food with the function of improving immunity according to claim 6, wherein when the medicine, health-care product or food is selected from oral dosage forms, the preparation method comprises the following steps:
(1) extraction: adding water which is 10 times of the total weight of the medicinal materials into the medicinal materials, boiling and extracting for 2 times, wherein the time for boiling and extracting each time is 1 hour, filtering through a 120-mesh filter, combining filtrates of the two times, concentrating the filtrate at 65 ℃ under reduced pressure until the relative density is 1.07 to prepare a water extraction concentrated solution, refrigerating the concentrated solution at 0-10 ℃ for 48 hours, and centrifuging at 7000r/m to obtain the composition;
(2) blending: mixing 8 times of the weight of the composition of water, sorbitol and citric acid with the composition, stirring for dissolving, boiling for 40min, cooling to room temperature, fine filtering with 200 meshes, fixing the volume, refrigerating the constant volume liquid at 0-10 ℃ for 48h, and centrifuging at 14000r/m to obtain a prepared liquid, wherein the mass percentage concentration of the sorbitol in the prepared liquid is 14%, and the mass percentage concentration of the citric acid is 0.1%;
(3) homogenizing: heating the prepared liquid to 60-70 ℃, homogenizing twice in a high-pressure homogenizer at the homogenizing pressure of 30-60 Mpa to obtain a homogeneous liquid;
(4) filling: filling and sealing the homogeneous liquid by an automatic filling and sealing machine, and rolling and covering;
(5) and (3) sterilization: filling, sterilizing in a sterilizer at 115 deg.C for 30 min.
